A new radiopaque surgical suture.

We have developed a new X-ray visible suture. It is a polyester suture containing platinum wires. The radiopaque suture was used clinically in 43 cases, and was useful in the following situations: (1) as a marker for post-operative angiography (post-operative graft angiography of aorto-coronary bypass or Brockenbrough's method), (2) as a marker for evaluation of valve reconstructive surgery or surgery of intracardiac anomalies, and (3) as a guideline for second radical repair after primary operation.

[Neural origin and modulation by hypothalamic catecholamines of pulsatile secretion of luteinizing hormone in ovariectomized rats (author's transl)].

The origin of pulsatile LH secretion and its modulation by hypothalamic catecholamines were studied in long-term ovariectomized rats. When the medial basal hypothalamus (MBH) was completely deafferentated, the suprachiasmatic nuclei being excluded from the island, episodic LH secretion with a lowered basal level persisted, and periods were unchanged. When the pituitary portal vessels were effectively lesioned electrically, pulsatile fluctuation in the plasma LH level was abolished with a lowered basal level. Various doses (from 10(-3) to 10(-6) M) of dopamine (DA), apomorphine (APO) and norepinephrine (NE) were injected into the lateral ventricle of the brain of ovariectomized rats. After injection of DA and APO separately, episodic LH secretion was markedly suppressed and LH levels were lowered. Similar effects were obtained after intraventricular injection of NE. When brain catecholamines were depleted by prior treatment with 6-hydroxydopamine (6-OHDA), basal levels in ovariectomized rats were elevated and the amplitude was augmented without noticeable change in the frequency of LH episodes. It is surmised that LH episodism originates in the MBH and both DA and NE exert an inhibitory action on it. The mode of action of the catecholamines appears not frequency-modulated, but of amplitude modulation of LH episodism.

[A case of psychomotor attack induced by the compression of carotid artery (author's transl)].

A patient with psychomotor attack beginning at the age of 60 showed no abnormal finding of EEG at rest, brain CT and other routine examination but cerebroangiography. Bilateral carotid angiography revealed the conspicuous course of anterior cerebral artery and the embryonic pattern of posterior cerebral artery. Vertebroangiography showed no image of the right posterior cerebral artery. Clinically the same as spontaneous attack was induced by the compression of right carotid artery, but the compression of left carotid artery did not provoke attack. From the finding of cerebroangiography above mentioned we guessed that the cerebral circulation impairment based on the abnormal form of Willis' circle and the process of aging caused the attack. This presumption might be supported by the fact the same as spontaneous attack was induced by the compression of right carotid artery. In this case furthermore, the evocability of attack, which the compression of carotid artery showed, was useful as one of indicators by which we judge the effect of antiepilepticus.

Identification of shellfish fatty acids and their effects on lipogenic enzymes.

The fatty acids which are common to and characteristic of shellfish, were identified by mass spectrometry and NMR spectral analyses as being: octadecatetraenoioc acid, eicosapentaenoic acid, docosapentaenoic acid and docosahexaenoic acid. When the fatty acids isolated by high performance liquid chromatography were separately intubated into rats, hepatic glucose-6-phosphate dehydrogenase (EC 1.1.1.49), malic enzyme (EC 1.1.1.40) and acetyl-CoA carboxylase (EC 6.4.1.2) were reduced more effectively as compared with linoleic acid intubation. These enzymes were reduced most markedly by eicosapentaenoic acid-intubation. The fatty acids seem to be effective components in reduction of triacylglycerol and lipogenic enzyme levels in rats fed on shellfish.

Enzymic determination of urinary free L-fucose.

Conditions necessary for the precise measurement of free L-fucose in urine by an enzymic method using L-fucose dehydrogenase have been studied. The normal urinary levels of L-fucose were 16.4 /+- 9.1 micrograms/ml in children (22.2 /+- 6.5 micrograms/mg of creatinine) and 17.7 /+- 8.5 micrograms/ml in adults (16.6 /+- 5.7 micrograms/mg of creatinine). There was a close correlation between the concentration of free L-fucose and that of creatinine. A close correlation was also found between the concentration of free L-fucose and alpha-L-fucosidase activity. Thus, it was suggested that the free L-fucose in urine reflected the metabolism of L-fucose or L-fucose-containing glycoconjugates. In a preliminary screening test, several urine samples which showed high concentrations of free L-fucose, namely cases of "fucosuria" were found.

Acute disseminated encephalomyelitis after influenza vaccination.

A 12-year-old, previously healthy boy was seen with typical signs of acute disseminated encephalomyelitis four days after influenza vaccination. Computerized tomography demonstrated an extensive subcortical low-density area in the left cerebral hemisphere. The CSF contained elevated levels of immunoglobulins, especially IgG, and an abnormal kappa:lambda chain ratio. Skin tests for hypersensitivity to egg protein were negative, but reaugmentation of CSF protein and immunoglobulin levels was observed within ten days after skin tests.

Specific perforation of muscle cell membranes with preserved SR functions by saponin treatment.

The effects of saponin on Xenopus and frog skeletal muscle fibres were examined. The twitch of Xenopus single fibres was first potentiated slightly and then irreversibly abolished by 5-10 microgram/ml of saponin. Treatment with saponin at 5-10 microgram/ml or higher concentrations for 30 min resulted in perforation of the muscle cell membrane, indicated by the following evidence. (i) Fibres became responsive to contractile activating solutions with a pCa-tension relationship similar to that of mechanically skinned fibres. (ii) Removal and re-introduction of MgATP became effective in bringing fibres into rigor and the relaxed state, respectively. (iii) After the saponin treatment large contractions due to Ca release from the SR could be elicited by substitution of Cl for methanesulphonate in the medium. (iv) The treatment decreased the optical path length across the fibre, indicating loss of soluble proteins. (v) The lattice spacing of myofilaments was wider after the treatment as in mechanically skinned fibres. Contractile response of mechanically skinned fibres and their SR responses such as Ca uptake, Ca-induced Ca release and Cl-induced Ca release were not affected by treatment with 50 microgram/ml saponin for 30 min, while 150 microgram/ml or higher concentrations severely impaired by the SR functions. It is possible, therefore, to make chemically skinned skeletal muscle fibres in which the functions of the SR are preserved by applying 10-50 microgram/ml saponin.

Application of the methylene blue staining method to the study of experimentally PNNG-induced intestinal metaplasia of the canine stomach.

To elucidate the developmental process of the intestinal metaplasia of the stomach, PNNG was orally administered to dogs and the sequential endoscopic examination was performed by using the methylene blue staining method. In the early period of the drug administration dogs showed the findings of erosive gastritis at the antral portion. In the 42nd experimental week, a clearly stained lesion was observed at the gastric angle. The histological findings of the biopsy specimens from the stained lesion showed the elongation of the foveolar epithelium and goblet cell-like mucous cells. Apparently stained lesion was observed at the antral portion in the 128th week. The close-up endoscopic appearance was very similar to that of stained intestinal metaplasia seen in the human stomach. The endoscopic methylene blue staining method as performed in the experimental dogs could provide useful results in the observation of minute lesions especially intestinal metaplasia in human stomach. This experiment, and further studies, may help to elucidate the pathogenesis of the gastric carcinoma in humans.
