Thermal damage threshold of brain tissue--histological study of heated normal monkey brains.

The thermal damage threshold of normal brain tissue was evaluated from immediate and delayed histological changes caused by hyperthermia treatment of normal monkey (Macaca fuscata) brains. A 2450 MHz microwave antenna and an antenna cooling system devised by our group were used for interstitial hyperthermia treatment. The antenna within the cooling system was inserted through a small craniectomy under general anesthesia. The temperature at a reference point, 4 mm radially away from the surface of the cooling system, was maintained at 42, 43, 44, 45, or 46 degrees C for 60 minutes. Eighteen animals were treated and sacrificed immediately after the treatment, while nine animals were treated and sacrificed 7 days after the treatment. The histological changes were studied microscopically on sections stained with HE or Kluver-Barrera's method. The non-survival experiment demonstrated that areas heated at 44 degrees C or below showed no obvious irreversible changes. The survival experiment showed areas heated at 44 degrees C or above developed coagulative necrosis. These histological findings indicate that thermal damage occurs in normal brain tissue after heating at 44 degrees C or above for 60 minutes, suggesting that the safety limit for brain hyperthermia is 43 degrees C for 60 minutes.

Multiple malignant fibrous histiocytoma of bone. A case report.

A 27-year-old man had tumors in both the femoral and the tibial metaphyses of the left knee. Although histological examination of the biopsy specimens revealed different findings in the 2 lesions, the final diagnosis was malignant fibrous histiocytoma in both.

[Ultrastructural consequence of shock wave exposure on VX2 cancer implanted to the urinary bladder].

In order to clarify the ultrastructural consequence of the VX2 cancer cell exposed to underwater shock waves. The VX2 cancer implanted to the urinary bladder of rabbit was received the shock waves (1000 shots, 100 MPa, 5/sec.) generated by the piezo-ceramics. Damage to the cancer cell were manifested by the three kind of findings: the one was the cell junction failure which caused the isolated cells; the second was the destruction of organelle which seemed to form the vacuole in cytoplasm; and the last finding was the emergence of moth eaten appearance in the exposed cell. We conclude that underwater shock wave exposure induces lethal injury in VX2 implanted cancer.

[Efficacy of benzimidazole-type drugs on alveolar echinococcosis (AE)].

AE patients can be cured only by complete excision of the lesions. In Europe and the United States, however, benzimidazole-type drugs have been reported to be effective. We treated 37 patients (20 males and 17 females, mean age of 45.9 years) with mebendazole (MBZ) (n = 17) or albendazole (ABZ) (n = 20) according to the WHO chemotherapy protocol, and compared adverse reactions, drug concentrations, serological parameters, and measurable lesions by CT scan and/or X-ray examination. In the MBZ group, serological parameters were unaffected. One patient each had hepatic disorder and alopecia (12%), and 5 lesions in the liver and 3 in the lung gradually increased in size (105% in 1 year, 112% in 2 years), and showed NC (no response) in 6, PD (progressive disease) in 2. One case of released biliary obstruction were judged as PR (partial response) (Response rate: 11.1%). In the ABZ group, no adverse reactions were found and ELISA level decreased significantly in 10 out of 12 patients. One lesion in the liver, 3 in the lung, 1 in the chest wall and 1 in the retropancreatic region reduced in size significantly (Mean: 56%) (PR 3, NC 3; Response rate: 50.0%). ABZ was safer and more effective than MBZ.

[A case of capillary hemangioma of the testis in a child].

Testicular tumors derived from connective tissue, blood vessels and musculature are uncommon and intratesticular tumors of vascular origin are extremely rare. A rare case of capillary hemangioma of the testis in a child is reported. An 11-year-old boy was admitted with the chief complaint of painless mass in the right scrotum. He was diagnosed with a right testicular tumor by physical examination and ultrasonography, preoperatively. Right high orchiectomy was done on December 10, 1990. The testis measured 2.5 x 2.3 x 1.5 cm. Pathologically, the tumor was diagnosed as a capillary hemangioma of the testis.

Clinical usefulness of computer-assisted diagnosis using combination assay of tumor markers for pancreatic carcinoma.

BACKGROUND: There has been extensive use of serum tumor markers in diagnosing pancreatic adenocarcinoma. There is no tumor marker, however, that alone has sufficient diagnostic accuracy. It is necessary to know which combination of tumor markers should be used to detect pancreatic cancer, with respect to clinical usefulness and cost effectiveness. METHODS: Serum levels of 17 kinds of tumor markers were determined in 145 patients and 40 healthy volunteers. Thirty-five patients with proven pancreatic adenocarcinoma, and 32 with benign pancreatobiliary disease (14 chronic pancreatitis and 18 biliary stones) were selected. For analysis of the usefulness of each tumor marker to differentiate these two groups, scatterplot and relative operating characteristic (ROC) analyses were used. A multivariate discriminant system to differentiate these two groups was developed using stepwise discriminant analysis by backward elimination selection. RESULTS: The significance of each tumor marker varied according to the tumor volume. By ROC analysis, the markers were divided into four subgroups according to their usefulness in discriminating pancreatic adenocarcinoma from benign pancreatobiliary disease. A discriminant system consisting of two different discriminant functions using nine tumor markers (CA 19-9, DUPAN-2, TPA, elastase-1, lipase, amylase, gamma-glutamyl transpeptidase, alkaline phosphatase, and lactate dehydrogenase) was developed and designated CAMPAS-P; it could differentiate between all 35 cases of pancreatic adenocarcinoma and 32 cases of benign pancreatobiliary disease. On the other hand, CAMPAS-P showed a low positive rate in pancreatic tumors of unusual histologic type, and poor organ-specific diagnostic ability in various digestive organ malignancies. CONCLUSIONS: CAMPAS-P may be very useful for differential diagnosis between pancreatic adenocarcinoma and benign pancreatobiliary disease.

Effects of morphine on release of acetylcholine in the rat striatum: an in vivo microdialysis study.

We examined the effect of morphine on the release of acetylcholine (ACh) in the striatum of freely moving rats using the in vivo microdialysis method. The basal level of ACh was 3.01 +/- 0.51 pmol/30 microliters/15 min in the presence of neostigmine (10 microM). Tetrodotoxin (1 microM), a selective blocker of voltage-dependent Na+ channels, rapidly decreased the release of ACh in the striatal perfusates. Morphine at a dose of 10 mg/kg (i.p.) caused a reduction of ACh release in the striatum at 90-150 min. However, a lower dose of morphine (5 mg/kg, i.p.) did not affect ACh release in the striatum. The reduction following intraperitoneal administration of morphine was abolished by naloxone (1.0 mg/kg). After microinjection of the neurotoxin 6-hydroxydopamine (6 micrograms/3 microliters, 7 days before) in the substantia nigra, the morphine (10 mg/kg)-induced decrease of ACh was attenuated, and a similar result occurred following reserpine (2 mg/kg, i.p.) 24 h before combined with alpha-methyl-p-tyrosine (300 mg/kg, i.p.) 2.5 h before. These findings indicate that morphine exerts an inhibitory influence on striatal ACh release in freely moving rats and that this inhibitory effect is mediated by the nigro-striatal dopaminergic system.

Polyostotic lesions compatible with osteofibrous dysplasia. A case report.

An 18-month-old girl with lesions in the cortex of the bilateral tibiae and ulnae and right fibula is reported. The lesion in the right tibia disappeared after curettage and xenogeneic bone grafting, and the other lesions disappeared spontaneously by the age of 12 years. All lesions involved the bone cortex. The right tibia lesion exhibited the histopathological features of "zonal architecture" and a osteoblast rim formation around the trabeculae. We consider that osteofibrous dysplasia can involve systemically any long bone.

Newly established human pancreatic carcinoma cell lines and their lectin binding properties.

Two human pancreatic carcinoma cell lines, designated HuP-T3 and HuP-T4, were established from the ascites of pancreatic cancer patients with carcinomatous peritonitis. The cell lines were grown in monolayer cultures and had population doubling times of 38.6 and 37.1 h, respectively. HuP-T4 secreted large amounts of CEA and CA19-9 into the medium, and HuP-T3 produced a small amount of CEA, but no CA19-9. Both cell lines showed tumorigenicity in nude mice. Histologically, the HuP-T3-derived tumor was poorly differentiated adenocarcinoma, and the HuP-T4-derived tumor was well differentiated papillotubular adenocarcinoma. In lectin histochemistry at both light and electron microscopic levels, the most striking difference between the lectin binding properties of the two cell lines and those of control normal pancreatic ductal cells was that soybean agglutinin (SBA) bound to both cell lines, but not to controls. These newly established cell lines should be useful models that will contribute to clarifying the biological and biochemical characteristics of pancreatic cancer.

An investigation into the effects of apomorphine on the release of acetylcholine in the striatum of freely-moving rat using in vivo microdialysis.

1. Using in vivo microdialysis, we studied the effects of apomorphine on the release of acetylcholine (ACh) in the striatum of freely-moving rats. 2. Basal release of ACh was 3.01 +/- 0.51 pmol/15 min in 30 microliters (containing of 10 microM neostigmine) of the striatal perfusate. 3. Ringer containing tetrodotoxin (1 microM), hemicholinium-3 (5 mM), or Ca(2+)-free Ringer was perfused into the striatum. Each of these treatments produced a significant attenuation in release content of ACh. High K+ (30 mM) produced a significant increase in ACh release. 4. In normal rats, apomorphine (0.05 mg/kg, s.c.) increased striatal ACh release. A high dose (1.0 mg/kg) significantly attenuated release. 5. In 6-OHDA-pretreated rats, although the lower dose of apomorphine did not increase striatal ACh release, the high dose produced a more significant attenuation of release. 6. It is concluded that apomorphine enhanced or attenuated the striatal ACh release and that this effect was regulated by nigro-striatal dopaminergic system in the freely moving rats.

Serotonergic, cholinergic and nociceptive inhibition or excitation of raphe magnus neurons in barbiturate-anesthetized rats.

Neurons in the nucleus raphe magnus were recorded extracellularly from barbiturate-anesthetized rats, and were classified by their responses to noxious mechanical stimulation as either pinch-excited, pinch-inhibited or biphasic (inhibited then excited). They were then subjected to iontophoresis of serotonin, some serotonergic agonists and antagonists, acetylcholine, and gamma-amino-n-butyric acid. Serotonin reduced the spontaneous firing of most pinch-inhibited cells (79%). Significantly fewer (P < 0.05) pinch-excited and biphasic cells were inhibited by serotonin (40% and 45%, respectively); in these two cell classes, the observed response was often excitation (30% and 14%), or inhibition for 10-30s followed by excitation for the next 1-2 min (25% and 36%). Acetylcholine showed a similar, statistically significant distribution of effects (P < 0.05), inhibiting all pinch-inhibited neurons (n = 10) but fewer pinch-excited (53%, n = 17) and biphasic neurons (20%, n = 10). Excitation, or excitation then inhibition, was again found frequently among the remaining pinch-excited and biphasic cells. The effect of gamma-amino-n-butyric acid was only inhibitory. In all three nociceptive classes, the serotonin-1A agonist buspirone (n = 15) was inhibitory (87%) and the serotonin-1C/2 antagonist ketanserin (n = 20) was excitatory (35%). The mixed serotonin-1/2 antagonist methysergide (n = 10) was inhibitory (50%) or excitatory (40%). 8-Hydroxy-dipropylaminotetralin (n = 3) was found to increase spontaneous activity (possibly because of partial serotonin-1A agonsim), and +/- propranolol (n = 4) to reduce it (possibly through beta-adrenoceptor antagonism, not serotonin-1A antagonism).(ABSTRACT TRUNCATED AT 250 WORDS)

Purification of hemagglutinin from Haemophilus paragallinarum using monoclonal antibody.

The purification of hemagglutinin from Haemophilus paragallinarum was attempted using affinity chromatography with monoclonal antibody. The antigen eluted from the affinity column using potassium thiocyanate buffer agglutinated chicken erythrocytes. In immunoblotting of the eluted antigen, a single band with monoclonal antibody was found as well as the crude antigen. When the chickens were immunized with the eluted antigen, they produced the hemagglutination inhibition (HI) antibody, and they showed protection against challenged exposure with H. paragallinarum strain 221. These results indicated that the HA antigen of H. paragallinarum was a protective antigen.

Bilateral carcinomas of the maxillary sinus.

We report a 68-year-old male with a rare case of synchronous bilateral carcinomas of the maxillary sinus. A CT scan revealed a large tumour mass that extruded from the left maxillary sinus; tissue of soft density filled the right antrum which had intact bony walls. A probe antrostomy on the right side disclosed a tumour which was diagnosed histologically as the same poorly differentiated squamous cell carcinoma as that in the left antrum. The incidence and aetiology of this disorder are presented, and its diagnosis and management are discussed.

Spindle cell hemangioendothelioma: a report of two cases.

Two cases of spindle cell hemangioendothelioma (SCH) are reported. One of the patients was a 16 year old Japanese female, who had been suffering from Ollier's disease (multiple enchondromatosis) since 3 years of age and had developed multiple SCH in the right leg at the age of 11 years. Spindle cell hemangioendothelioma lesions coincided with the site of enchondromatosis and increased in number thereafter. This is the first report of Ollier's disease complicated with multiple SCH. Another patient, a 33 year old Japanese female, who was a carrier of hepatitis B virus (HBV), developed solitary SCH in the lateral aspect of the right ankle where a lipoma was extirpated 10 years previously. Tumor cells of both cases were composed of four cell types: (i) spindle cells; (ii) epithelioid cells; (iii) vacuolated endothelial cells; and (iv) usual endothelial cells. Endothelia in the cavernous area and vacuolated cells reacted to Ulex europaeus agglutin 1 (UEA-I), factor VIII-related antigen and vimentin. Spindle cells and epithelioid cells reacted only to vimentin.

[Pharmacological studies of Houttuyniae herba: the anti-inflammatory effect of quercitrin].

Anti-inflammatory activities of quercitrin (Qu) were studied using various experimental models in mice, rats and guinea pigs. Qu (50, 100 and 200 mg/kg, p.o.) inhibited the rat hind paw edema induced by various phlogistics (carrageenin, dextran, histamine, serotonin and bradykinin) in a dose-dependent manner, and 200 mg/kg of this compound also inhibited the scald edema induced by hot water (54 degrees C). Qu did not show any significant inhibition of the ultraviolet light-induced erythema in guinea-pigs and of the increase of vascular permeability induced by acetic acid in mice. Qu did not affect the granuloma formation in a cotton pellet and the development of adjuvant arthritis in rats. These results indicate that Qu might have an inhibitory effect on acute inflammation.

Stepping analysis in patients with spinocerebellar degeneration and Parkinson's disease.

POLGON (Polarized light goniometer) was used to evaluate ataxia during stepping movements in patients with spinocerebellar degeneration (SCD) and Parkinson's disease. The measurements included mean angular change of shoulders (M.A.C.S.) and its coefficient of variation (C.V.). In patients with SCD, the values of M.A.C.S. were significantly larger at 1.0 step/s than those at other stepping rhythms. This results suggests that the stepping rhythm of 1.0 step/s is useful for the detection of cerebellar ataxia. The values of C.V. correlated with the degree of advancement of SCD. In patients with Parkinson's disease, the values of M.A.C.S. tended to decrease because of the restricted elevation of the knee, while those of C.V. were increased. The results showed that the stepping test using the POLGON was useful for estimation of the characteristic disequilibrium of SCD and Parkinson's disease.

[Staging pelvic lymphadenectomy for prostatic carcinoma].

Forty-two prostatic carcinoma patients with clinical stage A2, B or C underwent pelvic lymphadenectomy (limited node dissection) as a staging operation. The relationships among the presence of lymph node metastasis, tumor marker levels and histological findings of primary lesions were examined in 42 patients. Pelvic lymph node metastasis was noted in 18 (43%) of the 42 patients. The incidence of lymph node metastasis tended to be correlated with differentiation and Gleason's primary lesion sum. The preoperative PSA and PAP levels were significantly elevated in patients with positive lymph nodes for carcinoma than in those with negative lymph node for carcinoma. Patients with higher serum PSA (> or = 20 ng/ml) and PAP (> or = 10 ng/ml) levels were more frequently associated with lymph node metastasis. Thus, when serum PSA and PAP were markedly elevated before treatment in patients with prostatic carcinoma, they should be considered to have a potential of pelvic lymph node metastasis. Pelvic lymph node metastasis was observed in some cases even without high tumor marker levels, particularly those with poorly differentiated carcinoma.

[Combination therapy with low dose adriamycin for advanced or recurrent breast cancer. Hokkaido Breast Cancer Treatment Study Group].

Comparative clinical trials among 3 regimens for patients with advanced or recurrent breast cancer were performed as a multi-institutional joint study. Arm-I of 3 regimens consisted of a 3-day consecutive administration of adriamycin (ADM) at 10 mg/body every 4 weeks, a daily oral administration of cyclophosphamide (CPA), 5-FU and tamoxifen (TAM) at 100 mg, 200 mg, and 20 mg, respectively, and a once-a-week intramuscular or subcutaneous injection of OK-432 (OK) gradually increased from the initial dose of 1 KE to the maintenance dose of 5 KE. Arm-II contained methotrexate (MTX) at 10 mg/body for 3 consecutive days every 4 weeks in place of ADM in Arm-I. Arm-III contained neither ADM nor MTX. Of 69 cases registered, 52 were eligible, leaving 6 non-eligible and 11 incomplete cases. The results of the overall evaluation of 48 complete cases other than 4, in which the tumor was not exactly measured, were as follows. Arm-I resulted in 2 CR and 3 PR out of 19 cases, the response rate being 23.6% (5/19). Arm-II resulted in 1 CR out of 15, the response rate being 6.7% (1/15). In Arm-III, no response cases were found. No significant difference was observed among three treatment groups (p = 0.055), but in Arm-I, the response rate was higher than in the other treatment groups, suggesting that there is a probability of useful combined use of ADM.
