Egg white-specific IgE and IgG4 antibodies in atopic children.

IgE, IgG1, and IgG4 antibodies in egg allergy were evaluated. It was found that there was a correlation between clinical egg sensitivity and IgE RAST from birth to 5 years of age, although IgG1 and IgG4 antibodies tended to increase with age in egg-allergic subjects. The role of IgG subclass antibodies in this allergic disorder is also discussed.

Rapid change in mutation rate in a local population of Drosophila melanogaster.

The lethal and detrimental loads per second chromosome rapidly increased from 1968 to 1970 in a local population of Drosophila melanogaster in Japan (lethal load, from about 0.16 to 0.38; detrimental load, from 0.125 to 0.231 [Watanabe, T. K., Watanabe, T. & Oshima, C. (1976) Evolution 30, 109-118]). When the homozygous loads were measured in 1983, the lethal load had decreased to approximately the original amount (0.19) but the detrimental load had stayed high (0.241). The rise and fall of the lethal load can be accounted for by a P-type element that invaded a population with M cytotype, producing a high mutation rate. The mutation rate fell back to the earlier value after the cytotype became P. That the detrimental load did not decrease can be explained by assuming a longer persistence for detrimental mutations in the population. Evidence for a P-type mutator factor is that the mutation rate of the wild-type chromosomes differs between the different cytoplasmic and chromosomal backgrounds, being lower in the background from which the chromosomes were taken.

Effect of a chemically defined diet in liquid form on colon carcinogenesis in rats.

The effects of ad libitum feeding of a chemically defined diet in liquid form on the incidence and histology of colon cancer induced by 10 weekly sc injections of 7.4 mg/kg of azoxymethane [(AOM) CAS: 25843-45-2] were investigated in W-rats. The chemically defined diet was adjusted once every 24 hours from 4 weeks before injection of the carcinogen to the end of the experiment at week 40. Oral administration of the defined diet resulted in significant increase in the incidence of colon cancer at week 40. Histologic examination showed that unlike adenocarcinomas with high mucin-producing activity, which were common in rats on pellet diet, most of the adenocarcinomas that developed in rats fed on defined diet were highly or well differentiated, with a typical glandular pattern. Administration of the chemically defined diet also resulted in marked colon mucosal hypoplasia and reduced gastrin levels in the serum at weeks 4 and 40.

Effect of aculeacin A, a wall-active antibiotic, on synthesis of the yeast cell wall.

A wall-active, amphophilic antibiotic aculeacin A significantly but incompletely inhibited in vitro the activity of beta-(1,3)glucan synthase prepared from highly susceptible yeasts Saccharomyces cerevisiae and Candida albicans. In contrast, comparable cell-free preparations from S. cerevisiae active in chitin synthase or mannan synthase were insensitive to the antibiotic, suggesting selectivity of its action in synthesis of the yeast cell wall. An electron microscopic study of the effects of aculeacin A at 0.31 micrograms/ml, the optimally active concentration, on osmotically stabilized C. albicans cells revealed morphological alterations in both cell walls and cell membranes. Deformation in contour and derangement of the layered structure of the cell wall were prominent. In addition, massive fibrous material of beta-glucan-like microfibrils was occasionally extruded from the cell surface. Accompanying this effect on the cytology of the cell wall, ultrastructural and functional impairment of the cell membrane was demonstrated by transmission and freeze-fracture electron microscopic techniques. These data suggest that aculeacin A affects synthesis of the yeast cell wall through not only selective blockage of beta-(1,3)glucan synthase, as a result of a primary interaction with the cell membrane, but also inhibition of the fabrication of beta-glucan or other wall components into well-organized cell walls.

Intracranial pressure monitoring by a subdurally placed silicone catheter: technical note.

A silicone catheter manufactured originally for ventricular fluid drainage was used for continuous monitoring of intracranial pressure. It can be placed under the dura mater immediately before replacing the craniotomy flap or by introduction through an ordinary burr hole. Pressure is monitored isovolumetrically by connecting the catheter to a pressure transducer mounted at the bedside. This technique has been used in 34 neurosurgical patients for periods of up to 12 days without any untoward effects. This monitoring system by a subdurally placed silicone catheter is simple, safe, inexpensive, and acceptably reliable. It has a place in clinical practice, particularly when the lateral ventricle cannot be tapped because of its deviation or collapse.

[Percutaneous fine needle biopsy for malignant mediastinal tumors].

Seventeen patients with mediastinal malignant tumors (four neurogenic tumors, four thymomas, three germ cell tumors, two malignant lymphomas and one each of carcinoid, malignant melanoma, multiple myeloma and carcinoma simplex) were examined for the evaluation of percutaneous fine needle biopsy. In 16 cases of them, adequate materials for cytodiagnosis were obtained. Fifteen of the 17 cases (88%) were positive for malignant cells, and 1 case was negative. As for the histological types, 9 cytological diagnoses (60%) were compatible and 4 diagnoses (27%) were not. Regarding complications of percutaneous fine needle biopsy, 1 case of subcutaneous needle tract implantation was noted. Percutaneous fine needle biopsy is strongly recommended for diagnosis of mediastinal tumors.

Promotion by vagotomy of gastric carcinogenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine in Wistar rats.

The effects of truncal vagotomy after administration of N-methyl-N'-nitro-N-nitrosoguanidine on the incidence and number of gastric carcinomas and gastric acid secretion, gastrin secretion, antral pH, and duodenal reflux were investigated in inbred Wistar rats. Rats were subjected to truncal vagotomy after N-methyl-N'-nitro-N-nitrosoguanidine treatment. Vagotomy significantly increased the incidence and number of adenocarcinomas of the glandular stomach. It also resulted in significantly more atypical glandular hyperplasias, which are precursors of gastric cancer. Furthermore, it caused a decrease in gastric secretion and an increase in mucosal pH in the antrum but did not increase duodenal reflux. These findings indicate that vagotomy has a promoting effect on the development of gastric cancers. The reduced gastric acid secretion, but not duodenal reflux, may be related to this increased incidence of gastric cancer.

[Hematoma of the pineal region: a case report].

A 44-year-old man came to our clinic, complaining of slowly progressive disturbance of visual acuity and of ocular movement. This patient suffered from headache, narrowing of visual field and polyuria about 20 years ago, and received surgical and radiation therapy under the diagnosis of pituitary adenoma. Clinical symptoms and signs of this patient, except for bitemporal hemianopsia, almost completely disappeared after these treatment. The detailed information about the histology and radiation dose are not available at the present time. CT scan in our clinic revealed a round low-density area at the suprasellar region and a high density area at the left quadrigeminal cistern. Pineal calcification was compressed to the right about 2-3 mm from midline. This high density mass were not enhanced with contrast medium. Vertebral angiography showed a slight lateral displacement of the left medial posterior choroidal artery. Specimen of tissue removed 20 years ago was reexamined but definitive diagnosis could not be established. Presumptive diagnosis of an ectopic pinealoma in the suprasellar region treated successfully 20 years ago, and its recurrence in the pineal region was made. On May 24, the patient underwent a posterior fossa craniectomy and the pineal region was explored via the infratentorial supracerebellar approach. On sectioning the precentral cerebellar vein, yellowish mass was seen in the quadrigeminal cistern. Aspiration of this mass yielded dark red liquid hematoma. Incising into the capsule, a dark brownish mass of about 4g was removed en bloc.(ABSTRACT TRUNCATED AT 250 WORDS)
