[Tumor markers in renal cell carcinoma].

Erythrocyte sedimentation rate (ESR), lactase dehydrogenase (LDH), carcinoembryonic antigen (CEA), haptoglobin, fibrinogen, CH50, Blc-globulin (C3), B1E-globulin (C4), T-cell, B-cell and phytohemagglutinin (PHA) were evaluated in 82 patients with renal cell carcinoma. Positive rates of ESR, LDH, CEA, haptoglobin, fibrinogen, CH50, C3, C4, T-cell, B-cell and PHA were 58.9, 27.1, 33.9, 21.9, 55.8, 34.2, 33.3, 34.8, 15.3, 8.0 and 39.1%, respectively. Positive rates of all serological tests in high stage (stage III and stage IV) renal cell carcinoma were higher than those of the low stage (stage I and stage II) group. Positive rates of ESR, fibrinogen, CEA, haptoglobin, C3 and C4 in the high stage group were 86.1, 81.8, 54.1, 57.1, 53.3 and 60.0% respectively. The level of ESR, LSH, CEA, fibrinogen, haptoglobin and CH50 were significantly elevated (p less than 0.001) above the normal value. Each serological test was not significantly elevated pre- or postoperatively, but in all serological tests, postoperative values were lower than the preoperative values. ESR (less than 100 mm/hour), and CEA in the positive group correlated significantly with poor prognosis (p less than 0.005). Haptoglobin and C3 in positive group also showed a significantly poor prognosis (p less than 0.05). In conclusion ESR and fibrinogen were thought to be useful for screening renal cell carcinoma patients ESR, CEA, haptoglobin, fibrinogen and C3 for estimation of staging, and ESR, CEA, haptoglobin and C3 for estimation of prognosis.

Enzyme activities of cultured erythroblasts.

The enzyme activities of cultured early erythroid progenitor cells (burst-forming unit erythroid, BFU-E) were measured and were compared with the activities of mature erythrocytes. The enzyme activity of acetylcholinesterase was not detectable in the erythroblasts. The ratios of phosphofructokinase and glutathione peroxidase were low due to low enzyme activities in both the erythroblasts and erythrocytes. The ratios of triose phosphate isomerase, phosphoglycerate kinase, and adenylate kinase were low due to high enzyme activities in both the erythroblasts and erythrocytes. The ratios of hexokinase, glucose phosphate isomerase, monophosphoglyceromutase, pyruvate kinase, and adenosine deaminase were high due to high enzyme activities in the erythroblasts. The isozyme of erythroblast hexokinase was of the prototype isozyme I, while pyruvate kinase was predominantly of the prototype M2, with two hybrid isozymes to the anodal side by electrophoresis. These facts suggest that there is a greatly different metabolic pattern during the maturation of the erythroid cells.

Human-human hybridomas secreting antibodies specific to human lung carcinoma.

Human Namalwa cells were screened in serum-free medium and in 6-thioguanine, then fused with human lymphocytes from lymph nodes of lung adenocarcinoma cancer patients. Extensive testing using 14 lung cancer cell lines, 11 other cancer cell lines and 4 normal fibroblast lines identified monoclonal antibodies produced by 4 hybridoma clones that reacted specifically with lung adenocarcinoma cells. These monoclonal antibodies also reacted with lung adenocarcinoma tissues and not normal tissues or erythrocytes of any blood type. These hybridoma clones grew and stably secreted the antibodies in serum-free medium as well as in serum-containing medium.

Effect of reductones on cyclic 3', 5'-adenosine monophosphate phosphodiesterase.

The effect of some reductones such as ascorbic acid (AsA), triose reductone (TR), epinephrine (Ep) and their derivatives on cyclic 3', 5'-adenosine monophosphate phosphodiesterase (cAMP PDE) was studied in the presence or absence of Cu2+. AsA, TR, Ep and the reductones related to them inhibited cAMP PDE activity. Among the reductones, TR showed the highest inhibition. AsA, 5-methyl-3,4-dihydroxytetrone, pyrocatechol, p-hydroxyquinone and resorcinol had a relatively high inhibiting activity. The type of inhibition of AsA, TR and Ep was uncompetitive, competitive and noncompetitive, respectively. Cu2+ enhanced the inhibitory action of the reductones markedly and altered the type of inhibition of the reductones.

[Chemosensitivity testing using stem cell assay in urological and other malignancies].

We have attempted to grow several kinds of malignant tumors using human tumor stem cell assay. Formation of colonies in vitro was seen in 65 of 132 primary tumors (49%), including 25 of 41 (61%) uroepithelial cancers, 12 of 19 (63%) renal cancers, five of 12 (42%) testicular cancers, five of 21 (24%) gastrointestinal malignancies, five of 12 (42%) lung cancers, five of 11 (45%) hematopoietic malignancies and five of 16 (31%) other malignancies. Growth sufficient for in vitro chemosensitivity tests of CDDP developed from seven cases of uroepithelial cancer, three of them (43%) were sensitive to 2.5 micrograms X hour/ml of CDDP. The specimens from a metastatic testicular tumors that received several courses of PVB chemotherapy resulted in the resistance of the in vitro chemosensitivity test at a higher dosage of CDDP. Nine cases of renal cancer had sufficient growth for an in vitro chemosesitivity test of interferon. One of them was sensitive for alpha 2 type interferon. Three of seven cases were sensitive for alpha type of interferon. To predict clinical correlation, 19 patients were tested with the same drugs used in the in vitro chemosensitivity test. The predictability resulted in more than 60% of true positive, 91% of true negative and 86% of overall predictability.

Complement sensitivity of erythroid and myeloid precursors in paroxysmal in nocturnal hemoglobinuria.

To test the hypothesis that paroxysmal nocturnal hemoglobinuria (PNH) is a hematopoietic stem cell disorder, the growth of BFU-e and CFU-gm and the complement sensitivity of cultured cells from BFU-e and CFU-gm colonies, as well as of unipotential progenitor cells (CFU-gm and BFU-e), were examined in five PNH patients. BFU-e growth was reduced in the three patients examined, and poor CFU-gm growth was noted in three of the five patients. Compared to normals, BFU-e and CFU-gm colonies in all patients demonstrated an increased susceptibility to the lytic action of complement when the release of 59Fe and myeloperoxidase was measured as specific markers for monitoring membrane damage. Compared to the growth of normal bone marrow cells, CFU-gm growth was significantly inhibited by pretreatment of bone marrow mononuclear cells with monoclonal OKIal antibody and complement. These findings support the proposition that a membrane defect predisposing blood cells to complement-mediated lysis may occur at the level of unipotential progenitor cells.
