Transformation of Azotobacter vinelandii with plasmids RP4 (IncP-1 group) and RSF1010 (IncQ group).

Multicopy plasmid RSF1010 and four of its in vitro-constructed derivatives were mobilized by the self-transmissible RP4 plasmid into Azotobacter vinelandii UW. Modifications of the Escherichia coli transformation procedure of Cohen et al. (Proc. Natl. Acad. Sci. U.S.A. 69:2110-2114, 1972) allowed transformation of A. vinelandii strains UW and ATCC 12837 with purified RP4 or RSF1010 deoxyribonucleic acid.

HLA antigens in Jews with pemphigus vulgaris.

The association of HLA antigens with pemphigus in Jewish patients of various ethnic origins was investigated. Of 45 Ashkenazi patients, 31 (68.6%) had A26 (p less than 10(-5) and 32 (7175) had BW38 ( less than 10(-5)0. Among the 11 non-Ashkenazi patients, 7 (63.6%) had A26 and only 1 had BW38. The question whether the antigen A26 or BW38 is linked with the disease is discussed. Our findings may suggest that the primary association of pemphigus is with A26. This association may be stronger in Jewish male patients than in females. In view of the recent literature and our results it may be presumed that pemphigus in Jews is associated with antigens of Loci A and DR. No differences between pemphigus vulgaris and pemphigus erythematosus were observed.

[Peroperative plasma catecholamine levels during cardiovascular surgery].

Peroperative catecholamine levels were taken as a method for the assessment of automatic protection. Plasma adrenalin and nonadrenalin levels (spectro-fluometric technique) were measured at different pre-, per- and postoperative times in twenty eight surgical patientS: fifteen undergoing cardiac surgery and ten surgery involving the aortic bifurcation. Anaesthesia included high doses of fentanyl associated with droperidol. During aortic grafts variations in catecholamine levels were not significant, whilst during and after ECC there was a significant increase in the values. These variations were slight in comparison with those which may be seen during the excision of a phaeochromocytoma and were very similar to those obtained during effort. There were wide individual variations.

Acne with acute systemic reaction (acne fulminans?). Report of a case.

A 17-year-old girl, hospitalized because of severe cystic acne of six weeks' duration, developed symptoms of systemic disease, with fever, accelerated erythrocyte sedimentation rate, leukocytosis, thrombocytosis, arthralgia and anemia. Extensive investigation ruled out other possible causes for the systemic involvement, and there was a prompt response to steroid therapy. This case had features similar to those associated with "acute, febrile, ulcerative conglobate acne with poly-arthralgia," except that there was an absence of ulcerative lesions: nonetheless, the dominant picture was that of cystic involvement. We believe that the use of the term "acne fulminans" for this and similar cases would be appropriate.

Cell-mediated immunity in patients with less than pemphigus vulgaris greater than before, during and after prolonged treatment with steroids.

A study was made of the number of T lymphocytes and thier functional activity in 16 patients with pemphigus vulgaris, 5 with involvement of the mucosa, 3 with involvement of the skin and 8 with involvement of both; 2 patients had been suffering from the disease for 7 years, one for one year and the remainder for one to 6 months prior to this investigation. Patients were tested prior to institution of treatment, while receiving initially high doses of steroids and subsequently on maintenance doses. T lymphocyte number was determined by the E rosette technique and their functional activity by a local xenogeneic graft-versus-host reaction (GVHR) developed in our laboratories. Prior to therapy 50% of the patients showed impairment in T cell function, with no improvement during intensive steroid therapy; subsequently, when the disease was under control, there was a clear tendency for function to return to normal. There was no correlation between duration of the disease and immune competence but there appeared to be a partial correlation between the latter and the degree of involvement. Our study indicates that long-term steroid therapy not only benefits the clinical condition of the patient but is associated with an improvement in the immunological status.

Cell-mediated immunity in patients with mycosis fungoides in clinical remission: effect of thymic humoral factor on the immunocompetence of the lymphocytes.

In 10 patients with mycosis fungoides (MF) who were in clinical remission following local treatment with nitrogen mustard, cell-mediated immunity was investigated by means of tests for lymphocytes, forming rosettes in the peripheral blood, and the graft-vs-host reaction as well as intradermal skin tests. The effect of thymic humoral factor (THF) on T lymphocytes with impaired immunecompetence was also investigated. The results provided evidence of various kinds of damage to the immunologic system in most of the patients. Of the 10 patients, 4 had low percentages of T cells. Eight of the 10 had a graft-vs.-host reaction which was weak or negative, and in 4 of these, the administration of THF to the lymphocytes induced an improvement in immunoreactivity. These results together with other recently reported data indicate that the determination of cell-mediated immunity in patients with MF may be of prognostic importance. Those patients whose T lymphocytes respond to treatment with THF are probably the best candidates for immunotherapy.

[Correlation between the loss of plasmid DNA and the transition from virulent phase I to avirulent phase II in Shigella sonnei].

Transition from virulent phase I to non virulent phase II in five strains of Shigella sonnei was studied by immunofluorescence and plasmid electrophoresis. High frequency loss of phase I specific antigens was observed by use of fluorescent phase specific antibodies. Agarose gel electrophoresis of bacterial lysates showed that transition from phase I to phase II is associated with the loss of extrachromosomal DNA.

Identification of T & B lymphocytes on skin sections from patients with lymphoproliferative disorders of the skin.

The T & B lymphocytes in frozen sections of skin obtained from patients with various lympho-infiltrating disorders of the skin were identified using an immunofluorescent technique. Anti T-lymphocytes serum was used as a T marker and anti Ig GMA serum and HL-B alloantigens as B markers. The method confirmed to be efficient in preliminary experiments. Specimens were obtained from patients with lymphocytic infiltration of the skin (LIS), lymphadenosis benigna cutis (LBC), lymphoma cutis (LC) and mycosis fungoides (MF). In LIS and LBC the dominant cell was found to be the T lymphocyte, suggesting that a local immune reaction of the cell mediated type may play a role in their etiology.
