A simple and rapid method for HLA-DQA1 genotyping by digestion of PCR-amplified DNA with allele specific restriction endonucleases.

The second exon of the HLA-DQA1 genes was selectively amplified from genomic DNAs of 72 HLA-homozygous B cell lines by the polymerase chain reaction (PCR). Amplified DNAs were digested with HaeIII, Ddel, ScrFI, FokI and RsaI, which recognize allelic sequence variations in the polymorphic segments of the DQA1 second exon, and then subjected to electrophoresis in polyacrylamide gels. Eight different polymorphic patterns of restriction fragments were obtained, and seven were identical to patterns predicted from the known DNA sequences, correlating with each HLA-DQw type defined by serological typing. The remaining one pattern cannot be explained from the sequence data, suggesting the presence of a novel DQA1 allele at the nucleotide level. This PCR-RFLP method provides a simple and rapid technique for accurate definition of the HLA-DQ types at the nucleotide level, eliminating the need for radioisotope as well as allele specific oligonucleotide probes and can be extended and applied to HLA-DR, -Dw DP typing.

Specific inhibitory substance to the husband's HLA allotype in sera from patients with gestational trophoblastic disease detected by lymphocytotoxicity testing.

The sera of nine patients with gestational trophoblastic disease were investigated for the inhibitory substance specific to the HLA allotypes of the patients' husbands by microcytotoxicity inhibition testing using monoclonal antibodies (anti-A2, A9, A10) and HLA-typing serum to Bw52. The specific inhibitory substance was present in three of three choriocarcinoma patients, in three of four invasive mole patients, and in one of two hydatidiform mole patients and seemed to change in titer parallel with tumor growth. These facts suggest that the inhibitory substance could have some relation to the patient's immunological response against the tumor; that is, this inhibitory substance could be the reason why patients could not induce graft rejection response to trophoblastic tumor cells.

Short term effects of glucose and arginine on the preproinsulin messenger ribonucleic acid level in the perfused rat pancreas: comparison with insulin secretion.

To investigate the role of glucose and arginine in short term regulation of preproinsulin mRNA (ppImRNA) levels, the rat pancreas was perfused in the presence of glucose and/or arginine, and changes in ppImRNA levels in the pancreas were compared with the amount of insulin released during the perfusion. Perfusion of the pancreas with high glucose and arginine induced a significant increase in ppImRNA levels within 2 h, whereas perfusion with low glucose and arginine or high glucose alone had no significant effect during this period. The insulin release induced by perfusion of high glucose combined with arginine was 2 times greater than that induced by high glucose alone or low glucose with arginine. In conclusion, insulin gene transcription can be evoked during a short period in response to an extremely large secretory demand for insulin.

Studies on the antihemostatic substances in herbs classified as hemostatics in traditional Chinese medicine. I. On the antihemostatic principles in Sophora japonica L.

By following the antihemostatic activity of the material according to Tajima's method, the antihemostatic principle was isolated from dried buds of Sophora japonica L., and identified as isorhamnetin [2-(3-methoxy-4-hydroxyphenyl)-3,5,7-trihydroxy-4H-1-benzopyran-4-one]. The antihemostatic specificity of isorhamnetin toward antihemorrhagic compounds isolated previously from hemostatic herbs used in traditional Chinese medicine was examined.

[Studies on the anti-hemorrhagic substances in herbs classified as hemostatics in Chinese medicine. X. On hemostatic activities of the parched herbs for hemostatics].

The effect of heat treatment on the anti-hemorrhagic action of seven hemostatic herbs employed especially after parching in order to work effectively as an anti-hemorrhagic agent in traditional Chinese medicine, was examined. It was found that the anti-hemorrhagic activities of the following 5 herbs are apparently increased by parching: Kaika (Sophorae immaturus Flos), Renbo.(Nelumnbins Receptaculum), Gusetsu (Nelumnbins Rhizomatis Nodus), Chiyu (Sanguisorbae Radix) and Gaiyou (Artemisiae argyi Folium).

Maleness as risk factor for slowly progressive IDDM.

The effect of sex on longitudinal changes in serum C-peptide immunoreactivity (CPR) response to the oral glucose tolerance test (OGTT) was examined up to 48 mo in 30 islet cell antibody-positive (ICA+), non-insulin-dependent diabetes mellitus (NIDDM) subjects (15 men, 15 women) who were matched for age, duration of diabetes, and mode of treatment. The subjects were recruited from 2858 NIDDM patients screened for ICA between 1980 and 1984. In male NIDDM subjects, CPR levels to OGTT decreased insidiously, and 8 of 15 men developed the insulin-dependent state with abolished CPR. Only 2 female NIDDM subjects progressed to the insulin-dependent state (P less than .05, women vs. men). Thus, CPR in female subjects tended to decrease less than in male subjects. There were no significant differences between the two groups in human leukocyte antigens (HLA) or titer of ICA during the follow-up period. These results suggest that maleness is a major risk factor for slowly progressive beta-cell dysfunction in adult-onset insulin-dependent diabetes mellitus (IDDM).

Chicks' maze learning reinforced by visual pitfall extending downward.

The present study examined whether visually evoked fear of depth could reinforce a particular response of animals, i.e., to special maze learning. The maze was composed of four units of Y-shaped alley. In this maze, the visual pitfalls were set behind corners of the alley in place of a physical barrier. The experiments showed that eight of 13 male chicks could achieve the initial learning and that three successful ones could also achieve reversal learning. The results suggest that the visually evoked fear of depth provided by motion parallax can act as a reinforcer.

[Effect of azelastine on bronchial hyperresponsiveness].

We have investigated the effect of azelastine on bronchial responsiveness in 25 asthmatics, using methacholine inhalation challenge, (Astograph). Three parameters, initial respiratory resistance (RrsC), bronchial sensitivity (Dmin), and bronchial reactivity (SGrs/GrsC) were studied. After 8 weeks' treatment with 2 mg azelastine b.i.d., Dmin was increased significantly and after 4 weeks an insignificant increase was observed. RrsC and SGrs/GrsC did not change significantly during the 8 week treatment. Recent advances have revealed that various chemical mediators, especially leukotrienes, are closely related to bronchial hyperresponsiveness. Accordingly, the improvement of Dmin observed during azelastine treatment might be ascribed to its inhibitory action on the synthesis and release of leukotrienes.

[Effect of a protein-bound polysaccharide, PSK, on human hemopoietic progenitors].

Using in vitro clonal culture assays, we investigated the effects of PSK, a protein-bound polysaccharide derived from the cultured mycelium of CM101, Coriolus versicolor (Fr.) Quél in Basidiomycetes, on human hemopoietic progenitors. PSK alone did not stimulate colony formation by human bone marrow progenitors. Although 1-100 micrograms/ml of PSK had no effects on colony formation stimulated by erythropoietin and medium conditioned by phytohemagglutinin-stimulated leukocytes, more than 1 mg/ml of PSK inhibited all types of colony formation. In contrast, medium conditioned by PSK-stimulated leukocytes significantly stimulated formation of various types of colonies including erythroid bursts, granulocyte and/or macrophage colonies, eosinophil colonies, megakaryocyte colonies and mixed hemopoietic colonies. It is speculated that administration of the optimal dose of PSK can reduce the hematological suppression of antitumor drugs.

[Measurement of DNA damage in forestomach squamous epithelium by alkaline elution assay].

The alkaline elution assay were applied for assessing DNA damage of forestomach squamous epithelium in the male F344 rat. The animals were starved for 18 hr and then given a single oral dose of the sample in saline or corn oil. After 3 hr the stomach was removed, opened along the greater curvature and washed with cold 0.024M EDTA-0.075M NaCl (pH 7.5). The mucosa of the forestomach was scraped off gently with the edge of a cover glass, and the stratified squamous epithelium was collected by scraping off firmly with the edge of a cover glass. Since the collected epithelium cells adhere to the pipettes, these cells were transferred onto a polycarbonate filter by using a micro spatula. The dose-dependent DNA damages (5 approximately 150 mg/kg) were observed after administration of 1-methyl-3-nitro-1-nitrosoguanidine (MNNG), a forestomach carcinogen.

[Coexistence of abdominal aortic aneurysm and intraabdominal malignancy; two case reports].

We experienced two cases of abdominal aortic aneurysm which had intra-abdominal malignancy (early gastric cancer). Case 1 was a 72 year-old man who was treated by two-stage operation of them. Gastrectomy was performed about 7 months prior to the aneurysmectomy. Case 2 was a 70 year-old man who was diagnosed both lethal diseases, renal dysfunction, chronic respiratory failure and multiple ventricular arrhythmia. He was treated by one-stage operation and was discharged with no complications. Coexistence of abdominal aortic aneurysm and intra-abdominal malignancy is rare and it is difficult to decide whether to operate the malignancy first, the aneurysm first or both simultaneously. In Japan, 28 cases were reported and 18 cases could be analyzed in detail. In these cases, most frequent coincidental malignancy was the gastric cancer (13/18, 72.2%) and one-stage operation was performed in 8 (44.4%) cases. In general, the risk of infection during upper gastrointestinal surgery is less than that during lower abdominal surgery. We concluded therefore that concomitant resection of the upper gastrointestinal malignancy, especially early gastric cancer, should be considered.
