Immunoregulatory function and expression of OKT17 antigen of adult T-cell leukemia cells.

Research was carried out on neoplastic T-cells taken from 10 cases of Japanese adult T-cell leukemia (ATL) in regard to their immunoregulatory activity on pokeweed mitogen (PWM)-driven immunoglobulin (Ig) production and their differentiation antigens. ATL cells were reactive with OKT3, OKT4 and OKT11 in most cases; however, the ATL cells from one case did not react with OKT11 nor formed E-rosettes. ATL cells of three cases were reactive with OKT8 as well as OKT4. In six cases, the ATL cells were reactive with OKT17 and coincidentally displayed suppressor activity on Ig production. The cells of the remaining four cases did not have such reactivity with OKT17 or suppressor activity. Moreover, the OKT17(+) ATL cells from one case sorted by FACS 440 demonstrated suppressor activity, while the OKT17(-) ATL cells did not. These results indicated that ATL cells with suppressor activity had OKT3(+), OKT4(+), OKT17(+) phenotype.

Diverse properties of external and internal forms of yeast invertase derived from the same gene.

It has been shown by genetic analysis that the external and internal invertases from Saccharomyces cerevisiae share a common structural gene [Taussig, R., & Carlson, M. (1983) Nucleic Acids Res. 11, 1943-1954]. However, the only amino acid composition of these two forms of invertase reported to date has revealed extensive differences [Gascon, S., Neumann, N.P., & Lampen, J.O. (1968) J. Biol. Chem. 243, 1573-1577]. We have found from amino acid analyses of both enzymes and sodium dodecyl sulfate-polyacrylamide gel analysis of their cyanogen bromide peptides that they are most likely identical in their amino acid sequence. However, the invertases exhibit dramatically different physical properties, particularly in their stability. The most striking difference was in their renaturation following guanidine treatment where it was shown that inactivated external invertase could be renatured completely. Endo-beta-N-acetylglucosaminidase H treated external invertase was restored to 40% of its original activity while internal invertase remained completely inactive. The observed differences may be attributed to the presence and absence of the oligosaccharide moiety in the external and internal invertases, respectively.

Exploration of the pituitary stalk and gland by high-resolution computed tomography. Comparative study of normal subjects and cases with microadenoma.

By means of high-resolution CT, pituitary stalks and glands were demonstrated in 189 normal subjects and in six patients with microadenomas. In the horizontal view and in reconstructed coronal and sagittal projections, the normal stalks and glands showed homogeneous enhancement with the contrast medium. On metrizamide CT, the stalks and glands were demonstrated in high-density areas as a "defect". The detection-rate of the stalks and glands was more than 93.6% in all dimensions on enhanced CT and 100% on metrizamide CT. On reconstructed sagittal projections, there was a tendency for the glands to decrease in size with advancing age. Under the age of 29, especially in females, most of the glands were demonstrated as an oval area that filled the sella turcica. After the age of 50, the gland frequently was flattened with enlargement of the CSF space within the sella turcica, giving an appearance reminiscent of the "empty sella". Of six patients with microadenomas, five presented the appearance of oval gland, and one showed visualization of CSF space in the anterosuperior portion of the sella turcica. On reconstructed coronal projections, the angle of inclination of stalks was 1.5 +/- 1.2 degrees in normal subjects and 9.3 +/- 2.4 degrees in patients with microadenomas.

Differentiation of human pulmonary alveolar epithelial cells revealed by peroxisome changes in pulmonary proteinosis.

Regenerating areas of human lungs in pulmonary fibrosis were observed electron microscopically, and peroxidatic activity of catalase in lung peroxisomes were demonstrated cytochemically. Proliferation of Type II cells was prominent there, and some of the cells extended their cytoplasms to cover the denuded basement membrane. Unusual intermediate cells between Type II and Type I cells were observed. The extension of cytoplasmic processes with new generation of pinocytotic vesicles strongly suggested a Type I cell profile. However, catalase-positive peroxisomes were found in these cells simultaneously. From these results it was concluded that Type I cells may originate from Type II cells in human lungs as they do in experimental animals.

[Cerebellar cyst associated with cytoarchitectonic abnormalities in the cerebellar cortex].

The patient, 64-year-old female, had episode of sudden attack of severe vertigo, headache, nausea, and vomiting which lasted for about twenty minutes on May 20th in 1980. She had hypertension, polyp of stomach, diverticuli of duodenum in her past history. Neurological examination on her admission revealed fine horizontal nystagmus on bilateral gaze and slight clumsy movement on left F-N test. On plain skull and cervical X-P, atlanto-occipital fusion and Klippel-Feil syndrome (C2-C3 fusion) were seen. Plain CT scanning revealed a large cystic lesion which extended from the vermis to the left cerebellar hemisphere. No enhanced area was seen. The forth ventricle was seemed to be enlarged. And the left-sided dorsal part of the forth ventricle attached to the cyst. Metrizamide CT cisternogram showed there was no direct communication between them. Angiographically, the vertebrobasilar arteries were noted sclerotic changes and poor vascularities in the left cerebellar hemisphere was noted. On opening the dura during surgery, the left cerebellar hemisphere appeared bulging state and the bilateral cerebellar tonsils were hypoplastic. Outer thin membrane of the cyst was removed. The cyst has no communication with the subarachnoid space as well as with the forth ventricle. The cystic fluid was slightly yellowish, but had no Froin's sign. Reddish-gray color nodular area, which seemed to be similar to mural nodule macroscopically, was noted in the area of inner surface of the cyst. This part was removed. Histological findings of this area showed abnormal architecture with malarranged layer of cerebellar cortex.(ABSTRACT TRUNCATED AT 250 WORDS)

[Surgical management of peripheral hypoglossal nerve palsy caused by abnormal external carotid artery].

It is known that there are a number of variation and anomalies in the bifurcation of the external and internal carotid arteries. It has also been reported that the lateral position of the external carotid artery, which was formerly considered extremely rare, is not so unusual. However, there are few reports that the lateral position of the external carotid artery has caused neurological signs. In this paper, an unusual case of peripheral hypoglossal nerve palsy, attributed to the vascular compression by lateral position of the external carotid artery and abnormally high position of the bifurcation of the external and internal carotid artery was reported. A forty-nine year old man was admitted complaining of tongue deviation and dysarthria. Atrophy and fasciculation of the right lingual muscle were observed. In a right carotid angiogram, lateral position of the external carotid artery was noticed and the bifurcation of the external and internal carotid arteries was abnormally high (upper margin of C2 vertebra). The proximal portions of both arteries were very tortuous and dilated like a megadolichoartery. A right neck surgery was performed and the bifurcation of the external and internal carotid artery was exposed. The hypoglossal nerve was running just above the bifurcation. Owing to the lateral position of the external carotid artery and the tortuous dilatation of the proximal portion of the external and internal carotid arteries, the hypoglossal nerve was markedly compressed and extended laterally. The external carotid artery was ligated and cut at its origin. Inflammatory lymph nodes near the bifurcation were also removed.(ABSTRACT TRUNCATED AT 250 WORDS)
