Urinary fibrinolysis in toxemia of pregnancy.

Serial determinations of urinary plasmin level by radiocaseinolytic assay were performed in 40 healthy women and 29 patients with various types of toxemia throughout pregnancy, labor and the puerperium. The results indicate that: 1. The urinary plasmin level in normal pregnancy increased significantly from the third trimester to the early puerperium. 2. In mild pre-eclampsia, a significant rise in urinary plasmin level from 26 weeks of gestation to the 4th day post-partum. 3. In severe pre-eclampsia, a progressive decrease in late pregnancy was found. After delivery, in the early puerperium, the urinary plasmin level of the patients with a rapid clinical was significantly higher than that of the patients with the sequelae of toxemia. Our study showed that serial assays of urinary plasmin activity can provide a reliable and sensitive index of severity, progression and natural history of toxemia of pregnancy.

Mechanism of cardiovascular action of trapidil.

In anaesthetized, open-chest dogs N,N-diethyl-5-methyl[1,2,4]triazolo[1,5-alpha]pyrimidine-7-amine (trapidil) in doses of 0.3--3 mg/kg i.v. produced increases in coronary sinus outflow and heart rate and decreases in systemic blood pressure and coronary resistance in a dose-dependent manner. Trapidil produced an increase in myocardial oxygen consumption but virtually no change in coronary arteriovenous oxygen difference. At 1.8 mg/kg i.v. of the drug coronary resistance fell to half of the pre-drug value and coronary sinus outflow almost doubled, and so did myocardial oxygen consumption. In isolated, blood-perfused dog heart preparations, trapidil produced coronary vasodilator and positive inotropic and chronotropic effects. Theophylline produced similar effects. Trapidil was a more positive inotropic than positive chronotropic agent, and so was theophylline but to a lesser degree than trapidil. In producing vasodilator and positive inotropic effects trapidil was about 3 times more effective than theophylline. Trapidil and theophylline inhibited the cyclic AMP phosphodiesterase (PDE) activity in crude extracts prepared from the dog ventricular muscle. In this respect trapidil was nearly 3 times more potent than theophylline. It is suggested that PDE inhibition would be a fundamental mechanism of action of trapidil.

The high autoantibody activity of antibodies to rat skeletal muscle glycogen phosphorylase b produced in rabbits.

Antibodies to rat skeletal muscle glycogen phosphorylase b were prepared in six rabbits by weekly injection of the enzyme emulsified with complete Freund's adjuvant. All the antiserum preparations showed high autoantibody activities to react with the rabbit muscle enzyme in both inhibition of enzyme activity and precipitation. In Ouchterlony double diffusion in agar, the antiserum preparations were precipitable to give a distinct spur between the two precipitin lines formed with rat and rabbit enzymes. When the autoantibody index was taken as per cent cross-reactivity of rabbit enzyme with rat enzyme, the autoantibody indices of inhibition and precipitation of one of the antiserum preparations were as high as 98% and 52.3%, respectively.

Prostaglandin F2 alpha metabolite levels in plasma, amniotic fluid, and urine during pregnancy and labor.

Levels of 13, 14-dihydro 15-keto-prostaglandin F2alpha (dhk PGF2alpha) in the plasma of 30 patients as well as in the amniotic fluid of 17 patients, and 5alpha,7alpha-dihydroxy 11-keto tetranor-prostane 1,16-dioic acid (the main urinary metabolite of PGF2alpha [PGF2alpha MUM]) levels in the urine of 30 patients were measured by radioimmunoassay during pregnancy, labor, and the puerperium. During pregnancy, no increase in dhk PGF2alpha (ng/ml) in plasma was detected as the time of delivery approached. The levels of dhk PGF2alpha during the second stage (0.64 +/- 0.15) and also at delivery (0.88 +/- 0.27) were significantly elevated over those in the first stage (0.38 +/- 0.29) (P less than 0.025 and P less than 0.005, respectively). Its level 2 hours after delivery was reduced to predelivery levels. Its levels in umbilical arterial and venous blood were 0.71 +/- 0.26 and 0.67 +/- 0.26, respectively. A significant elevation (P less than 0.01) of dhk PGF2alpha from 0.89 +/- 0.21 before labor to 6.16 +/- 2.40 at delivery was found in amniotic fluid. The hourly excretion of PGF2alpha MUM (microgram/hour) increased significantly from pregnancy levels to 1.06 +/- 0.45 in the first stage (P less than 0.01), to 7.67 +/- 4.31 (P less than 0.005) for the first 2 hours after delivery, and 2.37 +/- 1.08 from 2 to 12 hours after delivery (P less than 0.01). The excretion of PGF2alpha MUM decreased to pregnancy levels 12 hours post partum. These data indicate that during labor the production of PGF2alpha is remarkably increased.

Polypeptide composition of the purified photosystem II pigment-protein complex from spinach.

The Photosystem II pigment-protein complex, the chlorophyll alpha-protein comprising the reaction center of Photosystem II, was prepared from EDTA-treated spinach chloroplasts by digitonin extraction, sucrose-gradient centrifugation, DEAE-cellulose column chromatography, and isoelectrofocussing on Ampholine. The dissociated pigment-protein complex exhibits two polypeptide subunits that migrate in SDS-polyacrylamide gel with electrophoretic mobilities corresponding to molecular weights of approximately 43,000 and 27,000. the chlorophyll was always found in the free pigment zone at the completion of the electrophoresis. Heat-treatment of the sample (100 degrees C, 90 s) for electrophoresis caused association of the two polypeptides into large aggregates. It is concluded that these two polypeptides, 43,000 and 27,000, are valid structural or functional components of Photosystem II pigment-protein complex.

Development of spinal motoneuron innervation of the upper limb muscle in the rat.

Horseradish peroxidase was injected into the biceps brachii muscle of rats at various stages of development, from 10 days to 50 weeks. The retrogradely labeled neurons were found in the ipsilateral ventro-lateral column of the cervical cord, C4--C8, of all stages studied, but the number of labeled neurons decreased according to exponential curve as the age advanced. A striking finding was that the contralateral ventral horn cells were also labeled in the 10- and 14-day-old rats.

Two electrogenic mechanisms contributing to the 560 nm absorption changes in intact Bryopsis chloroplasts.

Light -induced absorbance changes at 560 nm in dark-adapted intact chloroplasts of the green alga, Bryopsis maxima were studied in the time range of 200 ms. The initial rise of the 560 nm signals consists of two major components which are both electrochromic absorbance changes of the carotenoids, siponein and/or siphonaxanthin, but different in mechanisms of the field formation. The first component (component S) is related to electron transport since it was sensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) and 2,5-dibromo-3-methyl-6-isopropyl-p-benzoquinone (DBMIB) and showed at light-intensity dependence similar to that of electron transport in chloroplasts. In the presence of DCMU, component S could be restored on addition of proton-transporting electron donors such as reduced 2.6-dichlorophenol indophenol and phenazine methosulfate, but not on addition of N,N,N',N'-tetramethyl-p-phenylenediamine which does not carry protons with electrons (Trebst, A. (1974) Annu. Rev. Plant Physiol. 25, 423--458). We propose that component S is due to the electric field set up by the proton translocation across the thylakoid membrane. The second component (component R) was resistant to DCMU and DBMIB. The light-intensity dependency of component R was similar to that of cytochrome f photooxidation which showed saturation at a relatively low light intensity. The magnitude of component R was markedly reduced by phenylmercuric acetate, suggesting the participation of ferredoxin and ferredoxin-NADP oxidoreductase in the mechanism of the field formation responsible for this component. In the presence of DCMU and phenylmercuric acetate, time courses of the 560 nm changes paralleled those of cytochrome f changes. These results indicate that component R is due to the electric field formed between oxidized cytochrome f and other intersystem electron carriers located in the inner part of the thylakoid membrane and reduced electron acceptors of Photosystem I situated on the membrane surface. The complex natures of the 560 nm changes, as well as the contributions of Photosystems I and II to the absorbance changes, are explained in terms of the two electrogenic mechanisms.

The fine structural organization of the locus coeruleus in the rat with reference to noradrenaline contents.

Combination of glyoxylic acid perfusion and postfixation in permanganate was used in an electron microscopic study of the locus coeruleus (LC) of the rat to give good preservation of fine structure and a reproducible demonstration of noradrenaline (NA) storage granules. Medium-sized LC cells (18 x 30 microns) contained a moderate number of small granular vesicles (SGV) and a few large granular vesicles (LGV), mainly near the Golgi apparatus. Dendritic branches were identified by their SGV content up to the tip. Dendrites were occasionally in close contact with each other or with the soma of LC cells, forming dendro-dendritic or dendro-somatic contacts. Numerous axon terminals containing many SGV and some LGV were observed in the neuropil, and they tended to contact dendrites and somata of LC cells or dendrites of unknown origin. These neuronal contacts were devoid of synaptic specializations except for an array of dense perpendicular lines between the juxtaposed membranes. Small oval cells (10 x 15 microns) devoid of SGV occurred frequently in the peripheral part of the nucleus, and they were occasionally in direct contact with LC cells.
