[Acute toxicity study of cefpirome sulfate in mice and rats].

Acute toxicity of cefpirome sulfate (CPR) was examined in 6-week-old mice and rats and immature (5-day-old) rats. The LD50 values of CPR (mg/kg) were as follows: (1) mice: intravenous, 2420 (95% confidence limits, 2122-2758) for males and 2400 (2181-2640) for females; intraperitoneal, 3850 (3407-4351) for males and 4200 (3889-4536) for females; and oral, 16200 (14781-17755) for males and 18500 (17290-19795) for females. (2) 6-week-old rats: intravenous, 1900 (1784-2023) for males and 2080 (1953-2215) for females; intraperitoneal, 6550 (6179-6943) for males and 5800 (5311-6334) for females; subcutaneous, more than 10000 for both sexes; and oral, more than 8000 for both sexes. (3) 5-day-old rats: subcutaneous, between 1750 and 2500 for males and 2080 (1651-2621) for females. Major changes in general health conditions observed in 6-week-old mice and rats were decreased spontaneous activity, lying prone, tremor, respiratory changes (slow or deep respiration, gasping), clonic or clonic-tonic convulsions. In the 6-week-old rats dosed subcutaneously, vocalization, writhing and cutaneous changes at the injection site (dark reddening or blackening, swelling, exfoliation, depilation, induration) were also observed. In the 5-day-old rats dosed subcutaneously, the changes noted were slow respiration, writhing, cyanosis, and dark reddening and swelling of the skin at the injection site. After administration, transient depression of body weight gain or loss of body weight was observed in the mice and rats except the rats dosed orally. These changes disappeared at 7 days after administration at latest, and all surviving animals showed favorable body weight gain thereafter. Necropsies revealed hemorrhage under meninges in the brain in many of the mice and rats which died. Other findings included subcutaneous changes at the injection site in the 6-week-old and 5-day-old rats dosed subcutaneously (dark reddening, retention of dark red fluid, retention of red, white or dark red gelatinous material) and changes in the peritoneal cavity in the 6-week-old rats dosed intraperitoneally (red or dark red spots on the serous membrane, reddening of adipose tissues).

[A 17-year experience with ileal conduit urinary diversion--early and late complications].

A series of 185 patients, 133 males and 52 females, were treated by ileal conduit urinary diversion in the past 17 years. The patients ranged in age from 7 months to 81 years with an average of 59 years. Diversions were performed for malignant diseases in 174 patients, 85% of whom underwent a simultaneous radical surgery. The follow-up covered the postoperative period from 4 months to 16 years 8 months with an average of 4 years 8 months. Six patients (3%) died within 1 month of operation, and 43 of a total of 58 mortal cases died of cancer thereafter. The survival rates of 143 patients with bladder cancer were 84% for 1 year, 72% for 3 years, 67% for 5 years, 62% for 10 years and 54% for 15 years. Early complications were noticed in 38% of the patients. Delayed wound healing due to local infection (20%) and intestinal obstruction (10%) were the two major complications in this period. Late complications were encountered in 51% of the patients. Mild peristomal dermatitis (22%) and gradually developing renal complications (22%) are two major problems in the standard ileal conduit urinary diversion. The latter was significantly more frequent in patients who underwent the operation between 1973 and 1981 than in those who had the surgery between 1982 and 1989. Postoperative hydronephrosis was observed in 15 (13%) of 117 patients who showed normal urograms preoperatively. Ileoureteral reflux was observed in 50% of the cases with nonobstructing conduits, while it increased up to 70% along with obstruction of the conduit.(ABSTRACT TRUNCATED AT 250 WORDS)

Ontogeny of somatostatin in cerebral cortex of macaque monkey: an immunohistochemical study.

Distribution of somatostatin-immunoreactive cells in the cerebral cortex of macaque monkeys at embryonic day 120 (E120), E140, newborn, postnatal day 60 (P60) and adult stages were studied by the avidin-biotin-peroxidase immunohistochemical method. At all stages, there existed 3 types of cells in the gray matter: bipolar, multipolar and small-sized cells which stained only in perikaryon. Somatostatin-immunoreactive cells were observed from E120. The cell number increased between E120 and E140 and decreased until P60. At the newborn stage, a high density of cells was distributed in layer II of the prefrontal and parietal cortices (areas FD and PE). In layer I of the postcentral, parietal, temporal and preoccipital cortices (areas FA, PC, PE, TA, TE and OA), small numbers of horizontal cells were detected only at the embryonic and newborn stages. In adulthood, the number of somatostatin cells was much smaller than at the early stages (E140 and newborn). Compared to other cortical areas, in occipital cortex (area OC), there was little change in cell number during development. In occipito-temporal cortices, there were increases in cell number from posterior to anterior portion at all the stages. The large number of somatostatin cells in all layers of the cerebral cortex during the early stages indicates that somatostatin plays a role in the development of the monkey cerebral cortex.

Ontogeny of cholecystokinin-8 and glutamic acid decarboxylase in cerebral neocortex of macaque monkey.

Concentration of cholecystokinin-8 and the activity of glutamic acid decarboxylase were determined in the various cerebral cortical subdivisions of Japanese monkey (Macaca fuscata fuscata) at three different ages (embryonic 4 months, full-term and adult). The CCK-8 immunoreactive material extracted with 90% methanol from the cerebral cortex of the adult and foetal monkey were shown to be identical with synthetic cholecystokinin-8 by the criterion of co-elution on gel filtration chromatography (Sephadex G-50). The peptide concentration increased dramatically by about 30-80 fold (in terms of protein) and 17-28 fold (in terms of wet weight) between embryonic 4-month-old and full-term monkeys, while the level decreased 1/6-1/16 (protein) and 1/4-1/10 (wet weight) between full-term and adult monkeys. In adults, the highest levels of the peptide was observed in the association cortex, orbital prefrontal cortex and posterior parietal cortex. Glutamic acid decarboxylase activity, on the other hand, gradually increased about 4-10 fold (protein) between embryonic 4-month-old and adult animals and there was little variation in the increase rate among the cerebral subdivisions. In contrast to cholecystokinin-8, no reduction in the enzyme activity occurred between full-term and adult animals. The high level of cholecystokinin-8 in the embryonic period suggests that the peptide may participate in the regulation of the development of primate cerebral cortex.

[Growth stimulating activity of heparin-binding growth factor in the neural retina for retinal pigment epithelial cells].

Retinal pigment epithelial cells (RPE cells) participate in intraocular fibrocellular proliferation in proliferative vitreoretinopathy (PVR). The initial signal for proliferation and migration of RPE cells on PVR is not clear. However, water soluble extract from neural retina has been known to stimulate RPE cells, and fibroblast growth factor had a high affinity for heparin. In this experiment, we purified heparin-binding growth factor (HBGF) from neural retina using heparin-affinity chromatography. Then we examined the growth stimulating activity of HBGF for RPE cells proliferation. RPE cells were stimulated by HBGF, on the other hand heparin non-binding component could not stimulate RPE cells. This result suggests that the grwth-stimulating activity of retinal extract on RPE cells is related to HBGF.

Analysis of growth factors in renal cell carcinoma.

Tissue extracts prepared from human renal tissue, renal cell carcinoma and serum-free conditioned media of ACHN cells and A498 cells, cell line originated from human renal cell carcinoma, stimulated DNA synthesis of BALB/c 3T3 cells. The activity (growth factor activity) was significantly higher in renal cell carcinoma than in normal tissues. Radioreceptor assay revealed that the contents of epidermal growth factor and type alpha transforming growth factor in the tissue extracts from renal cell carcinoma and conditioned media from renal cell carcinoma cell lines were below detectable level. Most of growth factor activity of the tissue extracts and conditioned media showed high affinity for heparin-Ultrogel, indicating that the major growth factor activity was due to heparin-binding growth factor(s). In addition, renal cell carcinoma contained growth factor activity for ACHN cells, which did not show specific affinity for heparin-Ultrogel.

[Reproductive pharmacology of prostaglandins].

Effects of prostaglandins and their analogues in reproduction were investigated using rats and Japanese monkeys. Prostaglandin of the F series exerted uterine contractile, luteolytic, anti-nidation and abortifacient effects. Prostaglandins of the E series exhibited a potent uterine contractile effect, although their luteolytic, anti-nidation and abortifacient effects were much less potent than those of the PGF series. A comparison of animal and human data on prostaglandins and their analogues indicated a good cor-relationship between the uterine contractile effect in animals (rates and monkeys) and the abortifacient or labor inducing effect in humans.

Neuropeptides in cerebral cortex of macaque monkey (Macaca fuscata fuscata): regional distribution and ontogeny.

The concentrations of vasoactive intestinal polypeptide, somatostatin and substance P were determined in various cerebral subdivisions of adult and foetal Japanese monkeys (Macaca fuscata fuscata) by specific radioimmunoassays. In adult tissues, the highest level of vasoactive intestinal polypeptide was found in the somatosensory cortex and the lowest level in the occipital cortex. A high level of somatostatin was found in the association cortex (prefrontal, parietal and temporal cortex); the lowest level was noted in the occipital cortex. Substance P was found to be high in prefrontal and temporal cortex. The highest levels of substance P and somatostatin were obtained in the amygdala. Between embryonic 4 and 5.5 months, concentrations of peptides increased dramatically, and in the adult, all neuropeptides in cortical subdivisions significantly decreased. By the gel filtration method, only one immunoreactivity which coeluted with substance P and vasoactive intestinal polypeptide was demonstrated in extracts of 4-, 5.5-month-old and adult monkey cerebral cortex. In contrast, somatostatin immunoreactivity eluted as 3 peaks. Almost 80% of the immunoreactivity co-eluted with synthetic somatostatin, regardless of the age of the tissue. The molecular weights of two larger molecules were determined to be 13 and 3 kdaltons.

Temporary use of intraocular silicone oil in the treatment of complicated retinal detachment.

Complicated retinal detachments in 99 eyes were successfully managed by intraocular injections of silicone oil with or without pars plana vitrectomy. The silicone was then subsequently removed after panretinal photocoagulation. Any remaining anterior vitreous cortex near the vitreous base and/or preretinal proliferative tissue causing traction of the retina was also removed by a vitrectome or extracted by a hooked needle after the silicone-fluid exchange had been performed. The preretinal proliferation occurred in some cases even after silicone oil had been removed. Recurrent detachments were discovered in 8 of the 99 eyes and were managed successfully by reinjections of the silicone. Other silicone reinjections were required in two eyes with diabetic retinopathy and two nondiabetic eyes because of frequent rebleeding into the vitreous cavity.
