[Influence of the surgical stress on graft-versus-host reaction in mice--the suppressor activity to GVH reaction induced by the surgical operation].

The influences of surgical stress on the local graft-versus-host reaction (GvHR) in F1 mice were studied. Skin incision to the F1 mice inflicted to 1 day prior to injection of parental spleen cells produced impairment of popliteal lymph node enlargement, however, this effect was not observed when GvHR was induced on the day 3 and 5 after operation. Spleen cells from hind limb amputated F1 mice exhibited suppressor activity on local GvH reaction when injected into naive syngenic F1 mice together. This suppressor activity was markedly detected 3 and 24 hrs after operation, then gradually reduced and completely disappeared on the day 7. These suppressor activity completely disappeared by the treatment with anti-Thy 1.2. antibody and complement. These results showed that GvH reaction was suppressed by the surgical stress, and this suppression was due to suppressor T lymphocytes in the spleen.

[The tumor--immunological significance of splenectomy for cancer therapy].

In animal experiment, 5 X 10(5) MH-134 tumor cells were transplanted s.c. on the back of the C3H/He mice. Three, seven, 14 and 21 days after tumor transplantation, splenectomy or sham operation were performed and the tumor growth and survival days were examined in each group. As the results, the tumor growth was inhibited and the survival days prolonged not only in the group splenectomized three days but also 21 days after tumor transplantation. Clinically, the effect of splenectomy in combination with immunotherapy on cell-mediated immunity and the survival rates were studied in the gastric cancer patients of upper and middle stomach with 90 cases of stage III and 48 cases at stage IV, totalling 138 cases who underwent total gastrectomy during 1965 and 1981. Immunotherapy was conducted with immunomodulator levamisole at a daily dose of 150 mg, three consecutive days every other week. As a result, splenectomy was not effective for advanced gastric cancer at stage III and in the patients spleen should be retained for immunotherapy. Splenectomy for gastric cancer at stage IV, particularly in combination with immunotherapy, produced augmentation of cell-mediated immunity and longer survival as well. Complications caused by splenectomy were small.

Effect of 16,16-dimethyl prostaglandin E2 and ethanol on the constituents of gastric mucus.

The effect of ethanol, 16,16-dimethyl prostaglandin E2 (DMPGE2), and ethanol after DMPGE2 pretreatment on the solubilization of protein, glycoprotein, and glycolipid constituents of gastric mucus was investigated. The Lucite chamber stomach-flap preparation was used in dogs whose basal H+ secretion was inhibited by intravenous cimetidine. Graded concentrations (5-80%) of ethanol produced a dose-dependent decrease in potential difference (PD) which was accompanied by an increase in the content of proteins, glycoproteins, and glycolipids of the instillates. This effect was most pronounced at 40% ethanol. Exposure of gastric mucosa to DMPGE2 at 0.01 microgram/ml had no effect on the transmucosal PD, and the content of investigated components in the instillates increased only slightly over the saline control levels. DMPGE2 at 0.1 microgram/ml, although it did not induce an evident fall of PD, evoked a moderate increase in glycoprotein and glycolipid liberation. Higher doses (above 1.0 microgram/ml) of DMPGE2 were associated with the fall of PD and the increased solubilization of proteins, glycoproteins, and glycolipids. Pretreatment of the mucosa with DMPGE2 in a dose of 1.0 microgram/ml diminished the liberation of the investigated components from the gastric mucosa by 5 and 10% ethanol, but did not prevent the changes evoked by higher concentrations (20-80%) of ethanol. These data indicate that DMPGE2 applied topically protects in part the gastric mucosa against the ethanol-induced solubilization of mucus constituents.

Purification of dipeptidyl-aminopeptidase IV from human kidney by anti dipeptidyl-aminopeptidase IV affinity chromatography.

Dipeptidyl-aminopeptidase IV (DAP-IV) was purified 850 fold with a yield of 16.5% from human kidney cortex. Only two chromatographic procedures of DEAE-cellulose and anti DAP-IV Sepharose were used. The purified enzyme was shown to be homogeneous by polyacrylamide gel electrophoresis in the presence of sodium dodecylsulfate. Among various substrates with the structure of X-Y-p-nitroanilide, Lys-Pro-Gly-Pro- and Arg-Pro-p-nitroanilides were hydrolyzed very fast by DAP-IV. Optimum pH of DAP-IV in human kidney was pH 8.7, Km value for Gly-Pro-pNA was 2.51 +/- 0.01 x 10(-4) M and V max was 66.6 +/- 0.7 mumol/min/mg protein. Diisopropylfluorophosphate completely inhibited DAP-IV at 0.1 mM. However, the enzyme was almost unaffected by N-ethylmaleimide, p-chloromercuribenzoate, iodoacetate, phenylmethylsulfonylfluoride, and several metals. The amino acid composition of DAP-IV purified from human kidney was similar to that of DAP-IV purified from pig kidney, liver and intestine. These results indicate that the properties of DAP-IV purified from human kidney are almost same as those from pig kidney.

The promotion of adhesion by the infiltration of monomers into tooth substrates.

The effectiveness of 4-methacryloxyethyl trimellitate anhydride (4-META) on the adhesion of an acrylic rod with etched dentine and enamel was studied. Etching of tooth substrates with a 10% citric acid-3% ferric chloride solution prior to the adhesion proved effective. Monomers with both hydrophobic and hydrophilic groups like 4-META promoted the infiltration of monomers into the hard tissue. The infiltrated monomers polymerized in situ and good adhesion with the tooth substrates took place. The tensile adhesive strength was 18 MPa on the etched dentine. Scanning electron microscopic studies suggested that the monomers possess affinity with the hard tissue. The good adhesion was not provided by the interlocking at the tubules as had been considered previously.

Enkephalin biosynthetic pathway: a 5300-dalton adrenal polypeptide that terminates at its COOH end with the sequence [Met]enkephalin-Arg-Gly-Leu-COOH.

An enkephalin-containing polypeptide of 5300 daltons has been isolated from extracts of bovine adrenal medulla and purified to homogeneity, and its sequence has been determined by a combination of automated Edman degradation and enzymatic time-course hydrolysis. The polypeptide contains a single [Met]enkephalin sequence followed by Arg-Gly-Leu-COOH, which forms the COOH terminus of the molecule. The enkephalin sequence is preceded by a Lys-Arg linkage, typical of prohormone cleavage sites. Posttranslational processing of the polypeptide at this site would lead to release of the octapeptide [Met]enkephalin-Arg6-Gly7-Leu8.
