Origin of lingual proprioceptive afferents in Japanese monkey, M. fuscata fuscata. Studied by HRP-labeling technique.

The peripheral course of the axon of the lingual proprioceptive neurons in 5 Japanese monkeys was studied by injecting HRP into the distal part of the hypoglossal nerve through the proximal stump. The present authors confirmed that the HRP-labeled cell bodies appeared ipsilaterally in the C2, C3 and C4 spinal ganglia. The afferents to the C2, C3 and C4 ganglia pass through the ansa hypoglossi and direct neuronal anastomosis existing between the first cervical nerve and the hypoglossal nerve. It was confirmed that in the Japanese monkey, having abundant muscle spindles in the tongue, the lingual proprioceptive neuron soma exist in the C2, C3 and C4 spinal ganglia, their peripheral axons passing through the ansa hypoglossi and the direct neuronal anastomosis.

[Acute myocarditis with localized left ventricular aneurysm: a report of three cases].

Ventricular aneurysms are rarely observed in viral myocarditis. Three cases whose left ventriculograms showed localized left ventricular aneurysms in the chronic phase of myocarditis are reported. The etiology in one case was herpes simplex virus (Case 1). Two cases (Case 2, 3) of myocarditis were admitted to our Coronary Care Unit in the acute phase, when diffuse hypokinesis of the left ventricle was demonstrated by two-dimensional (2-D) echocardiography. Hypokineses progressed to localized left ventricular aneurysm formation, demonstrated by cine angiography. In the acute phase, ST segment elevation was observed in these two cases, but it resolved. Abnormal Q waves also resolved in the chronic phase. Negative T waves were nearly normalized in one of them (Case 3). Abnormal Q waves with ST segment depression were observed in another case (Case 1). Thus, there were no characteristic or consistent findings suggesting a left ventricular aneurysm on electrocardiography. 2-D echocardiography and cine angiography proved useful for diagnosing this uncommon complication. Long-term follow-up of these cases will be important, because viral myocarditis can develop into dilated cardiomyopathy. The mechanism of left ventricular aneurysm following acute viral myocarditis included: (1) direct viral injury of the myocardium, (2) localized injury due to immunological mechanisms, and (3) coronary thrombosis due to increased platelet aggregation by viral infection.

Monoclonal antibodies against human oral squamous cell carcinoma reacting with keratin proteins.

Two mouse monoclonal antibodies (MoAbs), B10 and 1H5, were generated by fusing mouse myeloma NS-1 cells with spleen cells from a BALB/c mouse immunized with Ueda-1 cells derived from human squamous cell carcinoma (SCC) of the floor of the mouth. Immunohistochemical analysis revealed that these MoAbs recognize the filamentous components of cytoplasm which were protein in nature. While the pattern of antigen distribution in various cell lines was not cell-type specific, reactivity of these antibodies with tissue sections was informative. MoAb 1H5 was preferentially reactive with well-differentiated squamous cell carcinoma, however, reaction with adenocarcinoma was observed infrequently. This antibody also preferentially reacted with the spinous layer of normal stratified squamous epithelium. MoAb B10, however, was reactive with nonepithelial tissues as well as with epithelial ones, and its level of binding bore no relationship to the grade of histologic malignancy. SDS-PAGE and Western blotting analysis, using cytokeratin extracts of Ueda-1 cells and human epidermis, demonstrated that MoAb B10 reacted with a wide range of keratin proteins of 65-67K, 58K, 56.5K, 56K, 52K, 50K, 48K, 45K, 40K, 38K, 36K, and 34K molecular weight (MW), while MoAb 1H5 reacted with keratin proteins of 65-67K, 58K, 56.5K, 56K, 52K, 48K, and 34K MW. These results suggest that MoAb 1H5 may recognize keratin subfamilies related to squamous differentiation, whereas MoAb B10 recognizes a wide range of keratin proteins, and may even react with other kinds of intermediate filament proteins (IFPs).

Isolation and characterization of ornitho-kininogen.

Ornitho-kininogen was purified from chicken blood plasma by a two-stage method using chromatography on columns of S-alkylated papain-Cellulofine and DEAE-5PW. The yield was 1.7 mg from 44 ml plasma. The isolated preparation gave a single band on sodium dodecyl sulfate/polyacrylamide gel electrophoresis (SDS-PAGE) with or without 2-mercaptoethanol and on disc/polyacrylamide gel electrophoresis. The relative molecular mass, Mr, of ornitho-kininogen was estimated as 74,000 on SDS-PAGE using the Ferguson plot method. Ornitho-kininogen was found to have the similar properties to those of mammalian high-Mr kininogen, in terms of the amino acid composition, molecular mass, and susceptibility to plasma kallikrein. No kininogen corresponding to mammalian low-Mr kininogen and rat T-kininogen could be detected in chicken plasma. In fact, ornitho-kininogen was degraded rapidly by bovine plasma kallikrein, liberating a kinin. This kinin was isolated from the digest by reversed-phase HPLC. The primary structure of the isolated kinin was determined as Arg1-Pro2-Pro3-Gly4-Phe5-Thr6-Pro7-Leu8-Arg9. The sequence of this peptide, named ornitho-kinin, was similar to that of bradykinin except for the substitution of Thr6 and Leu8 for Ser6 and Phe8. The isolated ornitho-kinin induced a contraction of chicken smooth muscle and had a strong hypotensive effect in the chicken. However, it did not contract the isolated rat uterus. It is suggested that this specificity difference is due to the replacement of Phe8 by Leu8. The sequence of residues 1-30 of ornitho-kininogen exhibited 43% identity with that of bovine kininogen.

A membrane-bound, calcium-dependent protease in yeast alpha-cell cleaving on the carboxyl side of paired basic residues.

Paired basic residues are known as a typical site for proteolytic processing of precursors of bioactive peptides. By using a fluorogenic substrate Boc-Gln-Arg-Arg-MCA, a unique endoprotease exhibiting hydrolytic specificity toward the carboxyl side of paired basic residues was partially purified (about 4600-fold) from the membrane fraction of yeast Saccharomyces cerevisiae alpha-cells. The enzyme is a calcium-dependent thiol protease, with optimal pH at 7.0. It is a glycoprotein, with an apparent molecular weight of about 100,000-120,000. It cleaves fluorogenic substrates and a synthetic model peptide at the carboxyl side of paired basic residues. From its unique substrate specificity, this enzyme may be involved in precursor processing in vivo.

Cortical dysplasia in a 23-week fetus with Fukuyama congenital muscular dystrophy (FCMD).

A 23-week fetus who is thought to be affected with Fukuyama congenital muscular dystrophy (FCMD) is reported. Cortical dysplasia of the cerebrum was extensive and could be categorized into three major types. The cerebral cortex was thoroughly covered by glio-mesenchymal tissue (extracortical glial layer), in which neuronal clusters were irregularly scattered. Radial bundles of neuroglial tissue frequently extended from the cortex into the extra-cortical glial layer through the focally defective molecular layer and pia mater. The deep cerebral structures, such as basal ganglia, thalamus and white matter, appeared normal in contrast with extensive malformation in the cortex. Glial fibrillary acidic protein-immunoperoxidase stain revealed: (1) presence of abundant radial glial fibers in the ventricular, subventricular and intermediate zones; (2) focal or diffuse lack of glia limitans; (3) focal derangement of radial glial fibers; and (4) proliferation of stellate glial cells in the extra-cortical layer. It is suggested that ectopic accumulation of neurons into the extra-cortical glial layer seems a cardinal pathogenetic process to generate cortical dysplasia in FCMD. Early development of superficial glio-mesenchymal tissue seems essential for upward displacement of migrating neurons.

Prostaglandin synthesis by murine salivary glands.

When submandibular-gland homogenate was incubated with radioactive arachidonic acid, three radioactive metabolites were detected by thin-layer chromatography; these were identical with the prostaglandins (PG) PGE2, PGF2 alpha and PGI2. Thromboxane A2 (TXA2) and PGD2 were not found. The major cyclo-oxygenase products of the parotid and sublingual glands were also PGE2, PGF2 alpha, and PGI2. The PGE2 and PGI2 synthetic activities of the submandibular gland were the highest of any tissue examined. This PGI2 activity was about 1.5 times higher in male than in female mice.
