[Non-Hodgkin's malignant lymphoma with central nervous involvement--report of 7 cases].

We encountered 7 non-Hodgkin's lymphomas involving the central nervous system (CNS) among 78 cases of lymphomas seen at the 3rd Dept. of Internal Med. between 1965 and 1980; 5 cases were diffuse histiocytic lymphomas (DH) and 2 were diffuse poorly differentiated lymphomas (DPDL). The age ranged from 27 to 66 years, there were 6 males and 1 female. In 4 cases, CNS involvement was the presenting symptom at the time of diagnosis of lymphoma. The diagnostic procedures in CNS lymphoma included complete neurological physical examination by a neurologist, spinal fluid cytology, surface marker study on lymphocytes of spinal fluid, if possible, computed tomography scan, and myelogram if spinal cord compression was suspected. The prognosis of patients with CNS involvement by lymphoma was poor even if early diagnosis was made. The conventional therapy for CNS lymphoma included cranial irradiation, systemic chemotherapy, and intrathecal instillation of methotrexate and/or cytosine arabinoside, or a combined modality; however, these treatments were only effective temporarily.

[Local hyperthermia combined with radiation in the treatment of refractory cancer].

Localized hyperthermia combined with radiation was applied to selected patients who had tumor known to be refractory to conventional treatment modalities. Hyperthermia was administered by 2,450 MHz microwave or 13.56 MHz radiofrequency according to the site of tumors. In most patients intratumor temperatures of 43 degrees C for 30 minutes was administered twice weekly immediately after irradiation with 4 Gy. A complete regression was obtained in 31 out of 54 lesions (57.5%). The results of our study have shown that localized hyperthermia in combination with radiation is an effective and safe modality for the treatment of radioresistant cancers.

Effect of guanidination on subunit interactions in hybrid isozymes from pig lactate dehydrogenase.

The thermostability of the isozymes from pig heart (H) and muscle (M) lactate dehydrogenase (L-lactate:NAD+ oxidoreductase, EC 1.1.1.27) decreased in the order of H4 greater than M4 greater than H3M greater than HM3 greater than H2M2, while the thermostability of the isozymes from guanidinated H4 and M4 increased as guanidinated H monomer was substituted by M monomer. The increased thermostability of H4 increased as guanidinated H monomer was substituted by M monomer. The increased thermostability of H4 on guanidination of five lysine residues per subunit was due to the decrease in the standard activation entropy, and no change in the standard activation enthalpy was observed. The more increased thermostability of H4 on further guanidination of lysine residues from 5 to 15 per subunit was due to the increase of the standard activation enthalpy which overcame the decrease in thermostability due to the increase of the standard activation entropy. The results indicate two different mechanisms of stabilization depending on the degree of guanidination. The increase of thermostability, as measured by the change of the standard activation free energy for thermal inactivation of H2M2, was almost the same as that of H4 on guanidination of five lysine residues in an H monomer. This result and the order of thermostability of the isozymes from unmodified and guanidinated H4 and M4 suggest that the increase of thermostability of hybrid isozymes on guanidination of H monomer is due to the change of the heterologous subunit interactions.

"Waldenström's macroglobulinemia" terminating in immunoblastic sarcoma. A case report.

An autopsy case of 69-year-old man with Waldenström's macroglobulinemia terminating in immunoblastic sarcoma is reported. In the course of this case, the appearance of plasmocytoid lymphocytes was initially observed in peripheral blood smears and bone marrow punctures, but lymph node biopsy and bone marrow aspirations performed in the terminal stage, as well as autopsy, revealed a diffuse monotonous proliferation of immunoblasts in the lymphoreticular tissues. With the immunofluorescence method and PAP method of Taylor,11 IgM kappa type of immunoglobulin was demonstrated in the cytoplasm of the proliferating cells, and the electron microscopic enzyme-labeled antibody technique identified localization of the immunoglobulin in the nuclear envelope and rough endoplasmic reticula of the immunoblasts. The results indicate that the initial appearance of plasmocytoid lymphocytes in this case and its terminal transformation into immunoblastic sarcoma are rare phenomena occurring in a single clone proliferation disorder of B-cell synthesizing IGM kappa immunoglobulins.

Lung phosphatidylcholine and fasting in male and female rats.

Lung from male and female rats fasted for 4 days were used. Phospholipid, phosphatidylcholine and its molecular species were analyzed in lungs frm these rats and effects of fasting upon the biosynthesis of phosphatidylcholine in lungs from both sexes were determined using radioactive choline. The molecular species of phosphatidylcholine in both male and female rats did not differ with fasting except the monoenoic species. Incorporation of choline into phosphatidylcholine in both male and female rats significantly increased after fasting, but distribution of radioactivity in phosphatidylcholine yielded similar values in each group. These results suggest that the decrease of saturated phosphatidylcholine content after fasting may not be due to specific change in saturated phosphatidylcholine.

Pathogenesis of fatty liver in rats fed a high protein diet without pyridoxine.

Fatty liver induced in rats by a high protein diet without pyridoxine was studied morphologically. The microscopic change was characterized by accumulation of fat in hepatocytes of the centrolobular and midzonal areas. Electron microscopic examination at an early stage showed marked accumulation of small osmiophilic particles in the granular endoplasmic reticulum and vesicles throughout the cytoplasm with similar particles in the spaces of Disse. After 4 weeks, numerous lipid droplets of various sizes were seen in pericanalicular lysosomes in hepatocytes with concomitant increase in the triglyceride level. The droplets gradually formed larger droplets in the cytoplasm. After 8 weeks, myelin figures together with fat droplets were seen in continuity with the endoplasmic reticulum and occasionally crystal clefts were observed within lysosomes of hepatocytes. These findings suggest that development of fatty liver results from impaired lysosomal degradation of lipid.

Simple purification method for anti-glycolipid antibody using polystyrene latex beads coated with gangliotetraosylceramide.

A simple method for the purification of antiglycolipid antibody is described. Polystyrene latex beads coated with gangliotetraosylceramide (GA1) were used as immunoadsorbent for the purification of anti-GA1 antibody. The antibody bound to the GA1-coated latex beads was released with 1 M NaSCN solution. The glycolipid-coated latex beads seem valuable not only for the purification of antibodies but also as a tool for investigating the function of glycolipids.

Absence of DNA in peroxisomes of Candida tropicalis.

Yeast peroxisomes were purified to near homogeneity from cells of Candida tropicalis grown on oleic acid for the purpose of examining the possible presence of DNA in this organelle. The purification procedure includes the effective conversion of cells to spheroplasts with Zymolyase and sodium sulfite and the separation of the organelles at extremely low ionic strength. The mitochondrial contamination was less than 1%, based on several criteria, and the yield of peroxisomes was about 40%. The purified peroxisomal fraction contained a very small amount of DNA, which yielded restriction fragments indistinguishable from those of mitochondrial DNA. The absence of DNA in peroxisomes was also supported by cesium chloride density gradient centrifugation of the organelles lysed with a detergent, staining of the organelles with a fluorescent dye specific to DNA, and labeling of the DNA with [3H]adenine.

The effect of hypoxia on insulin and glucagon secretion in the perfused pancreas of the rat.

It has been reported that insulin secretion decreases during hypoxia both in vitro and in vitro, while an increase in glucagon secretion is found only in vivo. The effect of acute hypoxia on the secretion of glucagon and insulin was studied in the perfused rat pancreas. Phentolamine, an alpha-adrenergic blocker, was perfused during the period of hypoxia to elucidate the role of alpha-adrenergic stimulation. Sodium ATP and dibutyryl cAMP were also administered to study their effects on insulin and glucagon responses during hypoxia. In the present experiments, insulin secretion was suppressed while glucagon secretion was increased during hypoxia. Phentolamine did not cause any change in insulin of glucagon secretion. When dibutyryl cAMP was added, the increased glucagon secretion was reduced to the basal level, whereas the decreases in insulin secretion were not altered. The addition of sodium ATP reversed the hypoxia-induced decrease in insulin and the increase in glucagon secretion. These results suggest that a decrease in ATP production, which leads to impaired cAMP generation, pays a role in, and that alpha-adrenergic stimulation does not participate in the changes in, insulin and glucagon secretion during hypoxia in vitro.

Effect of pyridoxal phosphate deficiency on aromatic L-amino acid decarboxylase activity with L-DOPA and L-5-hydroxytryptophan as substrates in rats.

This paper describes the distribution of aromatic L-amino acid decarboxylase (AADC) activities in fourteen tissues (eight peripheral tissues and six brain regions) of semicarbazide (SC)-treated rats, using both L-DOPA and L-5-hydroxytryptophan (L-5-HTP) as substrates. The distribution of pyridoxal phosphate (PLP) was also measured in control and SC-treated rats. SC-treatment decreased the PLP concentration in all tissues (about 50-60% of control). AADC activities towards L-DOPA and L-5-HTP as substrates were also decreased significantly in almost all tissues of SC-treated rats. After the addition of exogenous PLP in vitro, AADC activities were recovered only partially in most tissues, but the recovery patterns were not parallel between L-DOPA and L-5-HTP as substrates. L-DOPA decarboxylase activity was more sensitive to PLP-deficiency than L-5-HTP decarboxylase activity in the same tissues. Serum AADC activities were decreased drastically using both L-DOPA and L-5-HTP as substrates. No serum AADC activity was detected in SC-treated rats using L-DOPA as substrate, but low activity was detected in the same sample using L-5-HTP as the substrate; both activities recovered completely after in vitro addition of 10 microM PLP in the incubation mixtures.

Human pancreatic islet cell clones secreting insulin, glucagon and somatostatin: immunocytochemical and functional studies.

Human pancreatic A-, B- and D-cell clonal strains, named JHPG-1, JHPI-1 and JHPS-1, were established successfully from adult and fetal pancreata by the single cell plating feeder layer method using a modified Rose's chamber. This is the first time that insulin-, glucagon- and somatostatin-producing clonal strains have been separated into continuous clonal cell lines. The cultured cells are epithelial in nature, free of fibroblast contamination, and can be cloned. Under the phase-contrast microscope, the B-cell clone (JHPI-1) was generally oval or round in shape, the A-cell clone (JHPG-1) was bipolar, and the D-cell clone (JHPS-1) was nerve-like with cytoplasmic processes. By the use of immunocytochemical techniques, insulin-, glucagon- and somatostatin-like immunoreactivities were detected in each clone respectively. By radioimmunoassay it was revealed that each clone produced a single pancreatic hormone. The B-cell clone especially, was found to secrete insulin amply and continuously, for over 150 days. The glucagon release responses by the A-cell clone to insulin and glucose were also studied. The clonal strains obtained in this study provide useful systems for the investigation of the cell-biological aspects of human islet cells in vitro.
