Different susceptibilities of long-term potentiations in CA3 and CA1 regions of guinea pig hippocampal slices to nootropic drugs.

Effects of the nootropic drugs, piracetam and bifemelane, on long-term potentiation (LTP) of population spikes in the CA3 and CA1 regions of guinea pig hippocampal slices were investigated. Piracetam (10(-6) to 10(-4) M) and bifemelane (10(-8) to 10(-6) M) significantly augmented the LTP in the CA3 region. The effects of these drugs were inhibited by scopolamine (10(-6) M). However, the LTP in the CA1 region was not affected by piracetam and bifemelane even at highly effective concentrations (10(-5) and 10(-6) M, respectively). Thus, the LTP in the CA3 is more susceptible to nootropic drugs than in the CA1.

Evaluation of cryopreservation techniques of pancreatic fragments and islets in vitro and in vivo.

We evaluated cryopreservation techniques for pancreatic fragments and islets using rat tissue. After equilibration in 10% dimethyl sulfoxide (Me2SO), the tissue was frozen in a programmable freezer at 1 degree C/min down to -40 degrees C and at 3 degrees C/min down to -71 degrees C. The islets, when thawed, released abundant insulin in the presence of as little as 3.3 mM glucose, much more so than non-frozen islets did. Three additional procedures, prefreezing and post-thawing culture and the stepwise dilution of the Me2SO, lowered the non-specific insulin release of the thawed islets and improved their insulin response to 16.7 mM glucose. Thawed pancreatic fragments subjected to these additional procedures, transplanted into the peritoneal cavity of streptozotocin-induced diabetic rats, reduced their hyperglycemia significantly. The thawed fragments and islets did not differ from their corresponding non-frozen controls in 3H-leucine incorporation. The maintenance of tissue function was not satisfactory. However, our observations indicate that culturing pancreatic tissue before freezing and after thawing and the stepwise dilution of the cryoprotective agent reduce the damage induced by freezing the tissue.

Non-frozen cold storage is favorable for islet function and morphology.

Freezing has been shown to damage pancreatic islets and to disrupt their insulin release, probably because of intracellular ice formation. We compared frozen islets with fresh ones and with others stored at temperatures above freezing from a standpoint of insulin release response to glucose and transplantation. Group A islets, isolated from rats and immersed in 10% dimethyl sulfoxide in RPMI 1640, were stored at -2 degrees C, and group B islets at -196 degrees C, for 7 days. As for group B, the islets were cooled at 1 degree C/min from room temperature to -40 degrees C, subsequently at 3 degrees C/min to -80 degrees C and then put into liquid nitrogen to be rapidly frozen to -196 degrees C. The control islets were fresh. In vitro, basal release at 3.3 mM glucose was similar in group A to that in the controls, but was higher in group B than group A. Stimulated release against 16.7 mM glucose was lower in group A than group B. However, insulin responsiveness, i.e., the ratio of insulin release at 16.7 mM glucose to that at 3.3 mM glucose, was lost in group B. Freezing also caused damage to the group B cells visible under the light and electron microscopes, while group A islets were largely intact. In vivo, after 600 islets were transplanted into streptozotocin-induced diabetic rats, group A was better able to lower fasting blood glucose than was group B, and remained so for 4 weeks. Above sub-zero preservation in the non-frozen state thus seems adequate for the short-term storage for 7 days.

Release of oligosaccharides possessing reducing-end N-acetylgalactosamine from mucus glycoprotein in Streptomyces sp. OH-11242 culture medium through action of endo-type glycosidase.

A crude enzyme preparation from a culture medium of Streptomyces sp. OH-11242 contained endo-alpha-N-acetylgalactosaminidase activity. The activity could be induced by the addition of purified porcine gastric mucin to the culture medium. Oligosaccharides corresponding to approximately 2-14 glucose units were detected in the culture medium and also in an incubated reaction mixture of crude enzyme preparation and mucus glycoprotein. The resulting product with N-acetylgalactosamine at the reducing terminal implied the presence of a new type of endo-glycosidase liberating not only Gal beta 1-3GalNAc but also other larger oligosaccharides by hydrolysis of the O-glycosidic linkage between GalNAc and Ser (Thr).

Hepatic lesions induced by graft-versus-host reaction across MHC class II antigens: an implication for animal model of primary biliary cirrhosis.

To induce graft-versus-host reaction (GVHR), C57B1/6 (B6) spleen cells were injected into (B6 x bm1)F1, (B6 x bm12)F1, and (bm1 x bm12)F1 mice. Since the strains bm1 and bm12 are mutant at the H-2Kb and I-Ab regions of major histocompatibility complex (MHC), respectively, we can assess MHC class I- or class II-different GVHR. As reported earlier, immunological perturbations assessed by the number of immunoglobulin-producing cells and immune complex deposition in renal glomeruli were demonstrated in MHC class II-different GVHR. A conspicuous finding in this report is that epithelioid granuloma formation was observed in the portal area and around the central vein of liver of (B6 x bm12)F1 mice injected with B6 spleen cells. The epithelioid granuloma formation was not observed in (B6 x bm1)F1 nor (bm1 x bm12)F1 recipient mice. Degenerative changes resembling chronic nonsuppurative destructive cholangitis in primary biliary cirrhosis were also observed in the bile duct epithelium in (B6 x bm12)F1 and (bm1 x bm12)F1 mice. These lesions were already obvious at the 2 week postinjection of donor cells and were continuously observed up to 10 weeks when immunological perturbations subsided. Thus, class II-disparate GVHR in this experimental system might provide a novel animal model of protracted disease, primary biliary cirrhosis.

An improved method for the isolation of islets from the rat pancreas.

The conventional method for rat islet isolation was improved by injection of collagenase solution (1.3 mg/ml) into the bile duct and the concomitant dissection of the inferior vena cava, static incubation of the removed distended pancreas for 15 min at 37 degrees C and filtration of the digested tissue through two different meshes of 1190 and 590 micron in pore size. This new technique led to about 2.5 times higher yield of viable islets than the conventional method.

Two types of rat gastric mucus glycoprotein subunits.

Gastric mucus glycoproteins were extracted with 2% Triton X-100 from rat gastric corpus and antrum and purified by CsCl equilibrium centrifugation. Corpus mucus glycoproteins were degraded into what appeared to be two "subunits" (Mw 4.4 x 10(5) and 6 x 10(6)) by the reduction of disulfide bonds. Papain digestion of the latter produced glycopeptides with a molecular weight of approximately 4.4 x 10(5). This type of subunit had carbohydrate chains with about 9 sugars attached to every 2 amino acid residues. Papain digestion of the former type of subunit revealed no change in the elution profile on Bio-Gel A-15m. This type of subunit had carbohydrate chains with 17-19 sugars attached to every 3 amino acid residues. The subunit of antral mucus glycoproteins was essentially the same as the former type of corpus subunits in molecular weight (Mw 4.4 x 10(5)) and average oligosaccharide chain length. These results suggest that there are two distinct types of mucus glycoprotein subunits in rat stomach.

Mucus glycoprotein and mucosal protection.

For assessing the participation of mucus glycoproteins in the cytoprotective process, mucus glycoprotein content in the rat gastric mucosa was measured after treatment with 70% ethanol with or without prostaglandin (PG) E derivatives or 20% ethanol pretreatment. Oral administration of two synthetic PGE derivatives did not cause any significant changes in mucus glycoprotein content. Seventy percent ethanol administration caused marked reduction in mucus glycoprotein content (about 50% of control) as well as severe gastric mucosal damage. Treatment with two PGE derivatives (10-100 micrograms/kg) prior to 70% ethanol administration markedly inhibited the gross mucosal lesion, whereas the glycoprotein content under these conditions was significantly less than that in the untreated control group (ranging from 67-88% of control). Pretreatment with 20% ethanol markedly inhibited the gross mucosal damage caused by 70% ethanol dosing but the inhibition in the reduction of mucus glycoprotein content was restricted to about 80% of the untreated controls. In summary, cytoprotection induced by PG was not accompanied by entire conservation of intramucosal mucus glycoprotein in the gastric mucosa.

Comparison of proteinase activities in squamous cell carcinoma, basal cell epithelioma, and seborrheic keratosis.

The correlation between proteinase activities and invasive and metastatic potentials was investigated by comparing three different kinds of tumors. Extracts from tumor homogenate of 11 squamous cell carcinoma (SCC), 5 basal cell epithelioma (BCE), and 8 seborrheic keratosis (SK) were prepared in order to examine the activity of acid phosphatase and proteinases such as cathepsin B and D, type I and IV collagenase, and plasminogen activator (PA). There was no difference observed between acid phosphatase and cathepsin D activities among the three tumors. Cathepsin B and PA activities were slightly elevated in SCC. Type I collagenase activity of SCC was 9-fold higher than that of SK (p less than 0.01), and type IV collagenase was 3-fold higher per tissue DNA (p less than 0.05). Type I and IV collagenase of BCE were elevated per tissue protein but not elevated per tissue DNA. Correlation was found between the level of cell differentiation in SCC and the activities of cathepsin B, PA, and type I collagenase. Poorly differentiated SCC exhibited a tendency to have higher proteinase activities. Proteinases that showed high activities in malignant tumor homogenate may be related to the degradation of the surrounding cell matrix in addition to intracellular metabolism. Type I and IV collagenase, in cooperation with cathepsin B and PA, might play a major role in invading the dermal stroma and basement membrane.

Non-Hodgkin lymphoma of the skin excluding mycosis fungoides and cutaneous involvement of adult T-cell leukemia/lymphoma.

Forty-nine cases of cutaneous malignant lymphoma were reviewed in order to analyze the clinicopathological features of these neoplasms. Excluding 13 cases of mycosis fungoides and 4 cases of cutaneous involvement of proven adult T-cell lymphoma/leukemia, the remaining 32 cases were further classified according to their pathological and clinical features. There were 12 primary cutaneous lymphomas, 15 cases of secondary cutaneous involvement of systemic lymphoma, and 5 cases of concurrent skin and lymph node involvement. Histologically, large cell lymphoma predominated in both primary and secondary cutaneous lymphomas. Immunohistochemical study using monoclonal antibodies reactive with B- and T-cells in paraffin sections revealed the cellular lineage in 30 cases. Nineteen cases were of T-cell origin and 11 cases were of B-cell derivation. The prognosis of these patients was rather poor; 25 patients died within 5 years. The predominance of T-cell lymphoma contrasts with a higher frequency of cutaneous B-cell lymphoma in Western countries. As the clinicopathological features of cutaneous lymphomas are diverse, it is suggested that cutaneous lymphomas should be classified and studied in a similar way to their nodal counterparts.

Changes of rat gastric mucus glycoproteins in cytoprotection: influences of prostaglandin derivatives.

The effects of two synthetic prostaglandin E derivatives on mucus glycoproteins in the stomachs of rats were evaluated. Neither derivative caused change in mucus glycoprotein content in the gastric corpus or antrum, but both increased the biosynthetic activity of mucus glycoproteins in these regions (about 15-30%). Furthermore, the effects of the pretreatment of these derivatives on conserving mucus glycoproteins in ethanol-induced gastric lesions were examined. Treatment with these derivatives 60 min prior to 70% ethanol administration markedly inhibits the decrease in gastric mucus glycoprotein content caused by 70% ethanol. But the glycoprotein content under these pretreatment conditions was significantly less (12-19%) than that in the control group without any drug treatment.
