Preparation of seeding type immobilized microorganisms and their degradation characteristics on Di-n-butyl phthalate.

OBJECTIVE: To study the preparation of seeding type immobilized microorganisms and their degradation characteristics on di-n-butyl phthalate (DBP). METHODS: Diatomite, clinoptilolite, silk zeolite, and coal fly ash were chosen as reserved materials and modified. Their adsorption capacity and intensity in the bacteria were determined and the best carrier was picked out. The seeding type immobilized microorganisms were prepared by the best carrier and then it degraded DBP under different primary concentration, vibration rate, pH, temperature in the presence of metal compounds. RESULTS: The adsorption capacity of the modified coal fly ash, silk zeolite, clinoptilolite and zeolite was 44.2%, 71.6%, 84.0%, and 94.4%, respectively, which was 1.66, 1.49, 1.37, and 1.16 times as high as that of their natural state. Their adsorption intensity was 72.1%, 90.5%, 90.1%, and 91.1% in turn. The modified diatomite was selected to prepare the seeding type immobilized microorganisms. When the primary DBP concentration was 100 to 500 mg/L, the DBP-degraded rate of the immobilized microorganisms could be above 80%. The degradation activity of both the dissociative and immobilized microorganisms was higher in vibration than in stillness. When pH was 6.0 to 9.0, the DBP-degraded rate of the immobilized microorganisms was above 82%, which was higher than the dissociative microorganisms. When the temperature was between 20 degrees C and 40 degrees C, the DBP-degraded rate could reach 84.5% in 24 h. The metal compounds could inhibit the degradation activity of both the dissociative and immobilized microorganisms. The degradation process of the immobilized microorganisms could be described by the first-order model. CONCLUSION: The adsorption capacity of the diatomite, clinoptilolite, silk zeolite and coal fly ash on DBP-degrading bacteria can be improved obviously after they are modified. The modified diatomite is best in terms of its adsorption capacity and intensity. Its seeding type immobilized microorganisms could degrade DBP effectively and is more adaptable to DBP load, temperature, pH than the dissociative microorganisms. The metal compounds could inhibit the activity of both the immobilized and dissociative microorganisms. The degradation reaction of the immobilized microorganisms on DBP is consistent with the first-order model.

[Changes of orthodontic tooth in different healing extraction sites].

OBJECTIVE: To compare the canines root resorption and alveolar bone height when they were moved into different healing extraction sites. METHODS: Twenty patients with overjet were treated by extracting the maxillary first premolars. By random determination, distalization of different lateral upper canines started after 1 week and 12 weeks. Every 4 weeks use a force of approximately 100 g to distalize the canines. The experiment was ended when bilateral canines have contacted with the second maxillary bicuspids completely. Periapical radiography of the canine in the end of the distalization was made. RESULTS: External apical root resorption and alveolar bone height of the canines have no significant difference between the different healing extraction sites. CONCLUSION: Tooth extraction site healing has not much effect on the canine external apical root resorption and alveolar bone height, when it was distalization. In practice, tooth movement into extraction sites can start early.

[Research progress on cell apoptosis during embryonic development of the zebrafish (Danio rerio)].

Cells have a built-in death program through the regulation of genes. Apoptosis (programmed cell death) plays a very important role in the development of the vertebrate embryo. The zebrafish (Danio rerio) has become a useful and ideal model in molecular developmental biology for in vivo study on the function of apoptosis in morphogenesis, sex differentiation and so on, and has already been garnering more and more attention. At present, much progress has been made regarding apoptotic pathways in zebrafish development. Caspases and other regulatory apoptotic genes have been cloned successfully. Moreover, transgenic zebrafish embryos have been induced to undergo apoptosis in vivo to study apoptotic signaling pathways and the abnormal morphological changes during embryogenesis. Transgenic zebrafish has provided a powerful tool for illuminating the interaction between regulatory apoptotic genes and vertebrate development.

[Experimental study of acellular xenogenic dermal matrix and allogeneic sclera as wrapping materials for hydroxy apatite implantation].

OBJECTIVE: To observe the experimental results and histopathological changes of acellular xenogenic dermal matrix (X-ADM) and allogeneic sclera used as wrapping materials of hydroxy apatite (HA) ocular implants in New Zealand white rabbits. METHODS: Twenty-four rabbits received unilateral eye enucleating and the sockets were implanted with HA spherical implants wrapped with either acellular xenogenic dermal matrix or allogeneic sclera at random. The rabbits were examined for inflammation and implant exposure and sacrificed at 1, 2, 4, 6, 8 and 12 weeks after implantation. The sockets with the grafts were exenterated and the specimens were assessed histopathologically and ultrastructurally with light or transmission electron microscopy for the changes in inflammation reaction and vascularization. RESULTS: Compared to allogeneic sclera at the same stage of implantation, acellular xenogenic dermal matrix demonstrated more active and earlier growth of fibroblasts and new vessels with abundant collagen deposition. There were few inflammatory cells and no rejection was found. CONCLUSION: This experiment showed that the acellular xenogenic dermal matrix, with fast neovascularization and low immunity, can be an ideal material of ocular implant and a good substitute for allogeneic sclera.

[Inhibitory effect of diacerein on osteoclastic bone destruction and its possible mechanism of action].

AIM: To study the inhibitory action of diacerein on the formation of osteoclasts (OCLs) and their activity in bone resorption as well as the relationship between this action and the expression of osteoprotegerin (OPG) and receptor activator of NF-kappaB ligand (RANKL) in MC3T3-E1 cells. METHODS: A coculture system constituted with MC3T3-E1 cells and bone marrow cells for osteoclasts formation was established in vitro. TRAP-positive and multinucleated cells with three or more nuclei in each cell were counted as osteoclasts and the number of pits formed on the dentine slices was determined to judge the activity of osteoclasts. Western blotting, RT-PCR and flow cytometer were used to detect the expression of OPG and RANKL in MC3T3-E1 cells. RESULTS: Diacerein significantly inhibited the formation and function of the cultured osteoclasts stimulated by IL-1beta. sRANKL could reverse the effect of diacerein. Diacerein inhibited protein and mRNA expression of RANKL but enhanced those of OPG in MC3T3-E1 cells. CONCLUSION: Diacerein may inhibit osteoclastic bone destruction through the inhibition of RANKL expression and the increase of OPG expression in MC3T3-E1 cells.

[Molecular forms of TFF1 in normal gastric mucosa and its relationship with canceration].

OBJECTIVE: To study the molecular forms of TFF1 in normal gastric mucosa and its expression in normal, gastric carcinoma, atypical hyperplasia, and intestinalized gastric mucosa. METHODS: The molecular forms of TFF1 in normal gastric mucosa was observed by western blotting. The expression of TFF1 in normal, gastric carcinoma, atypical hyperplasia, and intestinalization gastric mucosa was assayed immunohistochemically. RESULTS: TFF1 existed in normal gastric mucosa in forms of monomer, dimer and 21-kD TFF1 complex, with the last being the richest. TFF1 was expressed mainly in the epithelial cytoplasm of the mucosa in the gastric body and antrum, especially around the nucleus, and the closer to the lumen, the higher the expression. TFF1 expression in the tissues adjacent to gastric carcinoma was higher than that in normal gastric mucosa (P<0.001), and the expression in gastric adenocarcinoma was positively correlated to differentiation of adenocarcinoma. No TFF1 was expressed in poorly differentiated adenocarcinoma. The expression of TFF1 in moderate and well differentiated adenocarcinoma was a little lower than that in normal mucosa (P>0.05). The gastric mucosa with atypical hyperplasia had significantly higher TFF1 expression than normal gastric mucosa (P<0.001), and TFF1 was not detected in intestinalized gastric mucosa. There was no significant difference in TFF1 expression between gastric mucosa around the intestinalized tissues and normal gastric mucosa (P>0.05). CONCLUSIONS: TFF1 plays an important part in protection and restitution of the gastric mucosa, and TFF1 may be related to suppression and differentiation of carcinoma.

[Effect of erzhi tiangui recipe on ovarian reactivity in elderly sterile women].

OBJECTIVE: To observe the clinical effect of Erzhi Tiangui Recipe (ETR), a traditional Chinese recipe for strengthening Shen and nourishing Tiangui, in improving the ovarian reactivity in elderly sterile women. METHODS: Sixty-six elderly sterile women receiving in vitro fertilization pre-embryo transfer (IVF-ET) were randomly divided into two groups: the ETR group treated with ETR plus follicle stimulating hormone (FSH) and the control group treated with FSH alone, 33 cases in each group. Changes of symptoms and the relative parameters were observed, including the dosage of FSH used, the serum hormone levels at different periods, endometrium thickness and arteriopalmus of uterus and ovary on the day of HCG injection, follicle number, follicular oocyte number, mature follicular oocyte rate and fertilization rate, prime embryo rate, and pregnancy rate. Moreover, the correlation of the symptom score of colpoxerosis (CR2) and serum estradiol (E2) level was analyzed after treatment (on the HCG injection day). RESULTS: Symptoms of Shen qi-yin deficiency were improved significantly in the ETR group, with the improvement better than that in the control group (P < 0.05). The total and daily FSH dosage in the ETR group were lower than those in the control group, showing significant difference between the two groups (P < 0.05). The effects were better in the ETR group than those in the control group in increasing serum E2 and endometrium thickness on the HCG injection day, and also in elevating the follicular oocyte number, the mature oocytes rate, the fertilization rate, and the prime embryo rate (all P < 0.05). CR2 analysis showed low negative correlation in both groups (r = -0.369 in the ETR group and r = -0.425 in the control group), suggesting E2 could be one of the factors but not the only factor for influencing colpoxerosis. And the score of colpoxerosis was lower in the ETR group after treatment than that in the control group (t = 2.422, P < 0.05). CONCLUSION: ETR combined with FSH can obviously reduce the dosage of FSH used, improve ovarian reactivity and pregnancy rate, and improve the quality of oocytes, the mechanism is possibly related with its effects in improving ovarian reactivity elevating blood estrodiol and regulating the hypothalamus-pituitary-ovary axis (HPOA).

[Study on fabrication of controllable microchannel structure scaffolds and rotating dynamic culture].

It is the critical technology for bone tissue engineering to fabricate scaffolds, especially to fabricate the internal microchannel of scaffolds. Utilizing stereolithography rapid prototyping indirect fabrication approach, suitable size microchannel structure scaffolds with orientation, branching, and interconnectivity under control was manufactured. Observation under light microscope showed that the arrange of microchannel size was 200-600 microm, the orientation and branching were accorded with designed structure, and all the microchannels were completely interconnected. Scaffolds seeded with osteoblast cells were put into 6-well plate and rotating 3D dynamic culture system which was developed in our lab. The morphology of osteoblasts and the development on the surface and internal microchannel of scaffolds were observed under scanning electron microscope (SEM). The results indicated that the dynamic culture permited an even distribution of cells across the surface of scaffolds, besides, a few cells grew into the lumen of microchannels.

Vanillin derivative 6-bromine-5-hydroxy-4-methoxybenzaldehyde-elicited apoptosis and G2/M arrest of Jurkat cells proceeds concurrently with DNA-PKcs cleavage and Akt inactivation.

Vanillin, a naturally occurring food component, has been reported to have anti-mutagenic and anti-metastatic potentials, and to inhibit DNA-PKcs activity. However, vanillin itself exhibits very weak antiproliferative activity. We explored the effects of bromovanin (6-bromine-5-hydroxy-4-methoxybenzaldehyde), a novel vanillin derivative, on survival and cell-cycle progression of human Jurkat leukemia cells. Treatment with >10 microM bromovanin significantly elicited apoptosis and G2/M arrest in Jurkat cells in a dose- and time-dependent manner. Bromovanin-induced DNA double-strand breaks (DSB) were demonstrated by means of comet assay as well as detection of phosphorylated H2AX, a sensitive indicator of DNA DSBs. Immuno-hybridization analysis revealed that the cleavage of procaspase-3 and DNA-PKcs occurred concurrently with bromovanin-induced apoptosis. Furthermore, phosphorylated Akt protein (Ser473), which is catalyzed by DNA-PKcs, as well as phosphorylated GSK3beta (a substrate of activated Akt), markedly decreased in bromovanin-treated Jurkat cells, suggesting that bromovanin leads to inactivation of Akt pathway via cleaving DNA-PKcs. These multiple effects, associated with the regimen of cancer therapeutic strategies, make bromovanin very appealing for future development as a novel anticancer drug.

Protective effect of resveratrol against oxidative damage of UVA irradiated HaCaT cells.

OBJECTIVE: To observe the photoprotective effect and possible mechanisms of resveratrol for ultraviolet A (UVA) irradiated HaCaT cells. METHODS: HaCaT cells under UVA irradiation with 5J/cm(2) were interfered with 0.01 mmol/L and 0.1 mmol/L resveratrol. The testing objects were divided into a control and a UVA irradiation group, and then we detected the proliferation capacity with methylthiazdyl tetrazolium (MTT) and superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activity, content of maleic dialdehyde (MDA) with hydroxylamine, colorimetric, thiobarbituric acid (TBA) methods. The ultrastructure was observed under electron microscope. RESULTS: Resveratrol could enhance the proliferation activity, SOD, GSH-Px activity of HaCaT cells under UVA irradiation, decrease the content of MDA in dose-dependent manner (P<0.05). The electron microscope revealed that resveratrol could relieve the injury of HaCaT cells' ultrastructure. CONCLUSION: Resveratrol can relieve the inhibition to HaCaT cell proliferation, injury of their ultrastructure and oxidation by UVA irradiation. The protection is dose-dependent. That resveratrol raises the oxidase activity and clears the oxyradical may account for these results.

Suspended particle effects on ClO2/ultraviolet light combined disinfection of effluent.

The concentration of suspended solids of effluent often varies in a wide range, therefore the dose of ultraviolet light (UV) in disinfection process needs to be adjusted to meet the disinfection criterion at a high frequency, and the desired disinfection effect is difficult to be ensured. The particles size and particle-associated fecal coliform (F.C.) contribution, and their influence on UV disinfection were investigated when ClO2 and UV combined disinfection process was used. The results showed that suspended solids content had a major impact on UV disinfection efficiency, especially the large particle size fraction. Particles (D>10 microm) associated F. C. were difficult to be disinfected and were the main part of the tailings of F.C. inactivation curve. Pre-ClO2 oxidation could reduce the number of particles in effluent, and make large particles decrease to small ones. Therefore, the influence of particles on UV disinfection could be reduced after pre-ClO2 oxidation, and the resistance ability to particle loadings of combined process was enhanced. Moreover, the combined process has a lot of advantages, such as low toxicity, low operational/maintenance costs; it is also convenient to be established in the existing wastewater plant or the new planned one.

[Extraperitoneal laparoscopic radical prostatectomy (report of 5 cases)].

OBJECTIVE: To discuss the clinical experience of laparoscopic radical prostatectomy by extraperitoneal approach. METHODS: Five patients with localized prostate cancer underwent laparoscopic radical prostatectomy by extraperitoneal approach. The surgical procedure included the excision of the prostate, seminal vesicles, ampulla ductus deferentis and part of the bladder neck, followed by urethrovesical anastomosis. RESULTS: All the operations were successful. The mean operation time was 350 minutes (ranging from 270 to 420 mm); the mean estimated blood loss was 480 ml (ranging from 250 to 600 ml). The bowel activity was recovered with 48 hours after surgery. The patients were ambulant between the 2nd and 3rd postoperative days. The mean hospital stay was 8. 5 days (ranging from 7 to 12 days). The 3-8 months follow-up found no incontinence of urine; of the 3 preoperatively potent patients, 2 were able to have sexual intercourse; strictured stoma was reported in only 1 case. CONCLUSION: The extraperitoneal laparoscopic radical prostatectomy, keeping the procedure out of the peritoneal cavity, with small incision and rapid recovery, may be considered as a promising surgical method for patients with localized prostate cancer.

[Isolation and characterization of 4 benzene/toluene-degrading bacterial strains and detection of related degradation genes].

Four bacteria with the ability of benzene or toluene degradation were isolated from the active sludge of a life sewage treatment farm, two of which degraded benzene, and named B1 and B2; and other two degraded toluene, and named J2 and J6, respectively. Sequence analysis of 16S rDNA showed that strain B1 and J2 belonged to Pseudomonas, and B2, J6 belonged to Acinetobacter. They all well adapted to the circumstances from 18 degrees C to 37 degrees C, 0% to 3% NaCl, and pH 7-10. The isolate J2 showed a high tolerance to organic solvent as it could grow well in the medium containing 70% (V/V) toluene, and degraded 92.4% of 0.1% (V/V) toluene within 72 hrs in a medium with toluene as the sole carbon source; while J6 presented 84.8% degradation under the same conditions to toluene. In the case of B1 and B2, they degraded 67.7% and 94.2% of 0.1% (V/V) benzene within 72 hours, respectively. Moreover, all strains were detected harboring the same benzene dioxygenase gene. In addition, J2 and J6 also had toluene dioxygenase genes which share 86.5% homology from each other. These bacteria are of potential in bio-treatment of benzene and toluene pollutants.

[Aberrant localization of beta-catenin in leukemia cell lines].

Beta-catenin plays a central role in Wnt signaling pathway. The aberrant localization of beta-catenin in nucleus causes the transcription of down-stream target genes, which is the pathogenesy of some solid tumours. As the expression of adheren junction on hemopoietic cells is very low, there are a few studies on beta-catenin expression in leukaemia. This study was aimed to investigate beta-catenin localization and beta-catenin mRNA expression levels in 4 leukemia cell lines so as to explore a new oncogenic mechanism and to find out a new therapeutic target. The beta-catenin localization in leukemia cell lines was detected by immunocytochemistry, the beta-catenin mRNA expression level was assayed by real-time quantitative RT-PCR. The results showed that there was aberrant localization of beta-catenin in Jurkat and Thp-1, and beta-catenin mRNA expression level was not increased in these two cell lines, however, the mRNA expression levels of Jurkat and Thp-1 were lower than those of Daudi and K562. The beta-catenin mRNA expression level was not correlated with beta-catenin aberrant localization in these 4 cell lines. It is concluded that the aberrant localization of beta-catenin may play a role in the development of some leukemia, and the mechanism resulting in beta-catenin aberrant localization not take place at transcription level.

[The effects of antiarrhythmic peptide AAP10 on ventricular arrhythmias in rabbits with healed myocardial infarction].

OBJECTIVE: To evaluate the effects of antiarrhythmic peptide (AAP10) on ventricular arrhythmias in rabbits with healed myocardial infarction (OMI). METHODS: Thirty rabbits were randomly divided into three groups (n = 10 each): Sham group, left thoracotomy was performed without coronary ligation; OMI group and OMI + AAP10 group, the circumflex coronaries were ligated. Three months post operation, the electrophysiological and antiarrhythmic effects of AAP10 were assessed in the arterially perfused rabbit left ventricular wedge preparation. Sham and OMI group were perfused with Tyrode's solution and OMI + AAP10 group was perfused with Tyrode's solution + AAP10 (80 nmol/L). Transmembrane action potentials were recorded simultaneously from endocardium and epicardium together with a transmural ECG by use of 2 separate intracellular floating microelectrodes. The stimulus-response-interval (SRI) of the epicardium and the incidence of ventricular tachycardia (VT) were observed. Whole heart and left ventricular weights, the left ventricular thickness at infarct border zone were measured. RESULTS: Whole heart and left ventricular weights as well as the left ventricular thickness at the infarct border zone significantly increased post infarction. VT was induced in 8 out of 10 rabbits in OMI group and in 2 out of 10 rabbits in OMI + AAP10 group (P < 0.05). SRI was also significantly shortened in OMI + AAP10 group compared to OMI group [SRI-1: (20.59 +/- 0.79) ms vs. (28.71 +/- 0.55) ms; SRI-2: (30.42 +/- 0.74) ms vs. (38.67 +/- 0.49) ms, all P < 0.01]. However, the action potential morphology and duration were similar between OMI and OMI + AAP10 groups. CONCLUSION: The antiarrhythmic peptide (AAP10) can increase gap junctional intercellular conductance without affecting the action potential morphology and duration and decrease the incidence of inducible ventricular tachycardia.

A variance component analysis on recombination rate in the COGA pedigrees.

We estimated the crossover frequency in 1,232 gametes from 356 subjects in pedigrees from the Collaborative Study on the Genetics on Alcoholism. We examined the effect of covariates including age, ethnicity, and years with ALDX1 on recombination rate, and found a positive correlation between recombination rate and years with ALDX1. By variance-component analysis, we estimated the heritability of recombination rate to be around 0.5, and provided suggestive evidence for a locus linked to recombination rate.
