[Amino acid resorption and protein metabolism following total gastrectomy in the rat].

The investigations were carried out in rats by means of the amino acid 15N-glycinee 6 to 12 weeks after the gastrectomy. 10 rats were operated by Graham and 9 by Longmire, Gütgemann. 12 rats as a control group were not operated. The animals got a single dose 15N-glycin with the food in the beginning of the experiment. The excretion of the isotope was measured every twelve hours for 5 days. All rats had a positive nitrogen balance. The rats operated by Graham excreted with the urine significantly less 15N than the rats of the other two groups. These differences were caused by different fast adsorption of the amino acids likely, at this the rats operated by Graham absorbed retardingly evident qualified by the absent duodenal passage. These disturbances of the amino acid absorption are not aggravating, because the nitrogen balance and the protein synthesis rate of the animals were normal.

Sports-medical studies on parachute jumpers with particular reference to the behavior of heart rate.

The aim of the experiment was to record the heart rate tracings of three groups of parachutists - with manually operated parachutes, automatically operated parachutes and formation parachutists (sky divers) - during the jump and to compare these with the findings obtained during bicycle ergometer stress tests. A total of 112 parachute jumps involving 31 subjects were evaluated. All the subjects exhibited a significant increase in heart rate during the exit, canopy opening and landing phases. Qualitatively, the heart rate curves of the formation, manual and automatic parachutists showed a similar trend. Rises in heart rate during the opening of the parachute (for the formation parachutists) and during landing (for the automatic parachutists) were, however, particularly pronounced. The increase in heart rate can be principally ascribed to emotional tension and a state of anticipation rather than to physical stress. The comparative evaluation of the ergometer tests showed that as a general rule stress factors in the submaximal region occur during parachuting, but that in extreme situations heart rates in the maximal region of physical stress may well be expected.

On the pyridoxal-5'-phosphate stimulation of aspartate aminotransferase and alanine aminotransferase in serum and erythrocytes of patients undergoing chronic haemodialysis and with kidney transplants.

Aspartate aminotransferase (AST) determinations in erythrocytes of patients undergoing chronic haemodialysis and with kidney transplants showed that patients receiving vitamin B6 had a smaller relative stimulation rate of AST by pyridoxal-5'-phosphate (P-5-P). In contrast to these results in erythrocytes, the apoAST and apo-alanine aminotransferase (ALT) activities in serum were increased in patients as compared with those of healthy persons. The corresponding relative stimulation rates of AST and ALT by P-5-P addition to the reaction mixture were not changed in the haemodialysis patients, but in renal transplant recipients the relative stimulation rate of AST was significantly smaller and that of ALT was greater.

Behavior of heart rate and incidence of arrhythmia in swimming and diving.

Heart rate behaviour and the incidence of arrhythmia were recorded in 29 young subjects aged between 16 and 20 years using radiotelemetry equipment. The study consisted of four regimens: 100m freestyle swimming with and without an aqualung, up to 50m under water swimming without a breathing apparatus (skin diving) and 100m under water swimming with an aqualung (scuba diving). In the course of the swimming experiments the heart rate tracing exhibits three phases, namely: (a) a sharp rise in the first 10 s; (b) a reduction in the rate of increase, and (c) a constant plateau at approximately 184 bpm without an aqualung and 168 bpm with an aqualung. During apnoeic diving there is a slight increase in heart rate, followed by a rapid drop (vasovagal diver's bradycardia) and finally a plateau at approximately 55 bpm. Diver's bradycardia does not occur when the diver uses an aqualung; the heart rates then correspond to those observed during swimming (maximal rate approximately 169 bpm). Cardiac arrhythmia occurred in 18 instances (3 each during swimming with and without a breathing apparatus, 5 during diving with an aqualung and 7 during diving without an aqualung). 15 were cases of supraventricular extrasystoles, 12 occurred in the plateau phase. A man with a thorough endurance training exhibited in addition to supraventricular extrasystoles a transient bigeminy, substitutive AV systoles and an electric alternans during diving without an aqualung. He experienced no subjective feeling of impairment of performance, nor was there any objective reduction in efficiency.

Improved determination of D-glucaric acid in urine.

Using a simplex optimization procedure, we have elaborated a sensitive and reliable micromethod for determining D-glucaric acid, based on inhibition of glucuronidase activity by 1,4-D-glucarolactone, produced from D-glucaric acid by heating. The maximum attainable difference between glucuronidase activity in the reaction mixture with and without D-glucaric acid was used as an optimization criterion. From the optimization scheme, 1 mmol/L 4-nitrophenyl-beta-D-glucuronide and pH 5.0 were found to be optimal conditions for determination of D-glucaric acid with this test. In contrast to other similar methods, in which a logarithmic calibration curve is used, we calculate D-glucaric acid with a linear calibration curve by using the Dixon (Biochem. J. 55: 170, 1953) plot. The coefficient of variation for the method (within-run and between-day precision) was from 3.1 to 5.8%. Men excrete more D-glucaric acid than do women per 24 hours, but we saw no sex-related differences in excretion of D-glucaric acid when it was expressed in terms of urinary creatinine.

Influence of inorganic phosphate on the activity determination of isoenzymes of alkaline phosphatase in various buffer systems.

The inhibitory effect of inorganic phosphate on the activity determination of isoenzymes of alkaline phosphatase (AP) in diethanolamine (DEA), glycine and 2-amino-2-methyl-1,3-propandiol (AMPD) buffer was studied. This effect depends on the buffer used and isoenzyme investigated. Especially the placental isoenzyme is inhibited; the inhibitory effect in DEA buffer is stronger than in the other buffers used. The requirement of purity for 4-nitrophenylphosphate with respect to its content of inorganic phosphate and conclusions for using control sera enriched with AP isoenzymes are discussed.

[Isoenzyme diagnosis in the clinical course of acute myocarial infarct].

For the observation of the course of the disease of patients after an acute myocardial infarction by the estimation of the isoenzymes of the LDH (LDH-H and LDH-M) the possibility of the repeated four- to eight-hour-measurement of these isoenzymes in larger groups of patients after myocardial infarction is given. Instances of the use during the observation of the course of the disease after acute myocardial infarction are discussed. In which cases at the same time the isoenzymes of the LDH (LDH-H and LDH-M) as well as of the CK (CK-MB) were repeatedly estimated.

Increased creatine kinase BB activity in rat plasma induced by hypoxia.

Rats show an elevated creatine kinase (CK) activity in plasma after a 20-hour exposure to hypoxia. The increase of CK activity is essentially caused by CK-BB activity attaining its maximum at the 3rd day after exposure to hypoxia. The possibility of establishing a potential brain damage by determining CK-BB activity in plasma is discussed.

Lecithin: cholesterol acyltransferase activity, HDL-cholesterol and apolipoprotein A in serum of patients undergoing chronic haemodialysis.

In comparison with a control group, significantly lower lecithin:cholesterol acyltransferase activities and HDL-cholesterol values were found in the serum of patients undergoing chronic haemodialysis, while apolipoprotein A concentration remained unchanged. Reduced activity of lecithin:cholesterol acyltransferase is an indication of the abnormal cholesterol metabolism in haemodialysed patients and could be, in addition to other factors, an explanation for the high incidence of cardiovascular diseases in this patient group.

Transferrin, C3 complement, haptoglobin, plasminogen and alpha 2-microglobulin in patients with urogenital tumors.

The serum levels of transferrin, haptoglobin, C3 proactivator, plasminogen and alpha 2-macroglobulin have been measured in patients suffering from urogenital cancer. In this randomized study, we found a frequent decrease of transferrin coinciding with the elevation of C3-proactivator and haptoglobin. The serum concentration of plasminogen and alpha 2-macroglobulin were rarely observed in the pathological range excluding cancer-associated changes. Tumor metabolites and/or the circulating immune complex may account for the alteration of transferrin, haptoglobin and C3-proactivator, more than primary cancer-related synthesis of these proteins.

An optimized assay of alanine aminopeptidase activity in urine.

Elaboration of an optimized kinetic test for urinary alanine aminopeptidase at 37 degrees C is described. As final concentrations, 50 mmol/L tris(hydroxymethyl)aminomethane x HCl buffer, pH 7.8, and 2 mmol/L alanine-4-nitroanilide are proposed. Five- to sixfold activitiy is obtained as compared with a generally used test procedure. Coefficients of variation for the method (within-run and between-day precision) ranged from 1.56 to 3.45%. Men excreted more alanine aminopeptidase than did women if urinary volume or urinary timed volume was used as basis of reference. We saw no sex-related differences in excretion of alanine aminopeptidase when enzyme excretion was related to urinary creatinine.

[Current state and perspectives of the use of the 15N-tracer technic in gastroenterologic studies].

The importance of 15N as tracer in medical research is increasing. Since 1965 many results were obtained in cooperation of surgeons (Surgical clinic of Karl-Marx-University; Bezirkskrankenhaus St. Georg, Leipzig) and coworkers of Central Institute of Isotope and Radiation research. Using 3 test models (15N-labelled amino acid solution, permanent infused; 15N-labelled glycine, given as one single dose; 15N-labelled casein, oral given) healthy persons, patients in stress condition and after gastric operations and rats after performing several types of gastric operations were studied. Possibilities of calculations with mathematical models are shown.
