An experimental study on the "double crush" hypothesis.

An experimental study investigated the vulnerability of peripheral nerves in the "double crush" situation. Canine sciatic nerves were studied by means of electrophysiologic and histologic examination. Compression was applied by the KEIO compression clamp, with compression force of 15 g (approximately, 27.6 mm Hg). An incomplete conduction block and mild axonal degeneration were induced by a single compression. However, a complete conduction block and severe axonal degeneration could be induced in half of the cases by the double compression. In these cases, the loss of nerve function after a double lesion was greater than the sum of the deficits after each separate lesion. A good therapeutic effect was obtained by removing all compression, but the effect was incomplete when either compression was retained. It is concluded that proximal compression of a nerve could lessen its ability to withstand further compression more distally.

Three cases of secondary leukemia. Clinicopathologic findings.

Three cases of secondary leukemia developing after chemotherapy and/or radiotherapy for myeloma, mycosis fungoides, and non-Hodgkin's lymphoma are reported. The first case was a 51-year-old man with IgG-lambda myeloma, treated with melphalan and prednisolone, who developed acute myelomonocytic leukemia 54 months after the diagnosis of myeloma. The second case was a 54-year-old woman with mycosis fungoides treated with radiation, predonine, and cyclophosphamide, who developed acute megakaryoblastic leukemia 298 months after the diagnosis of mycosis fungoides. The third case was a 35-year-old woman with stage IV non-Hodgkin's lymphoma treated with VEMP who developed acute myelogenous leukemia 26 months after the diagnosis of malignant lymphoma. All cases showed pancytopenia and two of three cases had morphologic abnormality in several hemopoietic cell lineages in the leukemic stage. There is a possibility that second malignancies are an increasingly recognized complication in the patients treated with a large amount of chemo-radiotherapy.

Hodgkin's disease accompanied with thymoma.

An autopsy case of Hodgkin's disease accompanied with thymoma is reported. The patient died of acute interstitial pneumonitis 9 years after the development of lymphadenopathy. On post-mortem examination thymoma was recognized. We believe that the present case is a very rare Hodgkin's disease complicating thymoma. The occurrence of second neoplasia in Hodgkin's disease is also discussed.

Immunoelectron microscopical study of non-lymphocytic leukemic cells. Expressing human granulocyte antigen defined by monoclonal antibody 1G10.

The cells from 5 cases of non-lymphocytic leukemia were investigated by immunoelectron microscopy using an anti-granulocyte antibody, clone 1G10 (New England Nuclear), to clarify the nature of immature leukemic cells. Reaction products on the surface of the leukemic cells were composed of two layers, an inner translucent and an outer dense granular zone. Although non-leukemic cells of the granulocytic series in various stages of maturation between promyelocytes and neutrophils were all positive for the antigen-antibody reaction, non-neoplastic monocytes and lymphocytes were all negative. Using this method, it was possible to diagnose the undifferentiated leukemic cells that were negative for myeloperoxidase. Furthermore, the results of our present study suggest the possibility that the antibody 1G10 may be useful for distinguishing granulocytic cells from cells of monocytic lineage.

Familial hemophagocytic reticulosis. Clinicopathologic findings and cytochemical, immunohistochemical and electron microscopic studies.

Clinicopathologic findings of a case of familial hemophagocytic reticulosis (FHR), for which splenectomy, and biopsy of the liver and mesenteric lymph nodes were performed, is reported. Histologically, the proliferated cells consisted of not only histiocytes showing severe erythrophagocytosis, but also lymphoid cells of various sizes. The lymphoid cells had atypical features including mitotic figures and convoluted nuclei, and were suspected to be T cells as a result of immunohistochemical studies. We therefore consider that FHR is a special type of lymphoproliferative disease accompanied by severe proliferation of erythrophagocytic histiocytes rather than being a true histiocytic disease. Cytochemical and electron microscopic findings of the involved spleen are also presented.

Effect of spergualin in autoimmune disease mice.

We investigated the effect of spergualin (SGL) upon the development of spontaneous systemic lupus erythematosus-like lesions in male MRL/MpJ-lpr/lpr mice. SGL was administered ip at doses of 2.5, 5 and 10 mg/kg to two groups of mice. One group received SGL prophylactically from 7 to 21 weeks of age. The other group received SGL curatively from 13 to 27 weeks of age. The occurrence of lupus lesions was characterized by enlarged lymphoid organs, high anti-DNA titer and blood urea nitrogen, and severe glomerular nephritis. In both groups these characteristics were significantly suppressed by SGL in a dose-dependent manner. This inhibitory activity was greatest at a dose of 10 mg/kg. These findings suggest that SGL has prophylactic and curative effects against lupus lesions in autoimmune disease in mice.

Enhancement of colony formation of mouse bone marrow cells by ubenimex.

Ubenimex enhanced colony formation of bone marrow cells from CDF1 mice induced by L920 cell supernatant, which shows a macrophage-colony-stimulating activity (M-CSA), and also enhanced the colony formation induced CDF1 mouse spleen cell conditioned medium, which shows a granulocyte and macrophage-colony-stimulating activity. The maximal effect was obtained at 0.01 microgram/ml. But, ubenimex showed no effect on the nature of the colonies induced by each CSA. By preincubation of the bone marrow cells with ubenimex, M-CSA-induced colony-forming and the M-CSA-binding activities of the cells were increased. These results suggest that ubenimex enhances the CSA-induced colony formation of bone marrow progenitor cells of CDF1 mouse by increasing the amount of the CSA-binding to the cells.

New monoclonal antibodies specific for the guinea pig line 10 hepatocarcinoma.

Monoclonal antibodies reactive to line 10 (L10) hepatocarcinoma cells, but not to L2C leukemia cells, of strain 2 guinea pig were produced and characterized. Complement-mediated cytolysis assay and quantitative absorption analysis revealed that one of the monoclonal antibodies, 3C4 antibody (IgG2a), was highly specific for L10. Neither normal guinea pig tissues including adult and fetal liver, nor other hepatomas of strain 2 guinea pig, line 1 (L1) and Hepatoma III, were reactive with the 3C4 antibody. Another antibody, 2E4 (IgM), was reactive to the liver, kidney and spleen cells but not to the thymocytes, lymph node cells, brain and red blood cells of the guinea pig. The 2E4 antibody reacted to Hepatoma III but not to L1 cells. Therefore, 3C4 antibody is specific to a tumor-associated antigen of L10 hepatoma, and 2E4 antibody is cross-reactive with a certain differentiation antigen of normal tissues. The 3C4 antibody showed antibody-dependent cytotoxicity. Since 3C4 antibody was different in the isotype and the antigen recognized from a monoclonal antibody (D3) which was reported from the NIH in the USA, 3C4 is a new monoclonal antibody reactive with L10 hepatocarcinoma.
