Selectivity of alinidine, a bradycardic agent, for SA nodal automaticity versus other cardiac activities in isolated, blood-perfused dog-heart preparations.

Selectivity of alinidine for SA nodal automaticity vs. other cardiac activities was investigated using isolated, blood-perfused SA node, AV node and papillary muscle preparations of the dog. The drug was administered intra-arterially. In SA node preparations alinidine (1 microgram-1 mg) produced a dose-dependent decrease of sinus rate down to about 74% of the basal value at 1 mg, but no atrial standstill. In AV node preparations alinidine, injected into the posterior septal artery (PSA) which supplies the AV node, prolonged AV conduction time up to about 124% of the basal value but only in large doses (30 micrograms-1 mg). In the same preparations, when injected into the anterior septal artery, which supplies structures from the His bundle down to the ventricular septum (the intraventricular conduction system), alinidine in large doses (300 micrograms-1 mg) also prolonged AV conduction time nearly to the same extent as when injected into the PSA. In both cases neither second- nor third-degree AV block occurred. In paced papillary muscle preparations alinidine reduced the force of contraction down to about 64% of the basal value but in large doses (100 micrograms-1 mg). In spontaneously beating papillary muscle preparations alinidine reduced the rate of ventricular automaticity down to about 78% of the basal value but in large doses (100 micrograms-1 mg). Alinidine (100 micrograms-1 mg) produced a transient increase in blood flow in these preparations but not in SA node preparations. The cardiac effects of alinidine were longer-lasting than its vascular effect and the reduction of sinus rate was particularly long-lasting. The order of selectivity of alinidine determined from the above results is as follows: SA nodal automaticity much greater than ventricular automaticity greater than intraventricular conduction divided by AV nodal conduction divided by cardiac muscle contraction divided by coronary blood flow. Such a cardiovascular profile of alinidine is different from that of AQ-A 39, another bradycardic agent, which is nearly equi-effective on both SA nodal and ventricular automaticity.

Cardiovascular actions of the dihydropyridine calcium antagonist nimodipine in the dog.

When injected i.v. into anaesthetized, open-chest dogs, isopropyl (2-methoxyethyl)-1,4-dihydro-2,6-dimethyl-4-(3-nitrophenyl)-3, 5-pyridinedicarboxylate (nimodipine, Bay e 9736) (0.3-10 micrograms/kg) produced an increase in coronary sinus outflow and decreases in mean arterial blood pressure, coronary resistance, arterio-venous oxygen difference and heart rate in a dose-dependent manner, but virtually no change in myocardial oxygen consumption. At 3 micrograms/kg i.v. of the drug coronary resistance fell nearly to half the pre-drug value, coronary sinus outflow nearly doubled and heart rate decreased by about 10 beats/min. Myocardial oxygen consumption was slightly reduced at 30 micrograms/kg i.v. and atrioventricular (AV) conduction time was slightly increased at 10 and 30 micrograms/kg i.v. of the drugs. When the coronary vascular and cardiac effects of nimodipine were assessed in isolated, blood-perfused dog heart preparations, i.e., sinoatrial node, AV node and papillary muscle preparations, by intra-arterial administration, the following was revealed. In nearly twice the dose doubling coronary arterial blood flow, nimodipine produced a 15% decrease in sinus rate and a 15% increase in AV conduction time. However, in reducing the force of contraction of the papillary muscle by half the pre-drug value was needed nearly 17 times the dose of nimodipine doubling coronary arterial blood flow. Suppression of AV conduction by large doses of nimodipine was evident only when it was injected into the artery supplying the AV node but not into the artery supplying the His-Purkinje-ventricular system.(ABSTRACT TRUNCATED AT 250 WORDS)

Analyses of the cardiac action of the bradycardic agent, AQ-A 39, by use of isolated, blood-perfused dog-heart preparations.

The cardiac and coronary vasodilator actions of AQ-A 39 were investigated in various isolated, blood-perfused dog-heart preparations. AQ-A 39 was injected i.a. In sinoatrial (SA) node preparations, AQ-A 39 (3 micrograms-3 mg) decreased sinus rate but rarely produced atrial standstill even in large doses (1-3 mg). In paced atrioventricular (AV) node preparations, AQ-A 39 injected into the posterior septal artery (which supplies the AV node) increased AV conduction time (i.e., AV nodal conduction time) in rather large doses (100 micrograms-1 mg), but produced neither second-nor third-degree AV block even with the largest dose examined (1 mg). In the same preparations, AQ-A 39 injected into the anterior septal artery (which supplies the His-Purkinje-ventricular system) prolonged AV conduction time (i.e., intraventricular conduction time) in rather large doses (0.3-1 mg). In paced papillary muscle preparations, AQ-A 39 reduced force of contraction only in large doses (1-3 mg). In spontaneously beating papillary muscle preparations, AQ-A 39 (10 micrograms-3 mg) decreased rate of automaticity and force of contraction only with large doses (1-3 mg). In all preparations, AQ-A 39 (30 micrograms-3 mg) increased blood flow. The order of effectiveness of AQ-A 39 on the above cardiovascular variables is as follows: ventricular automaticity not equal to SA nodal automaticity much greater than AV nodal conduction not equal to intraventricular conduction not equal to coronary blood flow not equal to ventricular muscle contraction.(ABSTRACT TRUNCATED AT 250 WORDS)

Changes in molecular forms of rat hepatic glutathione S-transferase during chemical hepatocarcinogenesis.

Changes in molecular forms of hepatic cytosolic glutathione S-transferases (GST) during rat chemical hepatocarcinogenesis were investigated. GST activities toward 1-chloro-2,4-dinitrobenzene and 1,2-dichloro-4-nitrobenzene increased with the increased area of gamma-glutamyltranspeptidase-positive foci and hyperplastic nodules induced by diethylnitrosamine followed by 2-acetylaminofluorene plus hepatectomy. Among GSTs with high activities toward 1,2-dichloro-4-nitrobenzene, which were separated by carboxymethyl Sephadex column chromatography, the activity of GST-A ( YbYb ) markedly increased with increased activity towards 1,2-dichloro-4-nitrobenzene in livers bearing foci and nodules and in isolated nodules and hepatomas, while activities of GST-C ( YbYb ') and -D (Yb'Yb') changed little. It was demonstrated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and two-dimensional gel electrophoresis that Yb as well as Ya , a subunit of ligandin ( YaYa ) and GST-B ( YaYc ), increased in livers bearing foci and nodules, while Yc as well as Yb' changed little. A new placental GST form (GST-P), which has a subunit molecular weight of 21,500 or 26,000, according to the marker proteins used, and neutral pls of 6.8 and 6.3, is immunologically different from any form of basic GSTs and is very low in normal liver; also, it was markedly induced in livers bearing foci and nodules and in well-differentiated hepatomas but not by short-term administration of drugs such as 2-acetylaminofluorene, in contrast to GST-A and ligandin. These results indicate that GST-A and more especially GST-P could be new preneoplastic marker enzymes for chemical hepatocarcinogenesis.

The placental form of glutathione S-transferase as a new marker protein for preneoplasia in rat chemical hepatocarcinogenesis.

A neutral form of glutathione S-transferase (GST-P), which has a subunit of 26,000 (or 21,500) daltons and charge isomers with isoelectric points of 6.7 (major), 6.3 and 6.0, was purified from rat placenta cytosol. Immunohistochemical staining using anti-GST-P rabbit antibody demonstrated that GST-P, which is hardly detectable in normal liver, is markedly increased and localized in preneoplastic hepatic lesions such as hyperplastic nodules, indicating that GST-P could be a useful marker proteins for preneoplasia in chemical hepatocarcinogenesis.

Human amniotic membrane and prostaglandin biosynthesis.

The human amnion has become a significant tissue for metabolism of chemical substances with biologic activities, such as steroid hormones and prostaglandins. In this paper our studies on the biosynthesis of prostaglandins in human amnion focused on the role of prostaglandins in initiating and maintaining labor. First of all, the source of prostaglandins released in amniotic fluid was investigated. The amnion was demonstrated to possess the highest activity of generating prostaglandins among amnion, chorion laeve, and decidua vera, obtained after normal vaginal delivery. Subsequently, the bioconversion of arachidonic acid into prostaglandins and related compounds using the homogenate of human amnion was carried out without the addition of cofactors in the incubation mixture to show the natural spectrum of various types of PGs formed. PGE2 was revealed to be a main product of the amnion; its conversion rate was 5% of incubated radioactivity. Other compounds catalyzed by cyclooxygenase were PGF2 alpha (0.2%), TXB2 (0.3%), and HHT (1-2%), whereas 15-HETE (2-3%) and a mixture of 9, 11, 12-HETE (1-2%) were formed by lipoxygenase in the amnion. And third, the activity to produce PGE2 in the amnion obtained at normal vaginal delivery was greater than that obtained at cesarean section prior to the onset of labor. Thus, these results suggested that the amnion played a significant role in prostaglandin biosynthesis in uterine tissues. To provide a better understanding of the metabolism of arachidonic acid in the amnion as regards initiation and maintenance of labor, we discuss prostaglandin biosynthesis in the amnion and the mechanism of arachidonic acid release.

NADPH-diaphorase: a selective histochemical marker for the cholinergic neurons of the pontine reticular formation.

Choline acetyltransferase immunohistochemistry has identified a large group of cholinergic neurons in the pontine tegmentum. By combined immunohistochemical and enzyme histochemical studies this particular cholinergic cell group was found to contain an enzyme, NADPH-diaphorase, that can be visualized histochemically. Thus NADPH-diaphorase histochemistry provides a simple, reliable method to selectively stain the cholinergic neurons of the brainstem reticular formation. The resolution obtained by this novel histochemical technique is similar to that found with the Golgi stain, and it should therefore be of great value in morphological studies of this cholinergic cell group.

[Viability of human chorionic villi in organ culture].

The method and technique of organ culture of human chorionic villi was elaborated in our laboratory. In this report, placental specimens obtained at 15 to 18 weeks of gestation were studied in organ culture for 7 days in terms of the maintenance of morphological integrity and the preservation of functions. The morphological aspect of the viability of the various villous elements with special emphasis on the trophoblast cells was described histologically and ultrastructurally. The functional aspect of the viability was discussed by analysis of the suppressive effect of the cultivated villi on plasminogen activator (PA) secretion by OK-432 elicited mouse peritoneal macrophages, by analyses of the activity of delta 5-3 beta-hydroxysteroid dehydrogenase coupled with delta 5-delta 4-isomerase (HSD) and of the radioactivity of [125I]-Iododeoxyuridine ( [125I]-IUdR) retained in the DNA of the trophoblast cells. Specimens of normal placenta were obtained at the time of induced abortion. The gestational ages were 15, 16 and 18 weeks. Specimens for organ culture were prepared under sterile conditions within one hour after expulsion of the placenta. Through gross dissection, the villi were isolated and minced into fragments of approximately 1 to 2 mm3. Incubation was carried out at 37 degrees C in a conventional static chamber with a gas mixture of 95% air, 5% CO2. The culture dishes and culture media were renewed every day. Data are the mean values of the duplicate incubations. In the first series of organ culture, placental fragments were removed at the end of each day of incubation, washed thoroughly and transferred to the new dishes with a culture medium freed from fetal bovine serum, where further incubation was performed for 24 hours. After this, placental fragments were fixed in 4% neutral buffered formalin, processed through paraffin embedding, serially sectioned at 5 micron and stained by periodic-acid Schiff (PAS) procedure. Culture medium obtained in this series at each end of incubation was put into a "macrophage plate" with the addition of plasminogen in the concentration of 2 IU/m1, in which OK-432 elicited mouse peritoneal macrophages with the capacity to secrete PA were cultured. Reaction was terminated at 24 hours unless otherwise indicated. Ten microliters of the medium in the "macrophage plate" was applied to the fibrin plate, and reaction was performed for 18 hours. PA activity was expressed by plasmin activity converted from plasminogen measured by the single radial immuno-diffusion method.(ABSTRACT TRUNCATED AT 400 WORDS)

[Regulation of prostaglandin F2alpha release from the pseudopregnant rabbit uterus in luteolysis].

In previous experiments it was found that PGF2 alpha originating from the uterine endometrium plays a physiologically important role in corpus luteum regression. The present study was undertaken to investigate the regulation of PGF2 alpha release from the uterus in luteolysis. The effect of ovariectomy on PGF2 alpha release was examined. Bilateral ovariectomy was performed on day 14 of pseudopregnancy. PGF2 alpha urinary metabolite excretion was almost constant from day 15 to 22, although a significant increase of PGF2 alpha UM excretion could be seen in the control rabbits. Ovariectomy was performed on day 17. Levels of PGF2 alpha UM excretion increased from day 15 to 17, however, an abrupt decrease occurred after ovariectomy. This indicates that the ovaries are necessary for PGF2 alpha release from the uterus. The effect of steroids on PGF2 alpha release was examined. No significant increase in PGF2 alpha UM excretion could be seen in the ovariectomized pseudopregnant rabbits by the administration of estradiol and/or progesterone, although they are reported to be involved in other species. The effect of the ligation of the vessels between the uterus and ovaries on PGF2 alpha release was investigated. The ligation was performed on day 9. No increase in the production of PGF2 alpha occurred after ligation. Luteal life span was prolonged by 6 days. Endometrial perifusion was carried out to investigate PGF2 alpha production activity on day 18 of the pseudo-pregnant rabbits after the ligation of the vessels between the uterus and ovaries. PGF2 alpha production activity in the treated rabbits was lower than in the controls. These results indicate that a non-steroidal regulator may regulate PGF2 alpha release via the circulation of the uterus and ovaries in the pseudopregnant rabbit.

Properties of the increased glutathione S-transferase A form in rat preneoplastic hepatic lesions induced by chemical carcinogens.

Glutathione S-transferase A form (GST-A) is increased markedly in rat preneoplastic hepatic lesions such as hyperplastic nodules induced by diethylnitrosamine followed by administration of N-2-fluorenylacetamide. GST-A was also significantly increased in livers of rats after short-term administration of some drugs. The increased activity and protein content of GST-A were demonstrated by CM-Sephadex C-50 column chromatography as well as by two-dimensional polyacrylamide gel electrophoresis following immuno-affinity column chromatography using antibody against GST-A. Immunologically, GST-A crossreacted strongly with GST-C, weakly with GST-C2, but not with ligandin, GST-B, or GST-AA. It was confirmed by subunit recombination that GST-C is a heterodimer composed of the subunits of homodimers, GST-A and GST-C2.

Effects of blindfolding a unilateral eye on so-called central nystagmus.

A bipolar stimulating electrode was chronically implanted at the mesodiencephalon in the rabbit to induce so-called central nystagmus. The nystagmus was remarkably facilitated in either darkness by blindfolding an eye ipsilateral to the stimulation site, but the nystagmus was almost never facilitated by blindfolding an eye contralateral to the stimulation site. This is a phenomenon related to photic inhibition. This phenomenon disappeared after ablation of the flocculus and paraflocculus ipsilateral to the stimulation site. The neural pathway responsible for it is considered to be by way of the flocculus.

Glycosaminoglycans and glycoproteins in bronchoalveolar lavage fluid from patients with pulmonary alveolar proteinosis.

Bronchoalveolar lavage fluid from two cases of pulmonary alveolar proteinosis were analyzed for glycosaminoglycans and glycoproteins. The clinical courses of the two cases were entirely different. In one patient, signs and symptoms recurred despite repeated therapeutic bronchoalveolar lavages. In the other patient, three successive bronchoalveolar lavages brought about complete recovery. It was found that the bronchoalveolar lavage fluid from the former case contained various subtypes of glycosaminoglycans [hyaluronic acid, chondroitin sulfate A(C), dermatan sulfate, and heparan sulfate] and glycoprotein. On the other hand, the bronchoalveolar lavage fluid from the latter case contained glycoprotein, but no detectable amounts of glycosaminoglycans. There was only a slight qualitative difference in glycoprotein of bronchoalveolar lavage fluid between the two cases. The presence or absence of glycosaminoglycans in bronchoalveolar lavage fluid may be related to the prognosis of pulmonary alveolar proteinosis.

In-vitro production of prostaglandin E, F and 6-oxo-prostaglandin F1 alpha by the uterine and ovarian tissues of pseudopregnant rabbits.

The production of prostaglandin (PG) E, PGF and 6-oxo-PGF1 alpha by the uterine and ovarian tissues of pseudopregnant rabbits was investigated at the various stages of pseudopregnancy. The production of PGE, PGF and 6-oxo-PGF1 alpha in the tissues tested was expressed as mean (+/-S.E.M.) ng/mg protein per 6h. The production of PGE by endometrial tissue was found to be slightly raised with levels ranging from 2.15 to 2.92 on days 13 - 19 compared with values on day 5 (P less than 0.01). Subsequently PGE production declined on day 22 to the same level as that on day 5. In the myometrial tissue the same tendency in PGE production was observed as in the endometrial tissue. A significant increase in PGF production in the endometrial tissues was observed on day 13 (4.08 +/- 0.56) compared with that on day 12 (1.28 +/- 0.17) (P less than 0.001). The highest activity was detected on day 19 (7.19 +/- 0.29). An abrupt decrease occurred on day 20. In the myometrial tissues there was a slight and gradual increase in PGF production from day 12 (1.88 +/- 0.18) until day 16 (3.22 +/- 0.21). A sudden decline in the activity was detected after day 17. A marked increase of the 6-oxo-PGF1 alpha production activity in the myometrial tissues was observed on days 12 - 20 ranging from 14.60 to 19.25 compared with that on day 5 (P less than 0.001). The maximum activity was detected on day 16 (19.25 +/- 2.76). In the endometrium significant increase ranging from 4.85 to 8.31 occurred continuously from days 14 - 20 compared with that of day 12 (P less than 0.01). The myometrial tissue was shown to be significantly more potent in 6-oxo-PGF1 alpha production than endometrial tissue (P less than 0.01). No significant increase of PGE and PGF production activity was detected in the ovarian tissues throughout pseudopregnancy. The corpora lutea were shown to have very low 6-oxo-PGF1 alpha production activity. The 6-oxo-PGF1 alpha production activity of the remaining ovarian tissue was demonstrated to be significantly (P less than 0.01) more potent than the corpus luteum. The endometrial tissue of pseudopregnant rabbits has marked PGF2 alpha production activity in the late stage of pseudopregnancy, whereas the myometrial tissue is remarkably potent in 6-oxo-PGF1 alpha synthesis. Conversely, ovarian tissues showed much lower PG production activity than the uterine tissues. These experiments suggest that the main site of PGF production in luteolysis may be the endometrium and not the ovary itself. The physiological significance of PG production in relation to luteolysis is discussed.
