Antibody-dependent cell-mediated cytotoxicity in humans. I. Characterization of the effector cell.

Mononuclear cells from human peripheral blood are able to destroy alloantibody-coated target cells of a human lymphoblastoid line. The effector cell is a nonadherent, non-phagocytic, medium sized lymphoid cell. The cell is not detectable in early human fetuses. Its activity is destroyed by x-ray treatment in vitro but is not inhibited by anti-immunoglobulin sera. Its efficiency in antibody-dependent cell-mediated cytotoxicity is rapidly lost in culture and this loss is prevented by stimulation with allogeneic cells.

Cystic hygroma: prenatal diagnosis and genetic counselling.

Six cases of cystic hygromas detected during second trimester ultrasound examination are reported: 4 fetuses (67 per cent) had a 45, X karyotype, 1 fetus had trisomy 18, 1 fetus had a normal karyotype (46,XX) and at autopsy multiple anomalies were observed. In the latter case the family history suggested an autosomal recessive pattern of inheritance. In order to reach a definite diagnosis and give proper genetic counselling when a fetus is found to have cystic hygroma, a fetal karyotype as well as a family and reproductive history should be obtained.

Partial trisomy 3q in a newborn female.

Partial trisomy 3q was observed in a newborn female with multiple malformations, who died in the first month of life. A balanced translocation t(3q--;15p+) was present in the mother and in other family members. The correlation between the cytogenetical and the clinical aspects is briefly discussed.

Circulating immune complexes in nephritis.

Serum samples from 115 patients with various kidney disease were studied by means of precipitation in 3.5% polyethylene-glycol (the PEG test) and by inhibition of agglutination of IgG-coated polystyrene particles induced by C1q, two tests developed for the detection of circulating immune complexes. Positive results were found in acute glomerulonephritis, particularly in its early stages, and also in chronic forms of glomerulonephritis: membranous, focal sclerosing, mesangiocapillary and focal glomerulonephritis. High levels of circulating immune complexes were found in lupus nephritis. In most cases the test results showed a good correlation with the clinical course of the diseases.

Atypical myeloproliferative disorder associated with a 21 trisomic clone.

The hematological and cytogenetical studies of a case of atypical myeloproliferative disorder are reported. The patient displayed moderate leukocytosis, anemia and thrombocytopenia. Reduced or absent growth of CFU-C was observed in the bone marrow and peripheral blood. The cytogenetical analysis revealed a trisomy 21 limited to the neoplastic clone. The overall results are discussed and compared with the rare cases of acquired trisomy 21 reported in the literature.

A new case of gamma-heavy chain disease (LIA protein) with deletion of the hinge region.

An IgG1K monoclonal component with abnormal covalent H and L chains structure (LIA protein) was identified during a systematic screening of myeloma proteins by means of non-reducing/reducing SDS-polyacrylamide gel electrophoresis. Using immunochemical and immunogenetic analysis the mutation was characterized as a hinge region deletion, with loss of L-H and H-H disulphide bridges and direct L-L bonds. Moreover, non-expression of the G1m(z) allotype suggested that the deletion might start at residue 216, a preferential site previously observed in other HCD proteins. This feature is in agreement with the discontinuous structure of immunoglobulin CH genes and suggests that an abnormal switch mechanism is responsible for the deletion.

[Value of echography in the measurement of changes in the total and visceral adipose mass].

Various methods to assess lean and fat body masses and abdominal-visceral adipose tissue were compared in 26 obese women on extremely hypocaloric diets. The following anthropometric parameters were measured before and after 15 days of extreme calorie restriction: arm circumference (A), waist circumference (W), hip circumference (H) and thigh circumference (T); W/H, W/T, A/H and A/T ratios; subcutaneous biceps , triceps , subscapular, abdominal and thigh folds; echographic thickness of abdominal muscle-aorta; area of visceral (VAT), total (AT) and subcutaneous adipose tissue measured using computed tomography, lean and fat body masses assessed by impedance measurement. Under standard conditions it was shown that echography, like impedance measurement, was the most useful method for assessing lean and fat body masses in obese subjects, whereas plicometry was not found reliable. On extremely hypocaloric diets, echography is able to assess the variations of lean and fat body masses, whereas impedance measurement overestimated the lean body mass. Localisation indexes of visceral fat based on body circumferences were shown not to be suitable for the evaluation of changes in visceral adipose tissue, at least when these were slight. Echography was also the most useful method to assess slight changes in visceral adipose tissue.

[Taste sensitivity in diabetic patients].

Taste sensitivity of 33 healthy subjects and 120 diabetic patients, either suffering from complication or not, was compared. Diabetic patients showed statistically significative reduction of sensitiveness for some tastes in some oral zones. Taste decrease looks to be related more to vascular of diabetes, than to neurologic ones.
