Differential effects of dietary eicosapentaenoic and docosahexaenoic fatty acids on lowering of triglyceride and cholesterol levels in the serum of rats on hypercholesterolemic diet.

This investigation was attempted in order to further clarify the effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on lipid metabolism in rats. Ninety-three percent ethyleicosapentaenoate (EPAconc) and 91% ethyl-docosahexaenoate (DHAconc) were used and supplied at the 3% level to a hypercholesterolemic basal diet which contained 5% lard. Four experimental diets were prepared from (1) 5% olive oil, (2) 2% olive oil +3% ethyl-linoleate, (3) 2% olive oil +3% EPAconc, and (4) 2% olive oil +3% DHAconc plus hypercholesterolemic basal diet. Young male rats were fed on these diets for respective periods of 10 and 20 days. Concentrations of serum lipids in rats fed on the diets (2), (3) and (4) were compared to those of the control group (diet (1]. Serum total cholesterol level was significantly lower only in the animals fed on DHAconc, whereas serum triglyceride concentration was significantly lower only in the animals fed on EPAconc. Serum phospholipid level was significantly lowered both in the animals fed on either EPAconc or DHAconc. Serum lipid peroxide concentration was elevated in these two groups of animals, while serum alpha-tocopherol concentration was lower in both groups of animals fed on EPAconc and DHAconc, respectively, although these findings were less marked in the rats on the EPAconc diet. From these findings, it is postulated that EPA and DHA have different effects on lipid metabolism in rats with hypercholesterolemia experimentally induced.

Malignant fibrous histiocytoma at site of bone infarction in association with DCS.

A malignant fibrous histiocytoma developed at the site of bone infarction in the right femur of a 51-year-old male who had worked as a helmet diver and had experienced two episodes of bends in his young days. The outcome was rapidly fatal despite amputation of the right thigh, and widespread metastasis was noted at the time of autopsy. Histopathological examination of the amputated femur disclosed an intimate relation between the old focus of bone infarction and the tumor tissue, which was composed of a mixture of fibroblast-like cells and histiocyte-like cells. In an analysis of the published cases of malignant fibrous histiocytoma associated with bone infarction, it was found that the patients with bone infarction who had worked under hyperbaric atmosphere are at the risk of developing such a malignant tumor. The prolonged reparative process following bone infarction seems to be an important factor in the pathogenesis of malignant fibrous histiocytoma.

[Therapeutic effect of Robaveron tablet (KN-7) on neurogenic bladder].

Therapeutic effect of Robaveron Tablet (KN-7) was studied on 128 patients suffering from neurogenic bladder. Robaveron Tablet was administered 6 tablets daily for 3 weeks. The effect was evaluated by the residual urine, subjective symptoms, cystometric findings, urethral pressure profile and uroflowmetry. Remarkable decrease of residual urine was observed regardless of position of injury. The pressure amplitude defined as the difference between the maximum voiding pressure and the maximum resting pressure increased on any group of neurogenic bladder. The effectiveness, rated as moderately improved or better was 76.9% for brain disorders, 58.3% for upper motor neuron lesion and 75.7% for lower motor neuron lesion of spinal injuries, 58.3% for peripheral nervous injuries and 100% for others. No remarkable abnormality was observed in the results of side effects or clinical laboratory tests.

[Comparative double-blind trial of KN-7 tablet and Robaveron injection in the treatment of neurogenic bladder].

The clinical effectiveness, safety and usefulness of KN-7 tablet as a new oral application of the prostatic extract, on urinary dysfunction of neurogenic bladder were compared with those of Robaveron injection by the double-blind test method. In the study, 2 tablets t.i.d. and a shot of intramuscular injection 1 ml a day were given successively for 3 weeks. A total of 233 cases were reported from 37 facilities belonging to the KN-7 Clinical Research Group. Some of them were excluded or dropped out. The number of cases used for analysing the effectiveness, safety and usefulness were 214, 232 and 215, respectively. There was no bias between the two groups with a significant homogeneity in the background. In the overall clinical effectiveness, the effective rate including excellent, moderate and slightly effective was 76.9% with KN-7 and 77.4% with Robaveron. In the clinical usefulness, the rate of usefulness of slightly useful or above was 75.0% with KN-7 and 75.7% with Robaveron. There was no significant difference between the two groups in the clinical effective and useful rates at a significant level of 5%. Side effects were observed in 1 of the 114 (0.9%) patients given KN-7 and 8 of the 118 (6.8%) patients given Robaveron. The incidence of adverse reactions with KN-7 was significantly lower than that with Robaveron. Based on the results, it was concluded that KN-7 tablets, 2 tablets t.i.d., would be as effective and useful as a Robaveron injection 1 ml daily and safer than the latter in the treatment of neurogenic bladder.

Identification of lactoferrin as an essential growth factor for human lymphocytic cell lines in serum-free medium.

A serum-free medium supplemented with growth factor(s) was devised to grow human lymphocytic cell lines. The medium was developed using human lymphocytic cell line, Bri 7 cells. In the process of constructing the medium, human lactoferrin was found to be an essential growth factor for the cell line. Human lactoferrin has higher growth stimulatory activity than human transferrin, and was sensitive to heat. Long-term cultivation of the cells was achieved in the defined medium supplemented with human lactoferrin only. The defined medium specifically supported the growth of various other human B- and T-lymphocytic cell lines but not the growth of various mouse lymphocytic cell lines. In lactoferrin-supplemented medium, the growth of some human cell lines were further stimulated by the addition of a combination of insulin, ethanolamine and selenium, or another combination of 2-mercaptoethanol and the above three factors. Bovine lactoferrin could be substituted for human lactoferrin.

Isolation and characterization of a new bacteriophage, KF1, immunologically cross-reactive with F-type pyocins.

Pseudomonas aeruginosa strain NIH S produced a bacteriophage, KF1, immunologically cross-reactive with F-type pyocins. Phage KF1 was neutralized by both anti-pyocin F1 and anti-pyocin F3 sera, although the efficiency was very low. About eleven polypeptides were detected by SDS-polyacrylamide gel electrophoresis of the phage. Most of the subunit proteins were different from those of F-type pyocins, but the molecular weights of minor subunit proteins P3 and P6 seemed to be the same as those of band 1 and band 5 of F-type pyocins, respectively. The head of the phage appeared to have an icosahedral structure, approximately 63 nm in diameter, with a long (190 nm, 11 nm wide and about 45 striations) flexuous tail connected to a fiber structure (about 53 nm in length). The density in CsCl and the sedimentation coefficient of the phage were 1.54 g/ml and 392S, respectively. Some other biochemical properties were described. The nucleic acid of the phage was linear, double stranded DNA of molecular weight 4 x 10(7). The density of the DNA in CsCl was 1.719 g/ml, the melting temperature was 95.4 degrees C. The guanine plus cytosine content was calculated to be 60 to 64%.

Phosphatidylcholine transfer activity in human erythrocyte hemolysate.

Phosphatidylcholine (PC) transfer activity was found in human erythrocyte hemolysate. The transfer activity was assayed by the ESR peak height increase when spin-labeled PC vesicles were incubated with egg yolk PC vesicles. The transfer activity was isolated from hemoglobin by an ion exchange chromatography followed by gel filtration. The partial purification resulted in a 405-fold increase in the specific transfer activity compared with that of the hemolysate. The molecular weight of the PC transfer protein was estimated to be 23,000 by gel filtration. The transfer activity was inactivated by heat-treatment at 75 degrees C for 10 min. Phosphatidylserine vesicles strongly inhibited the activity. Half-maximal inhibition occurred on addition of 0.24 mol% of phosphatidylserine vesicles to the incubation mixture. Ca2+ restored the activity. The transfer protein was quite similar to the PC transfer protein obtained from bovine liver cytosol.

Pharmacokinetics of cefotetan in normal subjects and patients with impaired renal function.

The elimination kinetics of cefotetan (YM09330), a new parenteral semisynthetic cephamycin derivative, were studied in eight healthy volunteers and 41 patients with renal insufficiency after the administration of a single 500-mg dose intravenously. Concentrations of cefotetan in serum and urine were determined by both bioassay and high-pressure liquid chromatography. The pharmacokinetic parameters for cefotetan were calculated on the basis of a two-compartment open model. Serum concentrations of cefotetan immediately after administration were approximately 180 micrograms/ml in all subjects regardless of function; however, serum concentrations during the beta-phase increased directly with the degree of renal impairment. The mean serum half-life during the beta-phase was 3.0 h in normal subjects as compared with 13.1 h in hemodialysis patients. There was a linear correlation (P less than 0.0001) between the elimination rate constant of cefotetan and creatinine clearance. The mean cumulative urinary recovery of cefotetan in the 24-h urine was 83.3% of the administered dose in normal subjects and decreased with reduced renal function.

Elimination kinetics of cefotaxime and desacetyl cefotaxime in patients with renal insufficiency and during hemodialysis.

The pharmacokinetics of cefotaxime, a new semi-synthetic cephalosporin for injection, was studied in 30 subjects with various degrees of renal function after a single 1-gram intramuscular injection. Serum and urinary concentrations of cefotaxime and desacetyl cefotaxime were determined by high pressure liquid chromatography. The pharmacokinetic parameters of cefotaxime were obtained using a one-compartment open model. The mean serum half-life of the parent compound (cefotaxime), 0.87 h in normal subjects, was prolonged to 2.35 h in hemodialysis patients. There was a significant linear correlation between the elimination rate constant of cefotaxime and creatinine clearance. The mean cumulative urinary recovery of the administered dose in the 24-hour urine was 51.7% as cefotaxime and 25.6% as desacetyl cefotaxime in normal subjects.
