Viscum album L. extracts reduce sister chromatid exchanges in cultured peripheral blood mononuclear cells.

Increasing concentrations of Viscum album L. extracts were shown to significantly reduce sister chromatid exchange (SCE) frequency of phytohaemagglutinin (PHA)-stimulated peripheral blood mononuclear cells (PBMC) of healthy individuals. This decrease of SCE could not be explained either by changes in lymphocyte subpopulations, by cytostatic effects of the drug or by accelerated proliferation of PHA-stimulated PBMC. Currently, no other cells tested have shown this effect. One therapeutic effect of these anti-mutagenic drugs could be a stabilisation of mononuclear blood cell DNA.

Focal-attentive and preattentive processes in letter- and figure-search tasks.

Evidence for preattentive and focal-attentive processes was obtained with two reaction-time tasks. In each case, effects of preattentive processes were identified by stimulating different proportions of the visual field. By varying the length of the search path, the effects of focal-attentive processes were identified. The first task, which required scanning arrays of letters, yielded short reaction times whereas the second task, which required the location of figures, led to long reaction times. The difference is explained by low automation and the strong demands of the latter task. The data do not indicate an interaction between preattentive and focal-attentive processes.

[Analysis of concepts, conditions and aims of replications].

There are different positions regarding the definition and the function of replication which lead to several alternatives when setting out to repeat a study. Classifications proposed for replications are of heuristic value but they do not take into consideration all the important conditions. Often replications are regarded as a means to control for errors or as a means for generalizing findings. But these aims of replications are only of secondary importance to the main function of replications: the demonstration of "scientific facts". Only direct or "identical" replications may serve this purpose well.

Application of intracellular ATP determination in lymphocytes for HLA-typing.

The amount of adenosine triphosphate (ATP) in human lymphocytes was determined using a technique based on light emission from a bioluminescent reaction with luciferin-luciferase. The amount of ATP changed when cells were incubated in the presence of specific HLA antisera and complement. For determination of intracellular ATP a modified method was applied, which was based on reduction of extracellular ATP by the addition of ATPase. The results of titration of an anti-human lymphocyte serum using the bioluminescence assay were in agreement with the results of fluorescence vitality staining. Bioluminescent HLA-determination in 57 cell samples each tested with 5 different antisera also gave good agreement (95.8%) with the conventional method. From these experimental data the calculated ATP content per lymphocyte was 0.135 +/- 0.058 pg ATP.

[Late death in congenital toxoplasmosis].

In spite of a known toxoplasmosis infection rate of up to 90% of the population of the Federal Republic of Germany published T-cases decrease steadily. T-infection during gravidity may cause more pathologic alterations in the infant cerebrum than is commonly assumed. Medico-legal aspects are very rare. A case of connatal toxoplasmosis in a 19 months old infant was found at exhumation (16 days p.m.). In the cerebrum T-parasites, terminal colonies and calcified spots were observed. Specific morphological and immunohistological findings lead to the diagnosis. The medico-legal problems of this case are discussed.

An improved method for preparation of samples for the simultaneous assay of some antiepileptic drugs by gas-liquid chromatography.

A new simple extraction method, e.g., for subsequent quantitative determination of some antiepileptic drugs by GLC is described. HCl and an internal standard are added to 100 microliter of plasma or any biological fluid. This mixture is applied directly to an Extrelut column. Lipophilic substances (i.e. the antiepileptic drugs diphenylhydantoin, primidone and phenobarbital) are then extracted into 20 ml of ether and the solvent is evaporated to dryness. Direct GLC analysis of the on-column methylated drugs is performed using a P-N-detector.

[Cryosurgical treatment of rectal carcinoma (author's transl)].

After comprehensive experimental especially morphological studies in animals, we started cryo-therapy of rectal carcinoma in 1972. We now present the first critical 5-years review concerning indication, clinical results and cryo-immunological features. In strongly indicated cases local tumour destruction by extremely low temperatures is superior to other palliative methods. Of 67 patients with rectal carcinoma treated with cryosurgery, 37 survived, 12 more than 36 months and 16 more than 24 months. Though there are only few hints indicating an increased immunological response following cryo-surgery, our investigations suggest a general influence on host's immunity by cryo-therapy.

Thiamine dependency in a patient with congenital lacticacidaemia due to pyruvate dehydrogenase deficiency.

A patient with congenital lactic acidosis, muscular hypotonia and severe ataxia is reported. The aetiology of his disease was found to be a deficiency of pyruvate dehydrogenase (E.C. 4.1.1.1.). Thiamine treatment (1.8 g/day) was successful in correcting biochemical and clinical symptoms. The mechanism of its action is probably based on activation of pyruvate dehydrogenase through interference in the physiologic regulation.

[Local cryosurgical management of colonic and rectal cancer].

Complex experimental investigations created the basis for cryosurgical treatment of colon and rectum cancer. 27 patients were chosen for cryotherapy according to strict indication. Especially developed instruments and technique are presented. A three year-follow up-study shows 19 patients alive, most of them without recurrence or troubles. Immediately after starting with cryosurgical treatment patients recover remarkably. In collaboration with the Dept. of Microbiology (Rhein.-Westfäl. Techn. Hochschule Aachen) the possibility of a special cryoimmunological response is investigated.
