Ultrastructural morphometric investigations on rat liver of young and adult rats after treatment with technical pentachlorophenol (PCP).

Age-dependent effects of technical pentachlorophenol (PCP) on male rat livers were investigated after a 15 day treatment with PCP, 30 mg/kg/d body weight. The liver tissues were investigated morphometrically at light and electron microscopical levels. Statistically significant alterations of nuclei and organelles of the hepatocytes were described.

Ultrastructural morphometric analysis of human myocardial left ventricles with mitral insufficiency. A comparison with normally loaded and hypertrophied human left ventricles.

15 biopsies of dilated and hypertrophied human left ventricles in mitral insufficient hearts were morphometrically investigated. On light and electron microscopical level the results were compared with those received from normally loaded human left ventricles and from hypertrophied human left ventricles found in hearts with aortic valve disease. The results demonstrate alterations when compared with the results from normally loaded left ventricles. The differences between normally loaded and volume loaded left ventricles are smaller than those in pressure loaded left ventricles from aortic valve diseased hearts.

[The presentation of the periodontal blood vessels. Histological studies in perfused rats].

Unbroken series of sections taken from various areas of the periodontium of vitally fixed rats were prepared as the basis for constructing a model for the morphology of the vessels and for planimetric evaluation. Using this material, we were successfully able to validate a technique for localization, course, and distribution of vessels and of capillary anastomoses.

[Treatment of anal fissure by lateral subcutaneous internal sphincterotomy].

50 patients have been followed up after lateral internal sphincterotomy for chronic anal fissure. The results are excellent. This operation has some advantages over against the posterior sphincterotomy or the sphincter stretching and should replace these latter procedures in the treatment of chronic anal fissure. The lateral internal sphincterotomy may probably be applied for other benign anal lesions.

[Ultrastructural morphometric analysis of normally loaded human myocardial left ventricles from young and old patients (author's transl)].

An ultrastructural and morphometric analysis was performed on human myocardial left ventricle, obtained during surgical heart operations on normally loaded ventricles. The diagnoses of the patients were a) Persistant foramen ovale b) Atrial septal defect stage (I.-II) c) Mitral stenosis (stage III.-IV.). The median values of left ventricular pressure were not pathologically elevated. The patients were divided in 2 groups, the younger one ranging from 5-15 years, the older from 42-78 years. It was shown that the volume density of the interstitial tissue does not differ with the aging process. The number of the nuclei per test area of the heart muscle cells decreased within age (P less than 0.001). This finding suggests a decreasing process in the number of the heart muscle cells and an increase in size of the individual myocardial cell. Increase in the volume density of myofibrils was demonstrated at electron microscopical level with aging (P less than 0.02). The volume density of mitochondria is the same in both groups, whereas the volume density of the remaining cytoplasm (without myofibrils and mitochondria) decreases (P less than 0.05). The numberical density of the mitochondria increased in the older patient group (P less than 0.001).

[Ultrastructural morphometric analysis of hypertrophied human myocardial left ventricles (author's transl)].

Biopsies of hypertrophied human myocardial left ventricles were investigated morphometrically. The diagnoses of the patients were stenosis of the aortic valve, aortic insufficiency or a combination of both lesions. The results were compared with those from normally loaded human left ventricles. There are no differences on light microscopical level between the volume densities of interstitial tissue and of heart muscle cells in the three groups of patients. A significant diminution of the volume density of the nuclei (P less than 0.001) and the number of nuclei per test area (P less than 0.001) when compared with normal groups suggests an increase in volume of the single heart muscle cell. The ultrastructural study shows marked increase in volume density of myofibrils (P less than 0.0001), with accompanying decrease in the volume densities of mitochondria (P less than 0.0001) and the remaining cytoplasm (P less than 0.001). A gross decrease in the surface area of mitochondria (P less than 0.001) and of cristae mitochondriales (P less than 0.0001) is found. The morphological equivalents of this result are numerous stages of mithochondrial destruction including cristolysis. All myocardial weights were beyond the "critical heart weight".

Epithelial rat liver cells have cell surface receptors recognizing a phosphorylated carbohydrate on lysosomal enzymes.

Receptor-mediated endocytosis of alpha-N-acetylglucosaminidase by cultured epithelial rat liver cells is inhibited by mannose, L-fucose and most effectively by mannose 6-phosphate. Endocytosis of alpha-N-acetylglucosaminidase is lost after treatment of the enzyme with alkaline phosphatase. These findings indicate that epithelial rat liver cells possess cell surface receptors that recognize a phosphorylated carbohydrate on alpha-N-acetylglucosaminidase, as was previously reported for cell surface receptors of human skin fibroblasts. Inhibition of alpha-mannosidase endocytosis by epithelial rat liver cells in the presence of mannose 6-phosphate and loss of enzyme endocytosis after treatment with alkaline phosphatase suggest that this enzyme is recognized by the same receptor.

Cycling aggregation patterns of cytoplasmic F-actin coordinated with oscillating tension force generation.

Isometric contracting protoplasmic veins of Physarum polycephalum show cycling patterns of cytoplasmic F-actin, dependent on their oscillating contraction behaviour (minute rhythms). The process of fibrillogenesis represents a parallel arrangement of F-actin chains ("plasma filaments, microfilaments") during the isometric contraction phase. A part of the results of the present work corroborates previous results on stretch-activated veins which showed that the fibrillar form of F-actin reflects the isometric contracted state. During isometric relaxation phase, a disaggregation of the fibrillar pattern takes place and is accompanied by a deparallelisation of F-actin chains. Therefore, the isometric relaxed state of cytoplasmic actomyosin is non-fibrillar in nature. Thus, the morphologically detectable fibrillar form of cytoplasmic actomyosin, according to physiological interpretation, is solely representative of the isometric contracted state. The question whether assembly-disassembly processes, e.g. G equilibrium F-actin-transformation, play a role in the contraction-relaxation cycle is discussed.

[Sensitivity to antibiotics of Pseudomonas aeruginosa isolated from patients in the years 1986-1988].

Pseudomonas aeruginosa strains (240 in all) isolated from various clinical materials in the years 1986-1988 showed a high sensitivity to amikacin (92.3-100%), colistin (84.6-100%) and ceftazidim (84.6-100%). Netilmycin acted on 75.4% of the strains, and penicillins (carbenicillin, ticarcillin and azlocillin) similarly as gentamicin, tobramycin, cefotaxim, cefoperazon and ceftriaxon were active only against 51.3-66.7% of the tested strains.
