[Effect of refined xuefu capsule on mRNA expression of nitric oxide synthase of hypoxic ischemic myocytes evaluated by northern blotting].

Comparative study on effect of refined Xuefu capsule, Xuefu Zhuyu capsule and diltiazem on mRNA expression of nitric oxide synthase (NOS) and release of LDH-L, CK, GOT of hypoxic hypoglycemic cultured myocytes were studied by using the serum pharmacological method, reverse transcriptase-polymerase chain reaction (RT-PCR) and Northen blotting. It was shown that the three drugs can reduce release of LDH-L,CK,GOT of myocytes (P < 0.01), the effect of refined Xuefu capsule and diltiazem were obvious than that of Xuefu Zhuyu capsule in reducing LDH-L release and NOS mRNA expression (P < 0.01), and effect of refined Xuefu capsule was the strongest (P < 0.01). These findings indicated that all 3 drugs have significant effect in protecting myocytes which related to their effect on NO production of myocyte particularly the refined Xuefu capsule and diltiazem.

[Effect of xueguantong on inhibiting experimental atherosclerosis in rabbits].

In this experiment, the model of experimental atherosclerosis in rabbits was established by feeding the cholesterol contained food for 12 weeks. The effect of Xueguantong (XGT) in inhibiting atherosclerotic formation in rabbits has been studied. It was found that XGT could decrease serum total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), platelet aggregation and platelet adhesion. The aortic atherosclerotic plaque intimal coverage and ratio of atherosclerotic plaque of aortic intimal coverage to aortic medium coverage in XGT groups were significantly smaller than that in the model group (P < 0.05). The results showed that XGT could inhibit experimental atherosclerotic formation in rabbits, it might be relevant to that XGT could reduce the TC and inhibit platelet aggregation.

[Effect of xue guan tong on PDGF-A, PDGF-B and c-myc gene expression of vessel wall in atherosclerotic rabbits].

In this study, the effect of Xue Guan Tong (XGT) on platelet-derived growth factor-A (PDGF-A), PDGF-B and c-myc gene expression of vessel wall was explored with dot blot and in situ hybridization. The results shown that the expression of PDGF-A mRNA in atherosclerotic plaque of the model group was higher, PDGF-A gene hybridizations were mainly distributed in smooth muscle cell which located in newly emerging plaque and edge of atherosclerotic plaque. In contrast, level of PDGF-A mRNA in the XGT group was lower than that in the model group, which indicated that XGT could influence the level of PDGF-A mRNA. The c-myc oncogene expression in the model group was significantly enhanced than that in the normal group. However, the expression in XGT group decreased less than that in the model group. From the results shown above, it suggested that one of the effects of XGT on preventing VSMC proliferation was its influence on PDGF-A and c-myc mRNA expression level.

[Experimental study on the effect of abstinence with herbal preparation composite dong yuan gao].

The effect of herbal preparation "Composite Dong Yuan Gao" (CDYG) on the animal model of drug dependence was studied. After the morphinistic models of white rats and mice were made by ever increasing doses of morphine each time, the animals were divided into large dose CDYG group, small dose CDYG group, sustained morphine group, and control group. The withdrawal symptoms were observed after naloxone was given to the animals intraperitoneally. The results showed that in the CDYG groups withdrawal symptoms of the addicted white rats were alleviated, the number of jumps of the addicted mice was reduced, the scores of the principal symptoms of the animals were decreased, while the body weights of the animals were gained. Comparing with the control group, the difference was significant (P < 0.01). The effects of the CDYG enhanced in the large dose group, which suggests that the CDYG has abstinence effects.

Salmonella pericarditis and empyema: a case report.

A 54-year-old female diabetic patient who had been under regular oral hypoglycemic agent control for 2 years was admitted because of having dyspnea and orthopnea for several days. Massive pericardial effusion and a moderate amount of bilateral pleural effusion were noted at the time of admission. Pericardiocentesis and thoracocentesis were performed; both cultures grew Salmonella typhimurium. Antibiotic therapy was started immediately, but pleural effusion persisted. After repeated thoracocentesis and pleural effusion drainage, the patient was discharged 51 days later with complete recovery. Pericarditis and empyema are rare complications of Salmonella infections. Coexistence is even more rare, and only three cases have been reported in the English literature. Therefore, this case is presented with a review of the previous literature followed by discussion.

Serine for glycine substitutions in type I collagen in two cases of type IV osteogenesis imperfecta (OI). Additional evidence for a regional model of OI pathophysiology.

Serine for glycine substitutions in type I collagen have been described in seven cases of lethal type II osteogenesis imperfecta (OI), and six cases of nonlethal OI. We describe here two cases of moderately severe type IV OI with serine substitutions at alpha 1(I) Gly352 and alpha 2(I) Gly922, respectively. In both cases, G-->A point mutations were detected by RNase A cleavage of RNA/RNA and RNA/DNA hybrids. These cases extend the location for serine substitutions producing the moderately severe OI phenotype to the alpha 2(I) chain and the amino-terminal end of the alpha 1(I) chain. Their location supports a regional model of OI pathophysiology for serine substitutions. The proband with alpha 2(I) Gly922-->Ser has both normal and overmodified forms of both type I collagen chains. The overmodified form has delayed migration of all CNBr peptides. Helix thermal stability is decreased 4 degrees C. The fibroblast collagen protein and RNA of her unaffected parents are normal. However, the father was demonstrated to be a mosaic carrier using leukocyte DNA. The fibroblasts of the proband whose serine substitution is at alpha 1(I) Gly352 synthesize type I procollagen chains with delayed electrophoretic migration; normally migrating forms are difficult to detect. Only alpha 1(I) CB 8 displayed delayed migration. Helix thermal stability is reduced 2 degrees C. Parental genomic DNA was normal.

Phagocytosis of platelet in coronary heart disease patients with blood-stasis syndrome.

Phagocytosis of platelets was studied under electronmicroscope in 36 healthy persons and 64 coronary heart disease (CHD) patients with blood-stasis syndrome (BSS). The result showed that both the number of latex particles phagocytized by platelet and platelets which phagocytized latex particles were decreased in CHD with BSS. It was also found in this group that most of the platelets having active-phagocytosis were of medium size. The proportion of large platelets containing plentiful glycogen granules was much more in CHD patients with BSS and the shapes of platelets were changed readily with active exocytosis and aggregations. Therefore the present findings of disability of platelet phagocytosis might be considered the basic change in CHD with BSS.

[Clinical and experimental studies on royal made ping an dan in preventing motion sickness].

Royal Made Ping An Dan (PAD) is a royal clandestine prescription of the Qing Dynasty Imperial hospital for emperors, empresses, ministers, imperial maids and eunuchs. Experimental study confirmed; (1) PAD had apparent peripheral effect in inhibiting vomiting and improving mental state (P < 0.05). (2) PAD possessed markedly sedative effect (P < 0.05). (3) PAD was able to strengthen the defence ability of gastric mucosa and decreased its damage induced by chemical irritation. (4) PAD could markedly relieve the spasm of intestinal smooth muscle in vitro. (5) PAD could inhibit the growth of common pathogenic bacteria in intestine and stomach such as B. coli and B. dysenteriae. (6) The study of toxicology suggested that PAD was safe for clinical use. The clinical results showed that PAD possessed the effect in preventing 274 persons on motion sickness. The total effective rate of PAD group was 83.9%, while that of Dramamine group was 60.8%. PAD revealed better effect than that of Dramamine. Therefore, the authors realize that PAD is a better preventive drug for motion sickness.

Ischemic stroke treated with Ligusticum chuanxiong.

Ligusticum Chuanxiong and its effective components were studied in the treatment of ischemic stroke, a common emergent disease in China. Some injections of the medicines, including Ligusticum, Ligustrazine, Ligustylid and ferulic acid, were tested clinically and experimentally. The results showed that the effects of the drugs were the same as or even better than those of the controls, such as papaverine, dextran and aspirin-persantin. They could improve brain microcirculation through inhibiting thrombus formation and platelet aggregation as well as blood viscosity.
