Effects of reactive oxygen intermediate scavengers on the antitoxoplasmic activity of activated macrophages.

Obioactin, Lonomycin A, muramyl dipeptide, and scavengers of hydrogen radicals and of singlet oxygen were used to study the participation of.OH and 1O2 in the killing of Toxoplasma in cultures of glycogen-induced peritoneal macrophages. Both the scavengers of OH (diazabicyclooctane and histidine) and those of 1O2 (mannitol and sodium benzoate) failed to inhibit the multiplication of Toxoplasma in macrophages that were incubated with either Obioactin, Lonomycin A, or MDP. The results of these experiments demonstrate the apparent lack of an inhibitory effect of.OH and 1O2 on the multiplication of Toxoplasma, whereas the scavengers alone inhibited the growth of the parasites.

Personal computer system for ECG ST-segment recognition based on neural networks.

A personal computer system for electrocardiogram (ECG) ST-segment recognition is developed based on neural networks. The system consists of a preprocessor, neural networks and a recogniser. The adaptive resonance theory (ART) is employed to implement the neural networks in the system, which self-organise in response to the input ECG. Competitive and co-operative interaction among neurons in the neural networks makes the system robust to noise. The preprocessor detects the R points and divides the ECG into cardiac cycles. Each cardiac cycle is fed into the neural networks. The neural networks then address the approximate locations of the J point and the onset of the T-wave (T(on)). The recogniser determines the respective ranges in which the J and T(on) points lie, based on the locations addressed. Within those ranges, the recogniser finds the exact locations of the J and T(on) points either by a change in the sign of the slope of the ECG, a zero slope or a significant change in the slope. The ST-segment is thus recognised as the portion of the ECG between the J and T(on) points. Finally, the appropriateness of the length of the ST-segment is evaluated by an evaluation rule. As the process goes on, the neural networks self-organise and learn the characteristics of the ECG patterns which vary with each patient.(ABSTRACT TRUNCATED AT 250 WORDS)

Gradient expression of neural cell adhesion molecule (NCAM) in the pontine migratory stream of fetal rats.

Immunohistochemical localization of a neural cell adhesion molecule (NCAM) was examined in the subpial pontine migratory stream of the fetal rat with a monoclonal antibody specific for the rat NCAM, MAb-AF11. Although the pontine cell strand showed weaker expression of NCAM than the parenchymal areas of the brainstem, within the cell strand, NCAM-immunoreactivity was somewhat greater in the anterior part of the stream near the neurons' destination than the caudal part. The weak NCAM gradient in the pontine migratory stream may contribute to formation of the pontine cell strand and the basal pontine gray.

Immunolocalization of alkaline phosphatase in osteoblasts and matrix vesicles of human fetal bone.

A monoclonal antibody raised against alkaline phosphatase (ALP) of human osteosarcoma was used to localize this enzyme in human fetal bone tissue. For light microscopy, the presence of alkaline phosphatase in osteoblasts and osteocytes was demonstrated by use of an avidin-biotin immunoperoxidase procedure. Electron microscopic immunolocalization was accomplished with an indirect immunoperoxidase method which revealed a concentration of the enzyme on matrix vesicle and osteoblast plasma membranes. In addition, many vesicular protrusions arising from areas of plasma membrane on the lateral surfaces of adjacent osteoblasts were strongly immunolabeled. Immunostaining for ALP was absent in vesicles which contained fine crystallites. Alkaline-phosphatase-rich matrix vesicles may play a significant role in the mineralization of the extracellular matrix.

Potentially lethal damage repair by quiescent cells in murine solid tumors.

We investigated potentially lethal damage repair by quiescent tumor cells in vivo. SCC VII tumor-bearing C3H/He mice were irradiated after being given 10 injections of 5-bromo-2'-deoxyuridine (BUdR) to label all the proliferating cells in their tumors, and the tumors were then excised and trypsinized. The tumor cell suspensions thus obtained were incubated with cytochalasin-B (Cyt-B, a cytokinesis blocker), and the micronucleus frequency in cells without BUdR labelling was determined using immunofluorescence staining to BUdR. The micronucleus frequency was then used to determine the surviving fraction of unlabelled cells on the basis of the regression line obtained for the micronucleus frequency and the surviving fraction of all tumor cells not labeled by BUdR, which can be regarded as the quiescent cells in a tumor for all practical purposes. Assessment performed 0, 3, 6, 9, and 24 hr after irradiation showed that quiescent cells had more potentially lethal damage repair capacity than the tumor cell population as a whole. Assays were also performed immediately after irradiation alone, 24 hr after the injection of cis-diamminedichloroplatinum(II) (CDDP), mitomycin C (MMC), or misonidazole [1-(2-nitro-1-imidazolyl)-3-methoxy-2-propanol] (MISO) following irradiation, and 24 hr after irradiation alone. It was found that CDDP and MISO (especially the latter) inhibited potentially lethal damage repair more strongly in quiescent cells than in the tumor cell population as a whole. This assay method thus appears to be quite useful for detecting the responses of quiescent tumor cells to various chemical agents.

Hepatocyte response to continuous low dose-rate radiation in radioimmunotherapy assessed by micronucleus assay.

The response of hepatocytes to low dose-rate irradiation was examined in mice following the injection of radiolabelled monoclonal antibody. Mice were injected intravenously with an 131I-labelled monoclonal antibody 196-14 which recognizes CA125 antigen, and the effect of continuous low dose-rate irradiation on hepatocytes was assessed using the micronucleus assay. The frequency of micronuclei increased in a dose-dependent fashion, but it was lower than the frequency induced by conventional external X-rays which was determined immediately after the irradiation. A linear quadratic model (micronucleus frequency = aD+bD2+c) showed that the value of b decreased with low dose-rate irradiation from the radiolabelled antibody. It is concluded that the micronucleus assay is useful for the evaluation of the response of hepatocytes to irradiation in radioimmunotherapy.

Hyperbaric oxygen therapy accelerates neurologic recovery after 15-minute complete global cerebral ischemia in dogs.

BACKGROUND AND METHODS: Although hyperbaric oxygen therapy is clinically used for the treatment of several types of ischemic brain injury, few basic animal studies are available that provide a rationale for this therapy for complete global brain ischemia. Therefore, we investigated the effect of hyperbaric oxygen therapy on neurologic recovery after 15-min complete global cerebral ischemia in a canine model. Complete global ischemia was induced in 19 dogs by occlusion of the ascending aorta and the caval veins. Nine dogs were randomized to treatment with hyperbaric oxygenation (3 atmospheres absolute, 100% oxygen for 1 hr) at 3, 24, and 29 hrs after ischemia under spontaneous respiration, while the other ten dogs served as the control group without hyperbaric oxygen therapy (group C). Neurologic recovery was evaluated based on the electroencephalogram (EEG) activity score (1 = normal; 5 = isoelectric) and the neurologic recovery score (100 = normal; 0 = brain death) over a 14-day postischemic period. RESULTS: The survival rates were 3/10 (30%) in the control group vs. 7/9 (78%) in the group treated with hyperbaric oxygen (p < .05). Over the 14-day postischemic period, the best (lowest) EEG scores of each dog were significantly (1.7 +/- 0.2 vs. 2.9 +/- 0.3; mean +/- SE, p < .01) lower in the hyperbaric oxygen-treated group. The best neurologic recovery scores of each dog were significantly (69 +/- 6 vs. 48 +/- 5; mean +/- SE, p < .05) higher in the treated animals. The number of dogs that recovered to a neurologic recovery score of > 65 (assessed as a slight disability) were 1/10 in the control group and 6/9 in the group treated with hyperbaric oxygen (p < .02). CONCLUSIONS: Hyperbaric oxygen therapy performed in the early postischemic period accelerated neurologic recovery and improved the survival rate in dogs after 15-mins of complete global cerebral ischemia.

Experimental spondylosis in the rabbit spine. Overuse could accelerate the spondylosis.

It has been suggested that overuse can accelerate the progression of cervical spondylosis. This assumption was derived from a study examining premature onset of cervical spondylosis in patients with athetoid cerebral palsy. To clarify the relationship between overuse and structural abnormalities of the spine, repetitive extension-flexion movement was located onto young rabbit spines through electric stimulation of the trapezius muscle. Repetitive loads of 200,000 cycles caused more severe delamination of the anulus fibrosus than control at the lower cervical spine. In addition, it was associated with early osteophyte formation at the same disc level. No severe degeneration of the nucleus pulposus, however, occurred through repetitive loading. It was found that repetitive movement could accelerate the progression of structural abnormalities, such as cervical spondylosis. This result suggests that overuse is an important factor in the pathogenesis of spondylosis.

Intraoperative measurement of lumbar spinal instability.

To justify lumbar fusion or stabilization, a quantitative assessment and definition of spinal instability are essential. To quantify spinal instability, the tensile stiffness of a motion segment (vertebra-disc-vertebra) was measured with a spinal distractor during spinal decompression surgery. Stiffness was indicated by the relationship between load and displacement between the two adjacent spinous processes where a vertebral spreader was suspended. A load-displacement curve was recorded at each step of surgical decompression and fixation while the motion segment was being distracted at a constant speed. The device used for measuring stiffness of a spinal motion segment is a lumbar spinal spreader with a load strain gauge and a displacement transducer. The stiffness of a spinal motion segment was reduced as disc degeneration developed. Degenerative spondylolisthetic discs showed the least stiffness (lowest, 3.9 N/mm; average, 5.4 N/mm). The stiffness of herniated discs, however, was relatively greater (average, 11.8 N/mm). The stiffness of normal motion segments was greater than affected segments. If the stiffness of a motion segment before decompression was graded as 100, it was reduced to 82% after partial laminectomy and facetectomy and to 65% after discectomy on average. After interbody fusion by iliac bone graft, it increased to 133% and to 184%, after Luque fixation.

Menogaril, an anthracycline derivative, inhibits DNA topoisomerase II by stabilizing cleavable complexes.

Menogaril, an anthracycline derivative, has been shown to possess antitumor activity in experimental animal systems, and is now under phase II clinical studies. However, its mechanism of action has not been elucidated. We have found that it inhibits the decatenation activity of purified DNA topoisomerase II using kinetoplast DNA from Crithidia fasciculata, its IC50 being 10 microM, which is comparable to that of etoposide. It does not, however, inhibit topoisomerase I activity at concentrations of up to 400 microM. Binding of topoisomerase II with DNA is not affected, but cleavable complex formation is stimulated by the drug. Cleavage site specificity differs from that of 4'-(9-acridinylamino)methanesulfon-m-anisidide. Menogaril was shown to possess a weak double-helix unwinding activity. These findings allow us to classify menogaril as a cleavable complex-stabilizing topoisomerase II inhibitor.

The physical properties of a new sealing cement.

This study was performed on a newly developed sealing cement, TS60, to evaluate its physical properties, namely sealing, setting time, compressive strength, solubility and ease of removal. It was compared with Temporary Stopping (GC Dental Industrial Corp., Tokyo, Japan), Eugedain (zinc oxide-eugenol based material, Showa Yakuhin Kako Co. Ltd, Tokyo, Japan) and Cavit-G (calcium sulphate based material, Espe Gmbh, Seefeld, Germany), which are typical commercially available sealing cements. The results of the study revealed that TS60 had excellent sealing properties, good compressive strength, low solubility, and was readily removable by heating and melting.

Synergistic action of cyclic GMP on catecholamine-induced chloride current in guinea-pig ventricular cells.

1. Effects of cyclic GMP on the catecholamine-induced chloride current (ICl) were studied using the whole-cell patch-clamp technique combined with internal perfusion in single ventricular myocytes dispersed from guinea-pig heart. 2. When ICl was activated by submaximal doses of isoprenaline (0.01-0.1 microM), adrenaline (0.5-1 microM) and histamine (0.2-0.5 microM), intracellular dialysis with cyclic GMP (10-100 microM) induced an extra increase of ICl. No further increase of ICl was induced by cyclic GMP when ICl was maximally activated. In the absence of agonists, cyclic GMP failed to induce ICl. 3. The enhancement by cyclic GMP was also observed when ICl was activated by external application of 0.2-1.0 microM-forskolin or by internal dialysis with a pipette solution containing 50-200 microM-cyclic AMP. 4. In contrast to cyclic GMP, 10-1000 microM-dibutyryl cyclic GMP and 8-bromo-cyclic GMP were ineffective in modifying ICl. 5. Milrinone (1-10 microM), a specific inhibitor of a kind of phosphodiesterase which is inhibited by cyclic GMP, also enhanced ICl activated by submaximal doses of isoprenaline. Milrinone itself did not activate ICl. 6. When ICl was enhanced by 5 microM-milrinone, an additional application of cyclic GMP failed to increase ICl. In the presence of cyclic GMP, milrinone failed to enhance ICl. 7. The above findings on ICl are analogous to the enhancement by cyclic GMP of the beta-adrenergic stimulation of the Ca2+ current reported in the same preparation, and support the hypothesis that in mammalian cardiac cells cyclic GMP potentiates elevation of cyclic AMP induced by beta-adrenergic agents, and thereby increases the amplitudes of ionic currents.

Phosphorylation restores activity of L-type calcium channels after rundown in inside-out patches from rabbit cardiac cells.

1. Rundown of L-type calcium channels was studied in inside-out patches made from single isolated rabbit ventricular myocytes, using barium as the charge carrier. 2. In the cell-attached patches single-channel activity was stable for more than 15 min after the patch pipette sealed. beta-Receptor stimulation by isoprenaline caused a characteristic increase in opening probability and the appearance of prolonged openings. When the patch was excised to the inside-out configuration and exposed to a simple ionic solution, channel activity disappeared within 1-2 min and never reappeared spontaneously. 3. After rundown of L-type channel activity in the excised patch, exposure of the inside face of the patch to MgATP and the catalytic subunit of the cyclic AMP-dependent protein kinase (PKAc) resulted in recovery of Ca2+ channel activity. Under these conditions channel activity could be even greater than under control cell-attached conditions, resembling channel activity after exposure to isoprenaline. This recovery of activity persisted many minutes, usually until the patch was lost. Addition of MgATP alone caused a small transient increase in channel activity in some patches. 4. Recovery of activity by MgATP and PKAc could be prevented by prior exposure of the excised patch to protein kinase inhibitor (PKI), or it could be abruptly terminated by exposure to PKI after recovery of activity. Addition to the pipette solution of okadaic acid, a protein phosphatase inhibitor, greatly slowed rundown. These findings support the proposal that dephosphorylation is an important component of rundown, and that phosphorylation is needed for channel opening activity. 5. Single-channel conductance was not altered by patch excision, but it was reduced after exposure of the excised patch to MgATP and PKAc. Mg2+ was responsible for this effect, probably by direct channel block from the inside, and Mg2+ also caused a negative shift in the channel activation, as expected from shielding of inside fixed negative charges.

Modulation of beta-adrenergic responses of chloride and calcium currents by external cations in guinea-pig ventricular cells.

1. The catecholamine-induced Cl- current and the Ca2+ current were recorded in the single ventricular cells of guinea-pig hearts, using the whole-cell patch clamp technique combined with internal perfusion. Dependence of the beta-adrenergic responses on external monovalent cations was investigated. The Cl- current was recognized by measuring the reversal potential of the agonist-induced current. 2. The amplitude of the Cl- current, activated by 1 microM adrenaline or 0.01-0.1 microM isoprenaline, was decreased when the external Na+ concentration ([Na+]o) was reduced by replacement with Tris+. The conductance of the catecholamine-induced Cl- current was proportional to the logarithm of the [Na+]o over a range of 15-140 mM. When the conductance was plotted against the concentration of Tris+, a dose-dependent inhibition of the Cl- response by Tris+ was suggested with a half-maximum concentration of 95 mM. 3. The inhibitory effect of the Na+ substitute TEA+ on the Cl- current was not affected by either increasing the buffer for the internal Ca2+ (10 mM BAPTA) or for the pH (50 mM HEPES). 4. In the relationship between agonist concentration and the Cl- conductance, the half-maximum concentration (K1/2) of isoprenaline was 0.013 microM in the control Na+ solution, and was shifted to 0.07, 0.08, 0.1 and 0.3 microM in the Li+, Cs+, TEA+ and Tris+ external solutions, respectively. The maximum slope conductance was not significantly affected, except for a slight depression on the Tris+ solution. When the current was induced by adrenaline, qualitatively the same finding was obtained; K1/2 was 0.15 and 3.2 microM in the Na+ and Tris+ solutions, respectively. 5. As a substitute for the external Na+, sucrose seemed to be inert. The activation of the inward Cl- current was conserved in the 300 mM sucrose solution ([Cl-]o = 8 mM) with a K1/2 value of 0.015 microM isoprenaline. 6. The Cl- current, when activated by either an external application of forskolin (0.2-10 microM) or an internal perfusion of cyclic AMP (100-500 microM), was not affected by replacing external Na+ with other cations. Activation of the Cl- current by 0.2-5 microM histamine was also insensitive to a substitution of Na+. These findings indicate that the inhibition by the Na+ substitute is at a point before the activation of GTP-binding protein. 7. The effects of Na+ substitution were not affected by varying the Na+ concentration (0-115 mM) in the internal solution, excluding an involvement of a change in the [Na+]i.(ABSTRACT TRUNCATED AT 400 WORDS)

Effectiveness of 6-month intermittent administration of natural human interferon-alpha against non-A non-B chronic hepatitis.

Interferon (IFN) was administered intermittently for 6 months to the patients with non-A non-B chronic hepatitis (CHNANB), and the effectiveness of the treatment for improving the hepatic function was evaluated. Of 26 patients with CHNANB, 16 received intermittent IFN therapy (IFN group), and 10 were treated by conventional therapies without IFN (non-IFN group). All patients were observed for 1 year. IFN was administered once a day at 3 MU in principle (1 MU in some patients and 6 MU in 1 patient) daily for 1 week immediately after the beginning of the therapy and 3 times a week for the subsequent 6 months at the outpatient clinic. The patients were followed up for at least 6 months after completion of the treatment. In the IFN group, the serum GPT level normalized in 11 (68.8%) of the 16 patients 1 year after the beginning of the treatment. In these 11 patients (normalized group), HCV-RNA was negative or became negative in 3 of the 6 patients in whom the serum HCV-RNA could be examined. Histological grades of inflammation in the liver were also markedly alleviated in the normalized group. The hepatic function did not normalize in any of the 10 patients in the non-IFN group. These findings indicate that IFN therapy is useful for CHNANB.

Retinal projections in the house musk shrew, Suncus murinus, as determined by anterograde transport of WGA-HRP.

Retinal projections in the house musk shrew (Suncus murinus) were determined by the anterograde transport of wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP). Unilateral injection of WGA-HRP into the vitreous body resulted in the terminal labeling of the optic projections in the suprachiasmatic nucleus (SCH), the ventral (CGLv) and dorsal (CGLd) lateral geniculate nuclei, the intergeniculate leaflet (IGL), the pretectum, the superficial layers of the superior colliculus (CS), and the dorsal terminal nucleus (DTN) of the accessory optic system (AOS). Labeling of the SCH was bilateral, with ipsilateral predominance, and covered the whole dorsoventral extent of the nucleus. Immunohistochemical studies revealed that VIP-like immunoreactive neurons and fibers were present in almost all parts of the SCH. No hypothalamic regions other than the SCH received the optic fibers. The ipsilateral projections to the CGLv, CGLd, and IGL were sparse, and a considerable number of uncrossed retinal fibers were found in the pretectal olivary nucleus. No retinal projections to the lateral posterior thalamic nucleus (LP) were found. Ipsilateral optic fibers projected sparsely to the medial part of the CS. The AOS consisted of a small DTN with a very few crossed retinal projections but no lateral and medial terminal nuclei. In addition, the AOS had no inferior fascicle.

Secretion mode of the harderian gland of rats after stimulation by cholinergic secretagogues.

We studied the morphological changes in rat Harderian glands 30 min after injection of cholinergic secretagogues. In controls, the glands exhibited a tubuloalveolar structure with relatively wide lumina, in which some osmiophilic dense droplets exocytosed from glandular cells were observed. Also two types of glandular cells (type A cells and type B cells sometimes showing exocytotic figures of lipid-secretory vacuoles) and myoepithelial cells were recognized. After injection of carbamylcholine chloride (subcutaneously, 0.1 mg/kg body weight), which has both nicotinic and muscarinic actions, many of the alveolar lumina dilated and contained a small number of osmiophilic droplets. Exocytotic figures in both types of cells and a pronounced decrease in the number of vacuoles in the glandular cells were observed. However, there was no evidence of apocrine or holocrine secretion. The injection of the higher dose of carbamylcholine (1.0 mg/kg) caused fusion of secretory vacuoles in the apical cytoplasm and contraction of myoepithelial cells. Most alveoli showed no clear lumina; their centers were jammed with cytoplasmic fragments and accumulated secretory products. Massive discharge of cytoplasmic fragments containing some secretory vacuoles was often observed. This may be classified as apocrine secretion. Bethanechol chloride (subcutaneous injection, 1.0 mg/kg), a muscarinic agonist, stimulated the Harderian-gland secretion, and enhanced exocytosis was observed. The discharge from the glandular cells, following injection of various doses of carbamylcholine, were almost inhibited by atropine sulfate, a muscarinic antagonist. The present results suggested that the cholinergic systems regulate the secretion of rat Harderian-gland cells which have muscarinic receptors.
