Anti-testosterone antisera produced in mice of different strains and sexes.

Specific anti-testosterone antiserum was obtained from mice of the strain C57B1/6. Testosterone 3(O-carboxymethyl)oxime-keyhole limpet hemocyanin conjugate was administered to male and female mice of the strains C57B1/6 (H-2b), DBA/2 (H-2d), (C57B1/6 x DBA/2)F1, (H-2b/d), and AKR (H-2k). Antisera obtained from male and female C57B1/6 mice were more specific for testosterone than those from the other strains, and the cross-reaction with 5alpha-dihydrotestosterone was 25.0 and 29.2%, respectively. The titer of antiserum from C57B1/6 mice was the highest and that of antiserum from DBA/2 mice was the lowest. On the other hand, the titer of antiserum from the female was higher than that of antiserum from the male in each strain. These results indicate that the mouse strain and sex differences are important factors for antibody formation from challenged antigen.

Enzyme immunoassay of serum testosterone.

An enzyme immunoassay of serum testosterone using the testosterone-glucoamylase complex is described. Testosterone was estimated by the enzyme immunoassay after extraction with hexan: ether (4:1) for serum from men and additional thin layer chromatographic step for serum from women. The within and between assay errors, measured as the coefficient of variation were 11.1 percent (n=8) and 12.0 percent (n=12). The sensitivity of this assay was 0.25 ng. The mean testosterone concentration (+/- SD) in 19 normal men and 4 normal cycling women were 5.3 +/- 1.8 and 0.52 +/- 0.12 ng/ml, respectively. The level of testosterone found by the present assay compared favorably with those obtained by other methods.

Determination of urinary dehydroepiandrosterone using 3beta-hydroxysteroid dehydrogenase.

A method for the determination of urinary dehydroepiandrosterone (DHA) using 3beta-hydroxysteroid dehydrogenase is described. The method includes extraction with ethylacetate, solvolysis, and color development using 3beta-hydroxysteroid dehydrogenase. A study of comparison with gas-liquid chromatography method and recovery test was made, and results were sufficient for routine tests.

Enzyme immunoassay of testosterone using the testosterone-glucoamylase complex.

A highly sensitive, reproducible method was established for enzyme-coupled immunoassay of testosterone, involving coupling of testosterone to glucoamylase [EC 3.2.1.3] with a water-soluble coupling reagen carbodimide. No enzyme activity was lost during this coupling procedure. The sensitivity of the method was comparable to that of competitive protein binding assay.

Binding of human chorionic gonadotropin by rat ovarian slices: dependence on the functional state of the ovary (38571).

Ovarian receptors specific for hCG-LH can be induced by PMSG-ovine LH priming. The marked increase in binding activity was accompanied by a rise in ovarian progesterone concentration and preceded by ovarian weight increase. The high binding activity of heavily luteinized ovaries was followed by a decline to prepriming levels during luteolysis. The ovarian progesterone content was decreased simultaneously and no change in ovarian weight was evident. The data demonstrate that ovarian hCG binding activity undergoes variation which depends on the functional state of ovaries.

Evidence for the effect of aging on tissue contents of gastrin, cholecystokinin and somatostatin in the rat.

The changes associated with aging in the tissue concentrations of cholecystokinin, somatostatin and gastrin in young (7-month-old), middle-aged (13-month-old) and old (25-26-month-old) rats were investigated. The concentrations and total contents of somatostatin and gastrin significantly decreased in the antrum in 13- and 25-26-month-old rats compared with the young controls. The cholecystokinin concentration in the proximal intestine and its total content significantly increased in the old rats. Somatostatin concentrations in the intestine and cerebral cortex did not change with age. Cholecystokinin content in the cerebral cortex decreased in old rats because of the decrease of tissue wet weight. We conclude that the effects of aging on the changes of peptide concentrations in tissues vary depending on peptide specie and in different tissues studied.

CCK pathway from supramammillary region to the nucleus anterior ventralis thalami of the young rats.

The existence of cholecystokinin-8-like immunoreactive (CCKI) pathway from supramammillary region (SUM) to the nucleus anterior ventralis thalami (AVT) was demonstrated in this study by means of experimental immunohistochemical manipulations in very young rats, because destruction of unilateral SUM, which contains CCKI neurons, resulted in a disappearance of CCKI fibers in the ipsilateral AVT.

Control of IkappaBalpha proteolysis by the ubiquitin-proteasome pathway.

It has recently been determined that the proteolytic destruction of IkappaB (inhibitor of NF-kappaB) by the ubiquitin-proteasome system plays a key role in the immediate elimination of IkappaB from the IkappaB-(NF-kappaB) complex which allows nuclear translocation of free NF-kappaB, thus leading to activation of a multitude of target genes. The SCF(Fbw1) (composed of Skp1, Cul-1, Roc1, and Fbw1) complex, identified as an IkappaBalpha-E3 ligase, binds and ubiquitylates IkappaBalpha phosphorylated by IkappaB kinase that has been activated in response to extracellular signals. The generating poly-ubiquitin chain is finally recognized by the 26S proteasome for ultimate degradation. In this NF-kappaB signalling pathway, it becomes clear that the SCF(Fbw1) activity is enhanced by a ubiquitin-like protein NEDD8 (equivalent to Rub1) that modifies Cul-1 in a manner analogous to ubiquitylation, and consequently, IkappaBalpha proteolysis is induced. NEDD8 is a new regulator of the SCF ubiquitin-ligase, functioning as a covalent modifier for proteolytic targeting at a physiological level.

Dynamic computed tomography in acute renal failure: analysis of time-density curve.

An analysis of dynamic CT curves in 12 patients with acute renal failure was performed. An oliguric patient who demonstrates corticomedullary differentiation on CT images will develop diuresis within 4 days. A corticoaortic junction time of more than 180 s associated with no differentiation between cortex and medulla is associated with prolonged oligoanuria. Impending diuresis is heralded by a corticoaortic junction time of less than 180 s. These preliminary results suggest that an analysis of dynamic CT curves in acute renal failure can predict the time of appearance of the diuretic stage.

Assay of somatomedin C by cartridge extraction prior to radioimmunoassay with antiserum developed against synthetic somatomedin C.

The whole molecule of human somatomedin C (SM-C) prepared by the total synthesis method was used as an antigen to produce an antiserum for a radioimmunoassay. Since plasma proteins that bind SM-C interfere with the assay, a method was developed that uses acid dissociation followed by C-2 cartridge extraction to strip SM-C from its binding proteins before assay. This assay has no cross-reactivity with human proinsulin or insulin-like growth factor II (IGF-II). The SM-C values in 339 normal subjects showed age-dependence, increasing from childhood to a peak at age 14 to 16 years and decreasing sharply before adulthood. In adults, the SM-C values decreased gradually with age. All 13 patients with acromegaly who were tested had an increased SM-C value, with no overlap with the normal range. The 12 patients with prolactinoma but non-growth-hormone-producing pituitary tumor had no increase in SM-C. Two children with pituitary deficiency had low SM-C values; one of these children received growth hormone therapy, and his SM-C value increased from undetectable to normal. By three weeks after discontinuation of the therapy, his SM-C value was again undetectable. Of 20 children with short stature and constitutional delay of growth and development, SM-C was below normal in 70 percent and normal in 30 percent. Two patients with malnutrition had below-normal SM-C values.
