[Effect of a calcium influx blocker, verapamil, on the sensitivity of human bladder cancer cells to adriamycin and mitomycin C].

We have investigated the effect of verapamil on the cytotoxicity of adriamycin and mitomycin C with human bladder cancer cell lines T-24 and JTC-30. The concentration of verapamil was 10 M, and adriamycin and mitomycin C were used at the concentrations ranging from 10(-2) micrograms/ml to 10 micrograms/ml. The efficacy of drugs were evaluated using cytotoxicity and colony forming assays. Judging from the results of these assays, no significant difference was observed between the drugs in combination with verapamil and drugs alone. The results indicate verapamil has no enhancement for the cytotoxic activity of the anticancer drugs when cell lines T-24 and JTC-30 are used.

Schmorl's nodes. Correlation of X-ray and histological findings in postmortem specimens.

Histological examinations were conducted on specimens of 368 intervertebral discs (T11/12-L5/S1), using X-ray photography and discography. Specimens were obtained from 61 individuals (36 males and 25 females) whose ages ranged from 25 to 85 years. Fifty-four Schmorl's nodes were found in 28 of the patients (19 males and 9 females), and in 43 discs. In the peripheral regions of Schmorl's nodes, where the vertebral bodies were in contact with the node, growth of cartilaginous cells was seen in many cases. Thickened bone trabeculae were also seen in three nodes accompanied by syncytia. These three nodes were detectable by X-ray photography of all discs containing Schmorl's nodes, and 35 were subjected to discography. Among these discs, 10 showed a limited form of shadow and 25 showed a diffuse form, and Schmorl's nodes were detected in 11 that showed a diffuse form of shadow. It was possible to detect Schmorl's nodes when they were contiguous with a degenerated annulus fibrosus adjoining the nuclear cavity. Pathogenetically, the presence of Schmorl's nodes in patients of middle and advanced age is interpreted to be one of the symptoms of age-related changes in the cartilaginous plate.

[Urinary bladder tumor manifesting multiple bone metastases similar to prostatic carcinoma--a case report].

A 72-year-old patient was found to have a non-papillary bladder tumor extending from the vesical floor to the retrotrigone and manifested multiple bone metastases without evidence of any hematogenic metastases in other organs. The pattern of the above metastases were very similar to that of a prostatic carcinoma and suggests that the so-called specific pattern of bone metastases observed in prostatic carcinomas might be caused by the location of the original tumor and not by features of the prostatic carcinoma cells themselves.

Sequential instillation therapy with mitomycin C and adriamycin for superficial bladder tumors.

From October 1983 to September 1985, 84 patients with superficial bladder tumor (Ta, Tl, Tis) were treated with sequential instillation of mitomycin C (MMC) and adriamycin (ADM). Doses of 20 mg MMC on day 1 and 40 mg ADM on day 2 were instilled into the bladder and retained for at least 2 h; this was repeated once a week for 5 consecutive weeks. Patients who achieved complete response (CR), were randomized and underwent prophylactic treatment taking the form of either intermittent instillation of MMC or daily oral administration of 5-fluorouracil. Of 79 evaluable patients, 72 (91%) had received prior treatment for superficial bladder tumors, 69 (87%) had high-grade tumors, and 18 (23%) had non-papillary Tis. The overall response rate was 68%, made up of CR in 43 patients (54%) and partial response (PR) in 11 (14%). Patients with either five or more tumors or tumors larger than 1 cm showed a significantly lower response rate than those with fewer than five tumors and tumors smaller than 1 cm, respectively. There was no correlation between tumor growth pattern, tumor grade and response rate, though non-papillary Tis appeared to respond better than papillary tumors. A history of prior instillation therapy or of toxicity to this treatment had no significant influence on the response rate. Although no systemic toxicity was observed, 62 patients (74%) experienced cystitis and the treatment had to be discontinued within 4 weeks in 13 of 33 cases with severe symptoms. The preliminary conclusion of prophylactic treatment was that intermittent instillation of MMC was superior to 5-FU medication in reducing the recurrence rate for at least 2 years after the treatment.

[N-acetyl-beta-D-glucosaminidase (NAG) activity in human semen: its relation to gamma-glutamyl transpeptidase (gamma-GTP) activity in seminal plasma and reproductive tissues, and relation between seminal mucoprotein concentration and seminal NAG, and gamma-GTP activities].

N-Acetyl-beta-D-glucosaminidase (NAG) and gamma-glutamyl transpeptidase (gamma-GTP) activities in seminal plasma obtained from male infertile patients and these activities in reproductive tissues obtained from cryptorchids and patients with bladder tumors were studied. Seminal mucoprotein concentration was also measured. The mean NAG and gamma-GTP activity in seminal plasma was 2,092 +/- 794 mU/ml and 10,942 +/- 4,179 mIU/ml, respectively. The mean seminal mucoprotein concentration was 28.0 +/- 5.7 mg/ml. In seminal plasma, a significant correlation was obtained between NAG and gamma-GTP (p less than 0.001). and between gamma-GTP and mucoprotein concentration (p less than 0.05). However, there was no correlation between NAG and mucoprotein concentration. In the reproductive tissue high gamma-GTP activity was observed in the testicular tissue and in the external gland of the prostate. The activity in the epididymis was higher in the tail than in the head. High NAG activity was observed in the epididymal head and in the internal gland of the prostate. In the testicular tissue, NAG activity was low. These findings showed that in the seminal plasma NAG and gamma-GTP activities were closely related but in the reproductive tissue, these activities located in different areas.
