Pseudomonas, the dominant polycyclic aromatic hydrocarbon-degrading bacteria isolated from Antarctic soils and the role of large plasmids in horizontal gene transfer.

Twenty-two polycyclic aromatic hydrocarbon (PAH)-degrading bacterial strains were isolated from Antarctic soils with naphthalene or phenanthrene as a sole carbon source, while no degrader was obtained from an unpolluted sampling site. Phylogenetic analysis showed that all belonged to the genus Pseudomonas except one that was identified as the genus of Rahnella. Some of them were closely related to previously reported cold-tolerant species, while some were separated in deeply rooted branches and represent new strains. All these strains showed a high efficiency to degrade naphthalene at 4 degrees C, and some additionally degraded phenanthrene. Using degenerate primers and polymerase chain reaction (PCR) amplification, ndo gene encoding naphthalene dioxygenase (NDO) was detected from all the isolates. Phylogenetic analysis grouped these genes into two clusters which shared 94% similarity to each other, and showed about 97% similarity within a cluster. However, no obvious difference was observed with mesophilic ndo genes; this indicates that the host cell is pivotal in cold adaptation. In addition, the mismatch between 16S rRNA and NDO phylogenetic trees strongly indicates horizontal gene transfer among these isolates and may have happened in situ. Further, Southern hybridization and plasmid curing confirmed that ndo genes were located on a large self-transmissible plasmid, which can be transferred to a mesophilic strains. The transconjugants acquired the ability to utilize naphthalene and phenanthrene. Results of this article imply that Pseudomonas plays an important role in PAH biodegradation in Antarctic soils, and the related genes might be originally transferred from outside Antarctica and spread among indigenous species.

Implications of previous subclinical dengue infection but not virus load in dengue hemorrhagic fever.

In a study comparing the virus load and immune reaction between patients with primary and secondary dengue-2 (DEN-2) infections in a hospital-based analysis, we found that 40.7% (55/135) of the 135 patients had secondary DEN-2 infection following a DEN-2 outbreak in southern Taiwan. Most of the secondary infections had subclinical primary dengue infections (78.2%; 43/55). Patients with secondary DEN-2 infections had lower platelet counts, and blood interferon-alpha and virus load, but significantly higher interleukin-10 (P=0.030) and anti-DEN-1 neutralization titers (P=0.013) than those with primary infection. Patients with secondary DEN-2 infection also had a higher rate of dengue hemorrhagic fever (DHF) (61.7% vs. 36.3%). A previous subclinical dengue infection is involved in the secondary DEN-2 infection associated with altered immune reaction and higher DHF rate, but lower blood virus load.

Effects of IgG and its F(ab')2 fragments of some patients with idiopathic thrombocytopenic purpura on platelet aggregation.

OBJECTIVES: To make humanized monoclonal antibodies by phage surface display technology, we screened out the specific anti-platelet glycoproteins (GPs) IgG antibody from patients with chronic idiopathic thrombocytopenic purpura (ITP), which can inhibit platelet aggregation. METHODS: We studied plasmas from 68 patients with ITP for the presence of IgG antibodies specific for GPIIb/IIIa and/or GPIb/IX using modified monoclonal antibody immobilization of platelet antigen assays. The IgG antibody and its F(ab')(2) fragments of the positive plasmas which could inhibit platelet aggregation function were prepared and purified. Their immunoreactivity to platelet GPs and effects on platelet function were further analyzed. RESULTS: GPIIb/IIIa- and GPIb/IX-specific antibodies were found in 21 and 19 patients, respectively. Six of them had antibodies against both GP complexes. Among the 34 positive plasmas, four with positive anti-GPIIb/IIIa autoantibody showed significant inhibition of platelet aggregation induced by adenosine diphosphate (ADP), whereas one with GPIb/IX-specific antibody inhibited ristocetin-induced platelet aggregation. The purified IgG and its F(ab')(2) fragments from two patients not only retained the ability to bind to platelet GPs but also impaired the in vitro ADP-induced platelet aggregation. CONCLUSIONS: F(ab')(2) portion of the IgG is a functional fragment, which is responsible for the autoantibody interaction with platelet GPs in ITP, and some of them also affect platelet function, which can be used to develop completely humanized anti-GPIIb/IIIa small molecular phage antibody.

Effective use of corticosteroids in treatment of plastic bronchitis with hemoptysis in Chinese adults.

AIM: To investigate whether corticosteroids are effective in the treatment of plastic bronchitis with hemoptysis. METHODS: A retrospective, clinical study was undertaken. Thirty two patients with only first episodes of plastic bronchitis with hemoptysis were divided into a steroid group (n=18) treated with glucocorticoids, and a non-steroid group (n=14). The supportive therapy was uniformly applied to both groups, except for glucocorticoids. Variables such as temperature and white blood cell counts were determined. Furthermore, the volume of hemoptysis and bronchial casts were evaluated in detail daily. RESULTS: There was no difference in the demographic data and variables at baseline between both groups (all P>0.05). On days 5, 6, 7 and 8, the volume of hemoptysis was significantly decreased in the steroid group compared with the non-steroid group (43+/-15 mL vs 117+/-33 mL on d 5, 29+/-12 mL vs 97+/-23 mL on d 6, 18+/-10 mL vs 80+/-20 mL on d 7, and 13+/-8 mL vs 66+/-14 mL on d 8; all P<0.05), and on d 10 after fibreoptic bronchoscopy, the cases with bronchial casts was reduced evidently in the steroid group in comparison with the non-steroid group (OR=5.69, 95% CI=1.76-43.6; P=0.005). There was no significance in mechanical ventilation and mortality between both groups. CONCLUSION: Despite some limitations of this study, it has been demonstrated that, on the basis of common supportive therapy, corticosteroids would be effective and safe for the treatment of plastic bronchitis with hemoptysis.

Isolation and biomimetic synthesis of anti-inflammatory stilbenolignans from Gnetum cleistostachyum.

One new stilbenolignan, gnetucleistol F (1), and four known stilbenolignans, gnetofuran A (2), lehmbachol D (3), gnetifolin F (4) and gnetumontanin C (5) were isolated from the lianas of Gnetum cleistostachyum C. Y. CHENG (Gnetaceae). Their structures and relative configurations were determined by means of spectroscopic evidence. Compounds 1, 2, 3 and 4 were synthesized for the first time on the basis of their biogenetic pathway, and their possible biomimetical synthetic mechanisms were discussed. The pharmacological activities of all stilbenolignans have been tested. Among them, 1, 2, 3, 4 and 5 showed moderate inhibitory activities on TNF-alpha and 1 also showed potent inhibitory activity on malondialdehyde.

The relationship between activation of TLR4 and partial hepatic ischemia/reperfusion injury in mice.

BACKGROUND: Toll-like receptors (TLRs) are a group of evolutionarily conserved pattern recognition receptors involved in the activation of the immune system in response to various pathogens. In this study, we elucidated the relationship between activation of TLR4 and liver injury in partial hepatic ischemia/reperfusion (I/R) injury in mice. METHODS: BALB/c mice were used in a model of partial hepatic I/R injury, and the changes of TLR4 gene expression in ischemic liver lobes were detected with real-time polymerase chain reaction (RT-PCR). The levels of plasma ALT and endotoxin in the portal vein were measured. TLR4-deficient mice (C3H/Hej) and wild type mice (C3H/Heouj) were used in a model of I/R injury; liver function impairment and the level of serum TNF-alpha were observed. RESULTS: After one hour ischemia, the expression of TLR4 mRNA increased at the 1st, 3rd hour of reperfusion, indicating the value of deltaCt (1st hour: 1.21+/-0.87 vs. 5.85 +/-1.07, t=13.72, P<0.01; 3rd hour: 0.85+/-0.92 vs. 6.11+/-1.24, t=16.33, P<0.01). No endotoxemia developed in every group of mice. At the 3rd hour of reperfusion, the level of serum TNF-alpha was significantly higher than that of sham group (Hej: 152+/-43 pg/ml vs. 18+/-10 pg/ml, t=5.26, P<0.01; Heouj: 249+/-52 pg/ml vs. 25+/-13 pg/ml, t=7.24, P<0.01). At the 1st, 3rd hour reperfusion, the level of plasmid ALT in Hej mice was lower than that in Heouj mice (1st hour 662+/-106 U/L vs. 1216+/-174 U/L, t=4.21, P<0.01; 3rd hour 1145+/-132 U/L vs. 2958+/-187 U/L, t=13.72, P<0.01). The level of serum TNF-alpha was lower than that in Heouj mice (152+/-43 U/L vs. 249+/-52 U/L, t=3.94, P<0.01) at the 3rd hour reperfusion. CONCLUSION: TLR4 activation causes partial hepatic I/R injury through release of TNF-alpha.

[Identification and evaluation promoter sequence and the transcription activation of Hcbp6 interaction with core protein of hepatitis C virus].

OBJECTIVE: To clarify the expression and regulation mechanism of the new target gene (human hepatitis C virus binding protein 6, Hcbp6) interaction with the core protein of hepatitis C virus (HCV). METHODS: Referring to the prediction online of the promoter region, 3256 base pairs (bp) upstream and downstream of the translation start site were selected as 5 putative promoter sequences which were amplified from the hepatoblastoma cell line-HepG2 cell genomic DNA by polymerase chain reaction (PCR). The amplified products were cloned into a pCAT3 vector. The HepG2 and NIH3T3 cell lines were transfected by pCAT3-Hcbp6-promoters. The CAT activity was detected using an enzyme-linked immunoassay (ELISA) kit. RESULTS: We found that 2 kinds of pCAT3-Hcbp6-promoters (1066 and 240) could direct the reporter gene expression. The expression of CAT was 3.1 times and 6.4 times higher than that of the pCAT3-basic control vector. CONCLUSION: These results indicated that pCAT3-Hcbp6-promoters have promoter activity.

[Effect of nitric oxide on Toll-like receptor 2/4 gene expression in the liver in acute hemorrhagic necrotizing pancreatitis in rats].

OBJECTIVE: To investigate the effect of nitric oxide (NO) on Toll-like receptors 2 and 4 (TLR 2/4) gene expression in the liver in acute hemorrhagic necrotizing pancreatitis (AHNP) in rats. METHODS: Seventy SD male rats were randomly divided into sham-operated group (n=10), AHNP group (n=30) and L-arginine (L-Arg) treatment group (n=30). Blood samples and liver tissues were obtained at 6 hours in sham-operated group, and 3, 6, 12 hours respectively in AHNP group and L-Arg-treated group. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), amylase in serum, NO, tumor necrosis factor-alpha (TNF-alpha) in liver tissue were determined. TLR2/4 mRNA expressions in the liver tissue were measured by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: TLR2/4 mRNA could be detected in the liver with low values in sham-operated group [(1.150+/-0.725)x10(-6), (11.450+/-1.724)x10(-4)], but they were markedly upregulated at 3 hours in AHNP group [(1.970+/-0.362)x10(-3), (175.000+/-0.111)x10(-3)], and peaked at 12 hours [(29.400+/-3.155)x10(-4), (267.300+/-8838)x10(-2), P<0.01). At the same time serum levels of amylase, ALT and AST increased, hepatic injuries were aggravated, the levels of TNF-alpha in the liver were increased and levels of NO in the liver were lowered (P<0.05 or P<0.01). Treatment with L-rg could effectively inhibit TLR2/4 mRNA expression [3 h: (3.510+/-1.528)x10(-4), (13.500+/-2.231)x10(-2); 6 h: (21.000+/-5.346)x10(-4), (18.700+/-2.685)x10(-2); 12 h: (26.200+/-2.076)x10(-4), 1.959+/-0.270, P<0.05 or P<0.01] and alleviate hepatic injuries. The levels of serum amylase, ALT and AST lowered, and the levels of TNF-alpha in the liver were lowered and the levels of NO were markedly increased (P<0.05 or P<0.01). CONCLUSION: These findings suggest that the expression of TLR2/4 mRNA is increased in the liver in AHNP, and the hepatic injuries are aggravated. NO could markedly inhibit TLR 2/4 mRNA gene expression in the liver in AHNP.

[Biodegradation of dibutyl phthalate by diatomite adsorptive immobilized microorganism].

OBJECTIVE: To study the biodegradation characteristics of seeding type immobilized microorganism on dibutyl phthalate (DBP). METHODS: The immobilized microorganism was made to adsorb DBP degradation dominant bacteria by using modified diatomite as carrier, then it degraded DBP under different primary concentration, vibration rate, pH, temperature and at the presence of metal compounds. The degradation kinetics was analyzed. RESULTS: When DBP primary concentration was 100-500 mg/L, the adsorptive immobilized microorganism could maintain relatively high activity and the DBP degraded rate was above 80% in 24h. Dissociative and immobilized microorganism could get higher degradation activity in vibration than in stillness. When pH was 6.0 - 9.0, the degradation rate of immobilized microorganism on DBP was above 82% in 24h and its activity is higher than dissociative microorganism. In the range of 20 degrees C to 40 degrees C, the DBP degraded rate by immobilized microorganism could reach 84.5% in 24h. If mental compounds existed in the DBP water sample, the degradation activities of dissociative and immobilized microorganism were inhibited obviously. The form of DBP degradation kinetics could be described as the first-order model. CONCLUSION: The immobilized microorganisms using diatomite as carrier could degrade DBP effectively. The adsorptive immobilized microorganism was more adapted to DBP load, temperature and pH than dissociative microorganism. The mental compounds could inhibited their activities. The degradation reaction of adsorptive immobilized microorganisms on DBP was according with the first-order model.

[Design of a quantitative keratoscope used for ocular microsurgery].

A new quantitative keratoscope used specially for ocular microsurgery to measure corneal astigmatism is reported. A combined cylinder-lens, which power can be changed by rotating lens in anti-direction equally, was designed according to the principal of the crossing cylinder lens. The axis direction is fixed when the lens is rotated. The corneal astigmatism can be measured by changing the power of the combined lens to adjust the corneal ring images. The keratoscope could be used to measure the corneal astigmatism quantitatively specially for ocular microsurgery. At the work-distance of 0.025 m, the maximum power of measured astigmatism was up to 8.50 D. The error of the measurement was less than 0.20 D. The keratoscope is a portable lens to be used easily during the ocular microsurgery. The reporting keratoscope is a simple combined cylinder-lens that can be used to measure corneal astigmatism, including the power and the axis of astigmatism, easily during ocular microsurgery.

Experimental study on denitrification using coated electrode of immobilized denitrifying bacteria.

OBJECTIVE: To develop a coated electrode of immobilized denitrificants and to evaluate the performance of a bioelectrochemical reactor to enhance and control denitrification. METHODS: Denitrifying bacteria were developed by batch incubation and immobilized with polyvinyl alcohol (PVA) on the surface of activated carbon fiber (ACF) to make a coated electrode. Then the coated electrode (cathode) and graphite electrode (anode) were transferred to the reactor to reduce nitrate. RESULTS: After acclimated to the mixtrophic and autotrophic denitrification stages, the denitrifying bacteria could use hydrogen as an electron donor to reduce nitrate. When the initial nitrate concentration was 30.2 mg NO3- -N / L, the denitrification efficiency was 57.3% at an applied electric current of 15 mA and a hydraulic retention time (HRT) of 12 hours. Correspondingly, the current density was 0.083 mA/cm2. The nitrate removal rate of the reactor was 34.4 g NO3- -N/m3 x d, and the surface area loading was 1.34 g NO3- -N / m2 x d. CONCLUSION: The coated electrode may keep high quantity of biomass, thus achieving a high denitrification rate. Denitrification efficiencies are related to HRT, current density, oxidation reduction potential (ORP), dissolved oxygen (DO), pH value, and temperature.

[Effect of ginkgolide B on the function of rat aorta smooth cells and U937 cells stimulated by oxLDL].

AIM: To investigate the effect of ginkgolide B on the proliferation of VSMC and the secretion of chemokines by U937 cells stimulated by oxLDL or PAF. In addition, to analyze whether the effect of oxLDL is mediated through PAF receptor. METHODS: Using 3H-Tdr incorporation assay, the proliferation of VSMC was measured. The protein and mRNA level of MCP-1 and IL-8 in U937 cells were determined by RT-PCR and ELISA. Using Western blotting the p65 and IkappaB was quantified. The binding of oxLDL to U937 cell was measured by a radio-ligand binding assay of 3H-PAF. RESULTS: Ginkgolide B inhibited, in dose-dependent manner, the proliferation of VSMC and the secretion of chemokines by U937 cells stimulated by oxLDL, and inhibited the oxLDL-induced p65 activation and depletion of IKappaB. oxLDL inhibited PAF binding to U937 cells. CONCLUSION: Ginkgolide B, as a PAF antagonist, possesses the effect of inhibiting the proliferation of VSMC and the secretion of chemokines by U937 cells stimulated by oxLDL in vitro. The effect of oxLDL is, at least in part, mediated through PAF receptor.

[Study on tetrahydrobiopterin deficiency in Northern Chinese population].

OBJECTIVE: To emphasize early differential diagnosis from patients with hyperphenylalaninemia (HPA) and to evaluate the treatment and long-term outcome of patients with tetrahydrobiopterin synthase (BH4) deficiency in Northern Chinese population. METHODS: From 1992 to 2005, a total of 618 patients with HPA were diagnosed and/or cared for in our outpatient clinic. Urinary pterin analysis, detection of dihydropteridine reductase (DHPR) activity in blood, and then BH4 loading tests were carried out to differentiate BH4 deficiency in these patients from classical phenylketonuria. BH4 deficient patients were treated with BH4, levodopa and 5-hydroxytryptophane (5-HTP) immediately while the diagnosis was done to disease. Patientso blood phenylalanine levels, psychomotor and intelligence development were followed up. RESULTS: A total of 38 cases were diagnosed as BH4 deficiency, all of them were revealed as 6-pyruvoyl-tetrahydropterin synthase (PTPS) deficiency from the extremely decreased urine biopterin, normal DHPR activities and drop down of blood phenylalanine level to normal range within 4 to 8 hours after BH4 loading. The most common manifestations were progressively psychomotor and mental retardation to patients even after taking early dietary treatment. The patients were diagnosed and treated with drugs at the ages of 2.1 months to 13 years. With 4 patients died of pneumonia, 7 patients refused to treatment, only 27 patients were under treatment and followed up. The average full scale development or intelligence quotient (DQ/IQ) of patients who were treated within and after 6 months were 86+/- 10 or 66+/- 7 respectively. Development was not even in different aspects. A significant negative correlation was observed between the level of the DQ and the age of treatment commenced (r was -0.714, P< 0.01). Eleven patients experienced the extrapyramidal movement disorders, 3 of them combined with epilepsy. The extrapyramidal disorders were controlled by administration of levodopa. CONCLUSION: The differential diagnosis for BH4 deficiency should be carried out in all patients with HPA. PTPS deficiency is the most common form of BH4 deficiency in Northern Chinese population. The long-term outcome of these patients benefits from diagnosis and treatment with BH4, levodopa and 5-HTP as early as possible.

[Calpain-I, calpastatin, caspase-3 and apoptosis in the human left atrium in rheumatic atrial fibrillation].

OBJECTIVE: The aim of the present study was to detect the expression of calpain-I, calpastatin, caspase-3 and apoptosis in the left atria of patients with rheumatic heart disease (RHD), and to find the association of these factors. Also, it was intended to investigate the effect of the above factors on the mechanism of atrial fibrillation (AF). METHODS: 43 patients with RHD undergoing valve-replacement were included, 15 patients with regular sinus rhythm (Group RSR), 8 patients with paroxysmal AF (Group AF1) and 20 patients with permanent AF (Group AF2). Western blot was used to examine the content of calpain-I, caspase-3 and calpastatin. The apoptosis index (AI) was measured by TUNEL. RESULTS: (1) Expression of calpain-I in group AF2 was increased to (344.0 +/- 101.9)%, and caspase-3 was increased to (394.0 +/- 99.4)% compared to group RSR (P < 0.01, respectively). Amount of calpastatin was reduced to (27.0 +/- 12.8)% (P < 0.01). The expressions of these proteins were unchanged in group AF1. (2) AI in group AF2 was higher than that in groups RSR and AF1 (P < 0.01). (3) In group AF2, the levels of calpain-I, caspase-3 and AI were positively relative to left atrial dimension and AF duration, P < 0.05 - 0.01, respectively, whereas calpastatin was negatively correlated with left atrial dimension and AF duration (P = 0.007 and P = 0.001, respectively). (4) The protein content of calpain-I was positively related with that of caspase-3 and AI (P < 0.01, respectively), and the content of calpastatin was negatively related with that of calpain-I and caspase-3 (P < 0.01, respectively). CONCLUSIONS: Apoptosis of atrial cell increased in left atria and the protein contents of calpain-I, caspase-3 and calpastatin significantly altered during AF in humans with RHD. The observed interactions suggest that these factors compose a system to cause the structural remodeling and dysfunction of atria. The course may play a key role in promoting the onset and maintenance of AF.

[Effect of antitachycardia pacemaker on atrial tachyarrhythmia detection, prevention and termination].

OBJECTIVE: The present study retrospectively evaluated the reliability of detecting atrial tachyarrhythmias (ATA), the efficacy of automatic atrial antitachycardia pacing (ATP) and the performance of atrial preventive pacing (APP) algorithms in an implanted antitachycardia DDDRP pacemaker for patients with sick sinus syndrome and paroxysmal ATA. METHODS: In all 24 patients, a DDDRP pacemaker (Medtronic AT500/AT501) was implanted. APP algorithms were switched on at the implanting physician's discretion. During each pacemaker follow-up, information was saved to disk and the ATA burden between those patients with APP algorithms switched "ON" and "OFF" were compared. Reliability of ATA detection was determined by reviewing the stored electrograms and ATP efficacy was also reviewed. Both the pacemaker memory data and manual EGM retrieval were used for the analysis. RESULTS: Complication-free survival at (17.63 +/- 8.79) months was 100%. In 12 patients APP was not turned "ON" until the latest follow-up, in 6 patients APP was switched "ON" at their first visit after implantation, and in another 6 patients APP was switched "ON" after a median follow-up of 9.29 months. There were 97 367 episodes of ATA detected by the devices, of those with stored atrial electrograms the correct classification of ATA was (76.77 +/- 20.52)%. The percentage of atrial pacing with APP algorithms turned on was (87.95 +/- 20.93)%, which was significantly higher than that in patients with APP "OFF" (50.73 +/- 34.46)% (P < 0.01). ATP efficacy was (50.27 +/- 19.29)%. However, the ATA burden (14.73% vs 16.52%, or 7.52 hours vs 6.58 hours per week, P > 0.05) and the longest duration of single ATA episode (27.27 hours vs 20.75 hours, P > 0.05) were not significantly different between those patients with APP "ON" and "OFF". No proarrhythmic effect or major cardiovascular event was observed. CONCLUSIONS: The antitachycardia DDDRP pacemaker correctly detects and diagnoses about 75% of the ATA episodes, while the ATP therapy successfully terminates atrial tachycardia or flutter in about 50% of attacks. However, there is no difference in ATA burden with the APP algorithms and high incidence of atrial pacing. As a non-curative therapy strategy, this high-cost device may only be used in strictly selected indication patients in addition to other treatments of ATA.

[The immunoreactivity of IgG and its fragments from ITP patients and their effects on platelet aggregation function].

OBJECTIVE: To prepare ITP plasma IgG and its F(ab')2 fragments and investigate their immunoreactivity to platelet GPIIb/IIIa and/or GPIb/IX and their effects on platelet aggregation function. METHODS: The ITP patients having inhibitory autoantibody to the platelet aggregation were selected by modified MAIPA and platelet aggregation test with turbidimetry. Plasma IgG and its F(ab')2 fragments were prepared by streptococcal protein A affinity column and pepsin digestion. The immunoreactivity and the effects on platelet aggregation function of the whole antibody and its fragments were detected by modified MAIPA and platelet aggregation test, respectively. RESULTS: (1) Anti-platelet GPIIb/IIIa and/or GPIb/IX autoantibodies were detected in 34 of 68 (53.6%) ITP patients' plasmas and that from 5 patients significantly inhibited the platelet aggregation induced by ADP or ristocetin. (2) By using protein A column combined with protease digestion, pure IgG and its F(ab')2 fragments were successfully obtained. (3) The purified IgG and its F(ab')2 fragments retained the ability to bind to their respective glycoproteins and inhibited the platelet aggregation function, whereas the IgG depleted plasma lost the ability of binding to the platelet GPs. CONCLUSIONS: F(ab')2 fragment of the IgG antibody is a functional fragment, which not only has the binding ability to the platelet GPs but also inhibits the platelet aggregation function in a dose-dependent manner.

[Study on thermal decomposition of HMX energetic materials by in-situ FTIR spectroscopy].

In-situ FTIR spectroscopy is a rising and dynamic technique. This technique, which combines the advantages of investigation in-situ and accurate structure analysis by FTIR spectsoscopy, can detect the chemical change of materials in different temperature with real time and obtain the relationship between micromechanism of materials and temperature. In the present paper, thermal decomposition of octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) heated with 5 degrees C x min(-1) was investigated by in-situ FTIR spectroscopy. The results demonstrate that C-N bond cleavage and N-N bond cleavage of HMX occur at 205 degrees C. With increasing temperature, the rupture rate of C-N bond is faster than that of N-N bond, which verifies that the cleavage of C-N bond is the dominant rupture form. The augment in tension of HMX cyclic confirms that intramolecular cyclization occurs in breakage of HMX. Eight kinds of gaseous products such as CO2, N2O, CO, NO, HCHO, HONO, NO2 and HCN were determined. According to the structure change in condensed state and gaseous products, the decomposition mechanism was deduced that HCHO, N2O, HONO and HCN were released due to the cleavage of C-N bond, and NO2 was released due to the cleavage of N-N bond.
