[Animal experiment and cell biology study of Saccharomyces cerevisiae Hansen CBS 5926 in the non-specific enhancement of resistance to infection].

In an animal experimental study the effect of Saccharomyces cerevisiae (S.c.) Hansen CBS 5926 (Perenterol) on bacterial infections has been investigated using different forms of application. A significant protective effect after oronasal, intragastric and subcutaneous application of S.c. Hansen CBS 5926 has been found by challenge with Streptococcus pneumoniae and Escherichia coli. In order to investigate the possible mode of action the effect of S.c. Hansen CBS 5926 on chemiluminescence activity of polymorphonuclear cells from porcine blood has been studied. Isolated cells as well as cells in whole blood showed a significant induction of chemiluminescence. In comparison to Zymosan A the yeast cells originating from the commercial formulation showed a less pronounced ability to stimulate the chemiluminescence of blood cells. However, the stimulation was significantly enhanced after inactivation by heat.

The role of detergent in labeling of Erysipelothrix rhusiopathiae bacteria.

The chelating agents oxine, acetylacetone, tropolone, and 2-mercaptopyridine 1-oxide were analysed for their suitability in labeling the bacterium Erysipelothrix rhusiopathiae (E.r.), strain B10, with 111In and 67Ga. The labeling conditions were improved. The bactericidal activity of the labeling process was investigated. The different chelates did not result in a significant difference in labeling yield. The addition of 0.01% detergent to the labeling buffer doubled the labeling yield up to 45%. Labeled bacteria were injected intravenously into rats. The percentage organ distribution was determined and compared with a control group.

[Synthesis of gestodene].

The synthesis of the new progestogen, 17 alpha-ethinyl-17 beta-hydroxy-18-methyl-4,15-estradien-3-one (gestodene, 6), starting from 18-methyl-4-estren-3,17-dione (1) can be accomplished by several methods. The oral progestational activity of gestodene is greater than that of levonorgestrel. Gestodene, in combination with ethinylestradiol, is contained in a recently developed oral contraceptive.

Intestinal and serum antibody response in gnotobiotic piglets to oral immunization with Escherichia coli.

The local and systemic immune response to a formolized E. coli oral vaccine was investigated in 13 gnotobiotic piglets. Beginning at ten days of age animals received a daily dose of 10(10) or 10(11) bacteria, on ten consecutive days. Intestinal loop tests with one animal of each group on day 26 showed protection which was more pronounced in the animal dosed 10(10) bacteria compared with the other immunized piglet. Immunoglobulin class-specific antibodies to O and K antigens were determined by ELISA technique. In serum no IgG or IgA antibodies were found, whereas IgM-anti O149 antibodies in both immunized groups reached their highest level at day 4 of dosing and decreased thereafter. IgM-anti K88 antibodies were first detected at day 10 of dosing. Both immunized groups had comparable serum levels at days 20 and 30. Also in gut secretion the IgM antibody response was predominant, and higher levels were found in the 10(10) group than in the 10(11) group. IgG and IgA antibody response were also detected in secretion.

Aldosterone antagonists. 1. Synthesis and activities of 6 beta,7 beta:15 beta,16 beta-dimethylene steroidal spirolactones.

Several derivatives of the highly active aldosterone antagonists dihydrospirorenone (2) and spirorenone (3) were synthesized. The purpose of these efforts was to prepare compounds exhibiting reduced endocrinological properties with the same or better aldosterone antagonistic activity than that of spirorenone. The 1 alpha,2 alpha-methylene derivative 20 has a similar aldosterone antagonistic potency compared to that of spirorenone but does not show decreased endocrinological side effects. Other substituents as in compounds 4-11, 15-19, and 21 sharply decreased the aldosterone antagonistic activity of 2 or 3, respectively.

The glycosphingolipid globoside as a serological marker on cytolytic T lymphocyte precursors and alloantigen-responsive proliferating T lymphocytes in murine spleen.

Biochemical analyses of murine lymphocytes have shown that the glycosphingolipid globoside (Glo) is present exclusively on alloantigen-stimulated murine T lymphocytes (Gruner, K. R., Van Eijk, R. V. W. and Mühlradt, P. F., Biochemistry 1981. 20: 4518). An anti-Glo antibody has now been raised in rabbits immunized with purified antigen. Most activity was recovered in the IgM fraction. The specificity of the antibody was ascertained in an enzyme-linked immunosorbent assay with purified glycosphingolipids bound to the solid phase. In antibody-dependent complement lysis experiments the anti-Glo eliminated about 20% of nylon wool-nonadherent splenic T cells of CBA/J mice. To determine the functional identity of these Glo+ cells, the effects of Glo+ cell elimination on mitogen stimulation with concanavalin A and lipopolysaccharide, as well as the effects on the mixed lymphocyte culture (MLC) reaction and cell-mediated lympholysis with mitomycin-treated DBA/2 splenocytes as stimulator cells were studied. Whereas lipopolysaccharide stimulation was not affected by elimination of Glo+ cells, there was a slight inhibitory effect on the concanavalin A stimulation, and a severe inhibition of the MLC reaction and the generation of H-2d-specific cytolytic T lymphocytes. Addition of interleukin 2 increased the MLC reaction, but interleukin 2-saturated cultures were also severely inhibited by anti-Glo and complement treatment. Combined treatment with anti-Glo and anti-Lyt-1 or anti-Lyt-2 antibodies, and determination of cytolytic T lymphocyte precursor frequencies in limiting dilution cultures after Glo+ cell elimination showed that a large proportion of T cells proliferating in a primary MLC are Lyt-1+,2+,3+Glo+, whereas in secondary MLC they are Lyt-1+,2-,3-,Glo+. Fifty % of the cytolytic T lymphocyte precursors in primary as well as secondary MLC are Glo+. The Glo marker is lost upon differentiation to cytolytic T lymphocyte effector cells. It is discussed herein that Glo is a marker for alloantigen-stimulated precursor T lymphocytes of both helper and cytolytic T cells.

[Critical evaluation of quantitative immunoglobulin determination in the calf].

From 3 calves fed with a single limited amount of colostrum 68 blood serum samples were tested comparatively by single radial immunodiffusion (RID) and by nephelometry for their IgG1-, IgA- and IgM-content. In comparison with nephelometry the commercial available RID kits seemed to be insufficient due to deficient agarose plates, difficulties in diluting, and weak reference sera. The results obtained by RID, therefore, were not comparable with those obtained by nephelometry which should be optimized, too.

Bronchoscopy and cytological examination of bronchial secretions of horses with chronic obstructive pulmonary disease (COPD).

On the basis of cytological similarities in chronic obstructive disease (COPD) in human beings and in horses during 1981 a total number of 144 horses with COPD were examined. Bronchial secretions were taken under endoscopic control and stained with a modified Hansel staining procedure. Whereas 26 horses did not exhibit signs of allergic reactions cytologically and 94 horses showed low amounts of eosinocytes and/or mast cells in their secretion; only the secretions of 24 horses (16,67%) were characterized by fairly high contents of these allergy associated cells and low numbers of neutrophilic granulocytes.
