A novel A-ring pyrazole steroid, 2,3-bisaza-A-nor-1,5(10)-estradien-17 beta-ol (3), was synthesized as a potential inhibitor of steroidal NAD(P)H-dependent oxidoreductases. Compound 3 proved to be a potent inhibitor of 3(17)beta-hydroxysteroid dehydrogenase (from P. testosteroni) exhibiting a Ki of 90 +/- 20 nM. The activities of 3 alpha,20 beta-hydroxysteroid dehydrogenase (from S. hydrogenans), steroid-5 alpha-reductase (from rat prostate), and 3 alpha-hydroxysteroid dehydrogenase (from rat liver) were unaffected by pyrazole 3. Dead end inhibition studies indicate an ordered binding of cofactor prior to substrate or pyrazole inhibitor.
Cardiac troponin T (cTnT) is being increasingly used as a blood marker of acute or ongoing cardiac injury in various laboratory animals although the range of species in which it is applicable and its tissue selectivity has not been demonstrated. To address this concern, cardiac and skeletal muscle biopsy specimens from various species were homogenised and diluted, and their reactivity was then determined in the first- and second-generation immunoassays for cTnT. Cardiac tissue reactivity was found for all species studies, being highest for rats and several-fold lower for chickens and fish, and intermediate for dogs, pigs, goats, cows, sheep, horses, rabbits, and turkeys. Skeletal muscle had 10 per cent of the reactivity of cardiac muscle in the first-generation assay and 1 per cent of the reactivity of cardiac muscle in the second-generation assay. In the absence of moderate to marked skeletal muscle injury, the second-generation cTnT immunoassay has sufficient reactivity and tissue-selectivity to serve as a blood test for the discrimination between cardiac and skeletal muscle injury in a wide range of species.
Lipoblastoma is a relatively rare tumor that occurs in infancy and early childhood and arises from embryonic white fat. Although a benign tumor, lipoblastomas tend to recur and may resemble myxoid liposarcoma. The authors report 26 cases over a 15-year period at Texas Children's Hospital. There was a slight female predilection (14F:12M). The most common symptom was a painless mass with or without increasing size. The trunk, extremities, head and neck, retroperitoneum, inguinal canal, peritoneal cavity, and lung were the tumor sites. Most tumors were circumscribed lipoblastomas and the minority were diffuse infiltrative lipoblastomatosis. Reexcision for residual or recurrent tumor was necessary more frequently in patients with lipoblastomatosis. Histopathologic examination and ultrastructural examination revealed cellular neoplasms composed of immature adipocytes with relatively well-defined septa, frequent lipoblasts, a fine vascular network, and often a myxoid appearance resembling myxoid liposarcoma. Cytogenetics was performed in 4 cases with chromosome 8q abnormality being most common. The major concern with lipoblastoma in children is to completely excise the tumor to avoid leaving residual tumor and to prevent recurrences. Confusion with myxoid liposarcoma, well-differentiated liposarcoma, and typical lipomas may occur. Although asymptomatic, lipoblastomas may cause dysfunction of other organ systems due to mass effect. Complete surgical excision with at least 2 years of follow-up is the preferred therapy.
