[The change in hyaluronic acid (HA) in auto-and allo-skin intermingled grafting in extensive full thickness burn in rat model].

OBJECTIVE: To elucidate the changes in hyaluronic acid (HA) contents in auto-and alloskin intermingled grafting. METHODS: HA was determined by radio-immunoassay in auto-and alloskin intermingled grafting (a large sheet of allograft inlaid with small stamps of autoskin) in experimental extensive full-thickness burn rats on 3rd, 5th, 7th, 14th, 21st, 28th postoperation days. RESULTS: In both areas of intermingled skin grafting, the contents of HA was reduced first and increased significantly thereafter, but the contents in auto-skin areas rose earlier and higher than those in allo-ones. CONCLUSION: The results suggest that adequate application of HA may be beneficial to the healing and remodelling of grafted area.

[Establishment and identification of B cell lines with PNH phenotype].

OBJECTIVE: To establish B cell lines from patients with paroxysmal nocturnal haemoglobinuria (PNH) and normal controls. METHODS: B lymphoid cells prepared from PNH patients and normal controls were infected with EB virus. Phenotype of the immortal cells was analyzed by flow cytometry. The immunomagnetic beads were used to select CD(55)(-) and CD(59)(-) B lymphoid cells, which were cloned by limiting dilution. The clonality of the cloned cells was confirmed by testing the pattern of Ig rearrangements using polymerase chain reaction (PCR). RESULTS: From 2 PNH patients and 2 normal controls, CD(55)(-) and CD(59)(-) B lymphoblastoid cell lines were established which had PNH phenotype and normal phenotype, respectively. CONCLUSION: B lymphoid cell lines from PNH patients can be obtained by EB virus infection.

[Study of the expression and function of c-kit receptor on the bone marrow mononuclear cells of the patients with myelodysplastic syndromes].

OBJECTIVE: To determine the expression and function of c-kit receptor on bone marrow mononuclear cells (BMMNC) of the patients with myelodysplastic syndromes. METHODS: c-kit protein (CD(117)) was detected by direct immunofluorescence assay, c-kit mRNA by reverse transcriptive polymerase chain reaction (RT-PCR) and the function of c-kit receptor by cell culture. RESULTS: c-kit protein expression in MDS was significantly higher than that in control [(8.58 +/- 5.28)% vs (3.04 +/- 1.49)%, P < 0.05]. c-kit protein expression in RA patients was significantly lower than that in RAEB/RAEB-t patients [(5.12 +/- 2.13)% vs (10.01 +/- 5.07)%, P < 0.05]. The rate of c-kit protein expression was (32.43 +/- 18.16)% in AML cases transformed from MDS (t-AML). c-kit mRNA expression in MDS was correlated with c-kit protein expression. Addition of interleukin-3 (IL-3), erythropoietin (Epo) and stem cell factor (SCF) to the semi-solid culture of BMMNC showed significant stimulating effects on CFU-GM and BFU-E formation in normal controls, but did not in MDS patients. CONCLUSION: The protein and mRNA expressions of c-kit receptor on BMMNC in MDS are higher than those in normal control, and the function of this receptor in MDS is abnormal.

[Signal transduction pathway for antral smooth muscle cell contraction induced by motilin and gastrin in rats].

Signal transduction pathway that mediates motilin and gastrin induced contraction of antral smooth muscle cells was investigated in rats. The results are as follows. (1) Motilin and gastrin induced contraction of isolated gastric smooth muscle cells in a dose dependent manner. (2) Motilin and gastrin induced antral smooth muscle cell contraction was inhibited by antibodies against G( i-3). Motilin and gastrin caused an increase of Guanosine 5 -(3- (35)S thio)triphosphate( (35)S GTPgammaS) binding of G( i-3). (3) Contraction of antral muscle cells induced by motilin and gastrin was inhibited by the phospholipase C (PLC) inhibitor U 73122 and the IP(3) receptor antagonist heparin. The results suggest that motilin and gastrin induced contraction of rat antral smooth muscle cells is mediated by stimulating G(I) protein coupled with PLC that produces IP(3).

Effect of total flavonoids of hippophae rhamnoides on contractile mechanics and calcium transfer in stretched myocyte.

OBJECTIVE: To study the role of total flavonoids of hippophae rhamnoides in improving contractile [correction of contactile] function of stretched cardiac myocyte. METHOD: Flavonoids were given to stretched myocytes which were proved their contractile function decline and then myocyte contractile mechanics characteristics and calcium transfer were measured. RESULT: Flavonoids increased myocyte contractility, as indicated by myocyte shortening, velocity of shortening, peak +dL/dt and peak -dL/dt during shortening, in a concentration-dependent manner (r>0.9, P<0.001), and with no relation to the intracellular calcium transfer in the myocytes. CONCLUSION: Flavonoids of the traditional Chinese drug hippophae rhamnoides is effective in improving the contractile function of stretched cardiac myocyte in low dosage.

[Exercise ECG test as a method for non-invasive detection of myocardial ischemia].

OBJECTIVE: To improve the exercise ECG analysis for non-invasively detection of myocardial ischemia. METHOD: The newly developed measuring system of exercise ECG defines ST segment area and STA/HR slope as the new characteristic parameters, thus can sensitively detect myocardial ischemia. Some methods were proposed to reduce noise and artifacts for effective sampling of exercise ECG. RESULT: 14 exercise ECG records were sampled and analyzed in clinical exercise test. It was found that the new parameters are very effective and sensitive to myocardial ischemia. The results show that the method is reliable and stable. CONCLUSION: The method improves one of the evaluating indexes of exercise ECG analysis. It is quite promising for clinical application in future.

[Influence of monkshood root-peony root combination on inflamation-induced agents and free radicals].

OBJECTIVE: To find out the effect of monkshood root and peony root on inflammation-induced agents and free radicals when used separately and in combination. METHOD: Two drugs were made into decoctions separately and in combination, i.p., qd, for 7 d, red blood cell SOD and serum LPO were analysed and the anti-inflammatory actions were observed. RESULTS: In an experimental rat model with inflamed paw edema induced by carrageenin or formaldehyde and in a mouse model with ear swelling induced by xylene, the anti-inflammatory effect appeared stronger when the two drugs were used in combination, especially in comparison with the use of monkshood root alone. The exudation of blood capillaries and the contents of inflammation medium PGE2 were lowered; The activity of SOD extracted from rat or mouse red blood cells was enhanced, and the serum LPO, which was high in level when monkshood root was used alone, could be declined when the two drugs were used in combination. CONCLUSION: The drug combination is more effective in inhibiting inflammation and scavenging free radicals.

[The correlation between anticardiolipin antibodies and disease activity in patients with systemic lupus erythematosus].

This study was conducted to determine the prevalence and clinical association of anticardiolipin antibodies (ACA) in serum of patients with systemic lupus erythematosus (SLE). ACA (IgG, IgA, IgM) was measured by enzyme-linked immunosorbent assay (ELISA) in the serum sample of 87 patients of SLE. The prevalence rates of ACAIgG, ACAIgA and ACAIgM were 62.1%, 52.9% and 35.6% respectively. The titers of ACAIgG and ACAIgM were positively correlated with the disease activity (r = 0.2763, P = 0.044; r = 0.3294, P = 0.018); ACAIgG, ACAIgA and ACAIgM were negatively correlated with complement C3 (r = -0.4737, P = 0.000; r = -0.2990, P = 0.017; r = -0.2758, P = 0.029); and ACAIgG was negatively correlated with complement C4 (r = -0.4079, P = 0.005). The titers of ACAIgA in patients with thrombosis was higher than that in patients without thrombosis. The patients with central nervous system involvement (CNS-SLE) had significantly higher ACAIgG and ACAIgM titers, compared with those without CNS-SLE. The results indicate there is a correlation between ACA titers and disease activity. In patients with thrombosis, the titer of ACAIgA is higher, and in patients with CNS-SLE, the titers of ACAIgG and ACAIgM are higher.

[Evaluation on usage of serum troponin-I, CK-MBmass and myoglobin measurements in diagnosing acute myocardial infarction].

The purpose of this study was to search the sensitivity of three indicators in early diagnosis of acute myocardial infarction (AMI) and analyze their window-time duration in serum, along with the decision on experimental golden indicator in later AMI inspection and effect assessment after recovery by testing the concentrations of CTnI, CK-MBmass and Mb in AMI patients' serum. By using corpuscle chemiluminescence, the serum concentrations of CTnI, CK-MBmass and Mb in AMI patients, UA (unstable angina) patients, SM (skeletal muscle) patients and a normal group were assayed and compared. A dynamic analysis was followed in five periods, which separately focused on 3-5, 8-14, 20-24, 72 hours and 7 days after heart aching occurred in AMI patients. The results showed the positive rates of CTnI, CK-MBmass and Mb in AMI patients were 65%, 70% and 70% respectively, and the positive rate of CTnI in UA and SM groups was 0%. The CTnI, CK-MBmass and Mb values in group AMI were significantly higher than those in groups of UA and SM. Separately, the CTnI value in AMI was 84 and 91.63 times (the CTnI values) in UA and SM; the CK-MBmass value in AMI was 8.82 and 4.67 times and the Mb value in AMI was 11.57 and 2.25 times as high as those in UA and SM. Significant difference was observed by the comparison between AMI group and UA, SM groups (P < 0.001). According to the dynamic analysis, CTnI, CK-MBmass and Mb rose significantly at the early stage of AMI onset (3-5 hrs), and then gradually increased with time. The climax occurred in the period of 8-14 hrs and Mb returned to its normal state in 72 hrs. On the 7th day, CTnI still existed in serum, but CK-MBmass approximated to normality. In conclusion, these data demonstrate a relatively high sensitivity of CTnI, CK-MBmass and Mb in early diagnosis of AMI. CTnI is specific to cardiac muscle for its special positive rate. Also, CK-MBmass and Mb are of higher clinical value in diagnosing cardiac muscle damage. CTnI has the longest window-time, which makes it the best special indicator for terminal inspection and recovery decision on AMI.

[Value of combined detection of tumor markers for the prediction of small cell and non-small cell lung cancer].

To evaluate the value of detection of 4 tumor markers(CEA, CA125, gastrin, and NSE) for histological types in patients with lung cancer and to improve the predicted efficiency of tumor markers for distinguishing between small cell lung cancer(SCLC) and non-small cell lung cancer (NSCLC), these 4 tumor markers in serum were determined in 51 patients (21 cases with SCLC, 30 cases with NSCLC) with confirmed primary diagnosis of lung cancer of different histology by radioimmunoassay. Linear learning machine method, PRIMA method and KNN method were used to classify SCLC and NSCLC. The levels of gastrin and NSE in SCLC were apparently higher than those of gastrin and NSE in NSCLC, but the levels of CEA and CA125 in SCLC were significantly lower than those in NSCLC. Smoking had an effect on the levels of CEA and CA125, but had little effect on those of gastrin and NSE. The total accuracy of the three methods was over 85% in distinguishing SCLC from NSCLC. So combined detection of the four tumor markers in serum might be useful in the prediction of histological types in patients with lung cancer.

[Comparative research on forms of dental and palatal arches between adults and children after rapid maxillary expansion].

OBJECTIVE: To study changes of dental and palatal arch forms between adults and children after rapid maxillary expansion (RME). METHODS: Trans-arch and trans-palatal widths, and molar and palatal angles on plaster models of 47 children and 47 adults were measured pre- and post-treatment with RME. RESULTS: 1. Trans-arch width: Increases of trans-molar and trans-cuspid widths were significantly greater in the child group than those in the adult group, except first and second premolars. In the child group, there was no significant difference of increased widths between posterior teeth and cuspids. But, in the adult group, increased widths in trans-arch of first molars, first and second premolars were significantly greater than those in cuspids. 2. Transpalatal width: Trans-palatal increases at mid-palates and the apex of palates were significantly greater in the child group than those in the adult group. 3. Molar angle: In the child group, no difference existed in the molar group between pre- and post-treament. But, in the adult group, statistical difference existed between pre- and post-treatment. Prior to RME, the molar angle was the same between the child group and the adult group. The molar angle was significantly greater in the adult than that in the child group after RME. 4. Palatal angle: The changes were not of statistical significance in child palatal angles between pre- and post-RME. Significant differences existed in the adult group between pre- and post-RME. Increases of palatal angles were significantly greater in the adult than those in the child group. CONCLUSION: Significant differences existed between the child and the adult group in forms of dental arches and palatal arches, which focused on the changes of molars and palatal angles after RME.

[A study of sensory pathway in central nervous system of experimental tooth movement in rats].

OBJECTIVE: To investigate whether ventral posteromedial thalamus nucleus (VPM) receive the stimulation from experimental tooth movement in rats through the conduction of spinal trigeminal caudal nucleus (Vc), thus to study the central sensory pathway of tooth movement. METHODS: 2% fluorogold (FG) was injected into rat's VPM using microinjector through retrograde tracing technique. After 4-6 days, the experimental tooth movement was conducted in contra-side against FG injection. 2 hours later, animals were sacrificed and FOS protein immunocytochemical reaction was conducted. The sections were observed under Olympus fluorescence microscope. RESULTS: FOS immunocytochemical-positive neurons were densely distributed in epilateral superfacial layer of Vc, being belt-like with more in dorsolateral 2 hours after experimental tooth movement; FG-labeled neurons were found in Vc and principle trigeminal nucleus complex, especially within the whole length of Vc, with more in layer I and the junction of layers I and II. There were both FOS immunocytochemically positive and FG-labeled neurons, approximately 5% of the whole FG-labeled neurons. CONCLUSION: As a special oro-facial stimulation, there are corresponding parts in the central nervous system according to tooth movement. The stimulating message integrated in Vc and Vc has projective neurons to VPM, and VPM might be a relay station of the stimulating message of experimental tooth movement in rats.

[Relationship of p53 gene mutation with pathological characteristics and prognosis of thyroid carcinoma].

This study was directed to the role of p53 gene in the carcinogenesis of thyroid carcinomas and to the correlation between p53 gene and the clinicopathological characteristics of the cancer. Single-stranded conformation polymorphism of PCR was used in detecting p53 gene point mutations in exons 7, 8. The result showed that ten of thirty-one thyroid carcinomas had mutations in exons 7 and 8 (32.3%). The frequency of p53 gene mutations was significantly higher in relapse group than in no relapse group (P < 0.01). No statistically significant differences in p53 mutation were found relating to metastasis, histological type and differentiation (P > 0.05). These data suggest that the mutation of p53 gene may play an important role in thyroid carcinoma and the mutations of p53 gene be associated with the prognosis of thyroid carcinoma.

[Distribution of peptidergic neurons in the rat suprachiasmatic nucleus].

The distribution of the peptidergic neurons in the suprachiasmatic nucleus(SCN) in five adult male SD rats was examined by multi-immunolabelling for vasoactive intestinal polypeptide(VIP), arginine vasopressin (AVP) and somatostatin(SOM). The immunoreactions were visualized with an Elite ABC Kit(Vector Lab). The substrates in the peroxidase reaction were diaminobenzidine(DAB), tetramethylbenzidine(TMB) and 4-chloro-1-naphtol. The results showed that the differences in distribution and population of VIP-, AVP- and SOM-like neurons existed in the rostral, intermediate and caudal regions of the SCN and these neurons were overlapped and SOM-like neurons were scattered among them in the area between VIP- and AVP-like neurons. These findings may provide new morphologic evidences for explaining the functional mechanism in the SCN.
