Direct cytotoxicity against chicken erythrocytes in mice. III. Degrees of radiosensitivity and cross-reactivity of cytotoxicity, delayed hypersensitivity and antibody production.

Exposure to 600 rad of X-irradiation 3 h after primary immunization with chicken erythrocytes (CRBC) abolished the production of antibodies and the generation of killer T cells, but scarcely affected the induction of delayed footpad reactions. Exposure to irradiation 3 h after secondary immunization reduced only slightly the generation of killer T cells and reduced slightly or substantially the production of antibodies. Delayed reactions persisted for long periods after elicitation in irradiated, boosted mice. Cross-reactivity between CRBC and quail erythrocytes (QRBC) was very weak with respect to the cytotoxicity, antibody and delayed reaction raised after primary immunization. Those raised after secondary immunization with the homologous antigen showed some degrees of cross-reactivity to another antigen. The booster with QRBC in CRBC-primed mice augmented the response to CRBC with respsect to the induction of cytotoxicity and delayed footpad reaction, but only weakly affected the response with respect to the production of antibody. Therefore, effector cells of cytotoxicity and delayed footpad reaction showed greater degrees of cross-reactivity than antibody-producing cells during an anamnestic response. The cross-reactive response raised after secondary immunization was radioresistant with respect to cytotoxicity and delayed footpad reaction but radiosensitive with respect to antibody production. Cytoxicity to immunizing antigen was inhibited by the unlabelled homologous antigen. Cytotoxicity to a cross-reacting antigen was inhibited by the same cross-reacting antigen and also by an immunizing antigen. There may exist heterogeneous populations of cytotoxic lymphocytes with different abilities to recognize antigens.

Biliary scintigraphy with a new scanning agent, 99mTc-(SN)-pyridoxylidene-isoleucine.

99mTechnetium-pyridoxylidene-isoleucine (hereinafter referred to as 99mTc-PI) is a new biliary scintiscanning agent. It has been evaluated in both health volunteers and patients with known surgical biliary tract diseases. It results in imaging of the liver in three minutes and the common bile duct and duodenum in 10-20 minutes. Its advantages as a diagnostic tool in disease of the hepatobiliary systems are in reducing the amount of radiotracer required, in shortening examination time and in producing scintigrams of higher resolution.

A cytologic study on small cell carcinoma of the esophagus.

The cytologic picture of small cell carcinoma primarily arising from the esophagus was studied with 7 cases which were confirmed by histologic examination. Cytomorphologic characteristics of small cell carcinoma of the esophagus are as follows: the arrangement of groups of tumor cells is irregular and overlapping with indistinct cell boundaries. The cytoplasm is small, or sometimes absent. The nuclei are round, oval or occasionally spindle shaped. Nuclear borders are thin. The chromatin of finely granular pattern has increased and is evenly distributed.

Inactive form of erythrocyte carbonic anhydrase B in patients with primary renal tubular acidosis.

Evidence was found for an inactive form of carbonic anhydrase type B in the erythrocytes of two children with primary renal tubular acidosis. The addition of zinc chloride to hemolysates from these patients resulted in a marked increase in the activity of this enzyme. No such effect was noted with hemolysates of control subjects. No significant differences were observed in the zinc levels of hemolysates of these patients and of normal individuals. However, the level of zinc in the carbonic anhydrase B isolated from one of these patients was low, suggesting a modified form of the enzyme. The restoration of activity upon the addition of zinc was reversed by ethylenediamine tetraacetate, but no such effects were noted for the carbonic anhydrase B of normal individuals. Thus the abnormal carbonic anhydrase B has decreased zinc binding. The ultraviolet difference spectrum of the carbonic anhydrase B of normal individuals and that of a patient showed a peak at 305 nm which decreased upon the addition of zinc. The abnormal form of carbonic anhydrase B was not distinguishable from that of normal individuals by either immunological or electrophoretic criteria.

Cellular immunity in rheumatic heart disease.

Cellular immunity to non-specific antigens (PPD, Candida albicans, and streptococcal antigens) and mitogen (PHA) was investigated in patients with valvular heart disease of rheumatic origin. The result disclosed that lowered response in cellular immunity exists in these patients. The possible significance of this finding in occurrence of rheumatic activity was discussed.

Hydraulic and hemodynamic studies on the blood flow through the cardiovascular system.

To investigate the hemodynamic aspects across a stenosis in the cardiovascular system, in vitro, in vivo, and clinical studies were performed. The results are summarized as follows: 1) Remarkable changes in the pressure profile were recognized downstream. The relationship between pressure gradient and flow across the stenotic orifice revealed an inverse linear relationship in in vitro studies, but showed a slight convex curve in in vivo studies. 2) The critical stenosis in vitro and in vivo studies 10% and 25%, respectively. 3) In the group of mitral valve area larger than 0.5 cm2, flow coefficient C was smaller than 1.0 and blood flow could be "vena contracta". 4) Gorlin's formula is considered to be appropriate in the cases with valve area larger than 0.5 cm2. But Gorlin's formula may over-estimate in the group with valve area smaller than 0.5 cm2.

Does the supplementary motor area play a part in modifying motor cortex reflexes?

Neuronal activity in the supplementary motor area was recorded from a monkey performing a trained motor task that required readiness for proper usage of sensory inputs. Thirty-two neurons exhibited activity changes, which supports the hypothesis that the SMA is part of the system involved in modulating responsiveness of the motor cortex to sensory inputs in association with learned movements.

Relationships among differentiated T-cell subpopulations. III. Radioresistance of delayed hypersensitivity to heterologous erythrocytes.

Effects of whole body irradiation with 600 rad on delayed hypersensitivity, direct cytotoxicity and antibody production were examined in mice immunized with chicken erythrocytes (CRBC) or sheep erythrocytes (SRBC) in saline. Delayed footpad reactions to CRBC were not affected by the exposure to irradiation 3 h before or after antigenic stimulation. On the other hand, direct cytotoxic activities in spleen cells and antibody titres to CRBC were reduced by such exposures. Additionally, delayed footpad reactions to SRBC were not affected by the exposure to irradiation. Generation of effector cells responsible for delayed footpad reactions proved to be resistant to irradiation.
