[The protective effects of paeonol on cultured neonatal rat heart cells during calcium paradox].

It has been found that the rapid (5min) and slow (120min) phases of 15Ca uptake by normal myocardial cells and their beating rate are remarkably inhibited by paeonol. And that paeonol can also decrease 45Ca uptake and lipid peroxide in calcium paradoxical heart cells, which appears in dose-dependence. This indicates that the mechanism of protection of paconol against calcium paradox is associated with its inhibition of Ca2+ influx and antioxidation.

[The inhibitory effects of sodium paeonol sulfate on calcium influx in the cultured neonatal rat heart cells during calcium paradox].

The calcium paradox model was made in cultured neonatal rat heart cells. The 45Ca uptake by the cultured neonatal rat heart cells and the contents of sialic acid (SA) of the cultured heart cells were measured to observe and evaluate the protective effects of sodium paeonol sulfate against calcium paradox in the cultured heart cells. The results demonstrated that sodium paeonol sulfate 50-400 micrograms/ml remarkably inhibited the rapid (5 min) and the slow (120 min) phases of 45Ca uptake by the cultured heart cells and myocardial beating rate (MBR) under normal states. Besides inhibition of 45Ca uptake by the cultured heart cells, this drug was found to elevate the contents of SA in the membrane of the cultured heart cells during calcium paradox. This indicates that the mechanism of protection against calcium paradox by sodium paeonol sulfate may be associated with blocking the slow calcium channel as well as increasing SA in the cell membrane.

[Effects of total saponins of Panax notoginseng on Ca2+ influx into myocardial cells].

Using simultaneous recording of action potential (AP) and contractile force (Fc) in right ventricle papillary muscle of guinea pig and measurement of 45Ca uptake by cultured myocardial cells of neonatal rat, the effects of total saponins of Panax notoginseng (PNS) on the Ca2+ influx into myocardial cells were studied. The duration of the fast AP was shortened, Fc was decreased and the maximal upstroke velocity and amplitude of the slow AP were depressed by PNS. The 45Ca uptake by cultured myocardial cells of neonatal rat was inhibited by PNS. The results indicate that PNS can inhibit Ca2+ influx into myocardial cells.

[Effects of total saponins of Panax notoginseng on increasing PGI2 in carotid artery and decreasing TXA2 in blood platelets].

Total saponins of Panax notoginseng (PNS) were given orally 100 mg/(kg.d) to rabbit for 8 wk. Aortic atherosclerotic plaque formation was restrained as compared to the control group. Radioimmunoassay was used to investigate the effects of PNS on the contents of prostacyclin in carotid artery and thromboxane A2 in blood platelets of rat. Oral administration of PNS 25,50,100 mg/(kg.d) for 10 d, the caused an increase of prostacyclin in carotid artery and a decrease of thromboxane A2 in blood platelets as compared to the control group. These results show that the anti-atherosclerotic action of PNS may be a result of the correction of the unbalance between prostacyclin and thromboxane A2.

[Cytogenetic effects of attenuated live measles vaccine on male mice].

The cytogenetic effects of attenuated live measles vaccine on somatic and germinal cells of C57BL/6J mice were studied. The frequencies of micronuclei of polychromatic erythrocytes, nucleated cells, chromatid breaks, fragments in bone marrow cells, and micronuclei of spermatids in testes were increased, which have dose- and time-dependent relationship. However, there were no statistical difference between the treated and control mice in chromosomal breaks and fragments of spermatogonia primary and secondary spermatocytes. The results and their possible biological significance were also discussed.

Establishment of a human B cell line that responds specifically to B cell growth factor.

A human B cell line (3D5) that responds specifically to B cell growth factor (BCGF) has been developed by a sequence of Staphylococcus aureus Cowen I activation, EB virus immortalization, and cloning. Proliferative response to PHA-stimulated T cell supernatant (PHA-T-Sup) and nonresponsiveness to rIL-2 stimulation were factors used to screen positive cells. Phenotype analysis with a flow cytometer indicated that: 1) 3D5 is a B cell line: 100% of the cells were positive for B1 marker and 59% were positive for sIg, while T3 and Mo 1 were negative; 2) 3D5 is an activated B cell line: both Tac and 4F2 markers of activated (but not of resting) B cells were 100% positive; 3) 3D5 expresses high molecular weight BCGF (HMW-BCGF) receptor-associated epitope BA5. 3D5 cells proliferated in response to cpBCGF stimulation in a dose-dependent manner. HMW-BCGF also induced 3D5 cells to proliferate. Interestingly, no proliferation could be detected in the presence of rIL-2, rIL-4, or rIFN-r. The data show that 3D5 cells are specifically BCGF-responsive B cells. Using 3D5 cells as target, BCGF activity was detected in crude BCGF preparation sedimented by 85% (NH4)2SO4 and chromatographed in a DEAE-Sephadex A-25 column from PHA-T-Sup. T24 cell supernatant with B cell differentiation factor (BCDF) activity could not induce 3D5 cells to differentiate into immunoglobulin-secreting cells.

Demonstration of kinetochores and centrioles in spermatocytes of two species of cockroaches by silver staining.

Light microscopy following silver staining of spermatocytes of German and Madagascar hissing cockroaches demonstrated: (1) the localization of a kinetochore in each autosomal synaptonemal complex during pachytene, and (2) visualization of centrioles in different stages of meiotic prophase. The presence of a "hairpin-like" twist and the nucleolus organizer region in the X-chromosome was observed only in the German cockroach.

Constructions of gene libraries of Rana nigromaculata and Mus musculus with cosmid pHC79.

Gene libraries of Rana nigromaculata and Mus musculus (C57J6) have been constructed by the cosmid system developed by Collins using BamHl and Pstl as cloning restriction enzymes. A simplified formula, which is a modification of Clarke and Carbon derived from approximation, was used for calculating the number of colonies or recombinants necessary for the construction of a gene library. Pro+ and leu+ complementary to the recipient cells (HB101) with the genetic constitution of pro- and leu- have been selected from the BamH1 gene libraries of both Rana nigromaculata and the mouse in the present work.

Quantitative determination of salidroside and specnuezhenide in the fruits of Ligustrum lucidum Ait by high performance liquid chromatography.

An accurate RP-HPLC method for the quantitative determination of two water-soluble biologically active compounds, salidroside (p-hydroxyphenethyl-beta-D-glucoside) and specnuezhenide in the fruits of Ligustrum lucidum Ait (a crude drug in Chinese traditional medicine) was developed. The reversed-phase column was Nova-Pak C18, saturated with water before the injection of the samples and eluted with a mobile phase of methanol:water (4:6, v/v). The detection wavelength was 230 nm. The recoveries of the two compounds were 96.1 and 97.0%, respectively. The contents of salidroside and specnuezhenide in the fruits of Ligustrum lucidum Ait and two traditional Chinese patent medicines in which this crude drug was an important component, Erzhi Pills and Anshenbuxin Pills, were determined.
