A case of simultaneous bilateral herpetic epithelial keratitis.

The patient, a 56-year-old man, presented with dendritic keratitis in the right eye and geographic keratitis in the left, with decreased corneal sensation in both eyes. The virus isolated from both eyes was identified as herpes simplex virus (HSV-1) by the indirect immunofluorescent method using anti-HSV-1 monoclonal antibody. Antibody tests of paired sera suggested that the epithelial lesions were not primary but recurrent. No significant difference was observed in DNA cleavage patterns between the virus isolates obtained bilaterally.

Human papillomavirus DNA in condylomata acuminata from Japanese males.

We examined condylomata acuminata from Japanese males for the presence of human papillomavirus (HPV) genomes by Southern blot hybridization. HPV 6/11-related DNA was found in 91% (32/35) of the condylomata. HPV 6a DNA was found in 40% (14/35), HPV 6c DNA in 6% (2/35), and HPV 11a DNA in 37% (13/35). HPV 6-related DNA, which had an unusual PstI-cleavage pattern, was detected in one sample. Types and subtypes of HPV DNA in the samples studied (HPV 6a, 6c, and HPV 11a DNA) were not correlated with the patients' ages nor outcomes of the disease. HPV 16 DNA was not detected in any condyloma acuminatum.

[Pharmacokinetics of high-dose cisplatin administration (relationship between pharmacokinetics and administration schedule)].

Three cases of urogenital malignancy were treated with 80-120/m2 of cisplatin by a 0.5-hour (rapid) or 4-hour (slow) infusion. Each patient received three courses of repeated chemotherapy. The kinetics of total platinum was measured up to 48 hours in the blood and urine. The blood peak concentrations were achieved 6.0 micrograms/ml by rapid infusion, 2.0 micrograms/ml by slow infusion. The time dependent urinary excretion rate against total administered dose was 40% in 60 min. by rapid infusion, 20% in 16 hours by slow infusion. The volume of distribution correlated with infusion time. Rapid infusion way of cisplatin could be better to treat the patient with bulky tumor mass or patients with uroepithelial cancer like CIS, because a higher blood and urine concentration can be obtained than by slow infusion.

Detection of herpes simplex virus type 1-IgM immune complexes in the brain of a patient with prolonged herpes encephalitis.

A 56-year-old man with herpes encephalitis died 5.5 months after disease onset. Herpes simplex virus was isolated from minced temporal lobe after one month of cocultivation with green monkey kidney cells. The virus was identified as type 1 by neutralization and the fingerprinting pattern on restriction enzyme digestion. By immunofluorescence, IgM deposition was seen in the cerebral vascular walls. After dissociation of IgM with 3.0 M NaSCN, viral antigen was noted at the same loci as the IgM deposition. Histopathology showed marked perivascular cuffings, approximately 80% of which consisted of T cells and widely distributed necrotic foci. When the patient's serum and spinal fluid were analyzed by the immunoblotting method, the spinal fluid contained antibodies that were reactive with two very-high-molecular-weight viral polypeptides.

Herpes simplex virus type 1-induced hydrocephalus in mice.

Adult ICR/Slc or BALB/c mice developed hydrocephalus when attenuated herpes simplex virus type 1 (HSV-1) (strain Ska) was injected intracerebrally 2 to 4 weeks earlier and then after mice were challenged with the same virus or virulent HSV-1. Initial inoculation of the Ska strain elicited acute meningitis and ependymitis with transient mild hydrocephalus. Viral antigen was seen in the meninges and subependymal areas, and the virus was titrated during the acute phase of infection. After the second virus inoculation, more prominent inflammation was evoked in the same area, and the animals developed hydrocephalus, although viral antigen and infectious virus were hardly detected. When the mice were immunosuppressed with cyclophosphamide, they ceased to develop hydrocephalus. BALB/c nude mice did not show the same pathology, even though they were treated in the same way. When irradiated mice, which had been infected with the Ska strain intracerebrally 2 weeks earlier, received syngeneic immune spleen cells, they developed hydrocephalus. The T-cell nature of the effector cells was confirmed by the elimination of the pathology after treatment of the donor cells with anti-Thy-1.2 plus complement. No hydrocephalic mice were observed after treatment of the donor cells with anti-Lyt-1.2 plus complement, which gave further evidence of the T-cell nature of the effector cells as the Lyt-1+.2+ antigen-bearing subsets. Intervals between priming and challenge virus inoculation could be more than 18 months. The presence of purified HSV-1 envelope protein was feasible for the development of the hydrocephalic animals.

Transforming potential of DNA of the human PLC/PRF/5 hepatoma cell line.

DNA of the human PLC/PRF/5 hepatoma cell line can induce the appearance of colonies on soft agar after transfection of BK-BK cells (BALB/c mouse kidney cells partially transformed by BK virus). Blot hybridization analyses indicate that the transformants contained the hepatitis B virus DNA sequence. This sequence disappeared during serial passage of transformants. The amplification and rearrangement of the integrated BK virus genome appears to be specifically associated with transformation.

[Clinical evaluation of Cernilton in benign prostatic hypertrophy].

Twenty patients with benign prostatic hypertrophy were treated with Cernilton, 6 tablets a day for an average of 13.2 weeks. Subjective effectiveness was observed in the improvement of sense of residual urine (92%), retardation (86%), night frequency (85%), strain on urination (56%), protraction (53%) and forceless urinary stream (53%). The overall subjective effectiveness was 80% of patients, and the overall objective effectiveness was 54% of patients. Night frequency, residual urine volume and tidal urine volume were improved significantly. The overall effectiveness was 80%. No side effects were observed.

[Investigation of the relationship between the function and light and electron microscopic structure of osteoblasts and changes in the tibial cortex in rachitic rats].

Four-week-old male Wistar rats were fed a low phosphorus diet without vitamin D and were injected with 1% lead acetate solution twice intravenously. Their tibias were then taken and decalcified in 5% formic acid solution saturated with hydrogen sulphate, in the cross-section of which the deposition of lead sulphate was observed under light microscopy as two concentric lines. The rate of matrix formation was calculated by measuring the distance of the two lines and compared with electron microscopic findings of the same sample. These results showed a decrease of the matrix formation rate and disturbance of mineralization in a low phosphorus diet. In the cortex of rickets, "resorption cavities" of various sizes were observed, and it was presumed that "resorption" came to the front with the progress of rachitogenic change, although whether resorption itself could be accelerated is still dubious.

Synthesis and secretion of biologically active mouse interferon-beta using a Bacillus subtilis alpha-amylase secretion vector.

Mouse interferon-beta (IFN-beta) cDNA, whose signal sequence had been removed by BAL 31 digestion, was introduced into a Bacillus subtilis secretion vector constructed by using the promoter and signal sequence of the B. subtilis alpha-amylase gene. The resultant chimeric plasmids were transferred into B. subtilis 207-25. Four kanamycin-resistant transformants were selected by both colony hybridization and a new immunoblot method for secretory proteins. They secrete the proteins which cross-react with sheep anti-mouse IFN-beta serum into the culture medium. One of them expressed a high IFN-beta activity as assayed by the L cell and vesicular stomatitis virus system, while the other three showed weak or little IFN activities. Based on our previous study [Ohmura et al., Nucl. Acids Res. 12 (1984) 5307-5319], it was suggested that the secreted IFN molecules are hybrid proteins in which the NH2-terminal region consists of part of the alpha-amylase signal peptide. Nucleotide sequence analysis revealed that plasmid pTUB502, which expressed high IFN activity, is joined to the mouse IFN-beta gene from the codon position 6 of its mature protein. The other three plasmids, pTUB506, pTUB509, and pTUB519, contain the mouse IFN-beta gene from the codon positions 3, 1, and -5, respectively. The NH2-terminal region of the mouse IFN-beta seems to be closely related to its biological activity.

[The effectiveness of chemotherapy and craniotomy for brain metastasis of non-seminomatous testicular tumor].

Four patients with non-seminomatous testicular tumor who already had brain metastasis were treated with combination chemotherapy. Three patients had received craniotomy in an effort to remove the metastatic lesion and intracranial hematoma. Two of them who were treated with PVB chemotherapy, which was effective against pulmonary and retroperitoneal metastasis but not against the brain metastatic lesions, died within 3 months; the other patient is receiving intense postcraniotomy chemotherapy using Cisplatin and large doses of Methotrexate administered with the Leucovorin rescue method which has shown a remarkable response against the brain metastasis of choriocarcinoma. The remaining patient has been receiving VAB VI protocol for 3 months. The metastatic lesion in the temporal lobe of his brain may consolidate through calcification, similar to the calcified change observed in the retroperitoneal lymph-node after chemotherapy. We discuss potential ways to induce improved therapeutic effects against brain metastasis of the non-seminomatous testicular tumor: It may be difficult to achieve an effective drug concentration level in the tissue immediately adjacent to the intracerebral tumor, because of the blood brain barrier. As induction therapy, a large dose of Cisplatinum (230 mg/body) or Methotrexate (10 g/body) was effective in attaining an effective drug concentration level in the tissue adjacent to tumor. Prior to the stem cell assay of the brain metastatic tumor, 1,100 mg Cisplatinum and 1,700 mg VP 16 were administered for treatment. The results of the stem cell assay in vitro showed a resistance to Cisplatinum and VP 16. Routine brain CT scanning is useful for detecting a metastatic lesion in its development. If detected, multidisciplinary chemotherapy should be performed.(ABSTRACT TRUNCATED AT 250 WORDS)

Irreversible conversion of the physical state of herpes simplex virus preceding inactivation by thermal or antibody treatment.

The buoyant density characteristics of infectious particles of herpes simplex virus types 1 and 2 were studied by centrifugation in sucrose and cesium chloride density gradients with a high resolution and satisfactory infectivity recovery. It was shown that two populations of infectious virions differing in buoyant density coexisted, the difference being slight but definite. The ratio of heavy (H) to light (L) viral particles varied depending upon the solute used, the strains of virus, and the cell origin. Circumstances favoring degradation of viral infectivity tended to increase the H portion. Incubation at 37 degrees C largely converted L to H, and heating at 45 degrees C converted all virions to H without infectivity. The L to H conversion was irreversible, and no populations intermediate between L and H were clearly observed. Inactivation by UV light irradiation did not affect the density pattern. That H was not an artefact due to penetration of solutes, osmotic pressure, viral aggregation, or loss of the envelope was shown experimentally. A difference in the outer shape of particles between negatively stained L and H populations was demonstrated by electron microscopy. Both cell-released and cell-bound herpes simplex virus particles gave essentially the same result with respect to the above characteristics. The effect of limiting dilutions of antiserum was similar to that of mild thermal treatment, in that denser virions increased parallel to a decrease in less dense virions. Sensitization with early immunoglobulin G, composed mainly of complement-requiring neutralizing antibody, caused the density transition, and subsequent addition of complement resulted in a further increase in the buoyant density of the sensitized virions. The DNA in virus particles neutralized with immunoglobulin G plus complement remained resistant to DNase treatment. Possible implications of the phenomena are discussed.

Selective staining by 4', 6-diamidine-2-phenylindole of nanogram quantities of DNA in the presence of RNA on gels.

4', 6-Diamidine-2-phenylindole.2HCl (DAPI) forms fluorescent complexes with double-stranded (ds) DNA but not with ds RNA as shown by fluorescence titration. The widely used dye ethidium bromide (EB) forms fluorescent complexes with both types of nucleic acids. Also, in contrast to EB, DAPI forms much weaker fluorescent complexes with single-stranded DNA than with ds DNA. These observations were utilized to develop staining procedures for the selective visualization of ds DNA on gels. The use of DAPI in addition to EB for staining makes possible the localization of ds DNA and other species of nucleic acids on a single gel.

Integration of herpes simplex virus type 1 DNA into the DNA of growth-arrested BHK-21 cells.

The ability of HSV-1 DNA to become associated with host cell DNA in an alkaline-stable form has been demonstrated following infection of a ts baby hamster kidney growth mutant (ts BTN-1), at the non-permissive temperature (39.5 depgrees C). After 8 h pre-incubation at 39.5 degrees C, ts BTN-1 cells infected at this temperature using m.o.i. ranging from 0.5 to 200 p.f.u./cell fail to replicate virus DNA even though transport of input virus genomes to the nucleus is the same at both permissive and non-permissive temperatures. Virions containing 3H-labelled DNA were used to infect ts BTN-1 cells at 39.5 degrees C, and the total cellular DNA isolated from these cells was resolved into host and virus material by repeated CsCl equilibrium gradient centrifugation. A significant amount of the input radioactivity was found as a distinct band in the host region in both neutral and alkaline CsCl gradients, strongly suggesting a covalent association between host and virus DNAs. Evidence for this association was strengthened by demonstrating that radioactive material (virus DNA) banding in the host region of CsCl gradients could be driven towards the density expected for virus DNA following degradation of the putative hybrid molecules by shearing.
