The frontal ganglion of Periplaneta americana L. (Insecta): an electron microscopic and immunohistochemical study.

The frontal ganglion, part of the stomatogastric nervous system, contains about 60 to 80 neurons, 25 to 30 micron in diameter. A well developed Golgi system, producing dense-core vesicles, lysosomes, multivesicular bodies and dense bodies are abundant. Glia elements are sparsely distributed. Many nerve fibres contain granules of different size and electron density. Five groups of fibres can be distinguished: Fibres with granules of about 200 nm (type A), fibres with granules of about 160 to 170 nm (type B), fibres with granules of about 80 to 100 nm (type C) and those with synaptic vesicles of 50 nm (type D) respectively. A fifth very small type contains neither vesicles nor granules. Special attention was paid to synaptic contacts. The divergent dyad seems to be the main type in the frontal ganglion. Frequently, neurosecretory endings are observed in presynaptic position. Immunocytochemical "staining" of neurosecretory material closely corresponds to the distribution of type A fibres, as observed electron microscopically. Immunoelectrophoresis of extracts from frontal ganglia with polyspecific anti-neurosecretion-serum reveals a single precipitation line, indicating that the immunocytochemical localization of neurosecretory material is due to reaction with a specific as well as a cross-reagent anitbody.

[The colonic carcinoma. Notes on clinical aspects and therapy].

1752 cases of colon carcinoma are reviewed. History and characteristics of these tumors are described and the results of operative therapy are shown. 50% of the curatively operated patients were still alive after five years, and it seems especially important to think of the possibility of such a carcinoma. Most of our cases have shown early symptoms. It is necessary to interrogate every patient in this respect.

[Struma. Results of surgical management].

Case report about 3620 struma operations and comparison of the results with the publications of other authors. The situations of malignant strumas are considered separately. Postoperative complications, causes of death and mortality are also considered. The necessity of postoperative hormon-substitution is pointed out.

[The rectal carcinoma. Characteristics of the tumor and its position within the framework of colonic neoplasms].

For the period of 15 years (May 1960 till the end of April 1975) all cases of colon carcinoma (1752) were checked. Of these 851 carcinoma were located in the rectum. Results concerning anamnesis, diagnosis and therapy between these groups are compared and discussed. The necessity of early recognition and the possibilities of practical use are underlined and described.

[Ectopic mammary gland on the back].

A supernumerary, fully developed mammary gland between both shoulder blades above the vertebral column was discovered in a 32-year-old woman when it caused symptoms. It was removed by operation. Histological examination revealed fibrous mastopathy. The only abnormal radiological feature was a spina bifida of T6 at the site of the ectopic breast.

Synthetic S-(2,3-dihydroxypropyl)-cysteinyl peptides derived from the N-terminus of the cytochrome subunit of the photoreaction centre of Rhodopseudomonas viridis enhance murine splenocyte proliferation.

Various lipopeptides representing the N-terminal part of the cytochrome subunit of the photosynthetic reaction centre from the purple bacterium Rhodopseudomonas virdis were prepared by solid-phase peptide synthesis. These lipopeptides consisted of a S-[2,3-dihydroxypropyl]-cysteinyl (Dhc) residue N-terminally coupled to the nonapeptide FEPPPATTT. Different numbers of palmitoyl (Pam) chains were attached to Dhc via ester and/or amide bonds. The lipopeptide Dhc(Pam)2-FEPPPATTT containing two ester-bonded palmitoyl residues and a free N-terminus was a potent polyclonal activator of murine (BALB/c) spleen cells at subnanomolar concentrations. The lipopeptide Pam-Dhc(Pam)2-FEPPPATTT containing three palmitoyl residues, the two-chain lipopeptide Pam-Dhc(Pam)-FEPPPATTT containing one amide- and one ester-bonded palmitoyl residue, and the N-terminally elongated lipopeptide SLVAG-Dhc(Pam)2-FEPPPATTT were less active. The nonapeptide FEPPPATTT and the decapeptide Dhc-FEPPPATTT were only marginal splenocyte activators, even at concentrations as high as 1 microM. Thus, lipopeptide Dhc(Pam)2-FEPPPATTT constitutes the first potent splenocyte stimulation Dhc-lipopeptide described so far that contains only two fatty acid residues.

Characterization of the proctolinergic and GABAergic systems in the terminal ganglion of the male and female cockroach, Periplaneta americana, and effects of insecticide treatment. Immunohistochemistry and computer-assisted image analysis.

The distribution of proctolin- and gamma-aminobutyric acid (GABA)-containing neurons in males and females of the American cockroach, Periplaneta americana, was evaluated by means of immunohistochemistry. Sex-related differences in the localization of proctolinergic cells are restricted to the median region of the terminal ganglion. No coexistence was found for GABA and proctolin. The high specificity of the GABA staining was confirmed by labelling of identical neurons with both anti-GABA and anti-glutamic acid decarboxylase (GAD) antisera. The volume fractions occupied by GABA- and proctolin-immunoreactive elements were evaluated using a computer-based method of planimetric area percentage. The GAD staining was markedly decreased after carbaryl insecticide treatment and the volume fraction of proctolin-immunoreactive neurons was significantly reduced after application of another insecticide, dieldrin.

Distribution of GABA-like immunoreactivity in the central nervous system of the cockroach, Periplaneta americana (L.).

Gamma-aminobutyric acid (GABA) is one of the most frequently occurring neurotransmitters in the central nervous system. Using an antiserum against a GABA-protein conjugate, this has also been demonstrated for insects. The distribution of GABA-like immunoreactive neuronal elements within the central nervous system of the insect Periplaneta americana has been investigated by means of immunocytochemistry. An overview of the distribution of neuronal elements with GABA-like immunoreactivity (GLIR) in the brain is given. The ventral nerve cord was extensively investigated. The highest number of neurons with GLIR was estimated in the metathoracic ganglion (ca. 1000). The function of the suboesophageal ganglion as an inhibitory centre for the central nervous system was further confirmed by the high number of GABAergic elements and descending GABAergic pathways within the ventral nerve cord. A GABAergic innervation of skeletal musculature by so-called common inhibitory neurons was revealed for the legs of cockroaches. Nevertheless, GABA mainly seems to be a neurotransmitter in central interneurons. Identical distribution patterns of neuronal elements after immunofluorescence double-staining with anti-GABA antibodies and antibodies against the GABA-synthesizing enzyme glutamic acid decarboxylase demonstrated the high specificity of the anti-GABA serum used in this paper.
