Caffeine resistance of Saccharomyces cerevisiae.

Four caffeine-resistant haploid isolates, two resistant to 50 mM caffeine and two resistant to 100 mM caffeine, were genetically analyzed. Complementation and tetrad analysis indicated that all four mutations are alleles of the same locus. All four isolates demonstrated incomplete dominance when hybridized to the wild-type strain and dominance of high to low resistance when hybridized to one another. Differences in caffeine resistance were found between wild-type grande cells and its petite derivative.

ESR determination of membrane order parameter in yeast sterol mutants.

ESR investigations designed to determine membrane order parameter in sterol mutants of Saccharomyces cerevisiae were conducted using the membrane probe, 5-doxyl stearic acid. These mutants are blocked in the ergosterol biosynthetic pathway and thus do not synthesize ergosterol, the end product sterol. They do not require exogenous ergosterol for growth and, therefore, incorporate ergosterol biosynthetic intermediates in their membrane. Increasing order parameter is reflective of an increase in membrane rigidity. Single mutants involving B-ring delta 8 leads to delta 7 isomerization (erg 2) and C-24 methylation (erg 6) showed greater membrane rigidity than wild-type during exponential growth. A double mutant containing both lesions (erg 6/2) showed an even greater degree of membrane rigidity. During stationary phase the order of decreasing membrane rigidity was erg 6 greater than erg 6/2 greater than erg 2 = wild-type. The increased membrane order parameter was attributed to the presence of substituted sterols rather than increased sterol content or altered fatty acid synthesis.

[Tomodensitometry and spinal pathology].

The authors describe the possibilities and the limitations of the "scanner" in spinal pathology. Use of the third dimension and the certainty of absolute measurements, make this a very interesting method that can be applied to several areas of spinal pathology, especially traumatology and detection of primary and secondary tumors. It seems that this technique may be used in particular in the study of canal stenoses. There are future possibilities in view for segmentary densitometry, which gives better results than overall scintigraphy, especially in locating metastases. At present, studies are underway using contrast solution in order to study the possibilities of interpreting the content-container relationship.

[Postoperative sciatica from epidural fibrosis and lumbar arachnoiditis. Results of 38 repeat operations].

Lumbar epiduro-arachnoiditis is a well-known complication of surgery of the intervertebral disc. The epidural fibrous scar is the normal outcome of inflammatory activity secondary to the mechanical tissue disturbance resulting from surgery. In certain individuals, perhaps genetically predisposed as excessive quantity of fibrous tissue is deposited in the epi and/or nitradural space. This abnormal situation, comparable to cheloid cutaneous scars, is perhaps at the origin of the clinical symptoms. The authors report the clinical and radiological signs observed in 38 patients having had at least one operation for discal hernia, and who underwent further operations in the aim of freeing the roots and the dural sack from fibrous compression. Results of excision of the epidural "cheloid" were good in 13 cases, and average in 8 others. There was a complete failure in 17 other patients. Three explanations were offered to explain the frequency of the failures: 1) formation of a new cheloid, 2) difficulty of neurolysis of the arachnoiditis, 3) possibility of intrinsic lesions of the nerve associated with the epiduro-arachnoiditis.

[Automatic determination of the alcohol level of dry wines].

An automated technique for the alcoholometric grade determination based upon measurement of dilution heat of ethanol in wine and appropriate reagent was tested. Interferences with wine other components was studied along with methods to minimize them. Two sets of determinations on more than 100 samples of various qualities (red wines, roses wines, white wines) and various geographical sources were performed and they were compared with the pycnometric reference technique. It is shown than this comparative study that for almost 95 p. 100 of the samples the alcoholometric grade can be determined with this automated technique with an accuracy better than 0,1 p. 100.

Further findings on assaultiveness and alcohol use in interpersonal disputes.

In a previous study uniform observational data were collected regarding family disputes managed by police officers; those data contradicted views commonly held by social scientists and by police about the roles of assaultiveness and alcohol use in these events. In the present study police officers employed systematic naturalistic observation during their interventions as third parties in disputes between nonfamily members as well as those between family members. Tentative conclusions about assaultiveness, based on the data of both studies, are that it: does not usually precede the arrival of police in such disputes; is associated with interpersonal closeness of the disputants; is associated with poverty rather than race; and is as likely in a middle-class small city as in a poor inner-city area. Tentative conclusions about the use of alcohol in police-managed interpersonal disputes are: that these disputes are not usually influenced by alcohol use; and that assaultiveness is not related to alcohol use in such disputes.

Sterol mutants of Saccharomyces cerevisiae: chromatographic analyses.

The sterols accumulated by ergosterol deficient mutants of the genes erg6, erg2, erg3, and erg5 (formerly po11, po12, po13, and po15) have been analyzed by gas liquid chromatography. Together with pure sterols obtained from the mutants, they were characterized on SE-30, OV-17, and OV-225. The effects of molecular structure on the retention characteristics of a range of C28 ergostane sterols have been studied. The double mutants obtained by crossing the single mutants were also analyzed and their sterols identified where possible. The effects of the erg mutations on the control of sterol biosynthesis in yeast are discussed.

[Xeroradiography : possibilities and value in osteo-articular pathology].

The authors report their first experiences with a new method of obtaining radiological information : radioxerography. Based on physical principles, replacing the older chemical basis, this new technique makes it possible to improve greatly, and with fewer exposures, the amount of information that can be obtained from radiological investigations. The advantages of the new method, based on the edge effect, the selectivity of the contours, and the spreading of the contrast, make possible much improved definition of both the bone structure and that of the neighbouring soft tissues. With reference to these first experiences, the authors summarize the possibilities and the preferred indications of the method, which although it remains a complementary and not a substitute method, appear very interesting and full of promise for the future.

[Gonalgia following meniscectomy. Arthrographic fingings. Review of 200 case records and radio-clinical comparison].

The authors have examined by arthrography two hundred knees which had remained painful after meniscectomy. In 170 cases this examination revealed a possible explanation for the pain; residual pathological posterior horn, lesion of the other meniscus, degenerative changes of the cartilages, lesions of the ligaments. 91 patients sustained a second surgical procedure. In 81 cases the arthrographic findings were confirmed, in 10 cases surgical exploration found more lesions than expected.
