Improvement of chemical analysis of antibiotics. XII. Simultaneous analysis of seven tetracyclines in honey.

An analytical system for the simultaneous determination of residual oxytetracycline, tetracycline, chlortetracycline, doxycycline, methacycline, demethylchlortetracycline and minocycline in honey has been established by a combination of simple thin-layer chromatographic (TLC) and precise high-performance liquid chromatographic (HPLC) methods. In this system, screening by TLC can detect tetracyclines (TCs) at a level of 0.1 ppm in honey without the need for special equipment, and the quantitative method by HPLC can determine TCs with good recovery (83.7-99.6%) and coefficient variation (0.9-4.3%).

Improvement of chemical analysis of antibiotics. X. Determination of eight tetracyclines using thin-layer and high-performance liquid chromatography.

Analytical methods for eight tetracyclines (TCs) were established using silica gel high-performance thin-layer chromatography (HPTLC), reversed-phase thin-layer chromatography (RP-TLC) and high-performance liquid chromatography (HPLC). Good separations of eight TCs were obtained using chloroform-methanol-5% disodium ethylenediaminetetraacetate solution (65:20:5) (lower layer) and methanol acetonitrile 0.5 M oxalic acid solution (1:1:4) (pH 3.0) on silica gel HPTLC and C8 TLC plates, respectively. A combination of HPTLC and RP-TLC made possible the identification of the eight TCs. Each calibration graph was linear between 0.1 and 1.0 microgram using UV densitometry except for rolitetracycline. For detection reagents, the diazonium salts including Fast Violet B gave variously coloured spots with the eight TCs and good sensitivities were obtained except with minocycline. In HPLC, the simultaneous analysis of the eight TCs on a C8 column was possible using methanol-acetonitrile-0.01 M oxalic acid solution (1:1.5:7) adjusted to pH 3.0 as the mobile phase. A linear relationship was obtained between 1.0 and 10 ng using the usual sample preparation except for rolitetracycline. The direct determination of rolitetracycline was possible using tetrahydrofuran, dimethyl sulphoxide and the mobile phase as solvents for preparation of the sample. For the determination of residual rolitetracycline, it was effective to measure the amount of rolitetracycline as tetracycline by HPLC, HPTLC and RP-TLC after conversion of rolitetracycline to tetracycline by incubating for 5 min in methanol at 50 degrees C.

Improvement of chemical analysis of antibiotics. IX. A simple method for residual tetracyclines analysis in honey using a tandem cartridge clean-up system.

A simple, rapid and precise analytical method for the residual tetracyclines in honey has been established using a tandem cartridge clean-up system (prepacked reversed-phase and ion-exchange cartridges) followed by high-performance liquid chromatography. The recoveries of oxytetracycline (OTC), tetracycline (TC), chlortetracycline (CTC) and doxycycline (DC) from honey spiked at a level of 1.0 ppm are 87.1, 85.3, 98.0 and 99.0%, respectively, with coefficients of variation of 1.1-3.9%. The detection limits in honey are 0.02 ppm for OTC and TC, and 0.05 ppm for CTC and DC, respectively. The time required for the analysis of four samples is only 1 h.

The complementary role of indium-111 labeled leukocyte imaging, ultrasonography and computed tomography in the evaluation of postoperative infection or abscess.

We report our experiences with the combined use of indium-111 labeled leukocyte imaging (In-III WBC scan.), computed tomography (CT) and ultrasonography (US) for evaluation of suspected postoperative infection or abscess, and discuss the complementary roles of these modalities. Postoperative abscesses or infections were diagnosed in 9 of 20 patients. All patients were correctly diagnosed by In-111 WBC imaging and 4 patients could not be diagnosed by US because of bowel gas. One false-positive CT examination and another artifact on CT images due to respiratory movements were obtained. The three modalities were found to be complementary: CT and US were efficient imaging methods for diagnosis and treatment of abscess. In-111 WBC imaging could estimate the activity of inflammation.

[A case of non-Hodgkin's lymphoma manifesting an eyelid tumor as a major symptom].

It is well known that a majority of non-Hodgkin's lymphoma (NHL) in the head and neck region arises from the Waldeyer's ring and neck lymph nodes. This report covers a rare case of a 69 year-old male with a symptom like the Mikulicz syndrome, who was diagnosed as having the NHL of B-cell type. This patient has been in complete remission following local radiotherapy for the tumor of the eyelid.

Relationship between chlordane and its metabolites in blood of pest control operators and spraying conditions.

Chlordane has been widely used to protect soil and house foundations against termite infestation. Pest control operators (PCOs) are occupationally exposed to chlordane. The relationship between chlordane and its metabolites in blood of PCOs and spraying conditions were investigated. Chlordane and its metabolites were detected in the blood of some chlordane-exposed PCOs, but not in that of the controls. Trans-nonachlor and the metabolites oxychlordane and heptachlor epoxide were detected in the blood of PCOs. Total concentration of chlordane and its metabolites in blood (trans-nonachlor + oxychlordane + heptachlor epoxide) was less than 5.6 ppb (mean: 0.89 ppb). The concentration of chlordane and its metabolites in blood of chlordane-exposed PCOs was significantly correlated with the number of spraying days and the amount of chlordane sprayed, particularly with a large correlation coefficient (r = 0.81, P less than 0.001) with the spraying days in the three months prior to the medical examination. The concentration of chlordane and its metabolites in blood is considered to be a useful indicator of biological monitoring for chlordane exposed workers (PCOs).

Selection of yeasts for breadmaking by the frozen-dough method.

Eleven yeast strains suitable for frozen dough were selected from over 300 Saccharomyces strains. All of these were identified as Saccharomyces cerevisiae from morphological, cultural, and physiological characteristics. The selected yeast cells accumulated a higher amount of trehalose than did commercial bakers' yeast cells.

[Performance of gamma camera collimators used for single photon emission computed tomography imaging with 123I-isopropyl iodoamphetamine].

123I Produced by 124Te(p, 2n)123I reaction is contaminated with 124I (less than 5%) and 126I (less than 0.3%). High energy photons from these mixed radioiodine compromise seriously image quality due to scattered photons and to septal penetration in the collimator. Four collimators of LEAP (for low energy all purpose), LEHR (for low energy high resolution), MESI (for medium energy made by Siemens) and MENU (for medium energy made by nuclear technology) mounted on a rotating gamma camera (Siemens, ZLC-7500), were examined in order to select a suitable collimator for 123I SPECT (single photon emission computed tomography) imaging. Sensitivities were measured with a plane source (5 X 5 X 0.5 cm) at the collimator face and distances from 2 to 30 cm in air. And, spatial resolutions in FWHM (full width at half maximum) and FWTM (full width at tenth maximum) were determined from line spread functions with planar and SPECT imaging. From the comparison of collimator performances with 99mTc and 123I, both collimators for low energy were not useful for 123I imaging. In other two collimators for medium energy, however, apparently the effect of septal penetration by the higher energy photons were also recognized, MENU with high geometrical resolution was more suitable for 123I SPECT imaging compared with MESI. And, it is important to perform the SPECT imaging with radius as short as possible.

Primary intramedullary spinal cord lymphoma: report of a case.

A case of primary intramedullary spinal cord malignant lymphoma is described. The patient was a 24-year-old female with a preceding history of a lupus-erythematosus-like disorder. The presenting symptom of the spinal cord tumor was spastic paresis of the lower extremities. Radiological examinations including magnetic resonance imaging (MRI) of the spinal cord revealed an intramedullary tumor from levels C2 to T6. A decompressive laminectomy was performed with the pathological diagnosis of poorly differentiated lymphocytic lymphoma with T-cell surface marker. Postoperative cerebrospinal radiation up to 40.5 Gy was performed. The patient is alive without disease 18 months after completion of the therapy. MRI proved to be a very useful method for evaluation of the response to therapy and for follow-up. Primary involvement of the spinal cord with non-Hodgkin's lymphoma is unusual and this is the ninth case of primary intramedullary spinal cord lymphoma reported.

Indium-111 leukocyte imaging in patients with rheumatoid arthritis.

This study evaluates the usefulness of labeled leukocyte imaging in patients with rheumatoid arthritis. In 33 patients, the incidence of pain and swelling in 66 wrist joints and 66 knee joints was compared with the accumulation of [111In]leukocytes. No accumulation of [111In]leukocytes was seen in any of the patients' wrists (0/12) or knee joints (0/14) when both pain and swelling were absent. In contrast, 93% (25/27) of wrist joints and 80% (24/30) of knee joints with both pain and swelling were positive by [111In]leukocyte scintigraphy. There was little correlation between the stage of the disease, as determined by radiography, and [111In]leukocyte accumulation. This study suggests that [111In]leukocyte imaging may be a reliable procedure for monitoring the activity of rheumatoid arthritis, especially for confirming the lack of an ongoing inflammatory response.

Improvement of chemical analysis of antibiotics. VIII. Application of prepacked C18 cartridge for the analysis of tetracycline residues in animal liver.

A simple, rapid and precise analytical method for tetracycline (TC) residues in the liver of slaughtered animals has been established. The recoveries of oxytetracycline (OTC), TC, chlortetracycline (CTC) and doxycycline (DC) from beef liver spiked at the level of 1.0 ppm were 87.7, 87.5, 79.6 and 67.5% with coefficients of variations of 1.01-2.87%. Detection limits in beef liver were 0.05 and 0.1 ppm for OTC and TC and for CTC and DC, respectively. It is also possible to apply this method to the analysis of residual TCs in various foods with the same recovery, accuracy and detection limits as in the case of beef liver.
