Isolation of non-cultivable acid-fast bacilli in sphagnum and moss vegetation by foot pad technique in mice.

In the former leprosy-endemic coastal area of Norway, 122 samples of sphagnum and moss vegetation were collected from 6 biotopes and examined for non-cultivable AFB by foot pad inoculation. Of the 759 foot pads examined, 20% contained non-cultivable AFB. A significantly higher frequency was found in a habitat where Sphagnum cuspidatum was preponderant, the sphagnum species from which the maximum yield was obtained. The bacteria were polymorphous, solidly staining AFB, which multiplied in passage in foot pads while they could not be cultivated on the conventional media for mycobacteria. Efforts are continuing to identify these AFB by biochemical methods and by inoculation into nine-banded armadillos.

[Selective caesarean section for breach presentation - bearings on morbidity of survivors neonatal period (author's transl)].

The authors have continued their own analysis of deliveries from breech presentation by studying childhood morbidity of 1,094 single infants who had survived the neonatal period, between 1966 and 1975. Particular attention was given to the impact of selective caesarean section on primiparae, beyond completion of the 36th week of pregnancy. The two reported periods between 1966 and 1971 as well as between 1972 and 1975 were compared for that purpose. -The frequency of caesarean sections went up from 11.6 to 52.2 per cent. Severe parturitional trauma, such as fractures or plexal paralysis, were prevented, but hypoxia was aggravating. No accurate assessment was possible of the full role played by hypoxia and of its importance. A conspicuously high number of children delivered by caesarean section exhibited neurological symptoms, during the first days after birth and even up to the date of leaving hospital. No relationship was found to exist between morbidity and parity. Conclusions with great relevance to practice were drawn from these findings.

[Effects of propranolol and metoprolol on beta-adrenergic metabolic responses (author's transl)].

In ten healthy volunteers infusion of 7 micrograms/kg orciprenaline produced typical changes in carbohydrate and fat metabolism due to beta-adrenergic stimulation. Pretreatment with intravenously administered propranolol or metoprolol significantly inhibited orciprenaline-induced changes in systolic blood pressure, heart rate and plasma FFA, potassium, glucose and glycerol levels. Propranolol (0.1 mg/kg) seemed to be more effective than metoprolol (0.2 mg/kg) even in inhibiting predominantly beta 1-adrenergic responses.

A membrane glycoprotein of aggregating Dictyostelium cells with the properties of contact sites.

Aggregating cells of Dictyostelium discoideum form EDTA-stable contacts which are blocked by a Fab (antigen-binding fragment) preparation from antisera raised against membranes. The target site of the blocking Fab fragments has been identified as a specific glycoprotein. In this paper its purification, carbohydrate and amino acid composition are described. Purification was 800-fold, starting with cells lysed by digitonin. The plasma membranes, preserved as ghosts by this treatment, were purified in a two-phase system and extracted with butan-1-ol. The water phase contained predominantly concanavalin-A-binding glycoproteins and was particularly rich in contact sites A. These were further purified on DE-cellulose and sucrose gradients. Sodium dodecylsulphate/polyacrylamide gel electrophoresis of the purified material revealed one major glycoprotein band in the molecular weight region of 80 000 to 90 000, depending on the acrylamide concentration. The sugars found in contact sites A were mannose, N-acetylglucosamine, fucose, and possibly glucose. The protein moeity contained 8% proline and was particularly rich in hydroxy amino acids.

Cultivable mycobacteria in sphagnum vegetation of moors in South Sweden and coastal Norway.

Intact sphagnum vegetation from moors in south Sweden and coastal areas of west Norway contained cultivable mycobacteria in 32% and 30% of the specimens, respectively. This frequency of specimens is lower than the 50% previously found in the partly altered moors of northwestern Germany, but the Scandinavian moors contained a larger variety of species. On both intact and altered moors M. chelonei and M. sphagni sp. nov. were found, the latter a homologous group of 151 strains. In south Sweden the highest frequency was found in S. balticum, S. recurvum. S. tenellum and S. compactum & molle. (40-65%). In coastal Norway the highest frequency was found in S. rubellum (48%) which offers favourable conditions for the accumulation of solar energy due to the red brown colour in the upper parts. Combined with a high humidity in coastal Norway in summer, this may contribute to the growth of mesophilic mycobacteria. A significant affinity of M. chelonei to S. tenellum was stated.

In vivo and in vitro studies on endothelial regeneration.

Endothelial regeneration was studied in rabbit aorta after intra-arterial balloon catheterization. Most of the regenerated endothelium originated from existing branches which was assessed by the Evans-blue uptake pattern and confirmed by transmission and scanning electron microscopy. Glucocorticoid treatment enhanced re-endothelialization whereas hyperlipemic diet inhibited. Sera from minipigs fed an atherogenic diet consistently have less ability than sera from control pigs to stimulate in vitro the regeneration of wounded endothelium-like monolayers of 3T3-B cells. The deficiency is probably due to an inhibitor which appears and disappears with changes in the diet.

[The effect of preventive cesarean section in breech presentation on perinatal mortality].

This study is based upon 1229 infants delivered from breech presentations who were investigated for the influence of the prophylactic Caesarean section carried out with primiparae after their 36th week of pregnancy in 1972, on perinatal mortality. The periods under report, i. e., 1966 to 1971 and 1972 to 1975, were compared with one another. The incidence of Caesarean sections carried out in breech presentations increased from 11.6% to 52.2%. With a premature birth rate of 22.1% and 20.9%, respectively, the uncleaned perinatal mortality decreased from 109.2% to 96.5%, whereas the cleaned one dropped from 73.3% to 64.2%. As to mature infants delivered vaginally or by Caesarean section, there was no significant difference in death rates. The worse prognosis for breech presentation infants given birth by pluriparae could be confirmed. The high portion of underweight infants amounting to 78.0% and 82.9%, respectively, in the total number of perinatal deaths illustrates the main problem of the breech presentation births.

[Results obtained from more intensive antenatal and intranatal attention to gemini pregnancy at gynaecological hospital of Karl-Marx-Stadt region (author's transl)].

Studies undertaken into gemini births between 1968 and 1977 have shown that reduction in perinatal mortality has been obtainable from high-intensity programmes of antenatal and intranatal attention to multiparae. Perinatal mortality had accounted for 13.4 per cent, between 1968 and 1974, and dropped to 7.4 per cent, between 1975 and 1977. This achievement is analysed and discussed. More improvement may best be expected from earlier detection of gemini pregnancy and earlier action of intensive care.

[Parturition of women with cardiac defect (author's transl)].

Reported in this paper are 115 childbirths of 98 women with concomitant cardiac defect hospitalised in the Obstetric Department of the Regional Hospital of Karl-Marx-Stadt, GDR. Coincidence of childbirth and defect accounted for 0.3 per cent, with mitral faults clearly taking first position with 40 per cent. Forty-seven per cent of the defects were congenital. The cases were subdivided by severity according to the N.Y.H.A. scale, with 76 per cent coming under State I, 19 under State II, and only five under State III.--Spontaneous births occurred in 56 per cent, while 27 per cent were forceps deliveries and 13 per cent caesarean sections, but no section performed for purely cardiac indications. No remarkable complications were experienced intra-operatively nor postoperatively. Five per cent (five cases) of all vaginal deliveries had to be completed by vaginal extraction for cardiovascular complications, including three cases of cardiac defects in State III.--No peripartum loss of cardiac patients was recorded. Two patients with State III mitral defects, however, died of aggravating cardiovascular decompensation, five or six years after delivery. Perinatal mortality was as low as 0.8 per cent. Premature births accounted for 13.6 per cent, while hypotrophic infants were found in 6.8 per cent of the cases under review.--The above findings and their implications are discussed and conclusions drawn for obstetric practice, with some emphasis being laid on programmed delivery as the optional method, more generous indication of primary section for State III cardiac defects, and long-term aftercare in the context of internal medicine.

[A new method for determination of cadmium in plasma (author's transl)].

Cadmium can be isolated from plasma by formation of negative charged iodide complexes bound to an anion exchange resin. Elution from the resin gives a cadmium solution with only a few interfering substances in an uniform matrix. Thus an undisturbed determination by flameless atomic absorption spectrometry is achieved. The mean of the natural cadmium level found in plasma was 1.9 +/- 0.8 ng/ml, within the normal range between 1.1 and 3.3 ng/ml.

A specific glycoprotein as the target site of adhesion blocking Fab in aggregating Dictyostelium cells.

During the acquisition of aggregation competence a new antigen appears on the surface of Dictyostelium cells. Univalent antibody fragments (Fab) against this antigen render the cells unable to form the specific type of cell adhesion which is characteristic of aggregating cells. This membrane constituent has been purified and identified as a concanavalin A-binding glycoprotein present at about 2 X 10(5) copies per cell.

The structure of human-plasma low-density lipoprotein B. An X-ray small-angle scattering study.

1. X-ray small-angle scattering of human plasma lipoprotein B of the low-density fraction (rho = 1.016--1.060 g.cm-3) has been recorded to high precision at different electron density contrasts. 2. The overall structure of the particles is characterized by a quasi-spherical shape and radial symmetry. A maximum diameter of 23 nm and a molecular weight of 2.4 X 10(6) have been determined. 3. The internal structure is described in terms of a model consisting of spherical layers with different electron densities indicating that the neutral lipids are arranged in the core of the molecule up to a radius of about 8 nm surrounded by a monolayer of free cholesterol, phospholipids and protein. The neural lipids are shown to be in an ordered, liquid crystalline state at 4 degrees C and to undergo a thermotropic transition into a disordered state at higher temperatures.
