Severe erosive stomatitis and giant lymph node hyperplasia of retroperitoneum (Castleman's tumor).

A severe, extensive, erosive stomatitis developed in a 33-year-old Japanese man who had had intraperitoneal giant lymph node hyperplasia (Castleman's tumor) of the hyaline-vascular type. Histologically, there was a dense lymphoid cell infiltration in the dermis with invasion of the mucosal epithelium. It was hypothesized that an autoaggressive attack of lymphoid cells against the epithelium of the oral mucosa played a role in the pathogenesis of this erosive stomatitis and it was suggested that there might be a correlation between the occurrence of stomatitis and the presence of Castleman's tumor.

Direct release of parathyroid hormone fragments from functioning bovine parathyroid glands in vitro.

To determine the origin of circulating parathyroid hormone fragments, hormonal peptides released from bovine parathyroid tissue in a physiologically responsive in vitro "perifusion" system were analyzed by gel exclusion chromatography and region-specific radioimmunoassays. When exposed to low Ca++, the tissue released large quantities of intact hormone (parathyroid hormone 1--84) as well as amino- and carboxyl-terminal fragments. Fragments of the hormone were also released when the tissue was exposed to high Ca++, but the carboxyl fragments comprised a much greater proportion of the hormonal peptides released. Control experiments indicated that fragmentation of the hormone occurred within the gland and not after it was secreted. These experiments provide direct evidence, therefore, that release of fragments from the parathyroid gland may contribute to the immunologic heterogeneity of the hormone in the circulation.

Pokeweed mitogen-induced B cell differentiation in idiopathic aplastic anemia associated with hypogammaglobulinemia.

Peripheral blood lymphocytes from 14 patients with idiopathic aplastic anemia were examined to determine their capacity to differentiate into immunoglobulin (lg) producing cells in the presence of pokeweed mitogen (PWM). Four patients had hypogammaglobulinemia. Lymphocytes from ten patients without and one with hypogammaglobulinemia were capable of differentiating into lg-producing cells. Lymphocytes from three patients with hypogammaglobulinemia, however, were unable to differentiate. Coculture experiments showed that in one patient helper activity of T cells was impaired, in another patient B cells could not be stimulated by PWM to differentiate even in the presence of normal T cells, and in a third patient both the helper function of T cells and the capacity of B cells to differentiate into lg-producing cells were affected. These results suggest that differentiation of lymphoid cells is impaired in some cases of idiopathic aplastic anemia associated with hypogammaglobulinemia.

RNA-reverse transcriptase complex from cultured human myeloma-leukemia cells.

A high molecular weight RNA-reverse transcriptase complex in the culture media of peripheral leukocytes obtained from two Japanese patients with myeloma-leukemia was detected by demonstration of a 3H-uridine peak and a peak of DNA polymerizing activity banding at a density of 1.15-1.19g/ml. The enzyme in the complex was able to utilize poly(rA)-d(pT)10 or poly (rC)-d(pG) 12-18, but not poly (dA)-d(pT) 10 or (dT) 12-18 as template-primers. The sucrose density sedimentation analysis revealed that RNA in the complex sedimented at a location of approximately 50s and 20-30s.
