Behavior pattern of rat ascites tumor cells arrested in liver sinusoids: an electron microscopic study.

The behavior pattern of tumor cells in the development of hematogenous liver metastasis by Yoshida sarcoma and two strains of rat ascites hepatoma, AH7974F and AH13, in Donryu strain rats was investigated by electron microscopy. Tumor cells of every strain in liver sinusoids stretched their cytoplasmic protrusions toward the vascular wall and were actually in contact with the endothelial cells with their tips to form various types of junction-like structures. The most distinct one was characterized by parallel and straight arrangement of the opposing cell membranes across a gap of about 100 A. The endothelial discontinuity was easily produced by the projection of cytoplasmic processes and the compression of tumor cells. Through the endothelial defect, tumor cells of Yoshida sarcoma and AH13 migrated locomotively into the dilated spaces of Disse or directly toward the hepatic cells, while AH7974F cells were frequently covered by elongated endothelial cytoplasm. Destruction of the hepatic cells by AH7974F and AH13 cells was found to begin with the insertion of their cytoplasmic processes into the coated vesicles in the hepatic cells. From these investigations, the particular interaction between tumor and normal cells seems to play an important role in the development of liver metastasis by these tumors.

[Biosynthesis of hyaluronic acid in parietal pleura of the rabbit (author's transl)].

The parietal pleura of rabbits was incubated with 14C-glucosamine. It was found that 14C-glucosamine was incorporated into the fraction of crude glycosaminoglycans. Then the crude glycosaminoglycans were fractionated by using specific mucopolysaccharide-lyases (hyaluronidase from streptomyces hyalurolytics, chondroitinase AC and chondroitinase ABC). As a result, evidence was obtained that hyaluronic acid was synthesized in parietal pleura and was released into the surroundings.

[Studies on medico-legal diagnosis in cold district. 2. Cadaveric phenomena in low temperature surroundings (author's transl)].

The authors have investigated 14 kinds of findings of 15 cadavers in low temperature surroundings which had remained at known postmortem interval and had been subjected to medico-legal autopsies in our laboratory, comparing with those at normal temperature. In low temperature surroundings, the onset and progress of the findings due to autolysis or putrefaction are retarded in a marked degree but the turbidity of the cornea which is considered to be due to physical or chemico-physical phenomenon is retarded merely in a slight degree. Hence, an erroneous determination of the postmortem interval can be avoided by taking care of the peculiarity. Even the cadavers at low temperature display in 3 to 5 months after death the findings whose degrees correspond to those of the cadavers at normal temperature in 4 to 7 days after death. Some fly-larvae pass the winter on cadavers demonstrating that the time of death was towards the end of autumn of the preceding year.

[Studies on medico-legal diagnosis in cold district. 3. Autopsy findings of the victims of cold exposure (author's transl)].

The authors have investigated the autopsy findings of 8 bodies died from cold. The conclusions are as follows: Pink colour of the blood in the left ventricle or the pulmonary veins, pink colour of the lungs and pink patches of the skin are fairly pathognomic findings of death from cold. Seven out of eight bodies have presented at least one of these findings. Subserous or submucous hemorrhage is less pathognomic and has been presented less frequently.

[Studies on medico-legal diagnosis in cold district. 4. An experimental study on the rise of temperature of dead body caused by the radiation of sunlight (author's transl)].

A sausage of 4 cm diameter and 15 cm length was chosen as a model of a part of human extremities. On the snowfield the model was stood on end and its temperature was recorded continuously by means of thermistor thermorecorder. The conclusions are as follows: In fine weather the temperature directly under the sunny surface of a model rose higher than the air temperature by 18 degrees C in the most remarkable case. Even in cloudy weather it rose fairly though lower than in fine weather. The rise of a model temperature became higher in February than in January. Even at a depth of 1.0 cm the temperature rose fairly though lower than directly under the surface. The temperature of the shady side of a model was equal to air temperature or snow temperature. It is presumed from these data that the temperature of a human dead body on snowfield becomes higher and reaches a optimum temperature for putrefaction at least partly by the radiation of sunlight. The presumption is supported by our experiences at the autopsies in cold district.

An endoscopic staining method for detection and operation of early gastric cancer.

The long term survival rate of gastric cancer has been much improved, and the 5-year survival rate in our institute was 42% with a significant difference between 29% in the advanced stage and 94% in the early stage. This result means that gastric cancer must be detected and treated early as possible. However, the diagnosis of early gastric cancer may remain extremely difficult even for an excellent endoscopist. Therefore, in order to clearly recognize early gastric cancer, an endoscopic staining method with methylene blue has been studied. In this method, one capsule of 150 mg methylene blue is swallowed with a small amount of proteinase solution 3 hours before endoscopy. Gastroscopy is performed routinely after this preparation. This procedure was performed on 153 gastric cancers and 137 of them (89.5%) have been successfully dyed in dark blue. In several cases, with this method, the actual borderline between the normal mucosa and the malignant extent was clearly recognized and resection line was decided. Introduction of methylene blue into the stomach could also stain the intestinalized epithelium of the gastric mucosa. Differential diagnosis of the dyed intestinal metaplasia and the dyed carcinoma seems to be very easy, because both gastric lesions have the characteristic dyed patterns. Mechanism of this phenomenon has been considered to be due to an absorption of the dye in the intestinal metaplasia, and in the gastric cancer, many factors may be involved, among which are the infiltration or diffusion of the dye into the cancerous tissue, the absorption from the abnormal epithelium, and the staining of the necrotic tissue.

Pagetoid malignant melanoma and lentigo maligna melanoma of toe. A study with the fluorescence method (Falck and Hillarp).

Two cases of malignant melanoma on the toe of middle-aged women were examined chiefly by the fluorescence method of Falck and Hillarp. In one of the patients, histopathology of the pigmented tumor on the left middle toe was a Pagetoid (superficial spreading) melanoma in situ, and the subungual granulomatous lesion on the right great toe in the other patient was a lentigo maligna melanoma. On fluorescence microscopy, characteristic findings of the pigment cells lying in the epidermis of both types may be summarized as follows: In the Pagetoid melanoma, the melanoma cells are ovoid, lack dendritic processes, and emit specific yellow fluorescence. In the lentigo maligna melanoma, the pigment cells clearly show dendritic processes, and emit specific green fluorescence.

Angiographic evidence of myocardial squeezing accompanying tachyarrhythmia as a possible cause of myocardial infarction.

The coronary arteriogram of a 52-year-old man with Basedow's disease and who was suffering from myocardial infarction following rapid atrial fibrillation, showed severe myocardial squeezing without organic stenosis. Angiographically, the functional obstructive lesion was always observed both at systole and diastole during atrial pacing, 150 beats per minute, and suggests that myocardial squeezing may be the cause of myocardial infarction following tachyarrhythmia.

Noninvasive assessment of aorta-coronary saphenous vein bypass graft patency using thermocouple.

Noninvasive monitoring of patency of aorta-coronary bypass grafts can be achieved with reasonable accuracy using the thermocouple. This method is consisted of attaching the thermocouple to the surface of grafted vein during surgery and judging the patency by rapid injection of iced solution into peripheral vein. Patient grafted vein will show the thermal change after injecting iced solution. On the contrary, occluded graft will show no thermal change. As this thermal curve is a thermodilution curve by a single injection and is measured from the outside of the vessel, thermal change depends on the temperature and the quantity of the injected solution, cardiac output, body temperature, and loss of indicator temperature along the way to the thermocouple. When the grafted vein is patent, the same thermal curves are recorded regardless of the amount of the flow, and zero flow resulted in disappearance of curve. Initially, the method was evaluated in dogs and the appropriateness was verified. Subsequently, this method was applied to 16 vein grafts of 12 patients undergoing aorta-coronary bypass surgery. Ten or 15 ml of iced solution was injected through inferior vena cava or antecubital vein and the thermal change of the grafted vein was recorded for 1-4 weeks after the operation. Fourteen grafted veins were judged to be patent and 2 were occluded. The comparison of the results by this method with coronary angiography undergone at 4 weeks after the operation showed no false positive case but 1 false negative case. This extravascular observation of the thermodilution curve is a convenient method for judging whether vein graft is patent or not.

Composition of acidic glycoconjugates (glycosaminoglycans and glycoprotein) in myometrium of rabbit uterus under estrogenic condition.

Acidic glycoconjugates (glycosaminoglycans and glycoprotein) were obtained, from myometrium of ovariectomized rabbit under estrogenic condition, by pronase digestion, fractionation with cetylpyridinium chloride and Dowex I column chromatography, in succession. Composition of acidic glycoconjugates was determined enzymatically, employing Streptomyces hyaluronidase, chondroitinase AC II, chondroitinase ABC and crude heparinase. Each glycoconjugate was distributed in 3 approximately 8 fractions obtained by Dowex I column chromatography, indicating its charge and/or molecular heterogeneity. Acidic glycoconjugates consisted of hyaluronic acid (13.4%), chondroitin sulfates A plus C (39.4%), dermatan sulfate (24.6%), heparan sulfate (18.7%) and acidic glycoprotein (most probably sialoglycoprotein) (3.9%). Composition of acidic glycoconjugates in myometrium differed remarkably from that in whole uterus. Myometrium was abundant in chondroitin sulfate isomers (chondroitin sulfates A plus C plus dermatan sulfate), but lacked sulfated glycoprotein. The present results suggested that myometrium and endometrium of uterus may play quite different roles in reproduction.

Changes in free amino acids of cerebrospinal fluid and plasma in various neurological diseases.

By means of liquid chromatography we investigated free amino acids in cerebrospinal fluid (CSF) (161 subjects) and in plasma (138 subjects) in various neurological diseases and in controls. A general increase in CSF amino acids was found in Group C (samples exhibiting both pleocytosis and increasing CSF protein concentration) and in "spinal canal block" and "neuro-Behçet's syndrome". Also, some abnormalities were detected in these groups with regard to changes in CSF cells, protein or IgG%. In the case of "Guillain-Barré syndrome" we noted an increase in CSF alanine, glycine, methionine, arginine and the branched-chain group. In "multiple sclerosis" an increase was evident in CSF alanine, in plasma alanine and arginine, and in the plasma: CSF ratio of tyrosine. In patients of "motor neuron diseases (MND)" CSF glutamic acid, alanine, histidine and arginine all indicated an increase, along with the plasma: CSF ratio of tyrosine. In Group C and in "neuro-Behçet's syndrome" glycine, methionine and lysine were lowered in the plasma: CSF ratio. Also, the ratio of valine in the former and histidine in the latter indicated a decrease. These results can be supposed to reflect either biochemical abnormalities of plasma and/or the CNS, or changes in the transport systems of the blood-brain barrier.
