The mode of binding of potential transition-state analogs to acetylcholinesterase.

Phenylacetone, 4-phenyl-2-butanone, and 4-oxopentyltrimethylammonium chloride were tested as potential transition state analogs for eel acetylcholinesterase (acetylcholine hydrolase, EC 3.1.1.7). Phenylacetone is a competitive inhibitor of the enzyme but not a transition state analog, since its binding constant is similar to that for the substrate phenyl acetate. 4-Phenyl-2-butanone binds 6-18 times more tightly than the inhibitors 4-phenyl-2-butanol and N-benzylacetamide and the substrate benzyl acetate and also blocks inactivation of the enzyme with methanesulfonyl fluoride. However, its binding is independent of pH in the range 5-7.5, whereas both V and V/Km for benzyl acetate hydrolysis decrease with decreasing pH in this range. These data indicate a specific but weak interaction between the ketone carbonyl and the enzyme, but probably do not justify considering this compound a transition state analog. 4-oxopentyltrimethylammonium iodide has previously been shown to bind about 125 times more strongly than the substrate acetylcholamine. It also binds about 375 times more strongly than the alcohol 4-hydroxypentyltrimethylammonium iodide. Furthermore, the ketone protects the enzyme from inactivation by methansulfony fluoride, while the corresponding quaternary ammonium alcohol accelerates this inactivation reaction. This additional information confirms that the ketone is a transition state analog.

Symptomatic cataplexy.

The case is described of a man who developed attacks of cataplexy, narcolepsy, and sleep paralysis because of microglioma which infiltrated the walls and floor of the IIIrd ventricle and the upper brain stem. The mechanisms by which the pathology is related to the symptoms are discussed.

The driniking driver: a blueprint for some further action.

Statistical information relating to the offence of drunken driving in New Zealand is reviewed. The relationship between alcoholism and this offence is discussed. Two groups of offenders who should be dealt with separately can be identified. A plan is put forward with an outline of methods which could be used for dealing with this problem.

Isolation, preparation and the amino acid composition of 4 milk-fat globule membrane proteins solubilized by treatment with sodium dodecyl sulphate.

Milk-fat globule membrane (MFGM) proteins were solubilized by treatment with SDS. Four of the major proteins were isolated as SDS complexes using column chromatography. The purity of each isolate was determined by SDS polyacrylamide gel electrophoresis and sufficient of each protein was obtained for amino acid analysis. The amino acid compositions of the isolated MFGM proteins and a total MFGM protein extract were determined. Differences in amino acid composition were found in particular between the major MFGM glycoprotein and the other 3 membrane proteins. The relationships of the amino acid composition to protein properties and structure are discussed.

Electrical activity of the proventriculus of the polychaete worm Syllis spongiphila.

The straited myoepithelial cells of the proventriculus of Syllis spongiphila are composed of only one or two sarcomeres that may reach 40 mum in length. Experiments were performed to study some of their electrophysiological properties and their synaptic control. The mean resting potentials recorded in two different bathing media were 59-1 +/- 5-5 mV (S.D., n=91) and 62-5 +/- 6-3 mV (S.D., n=98). At rest the membrane potential is determined largely by permeability of the membrane to K+ ions, but the membrane is also permeable to other ions. On a semilogarithmic plot of membrane potential v. [K]o the mean slope of the data points from 9 to 90 mM-[K]o was 48 +/- 3 mV for a 10-fold change in [K]o. The anterior end of the animal was stimulated with a suction electrode to elicit activity of nerve fibres that innervate the proventriculus. Single indirect stimuli usually evoked hyperpolarizing or biphasic responses, and occasionally depolarizing responses, from the myoepithelial cells. The depolarizing synaptic potentials exhibited a faster time course than the hyperpolarizing ones. The rise time to peak ranged from 20 to 35 ms for simple depolarizations (n=32) and 25-75 ms for simple hyperpolarizations (n=103). Time to decay to half amplitude ranged from 20 to 55 ms for depolarizations (n=29) and 62-135 ms for hyperpolarizations (n=87). Low frequency (is less than or equal to 4 Hz) trains of indirectly applied stimuli elicited mainly hyperpolarizing responses; higher frequency (5-40 Hz) trains elicited complex responses composed of hyperpolarizations and depolarizations. Hyperpolarizations were selectively and reversibly abolished in chloride-free solutions. The reversal potential of the hyperpolarizing synaptic potential was -104 +/- 3 mV (S.D., n=8, 2 preparations). In calcium-free solution both hyperpolarizations and depolarizations were almost completely abolished. 4 mM-Mn2+ added to the bath almost completely abolished the depolarization but not the hyperpolarization. It was not clear whether Mn2+ acted at the presynaptic membrane, the postsynaptic membrane or both. The myoepithelial cells are electrically coupled. The mean space constant of five preparations was 0-52 mm (range 0-40-0-66 mm).

Very early termination of pregnancy (menstrual extraction).

Very early termination of pregnancy was performed on 424 women in three London teaching hospitals. Altogether 90% of the women were no more than 14 days overdue, and 67% of these had histological evidence of pregnancy. The procedure differed little in technique or its acceptability to the patient from termination done later in the first trimester. The similar incidence of complications suggested that it is not an alternative to conventional contraception. The response of patients, general practitioners, and referral agencies, however, indicated that there is a definite need in the community for a very early termination service.
