Sequential passages of human rotavirus in MA-104 cells.

Starting with a small amount of diarrheal feces containing human rotavirus (HRV), we succeeded in propagation of the virus using the roller culture technique with MA-104 cells. Furthermore, we made a successful adaptation of HRV to a stationary culture and developed a plaque assay for the cell culture-adapted viruses. The 3 culture-adapted virus isolates, KU, YO, and 44 produced plaques (about 0.5-1.0 mm in diameter) under the overlay medium consisting of 0.6% purified agar, 3 micrograms of acetyl trypsin/ml and 50 micrograms of DEAE-dextran/ml. Subsequent plaque purification resulted in the formation of clear, larger plaques. It was shown from the results of cross neutralization tests using the fluorescent focus reduction method that the three culture-adapted HRV isolates were clearly different antigenically from ovine rotavirus (NCDV) and, further, that a noticeable difference in antigenicity also existed among the HRV isolates.

The secondary cytotoxic response to allogeneic cells: the specificity of secondary cytotoxic effectors differs from that of primary effectors.

Following priming with alloantigen in vivo, a secondary cytotoxic activity of AKR mice (H-2k) was generated by in vitro stimulation with the same alloantigen. Such secondary effectors could lyse not only targets of a sensitizing haplotype but also third-party targets unrelated to sensitizing or responding cells. Such cross-reactive cytotoxicity of AKR secondary effectors were demonstrated not only by stimulation with C57BL/6 (H-2b) cells but also with C3H/He (H-2k) cells. It was observed that such cross-reactive secondary cytotoxicity was mediated by T lymphocytes, and required protein synthesis in secondary culture. These results suggested the possibility that the antigen receptors on such secondary cytotoxic effector cells have different recognitive specificity from that on primary cytotoxic effector cells.

Analysis of differentiated T-cell subpopulations: difference in radiosensitivity of immune responses to allogeneic antigen.

The radiosensitivity of immune responses to allogeneic tumor cells was compared. Delayed hypersensitivity as demonstrated by macrophage migration inhibition and antibody production as detected by immune adherence test were extremely radiosensitive. The radiosensitive nature of macrophage migration inhibition was in marked contrast to he radioresistance of the delayed footpad reaction. The generation of cytotoxicity was relatively radioresistant.

Analysis of elements determining the generation of cytotoxic T lymphocytes. I. Irradiated F1 recipients in which antigenic stimulation occurs consistently at an excess dose.

Specific cytotoxicity against histocompatibility antigens of a parental strain was induced in the spleens of sublethally irradiated F1 mice after intravenous transfer of lymphoid cells from another parental strain. Such cytotoxicity reached the highest level at 10 days after the transfer of 3 to 5 X 10(7) spleen cells and was ascribed to be T lymphocytes of donor parental origin. Secondary response of the generation of cytotoxicity was compared among in vivo rechallenge, in vitro MLC and transfer to irradiated F1 mice. Cytotoxicity was not so augmented by in vivo rechallenge. It may be attributable to early elimination of grafts. In in vitro MLC and transfer to irradiated F1 mice, the generation of cytotoxicity appeared earlier with higher magnitudes as compared to primary response. The transfer system to irradiated F1 recipients appears to be more suitable for analyses of the generation of cytotoxic T lymphocytes from memory cells than in vivo rechallenge.

Diffuse leiomyomatosis of the esophagus.

A case of diffuse leiomyomatosis of the esophagus which was successfully resected is presented. The patient was a 44-year-old man who had an abnormal filling defect, which was incidentally discovered during esophagography, and was observed for two years. Although he remained asymptomatic, an operation was performed because of increase in the tumor's size. The lesion consisted of many confluent myomatous nodules throughout the thoracic portion of the esophagus, mostly seen in the circular layer of the muscularis, and some in the muscularis mucosa. Histologically, these nodules were leiomyomas. This is a very rare condition, and only a dozen similar cases have been reported so far. A brief discussion is made on the definition of diffuse leiomyomatosis of the esophagus.

Effect of anaesthesia and surgery on endocrine function in elderly patients.

The effect of halothane-nitrous oxide anaesthesia and abdominal surgery on endocrine function was investigated in 18 elderly patients whose ages ranged from 65 to 77 (mean 69 years). Cortisol levels in plasma increased significantly during anaesthesia and surgery, and the increases were slightly higher than those in young adults. Plasma aldosterone increased and insulin levels did not change during the procedures, and these levels were lower than in younger adults. The endocrine responses in elderly patients were similar in magnitude to those in young adults. Therefore it appears that no special hormonal treatment is necessary for elderly patients undergoing routine surgical procedures.

ATP dependent reversible conformational change of Na+,K+-ATPase modified with N-(p-(2-benzimidazoly)phenyl)maleimide.

Na+,K+-ATPase [EC 3.6.1.3] from pig kidneys was treated with the fluorescent reagent N-(p-(2-benzimidazolyl)phenyl)maleimide (BIPM) in the presence of CKl. The resultant preparation showed 70% of the activity with only a small change in the apparent affinity for ligands of the enzyme. The addition of Na+ to the treated preparation induced a -2.1 +/- 0.1% change of the total fluorescence intensity observed in the absence of Na+. Further addition of both Mg2+ and ATP transiently increased the fluorescence to +0.5 +/- 0.1%. After the exhaustion of ATP, the fluorescence decreased to -3.1 +/- 0.1%. This cycle can be repeated by the readdition of ATP but not by ADP. Ouabain inhibits the fluorescence change. The ligands used reduced the fluorescence intensity as follows: Mg2+ + Na+ + ATP approximately K+, none, Mg2+ approximately ATP, Na+ + ATP, Na+ approximately Na+ + Mg2+, Na+ + Mg2+ + ADP approximately Na+ + ADP. The data indicate the presence of multiple conformational states of the enzyme.

Supplementary motor area: neuronal response to motor instructions.

1. Single-unit recordings were obtained from the supplementary motor area of the cerebral cortex of two monkeys during execution of learned movements 2. Monkeys were required to push or pull a cast attached to the right forelimb in response to a sudden perturbation delivered via the cast. An instruction as to the direction of the monkey's movement was delivered 2.5--5 s prior to the occurrence of the perturbation and correct performance, therefore, required the animal to develop a preparatory state prior to the perturbation. 3. Of many hundreds of neurons recorded, 201 exhibited instruction-induced changes of activity during the period intervening between the instruction and the perturbation-triggered movement. 4. In 94 neurons, effects of the instruction were differential depending on which of the two instructions was given, whereas in 107 neurons, effects were nondifferential. The latencies of the differential responses appeared to be shorter (starting as early as 140 ms after the instruction). 5. The magnitude of the instruction effects varied in parallel with development of enhanced motor skill as the monkeys gained more experience in responding to the triggering stimulus. 6. These observations substantiate the hypothesis that the supplementary motor area plays a part in modifying a sensory-triggered motor output.

The influence of low frequency acupuncture on a demand pacemaker.

Various complications with demand pacemakers have been known to originate from their own sensing mechanisms. In these complications, electromagnetic interference is one of the well known causes of oversensing with demand pacemakers. The influence of low frequency acupuncture as a specific type of electromagnetic interference on a demand pacemaker was examined during a cervical operation. It was found that low frequency acupuncture is a cause of electromagnetic interference with demand pacemakers. Therefore, caution should be exercised when using the low frequency acupuncture on a pacemaker patient.

Differences in thymus-dependency among various T-cell functions.

Effects of thymectomy of various times after birth on cytotoxicity, delayed footpad reaction and antibody production were examined in 6 week old mice immunized intraperitoneally with chicken erythrocytes. Cytotoxicity and antibody production were abolished by thymectomy within 3 days after birth. Mice thymectomized at 5 days of age or later showed appreciable degrees of cytotoxicity and antibody production. On the other hand, a delayed footpad reaction could be elicited even in mice thymetocmized within 24 h after birth, although the degree of the reaction was reduced by such a treatment. Therefore, T cells responsible for delayed hypersensitivity were less thymus-dependent and required the presence of the thymus for a shorter period in their development that those responsible for cytotoxicity or antibody production.
