Adrenal opioid proteins of 8600 and 12,600 daltons: intermediates in proenkephalin processing.

[Met]Enkephalin-containing proteins of 8600 and 12,600 daltons have been isolated from acid extracts of bovine adrenal medulla and purified to homogeneity, and their sequences have been determined by a combination of automated Edman degradation, tryptic mapping, and enzymatic time-course hydrolysis. The 8600-dalton protein contains one copy of the [Met]enkephalin sequence at the COOH terminus and the 12,600-dalton protein contains three copies of [Met]enkephalin, of which two are internal and the third is at the COOH terminus. They possess identical NH2-terminal amino acid sequences, suggesting that the 8600-dalton protein is derived from the 12,600-dalton protein by intracellular proteolytic processing. This is supported by results from tryptic maps of both proteins. Furthermore, chemical analysis of the tryptic peptides obtained from the 12,600-dalton protein indicates that it also contains the amino acid sequence that corresponds to a previously characterized enkephalin-containing polypeptide of 3800 daltons (peptide F) [Jones et al. (1980) Arch. Biochem. Biophys. 204, 392-395]. All three polypeptides appear to be intermediates in posttranslational processing of a still larger polyenkephalin precursor molecule, proenkephalin, and part of a biosynthetic pathway leading to smaller enkephalin-containing polypeptides and free enkephalins.

An 18,200-dalton adrenal protein that contains four [Met]enkephalin sequences.

An 18,200-dalton adrenal enkephalin-containing polypeptide was purified to homogeneity and shown to contain four internal [Met]enkephalin sequences. Protein sequence analysis and mapping of trypsin-derived peptides demonstrated that the previously described 12,600-dalton and 5,300-dalton adrenal enkephalin-containing polypeptides compose the amino-terminal and carboxyl-terminal sequences, respectively, of this larger protein. They are linked by two basic amino acid residues to form the entire sequence of the 18,200-dalton protein. This protein is the largest enkephalin-containing polypeptide yet isolated from beef adrenal medulla and serves as the precursor for several other previously characterized adrenal enkephalin-containing proteins and peptides.

[Studies on dental self-curing resins (19) - adhesion of 4-META/MMA-TBB resin to pretreated dentine (author's transl)].

A new 4-META/MMA-TBB adhesive resin has been studied and the effectiveness on adhesion to tooth substrates and dental alloys was reported previously. Here we are showing affection of cleaner or etchants on the adhesion between dentines and the 4-META/MMA-TBB resin. The adhesive strength becomes superior, 18 MPa, when dentines have been cleaned with a 10% citric acid-3% ferric chloride solution. On the other hand an etchant, phosphoric acid or citric acid alone, is not so effective as the mixed solution and the strength decreases to 6 MPa. Scanning electron microscopic study on pretreated dentine with several solutions is also carried out and it is found that debris on dentine should be removed but that in tubules is not necessary to be cleaned up to get good adhesion.

[Studies on dental self-curing resins (20) - adhesion mechanism of 4-META/MMA-TBB resin to dentine (author's transl)].

Scanning electron microscopic studies on dentine surface cleaned with a mixture of citric acid and ferric chloride, fractured surfaces parallel to the tubules of dentine and dentine with the cured adhesive and the partially demineralized fractured surfaces, tensile fractured surface of the joint on the dentine and cured adhesive resin tags impregnated in tubules were carried out to clarify the adhesion mechanism. Infiltration of monomers into dentine is more important to get good adhesion with dentine than the interlocking at dentinal tubules due to impregnated resin. Monomers with both hydrophobic and hydrophilic groups like 4-META promote the monomer infiltration. The infiltrated and cured adhesive layer in dentine is observed at the subsurface dentine zone. This zone has excellent resistance against acid demineralization.

[Studies on dental self-curing resins (22) - adhesion of 4-META/MMA-TBB resin to enamel (author's transl)].

Bovine enamels cleaned with citric acid and FeCl3 which had been brushed or ground were joined with PMMA rods with the 4-META/MMA-TBB cement. Their tensile adhesive strength was measured after post-treatments. Stable adhesive strength such as 6.2 MPa was obtained on an enamel which was brushed and washed with an aqueous solution of 10% citric acid-3% FeCl3 for 30 sec. even after a 60 times percolation test between 4 degrees C and 60 degrees C. It is adequate to clinics. The value increased to 16.1 MPa on ground enamel. These results suggest that phosphoric acid etching, essential nowadays, is not necessary for good adhesion to enamel. Monomers with both hydrophobic and hydrophilic groups like 4-META possess affinity with tooth substrates and promote the infiltration of cements into the enamel. The infiltrated monomers polymerized in situ and strong adhesion with the enamel took place. Comparison of the tag length formed in the enamel by cements, MMA-TBB, HNPM/MMA-TBB, Phenyl-P/MMA-TBB and 4-META/MMA-TBB supports this consideration.

[The antigastric ulcer activity of succinic acid mono-3-guaiazulenamide (TPH-3) (author's transl)].

Effects of TPH-3 on various experimental gastric ulcers in rats and guinea-pigs were studied and the following results were obtained. In the preventive experiments, such as the Shay's ulcer, pylorus-ligated aspirin ulcer and restraint and water immersion stress ulcer, TPH-3 (200 mg/kg i.d. or p.o.) showed a statistically significant inhibition. TPH-3 markedly inhibited the histamine-induced gastric ulcer in guinea-pigs and a dose-response relationship was obtained for doses of 12.5, 25 and 50 mg/kg p.o. TPH-3 showed the weak inhibition regarding therapy for serosa-seared gastric ulcer and the recovery process of restraint and water immersion stress ulcer in rats. TPH-3, dosing 100 mg/kg intraduodenally, significantly inhibited the gastric secretion in the pylorus-ligated rats. TPH-3 significantly inhibited the histamine, pentagastrin and carbachol-induced acid secretion in the stomach-perfused rats. In particular, TPH-3 showed strong sensitivity to the action of histamine. TPH-3 had no anti-ACh or anti-H1-receptor effects.

A normalized protein map of human plasma, and characterization of the normalized spots by microscale amino acid analysis.

A "normalized map" of human plasma proteins, which shows the standard distribution pattern of the proteins separated two-dimensionally in the absence of denaturing agents, is described. The pI and molecular mass of the protein represented by each spot in the normalized map are shown, and 84 of 128 spots are identified or tentatively identified as known plasma proteins. Of the several methods we used for the identification, the method of micro-scale amino acid analysis of the proteins extracted from the fixed, stained two-dimensional gels is described in detail. Patterns for human cerebrospinal fluid and urinary proteins are compared with the normalized map, the positions of proteins characteristic of each body fluid sample being shown.

Glyceroglucolipids of the mucous barrier of dog stomach.

Distribution of glyceroglucolipids in the mucous barrier of dog stomach fundus, body and antrum was investigated. Surface mucus lining and preformed intracellular mucus were obtained by instillation of the ligated stomach compartments with 2 M NaCl. Lipids were extracted from the dialyzed and lyophilized samples, and the glycolipids were separated into neutral and acidic fractions. The glyceroglucolipids contained in each fraction were purified into individual components by thin-layer chromatography and were quantified. The content of glyceroglucolipids (mumol glucose/g protein) in the antral portion of the stomach was 2.1 times greater than that of the fundus and 3.4 times greater than that of the body. All three areas of the stomach contained neutral and sulfated glyceroglucolipids. However, the level of sulfated glycerolglucolipids (mumol glucose/g protein) was three times higher in the antrum as compared to the fundus and four times higher as compared to the body. The neutral and sulfated glyceroglucolipids were present in a molar ratio of 1.0 : 0.7 in the fundus, 1.0 : 1.1 in the body and 1.0 : 1.5 in the antrum.
