Determination of tetracycline hydrochloride in presence of anhydrotetracycline by differential pulse polarography.

A differential pulse polarographic method is described for the determination of the antibiotic tetracycline HCl in the presence of its degradation product anhydrotetracycline. The method utilizes the large difference in their differential pulse polarograms at a peak potential of -1.39 V in 0.1 M phosphate buffer as the base electrolyte (pH 6.8). The assay was evaluated using synthetic mixtures and applied to the analysis of commercial tetracycline HCl samples. The results obtained with this method are in close agreement with those from the spectrophotometric absorbance ratio method.

Neonatal lupus erythematosus, late onset hypocalcaemia, and recurrent seizures.

A 6 day old infant had neonatal lupus erythematosus manifested by rash, haemolytic anaemia, and hepatosplenomegaly. His mother was asymptomatic until eight months of pregnancy. Between 7 and 10 weeks he had recurrent seizures with hypocalcaemia. Other causes of convulsions were excluded. By 14 weeks various abnormalities had largely disappeared.

Acute viral hepatitis, intravascular haemolysis, severe hyperbilirubinaemia and renal failure in glucose-6-phosphate dehydrogenase deficient patients.

Five patients with acute viral hepatitis developed severe intrasvascular haemolysis and unusually high levels of serum bilirubin (427 to 1368 mumol/l). All 5 had high fever, marked anaemia, reticulocytosis and neutrophilic leucocytosis. Three of them developed acute renal failure, which was of non-oliguric type in 2. The clinical course was protracted, but complete recovery occurred in 4 patients between 4 to 10 weeks. One patient with hepatic coma and oliguric renal failure died. Deficiency of the enzyme G-6-PD was confirmed in 4 cases. Massive haemolysis in the patients was probably induced by the administration of chloroquine and other drugs. Intravascular haemolysis should be suspected in patients with acute viral hepatitis, if they show unexplained anaemia and very high serum bilirubin levels, and measures to prevent renal failure should be instituted in such cases.

2-(2'-Hydroxyphenyl)-4-aryl-1,5-benzodiazepines as CNS active agents.

1-Aryl-3(2'-hydroxyphenyl)-propane-1,3-diones (III) have been prepared by the Baker-Venkataraman transformation of the corresponding 2-substituted benzoyl-oxyacetophenones (II). The aforesaid propane-1,3-diones (III) were condensed with o-phenylene diamine to give eight benzodiazepines (IV) which have been found to be very good MAO inhibitors (in vitro), and anticonvulsant and hypnotic (in vivo) agents. Some chemical leads on pharmacological activities have been observed.
