Experiments on rabbits proved that amidopyrin, sulphatizonicotinoilamine-guanidine, chlorpromazine, triphthazine, euphylline and tedamine inhibit aggregation of thrombocytes and unidirectionally have an effect on the carbohydrate metabolism enzymes in these cells, both in vitro and in vivo following their single and coursewise introduction. They also activate the lactate- and glyceraldehide-3-phosphate-dehydrogenase and reduce the activity of glucose-6-phosphate-dehydrogenase.
Tests set up on rats and rabbits showed the ability of phospho-enolpyruvate and pyruvate-kinase to lower the aggregation of thrombocytes and prevent formation of experimental microthrombosis. The joint introduction of these compounds in most experiments resulted in potentiation of the antithrombotic effect.

The capacity of 25 different amidines, guanidines and biguanidines to suppress the thrombocytes aggregation, induced by adenosinodiphosphate, was studied. Good prospects of a search for antiaggregation agents in this series of compounds are shown. N-benzyl-N1-propionylguanidine was found to reduce by more than 4 times the induced adhesiveness of the blood platelets.
It has been shown in experiments on Wistar rats that adrenaline inhibits both in vitro and in vivo the capacity of the arterial wall for synthesizing an antiaggregating substance (prostacycline). It is suggested that adrenaline takes a direct part in body thrombogenesis during emotional stress and in conditions associated with the increased catecholamine output in the blood.
Experiments on Wistar rats have shown that single oral administration of acetylsalicylic (300 mg/kg), nicotinic (3 mg/kg) acids and complamin (2 mg/kg) inhibits platelet aggregation. It has been established that acetylsalicylic acid inhibits, as an antiaggregation agent, platelet aggregation and synthesis of a prostacycline-like substance by the aortal wall, and decreases its antiaggregation activity. Nicotinic acid and complamin inhibit platelet aggregation but do not lower antiaggregation activity of the aortal wall. Therefore, these latter drugs compare very favorably with acetylsalicylic acid as antiaggregation agents.

The authors analyze the results of research on clinical pharmacology carried out at various institutions in this country. The overwhelming majority of works are concerned with clinical studies on the pharmacodynamics and pharmacokinetics of the drugs, their interaction, and detection of drug side effects. Special attention is paid to the improvement of the methods for determination of the pharmacological agents in biological fluids, and to the design of the maximally adequate parameters of assessing the pharmacodynamics of the main groups of the drugs. Considerable part of investigations is devoted to clinical pharmacogenetics, chronopharmacology, age pharmacology, prediction of the drug effects, approaches to the extrapolation of experimental data to the clinical trials of the drugs. The authors also provide evidence on the system of clinical trials of new drugs and detection of untoward effects of the medicines used.
Two mathematic models depicting the kinetics of induced platelet aggregation are suggested. The mechanisms of platelet aggregation involving the reactions of the inductor molecules with membrane receptors and intercellular interactions of platelets are discussed. Analytical expressions depicting the changes in the amount of platelets in the aggregation population and in the course of aggregation are offered. A procedure for calculating the kinetic parameters according to the experimental data is presented. The parameters of aggregation induced by different substances are calculated with the use of computers. It has been shown that application of mathematic stimulation provides valuable information both during the study of the mechanisms and for quantitative analysis of platelet aggregation.
