Three autopsy cases of adult T-cell leukemia (ATL) complicated by a severe hypercalcemia are presented. In two of them, the hypercalcemia itself was the direct cause of death. There was no evidence of an increase of serum PTH in all three cases. Prostaglandins were within normal range in two of them. By a bioassay, a bone-resorbing factor, osteoclast activating factor (OAF)-like substance, was demonstrated in the culture medium of leukemic cells from one patient. Also, various degrees of osteoclastic activation were found in the bones of all three patients by the postmortem examination. It may be that the hypercalcemia occurring in cases of ATL is partly caused by the humoral factor, which is released from leukemic cells and activates a bone resorption by osteoclasts. Similar cases in which a significant activation of osteoclasts was practically demonstrated by histopathologic examination in addition to the detection of a bone-resorbing factor have been rarely reported.

Immunohistochemical staining using antirat glutathione S-transferase placental form (GST-P) rabbit antibody demonstrated marked binding to cells of putative preneoplastic lesions induced in both the liver and pancreas of Syrian hamsters by dihydroxy-di-n-propylnitrosamine treatment, whereas background normal-appearing tissue was negative. Thus, a protein showing immunological cross-reaction with rat GST-P is also elevated during pancreatic carcinogenesis and hepatocarcinogenesis in the hamster, suggesting its usefulness as a marker with potential relevance to the mechanisms underlying the neoplastic process.

A cluster of neurons in the pontine tegmentum thought to correspond with the micturition reflex center was retrogradely labeled following the injection of the fluorescent tracer True Blue into the sacral spinal cord. Simultaneous immunofluorescence staining indicated that these neurons display corticotropin-releasing factor (CRF)-like immunoreactivity. Also, CRF-positive varicose fibers were detected in the intermediolateral column of the sacral cord. The results indicate the presence of CRF or a CRF-like peptide in a descending projection to the spinal cord that may be involved in regulating the micturition reflex.
Activated partial thromboplastin time (APTT) was examined in Brown Norway (B/N) Katholiek rat, which was previously reported as high molecular weight kininogen deficient. APTT of B/N Katholiek was prolonged to 35 sec in comparison with B/N Kitasato and SD rat, showing APTT of 22-24 sec. The mixture of B/N Katholiek plasma and B/N Kitasato plasma (1:1) showed normal APTT value. B/N Katholiek plasma corrected the abnormally prolonged human coagulation factor deficient plasmas, such as XI, XII and prekallikrein deficient plasmas, while it did not correct the APTT of HMW kininogen deficient, Fitzgerald plasma. Intravenous injection of bromelain, which was previously reported to produce prolonged hypotension through the activation of factor XII to release bradykinin, induced slight effect in Katholiek rat, while in Kitasato rat it showed prolonged hypotension in similar degree as SD rat. Contents of coagulation factors in B/N Katholiek thus measured as well as the values of prekallikrein and HMW kininogen previously reported were summarized and suggested that B/N Katholiek rat could be similar deficiency as Fitzgerald trait.
The activity of choline acetyltransferase (ChAT) in the cerebellar lobules of the rat was determined after bilateral destruction of either the superior, middle, or inferior cerebellar peduncles. A significant, 40-60% decrease in ChAT activity occurred in all subdivisions of the cerebellum (anterior lobe, posterior lobe hemisphere, posterior lobe vermis/nodulus and flocculus/paraflocculus) following bilateral inferior peduncle lesions. In contrast, bilateral lesions of the superior or middle peduncles did not result in significant reductions of enzyme activity. These findings indicate that the cholinergic afferents to the cerebellum are contained predominantly in the inferior peduncle, from which they emerge to innervate all of the gross subdivisions of the cerebellum.
The extent of 67Ga accumulation in the two lacrimal glands in patients with keratoconjunctivitis sicca (KCS) of Sjögren's syndrome was studied. Of the two main groups one consisted of 69 subjects without ophthalmic complaints (control group), the other consisted of 26 patients with KCS of Sjögren's syndrome. Of the 26 patients with KCS, 7 had been diagnosed as probable KCS (probable sub-group) and the other 19 had been diagnosed definite KCS (definite sub-group). About 3 mCi (111 MBq) 67Ga-citrate was injected IV into each subject and this was followed by scintigraphy at 24, 48, and 72 h after the injection of 67Ga. A positive finding in the lacrimal gland was noted in 64 of 69 subjects (92.7%) in the control group and in 7 of 7 patients (100%) with probable KCS. Three of 19 patients with definite KCS (15.7%) showed positive findings under scintigraphy. When the scintigraphic finding in the lacrimal gland is not positive in patients with suspected KCS of Sjögren's syndrome, they can then be diagnosed with little risk as definite KCS cases. Shirmer's test was performed on subjects in the probable and definite groups. There was statistical significance between the positive and equivocal or negative scintigraphic finding and Schirmer's values. These results suggest a correlation between gallium accumulation in the lacrimal gland and the tear production.
Bay k 8644 is suggested to increase calcium influx through the voltage-dependent calcium channel. In isolated, blood-perfused sinoatrial node preparations of the dog. Bay k 8644 injected intra-arterially increased sinus rate as isoprenaline did. In isolated, blood-perfused papillary muscle preparations of the dog isoprenaline injected intra-arterially increased both the force of contraction of the papillary muscle and its spontaneous beating rate. In these preparations, however, Bay k 8644 failed to increase the beating rate, although it increased the force of contraction. These results are consistent with the concept that the slow inward current plays a role in sinoatrial nodal automaticity but not in automaticity of Purkinje fibres which was reflected as ventricular automaticity of the papillary muscle preparations used here.
The viability of the placental villi in organ culture was investigated both from the morphological and functional points of view. Morphological viability was judged on the appearance of the villi with special emphasis on the trophoblastic cells and the morphological integrity of the placental villi could be maintained for at least 96 h in organ culture. Functional viability was judged by analyses of delta 5-3 beta-hydroxysteroid dehydrogenase; delta 5,4-isomerase (3 beta-HSD) activity with 14C-pregnenolone as the substrate and by the suppressive effect of the cultivated villi on the secretion of plasminogen activator (PA) obtained from mouse peritoneal macrophages stimulated by OK-432. It was found that the 3 beta-HSD activity and suppressive effect on PA secretion could be preserved for at least 120 h in organ culture.
