Spatial distribution and temporal change of cytoplasmic free calcium in human platelets.

Our digital imaging microscope equipped with a microspectrofluorometer revealed in single resting human platelets the existence of continuous Ca2+ gradient increasing towards the plasma membrane (frequency; 100%) and discontinuous ones (Ca2+ plateaus) in the endoplasmic regions (frequency: 70%). An average cytoplasmic free Ca2+ concentration ([ Ca2+]i) in a whole cytoplasm was 72 +/- 7 nM, ranging from 30 nM in the lowest to 150 nM in the highest region just beneath the plasma membrane. When stimulated with thrombin, [Ca2+]i uniformly increased to the average [Ca2+]i of 300 nM and these gradients disappeared. This [Ca2+]i transient was followed by the sustained increase in [Ca2+]i in both single cells and cell suspension.

Common interneurones mediating cortical and tectal excitation of abducens motoneurones in the cat.

Tectal and cortical effects on abducens motoneurones were examined with intracellular recording techniques in cats under chloralose anaesthesia. Abducens motoneurones exhibited disynaptic EPSPs after stimulation of the contralateral superior colliculus and cerebral peduncle. The tectal disynaptic EPSPs were observed invariably in all motoneurones tested, while the peduncular EPSPs were observed only in 40% of motoneurones after stimulation of the contralateral cerebral peduncle. However, the tectal disynaptic EPSPs were consistently facilitated by conditioning peduncular stimulation in all motoneurones tested. These results indicated that the disynaptic excitatory tecto-abducens and cortico-abducens pathways shared common premotor interneurones. The common interneurones which mediated the tectal and cortical disynaptic excitation of abducens motoneurones were explored in the pons. These interneurones were identified by the criteria that they were fired monosynaptically from both the tectum and the cerebral peduncle and were activated antidromically from the abducens nucleus. Systematic threshold mapping for the antidromic activation in and around the abducens nucleus indicated that they gave off many collateral branches in the nucleus. Such neurones were found in the nucleus reticularis pontis caudalis, being distributed in the area extending 0.8-3 mm rostral to the rostral pole of the abducens nucleus, 1.3-2.7 mm deep from the dorsal surface of the brain stem, and 0.8-1.8 mm lateral from the midline. The present experiments strongly suggest that a group of neurones in the paramedian pontine reticular formation make direct excitatory connexions with abducens motoneurones and play a role of common interneurones that transmit both tectal and cortical commands.

The psychiatric acute observation unit in a general hospital.

A general hospital based psychiatric acute observation and treatment unit is described, and its operation over a one year period is reviewed. This seven-bed unit is designed to provide constant nursing observation and management, as well as intensive psychiatric treatment, in a secure, safe, and quiet environment for the most acutely psychiatrically ill patients. During the year under review, the unit had 330 admissions. Demographic, clinical, and treatment data are presented, and management of disturbed behavior, staff selection, and acceptance of the unit are discussed.

Enhanced multiplication and transmission of Streptococcus pneumoniae in guinea pigs by Sendai virus infection.

It was demonstrated that the transmission S. pneumoniae in guinea pigs was remarkably promoted by the combined infection with Sendai virus in the following experiments. When guinea pigs infected with S. pneumoniae alone (infector) were cagemated with non-treated guinea pigs (contact) for 2 and 4 weeks, only 2 of 30 contacts were infected with the organism. On the contrary, when the contact guinea pigs were infected with Sendai virus and immediately cage-mated with the infectors, the pneumococcal infection occurred in 25 of 30 contacts during 2 to 4 weeks period. In the experiment in which 30 non-treated contacts were cage-mated with pneumococcal infectors for 4 weeks and then infected with Sendai virus, no pneumococcal infection was demonstrated in the contacts, suggesting no presence of latent infection of the organism in the contact guinea pigs. Twenty-five of 30 contacts suffered from pneumococcal infection when they were exposed to Sendai virus for 2 weeks and then cage-mated with infectors. The multiplication of S. pneumoniae in the respiratory tract of the guinea pigs was remarkably enhanced by combined infection with Sendai virus. Namely, a 1000-fold increase in the number of organism resulted in the guinea pigs suffered from combined infection as compared with that in the animals received pneumococcal single infection.

Antinociceptive activity induced by tizanidine and alpha 2-adrenoreceptors.

Tizanidine [5-chloro-4-(2-imidazolin-2-yl-amino)-2,1,3-benzothiadiazole] is able to increase the pain threshold in the tail-flick test in mice. The effect of tizanidine was investigated after treatment of mice with drugs influencing central monoaminergic and GABAergic mechanisms. A drug that inhibits the synthesis and storage of monoamines and drugs that cause specific lesions of monoaminergic neurons had no consistent effect on the antinociceptive action of tizanidine. The action of tizanidine was antagonized by the alpha 2-adrenoreceptor antagonist, yohimbine, but not by the alpha 1 antagonist prazosin, nor by dopamine, serotonin and GABA receptor antagonists. These results indicate that the antinociceptive action induced by tizanidine may be mediated by alpha 2-adrenoreceptors.

Epizootiological studies of Salmonella typhimurium infection in guinea pigs.

In two natural outbreaks of S. typhimurium infection in guinea pigs, frequent isolations of the organism from the conjunctiva and cervical lymph nodes and high incidences of the conjunctivitis and abscess formation in the cervical lymph nodes were shown, suggesting more importance of the conjunctiva as infection route than oral route. These features in salmonellosis of guinea pigs were tried to reproduce in experimental infections by conjunctival and oral inoculations of 10(2) and 10(6) cells of 4 different strains of S. typhimurium and also by contact infection simulating natural conditions. As the results, it was demonstrated that guinea pigs were more susceptible to the conjunctival infection than the oral infection, because higher infection rates and more frequent incidence of abscess formation in the cervical lymph nodes as well as conjunctivitis were produced by the conjunctival inoculation than the oral inoculation of the organism. Main localization sites of the organism were the cervical lymph nodes, conjunctiva and upper respiratory tract in conjunctivally inoculated guinea pigs but more widely distributed in orally infected ones. These findings were common in animals infected with 4 different strains of S. typhimurium and also in contact infection. Thus the conjunctiva was regarded as an important route of S. typhimurium in guinea pigs.

Comparison of alpha-adrenoceptor involvement in the antinociceptive action of tizanidine and clonidine in the mouse.

The effect of drugs that influence the opioidergic and monoaminergic neuronal systems on the antinociceptive action of tizanidine [5-chloro-4-(2-imidazolin-2-yl-amino)-2,1, 3-benzothiodiazole] was compared with their effect on the action of clonidine. The potency of the clonidine-induced antinociceptive action was 1.83 and 7.75 times greater than that of tizanidine in the tail-flick and acetic acid-induced writhing tests, respectively. The action of tizanidine and clonidine was completely antagonized by pretreatment with yohimbine, an alpha 2-adrenoceptor blocker, but not by prazosin, an alpha 1-adrenoceptor blocker. Other alpha-adrenoceptor blockers, phenoxybenzamine and phentolamine, also attenuated the action of tizanidine and clonidine but the potency of these drugs was less than that of yohimbine. An opioid antagonist (naloxone), drugs influencing the serotonergic neuronal system (p-chlorophenylalanine, 5,6-dihydroxytryptamine, cyproheptadine), and drugs influencing the catecholaminergic system (alpha-methyl-p-tyrosine, diethyl-dithiocarbamate, 6-hydroxydopamine, haloperidol) showed no effect on the action of tizanidine and clonidine. From these results, it appears that alpha 2-adrenoceptors might be of importance in mediating the tizanidine and clonidine antinociceptive action in the tail-flick test.
