Effects of nonhypotensive hemorrhage on renal organ and urinary clearances of vasopressin in the dog.

A study was undertaken to investigate the effects of moderate nonhypotensive hemorrhage on the renal organ and urinary clearances of vasopressin in anesthetized dogs. A nonhypotensive hemorrhage was conducted in nine dogs by withdrawal of 12 ml/kg arterial blood over 10 min. This reduction in blood volume increased the plasma vasopressin concentration from a prehemorrhage value of 4.3 +/- 0.5 to 12.5 +/- 3.4 microU/ml (P less than 0.01) in the 15-min period immediately after hemorrhage and to 8.1 +/- 1.4 microU/ml (P less than 0.01) in the 45- to 60-min period after hemorrhage. The increased plasma vasopressin concentration was not associated with changes in either plasma osmolality or mean arterial blood pressure. The urinary excretion of vasopressin increased significantly after hemorrhage (P less than 0.01, 0-15 min after hemorrhage; P less than 0.05, 45-60 min after hemorrhage) and correlated significantly with the plasma vasopressin concentration (r = 0.92; P less than 0.001). However, this moderate nonhypotensive hemorrhage did not change the renal vasopressin extraction ratio or the renal organ and urinary clearances of vasopressin from their prehemorrhage values of 0.27 +/- 0.02, 2.00 +/- 0.20 ml/min X kg, and 1.43 +/- 0.13 ml/min X kg, respectively. At no time were these values different from those in nine time control dogs. Thus, changes in the renal handling of vasopressin do not contribute to the increase in the plasma vasopressin concentration after moderate nonhypotensive hemorrhage.

Regulation by estrogen and progesterone of protein kinases in rabbit myometrium.

We report on hormonal regulation of protein kinases in the rabbit myometrium. The injection of estrogen or progesterone produced 5.2 +/- 0.7- or 4.7 +/- 0.4-fold increases, respectively, of the total activity when histone was used as the substrate in the cytosol fraction of the tissue. The analysis of the activity by DEAE-cellulose column chromatography revealed that estrogen had no effect on increases in the levels in both types I and II cAMP-dependent protein kinases, on the basis of mg protein, while progesterone produced a 50% decrease in type I cAMP-dependent protein kinase and a 50% increase in type II cAMP-dependent protein kinase. Thus, the activity ratio of types II to type I did not change by treatment with estrogen and increased from 2.8 to 7.9 with progesterone administration. As the activity patterns by treatment with human chorionic gonadotropin closely resembled those seen with progesterone, our findings in case of the latter hormone are probably valid. The results indicate that the levels of protein kinases in the myometrium are altered in response to estrogen and progesterone and may possibly be involved in the physiological functions of the tissue.

[Studies on the behavioral pharmacology of TRH-T in cytosine arabinoside-induced ataxic mice: a comparison with genetically ataxic mice].

The effects of thyrotropin releasing hormone tartrate (TRH-T) on the behavior of cytosine arabinoside (Ara-C) induced ataxic mice were studied. The ataxic mice were prepared by injecting 50 mg/kg/day subcutaneously on the 2nd, 3rd and 4th postnatal days. Spontaneous motor activities were measured with a movement spectrum analyzer (ANIMEX-2) and the degree of ataxic gait determined by an open-field study. Cerebella from these animals were subjected to histopathologic examination at 4, 8 and 12 weeks of age. Ataxic gait became improved moderately and spontaneous motor activities increased slightly following administration of TRH-T. The effects of TRH-T on ataxia and spontaneous motility in the Ara-C injected mice varied with the age, and resembled those seen in weaver mice but not those in the Rolling mouse Nagoya. The results also suggest that the effects of TRH-T on the behavior of ataxic mice correlate with the extent of pathologic changes evoked in their cerebella.

Hormone treatments and pregnancy alter myosin light chain kinase and calmodulin levels in rabbit myometrium.

Myosin light chain kinase activity and the amount of calmodulin were measured in rabbit myometrium after the rabbits had been treated with oestrogen, progesterone and human chorionic gonadotrophin, and also during pregnancy. Injections of oestrogen and progesterone produced significant increases of of 2.8- and 2.1-fold in the enzyme activity/mg protein and of 1.4- and 1.2-fold in the calmodulin content/mg protein respectively. The increases seen with oestrogen treatment were inhibited by cycloheximide administration, suggesting that protein synthesis is involved in the process. Calmodulin increased about twofold in the particulate fraction under conditions of oestrogen and progesterone treatments and the raised level of calmodulin detected in the cytosol fraction was not due to translocation from the particulate fraction. During pregnancy the specific enzyme activity significantly increased in comparison with the values seen in samples from the non-treated rabbits. These findings suggest that myosin light chain kinase and calmodulin in the myometrium are under hormonal regulation and may play a role in expulsion of the fetus at the latter phase of pregnancy.

[Conditions suitable for chemotherapy of lung cancer].

The influence of several factors for response to Chemotherapy and survival was analysed in 310 patients with inoperable lung cancer who were treated by single or combination chemotherapy. In small cell lung cancer, survival was strongly affected by the results of induction chemotherapy, and combination chemotherapy with three or four drugs was most effective. In non-small cell lung cancer, performance status and body weight loss on admission were important factors for response to chemotherapy and survival. Combinations did not improve the response rate in non-small cell lung cancer.

Flexibility and rigidity of left-handed Z-DNA.

The local variation of torsional angles and helical parameters in Z-DNA was analyzed. The sugar phosphate backbone is fairly rigid but the angles at GpC linkage are more changeable than those at CpG linkage in order to form a variety of structures. The water channel at minor groove is important to stabilize and retain the novel Z-DNA helix.

[Evaluation of steroid hormone sensitivity of human breast cancer cells in culture (MCF-7) by a colony formation method].

The effects of steroid hormones on the colony forming activity of a human breast cancer cell line (MCF-7) were studied. Endrogeneous estrogens in the fetal calf sera used for culture were deprived using the dextran-coated charcoal, and only batches of E2-deprived serum were chosen with which there was a significant increase in the plating efficiency after E2 (estradiol-17 beta, 10(-8) M) treatment. Progesterone showed little effect on the growth of the cells. not being influenced by the addition of E2. The capacity of the cells to form colonies was stimulated by androgens (5 alpha-dihydrotestosterone, testosterone) only in the absence of E2. Dexamethasone had suppressive effects regardless presence of E2. Thus, MCF-7 cells showed variable responses to these steroid hormones, although the cells have specific receptors to the hormones. This colony formation method of MCF-7 cells may be a useful model system for the study of hormonal and non-hormonal anti-cancer agents.

Nephropathy induced in rats by repeated intravenous injections of rabbit anti-FX1A antibody.

Repeated intravenous injections of small amounts of rabbit anti-rat FXIA antibodies (RAFA) induced a renal damage with the morphological features of epimembranous glomerulonephritis with significant and persistent proteinuria. These findings indicated that repeatedly injected RAFA accumulated in glomeruli and that there seems to be a threshold in quantity of RAFA localized in glomeruli to induce renal damage with proteinuria.
