Production, characterization, and application of monoclonal antibodies which distinguish three glucosyltransferases from Streptococcus mutans.

Thirty-three murine monoclonal antibodies (MAbs) against the three glucosyltransferases (GTFs) (GTF-I, -SI, and -S) from Streptococcus mutans were obtained by the fusion of murine myeloma cells (P3X63-Ag8-U1) with spleen cells of BALB/c mice immunized with pure GTF-S or partially purified GTF-I from serotype c S. mutans PS14. The immunoreactivities of these MAbs were tested by enzyme-linked immunosorbent assay and Western blotting (immunoblotting) with various GTF preparations. GTF-I and GTF-SI were expressed from two Streptococcus milleri or Escherichia coli transformants harboring gtfB or gtfC, respectively. All of the five MAbs raised against the GTF-S from PS14 reacted only with the homologous enzymes. Of these, 8 MAbs reacted only with the gtfB gene product (GTF-I), 4 MAbs reacted only with the gtfC gene product (GTF-SI), and the remaining 16 MAbs reacted with both gene products. The existence of GTF-SI in the purified GTF-I from PS14 was demonstrated by Western blot analysis using the representative monospecific MAbs. Further, the relative levels of the three GTFs in the extracellular and cellular fractions of S. mutans clinical isolates were examined by immunoblot analysis. The findings indicated that the relative level of GTF-SI, unlike that of GTF-I or GTF-S, differed markedly among isolates although the three GTFs were synthesized extracellularly by all the strains.

Hepatocellular carcinoma: correlation of CT, angiographic, and histopathologic findings.

PURPOSE: To elucidate the causes of various enhancement patterns of hepatocellular carcinomas (HCCs). MATERIALS AND METHODS: Computed tomographic (CT) scans of 68 surgically resected HCCs (62 patients) were obtained 45 seconds and 6 minutes after administration of contrast material. These scans were compared with angiographic and histopathologic findings. RESULTS: On the early images, 24 HCCs (35%) were hypoattenuating, 33 were totally or partially hyperattenuating (48%), and 11 were isoattenuating (16%) compared to adjacent liver. On the delayed images, 55 HCCs (81%) were hypoattenuating and 13 (19%) were isoattenuating. Twenty-nine of the 48 very hypervascular or hypervascular tumors (60%) were hyperattenuating or partially hyperattenuating at early CT. Sixteen of the 20 slightly hypervascular or hypovascular tumors (80%) were isoattenuating or hypoattenuating. CONCLUSION: Although there was relatively good agreement between tumor vascularity and enhancement pattern (60% correlation for hypervascular tumors, 80% correlation for hypovascular tumors), hyperattenuation of the large HCCs (> or = 5 cm) at CT appeared to be a function of dilated sinusoids within the tumor (peliotic changes) as well as vascularity. In patients with advanced cirrhosis, hypovascular HCCs could be hyperattenuating at CT.

Pathomorphologic findings of lead poisoning in white-fronted geese (Anser albifrons).

Nineteen lead-poisoned white-fronted geese (Anser albifrons), including nine immature birds, were examined pathologically. Subacute lead poisoning due to ingestion of spent lead shots was diagnosed pathologically and confirmed by demonstrating high lead concentration in the liver. The liver lead concentration ranged from 6.9 to 67.7 mg/kg wet weight. The most suggestive gross lesions were mottled bile-stained liver in eight geese and proventricular impaction and/or the presence of lead pellets in the gizzard. Histologic lesions of the liver consisted of Kupffer cell hemosiderosis, large bile plugs in dilated canaliculi, bile pigmentation in hepatocytes, and bile extravasation and associated hepatic necrosis. Seven geese of the remaining 11 birds also had hepatic necrosis in the liver, the greenish discoloration of which was obscure macroscopically. The liver discoloration was considered a jaundice due to both rapid overproduction of bile from increased breakdown of erythrocytes and intrahepatic impaired excretion of bile. The severity of lesions was not correlated to the liver lead concentrations. All examined geese had hemosiderosis of mononuclear phagocytic system cells in the spleen and hypoplasia or edema of the bone marrow with increased numbers of polychromatic erythroblasts. These prominent changes probably resulted from excess breakdown of erythrocytes, hypercholia followed by intrahepatic cholestasis, and disrupted erythropoiesis in bone marrow caused by lead.

Direct ABC immunohistochemistry to Encephalitozoon cuniculi.

Among the sera from 9 rabbits spontaneously infected with Encephalitozoon cuniculi, a serum which revealed a high titer for E. cuniculi by indirect protein A-gold (IPAG) immunohistochemistry and reacted with the outer layer of the shell of E. cuniculi spores on immunoelectron microscopical examination, was biotinylated. The biotinylated rabbit anti-E. cuniculi IgG reacted immunohistochemically with E. cuniculi, but not with other protozoa tested, namely Neospora caninum, Toxoplasma gondii and sarcocystis. The direct avidin-biotin-peroxidase complex (ABC) immunohistochemistry using biotinylated rabbit anti-E. cuniculi IgG in this study is a useful tool for the diagnosis and study of encephalitozoonosis.

Dual infection with canine distemper virus and infectious canine hepatitis virus (canine adenovirus type 1) in a dog.

A 72-day-old, female, Golden Retriever dog showed anorexia, coughing, nasal discharge, diarrhea and hematochezia, and died on the 15th clinical day. Pathological examination revealed dual infection with canine distemper virus (CDV) and canine adenovirus (CAV). CAV inclusion bodies occurred only in the liver, and biliary and respiratory system, whereas CDV inclusions were demonstrated in the visceral organs systematically. The CAV inclusions were associated with multifocal hepatocellular necrosis and edematous swelling of the wall of the gall bladder, suggesting infectious canine hepatitis virus (canine adenovirus type 1) infection.

Ca2+ sensitivity of skinned uterine muscle fibers in unilaterally pregnant rats.

We prepared chemically skinned uterine muscle fibers from both horns of unilaterally pregnant rats and investigated the effect of gestation on the Ca2+ sensitivity of the Ca(2)-induced contraction. The Ca2+ sensitivity was defined as the pCa (minus logarithm of free calcium concentration) required for half maximum activation of force production. Gravid horns did not show any statistical difference in the Ca2+ sensitivity between normal pregnant and unilaterally pregnant rats. Non-gravid horns of unilaterally pregnant rats before Day 14 of gestation showed the same behavior in the Ca2+ sensitivity as horns of normal pregnant rats. After Day 15 of gestation the Ca2+ sensitivity of non-gravid horn also increased but significantly less than that of gravid horn. Taking into account the hormonal situation, the present results suggest that some local factors, such as an increased size of conceptus (fetus and placenta) and/or intercellular binding conditions of myometrium modulated by the gap junction formation and the appearance of connexin molecules, may also be requisite for the regulation of the contractile activity associated with parturition and delivery of the fetus and placenta.

Cell cycle perturbations of platinum derivatives on two ovarian cancer cell lines.

Cisplatin continues to be one of the most commonly used cytotoxic agent. Problems of drug resistance and nephrotoxicity have generated interest in new platinum derivatives. In this study, we used flow cytometry to study their effects on cell kinetics and to see if the extent of cell cycle perturbations can be used to determine relative potency. The following four platinum derivatives were tested: cisplatin, carboplatin, 254S, and NK121 on two human ovarian cancer cell lines: BG1 and CAOV3. Flow cytometric analysis revealed a dynamic spectrum of cell kinetic perturbations, which included sequential S-G2 block, concomitant S-G2 block, and a dominant S block with abolition of G2 block. Platinum derivatives NK121, 254S, and CARBO induced an average of 54.5 +/- 5.6, 21.2 +/- 5.5, and 2.5 +/- 2.8% more S-G2 blocks than cisplatin, respectively. When comparing the severity of S-G2 blocks and requiring a p-value of 0.05, the order of increasing potency was: cisplatin, carboplatin, 254S, and NK121.

A survey of lead poisoning in wild waterfowl in Japan.

Ingested shotgun pellets were found in 13 of 56 hunter-killed wild waterfowl between October and December 1991 from two hunting grounds in Japan. Four of 33 other waterfowl found dead in lightly hunted areas between January 1991 and March 1992 were diagnosed as having lead poisoning. We propose that lead poisoning maybe a threat to waterfowl in Japan.

Nutritional secondary hyperparathyroidism occurring in a strain of German shepherd puppies.

Five German shepherd puppies, aged between 66 and 174 days (4 of which had the same parents) had ananastasia with deformation of limbs. Histopathologically, all the puppies showed moderate to marked fibrous osteodystrophy with parathyroid gland activation. These puppies were fed a diet consisting of 80% steamed rice and 20% raw meat. It was implied that though the diet contained the required level of calcium, it contained more than the required level of phosphorus. From the results, the present cases were diagnosed as nutritional secondary hyperparathyroidism caused by a diet containing too much phosphorus. Based on the clinical aspects concerning the development of this condition a familial factor was suggested.

[Studies of prognostic factor and chemotherapeutic effect of epithelial ovarian cancer using Cox's proportional hazard model].

To make clear the prognostic factor and chemotherapeutic effect of epithelial ovarian cancer, a multiple-center study involving 22 hospitals in Japan was conducted using Cox's proportional hazard model. A total of 1,181 cases were reviewed. Clinical stage, histologic type, and residual tumor diameter were significant prognostic factors, but the degree of tissue differentiation was not. The effect of remission induction chemotherapy was assessed with or without CDDP, and a distinct prognostic difference was noted. Among the patients receiving CDDP + ADM + other chemotherapeutic agents (PA group), CDDP + other chemotherapeutic agents (PO group) and CDDP only (P group), the prognosis of the PO group was better than for the P group. The long-term prognosis improving effect of chemotherapy was assessed. Neither maintenance chemotherapy based on oral administration of pyrimidine fluoride nor immunotherapy had any long-term prognosis improving effect, while intermittent chemotherapy based on CDDP resulted in improved prognosis.

[Strategy for the ovarian cancer treatment].

The morbidity and mortality rate of ovarian cancer patients are significantly inclined recently in Japan. FIGO stage is a powerful prognostic factor, hence treatment schedule must be based on precise staging. For stage I patients, total abdominal hysterectomy (TAH), bilateral salpingo-oophorectomy (BSO), partial omentectomy (p-OMTX) and appendectomy (APX) are included to standard surgical procedure. Adjuvant chemotherapy consisted with CAP (cyclophosphamide, adriamycin, cisplatin) or CP is given for 5 courses postoperatively. However, those who are stage Ia (grade 1) and desire child bearing could have unilateral salpingo-oophorectomy and conserve conceptability. Adjuvant chemotherapy for those patients is now on prospective randomized trial in Japan. One arm is 50 mg/day of cyclophosphamide for 3 months and the other arm is cyclophosphamide 500 mg/m2 + CDDP 75 mg/m2 every 4 weeks for 3 courses. For stage II or more patients, total omentectomy and retroperitoneal lymphadenectomy are added to above mentioned procedure. Cisplatin (15.75 mg/m2/week or more) based adjuvant chemotherapy is given for 5-10 courses. Therapeutic significance of retroperitoneal lymphadenectomy is also under study in patients who are optimally debulked (< 2 cm residual tumor). They are randomly divided into two groups: lymphadenectomy group and no-lymphadenectomy group. They receive 5 courses of CP therapy (cyclophosphamide 600 mg/m2 + CDDP 75 mg/m2) afterwards. Survival rate and time to progression are the primary endpoints.

[Gynecologic oncology and clinical trials].

Clinical trials have been developed tremendously in oncology because of its clear endpoints. In Japan, however, a concept of clinical trial has not been so accepted for long time. Recently, Minister of Health and Welfare announced guide lines of clinical evaluation of anticancer drug and statistical analysis. Elements essential to design of clinical trials are following; the first element is a clearly stated hypothesis. The hypothesis must be testable, that is, the investigator must have a clear idea of how these differences will be determined in this study subjects. The trial must have a well documented protocol and have a well defined primary endpoint by which the anticipated effect of the new treatment will be evaluated. Although one or two secondary endpoints may also be identified in the trial, the primary endpoint is the major focus of the trial and determines trial design, size, and early stopping rules. The clinical trial must be appropriately designed so that the hypothesis can be adequately tested and evaluated. The study may compare the new therapy to a standard treatment or assess the basic characteristics of the treatment. The clinical trial must be designed so that adequate sensitivity (significance level) and power to provide a high degree of confidence that results are not spurious. The next critical aspect to the performance of a successful clinical trial is the ability to accrue adequate numbers of patients within a reasonable period of time for the study to be completed. The next essential element to the design of a clinical trial is the prospective selection of statistical tool appropriate to the data to be collected.(ABSTRACT TRUNCATED AT 250 WORDS)

[Effect of cytokines on eosinophil differentiation and proliferation].

IL-3, IL-5, and GM-CSF are eosinopoietic cytokines that induce the proliferation and differentiation of eosinophils. However, the regulatory mechanism of eosinopoiesis is largely unknown. A combination of IL-3 and IL-5 induces selective proliferation and differentiation of eosinophils from mononuclear cell fractions of human cord blood. IFN-gamma decreases the IL-3- and IL-5-induced eosinophil differentiation in a dose-dependent manner. IL-4 and IL-2 also decrease the IL-3- and IL-5-induced eosinophil differentiation. These observations indicate that IL-2, IL-4, and IFN-gamma have a direct or indirect inhibitory effect on eosinopoiesis.

[Twenty one multicentral studies on prognosis of advanced ovarian cancer; focused on correlation between primary surgical procedure following adjuvant chemotherapies and 4 year survival rates].

The treatment outcome of 778 patients with stage III ovarian cancer treated by surgery and chemotherapy between 1976 and 1990 in 21 institutions was analyzed retrospectively. Survival of patients treated between 1985 and 1990 was significantly superior to that before 1985 (Kaplan-Meier, p < 0.01). Four year survival rates were found to be distributed from 5 to 52% among institutions (p < 0.001, log-rank test). When survival rates were compared according to the relative dose intensity (RDI) of CDDP administered, patients who were given CDDP RDI > or = 0.9 had a favorable prognosis. The twenty-one institutions were divided into two groups: Standard (total abdominal hysterectomy and bilateral salpingo-oophorectomy) vs. standard+debulking (of as much gross tumor as can safely be performed) according to the operative procedure which was performed in each hospital. It was found that the four-year survival rate for patients treated in hospitals where standard surgery was done was 22.9 +/- 12.2%, whereas that in hospitals adopting standard+debulking surgery was 34.4 +/- 10.1% (p = 0.03, t-test). When the hospitals were divided into 4 groups according to combination of the operative procedure and CDDP RDI, the four year survival rates were 17.5 +/- 8.3% for standard+debulking+CDDP RDI < 0.9, 39.0 +/- 1.4% for standard+CDDP RDI > or = 0.9, 28.8 +/- 11.5% for standard+debulking+CDDP RDI < 0.9 and 37.8 +/- 8.0% for standard+debulking+CDDP RDI > or = 0.9.(ABSTRACT TRUNCATED AT 250 WORDS)

Epidermal Langerhans cells from normal human skin bind monomeric IgE via Fc epsilon RI.

Human epidermal Langerhans cells (LC) bearing IgE are found in disease states associated with hyperimmunoglobulinemia E. When studying the mechanism(s) underlying this phenomenon, immunohistology revealed that a majority of epidermal LC from normal skin of healthy individuals can specifically bind monomeric IgE. IgE binding to LC could neither be prevented by preincubation of the tissue with monoclonal antibodies (mAb) against either Fc epsilon RII/CD23 or Fc gamma RII/CD32, nor by the addition of lactose. However, binding could be entirely abrogated by preincubation with the anti-Fc epsilon RI alpha mAb 15-1, which interferes with IgE binding to Fc epsilon RI alpha gamma transfectants. These observations indicated that IgE binding to epidermal LC is mediated by Fc epsilon RI rather than by CD23, CD32, or the D-galactose-specific IgE-binding protein. This assumption gained support from our additional findings that: (a) the majority of LC exhibited distinct surface immunolabeling with the anti-Fc epsilon RI alpha mAbs 15-1 and 19-1, but not with any of eight different anti-Fc epsilon RII/CD23 mAbs; and (b) transcripts for the alpha, beta, and gamma chains of Fc epsilon RI could be amplified by polymerase chain reaction from RNA preparations of LC-enriched, but not of LC-depleted, epidermal cell suspensions. In view of the preeminent role of Fc epsilon RI crosslinking on mast cells and basophils in triggering the synthesis and release of mediators of allergic reactions, the demonstration of this receptor on epidermal LC may have important implications for our understanding of allergic reactions after epicutaneous contact with allergens.

Detection of rabbit haemorrhagic disease virus particles in the rabbit liver tissues.

Liver tissues from rabbits experimentally infected with rabbit haemorrhagic disease virus (RHDV) were studied electron microscopically. The earliest change in hepatocytes of the rabbits infected with RHDV was hydropic degeneration. Rough endoplasmic reticulum was dilated with a mild increase in polysomes and cytoplasmic cisternae in degenerated hepatocytes. Characteristic cytopathological changes of necrotic hepatocytes included shrinkage of the cell body, formation of cytoplasmic vesicles, vacuoles or cisternae and karyolysis. A large number of viral particles resembling a calicivirus in size and morphology was demonstrated in the cytoplasm of many necrotic hepatocytes. The particles had accumulated mainly in the membrane-bound cisternae or scattered around the membrane-bound vacuoles of the necrotic hepatocytes. Western blot analysis demonstrated that RHDV antigen was present in the infected hepatocytes. RHDV particles were also detected by immunoelectron microscopy. Replicating patterns of RHDV particles and subsequent cytopathology resembled those in other calicivirus infections.
