[Diphyllobothrium latum infection in Akita Prefecture. Infections occurring from 1974 to 1981 and the clinical effect of paromomycin sulfate].

Forty-one patients, aged 2 to 68 years old, infected with Diphyllobothrium latum in Akita prefecture were treated using paromomycin sulfate from 1974 to 1981. Paromomycin sulfate was administered orally in a single dose of 20, 30 mg/kg or 50 mg/kg. Twenty-three cases expelled tapeworms. However, only 9 (28.1%) out of 32 tapeworms expelled had their scolex. It can be said that all cases were successfully treated with paromomycin sulfate, since they showed the egg-negative on 3 weeks or 1 month after the treatment. The drug was well tolerated and no side effects were encountered in any patients. It was suggested that paromomycin sulfate was effective and also safe therapeutic agent for the treatment of D. latum infection.

The periodic change in adenosine 3',5'-monophosphate concentration in sea-urchin eggs.

The level of adenosine 3',5'-monophosphate (cyclic AMP) in the eggs of the sea urchin, Anthocidaris crassispina, was found to change periodically after fertilization. The minimum and maximum levels of cyclic AMP were 1.0 X 10(-7)M and 1.5 X 10(-6)M, respectively. The activity of adenylate cyclase in a 105 000 X g precipitate reached a plateau at 20 min after fertilization and stayed constant for at least 2 h. It was also found that 1.0 mM CaCl2 increased the activity of adenylate cyclase in the same precipitate from unfertilized eggs. In contrast, phosphodiesterase activity changed periodically and correlated with cyclic AMP levels in the eggs. Up to a concentration of 1.5 X 10(-6)M cyclic AMP, phosphodiesterase activity was low, but it became activated when the level of cyclic AMP rose beyond this level. These results indicate that the change in the intracellular level of cyclic AMP is regulated mainly by the change in phosphodiesterase activity.

Variables in demonstrating methicillin tolerance in Staphylococcus aureus strains.

Certain technical considerations which affected the status of methicillin tolerance in Staphylococcus aureus strains were studied. Methods which consistently demonstrated tolerance or intolerance of a given strain were avoidance of inoculum splashing, use of stationary-phase inoculum, 24-h tube incubation, and minimization of antibiotic carry-over. These studies suggested a need for the establishment of a standardized reference for the determination of tolerance.

Therapy of experimental infective endocarditis due to antibiotic-tolerant Lactobacillus plantarum-bactericidal synergy of penicillin plus gentamicin. Correlation of in vitro susceptibility studies with in vivo efficacy.

Infective aortic valve endocarditis (IE) was induced in 40 rabbits with a penicillin (PNC)-tolerant, gentamicin (GM)-resistant strain of Lactobacillus plantarum; this isolate was synergistically killed in vitro by PNC + GM. The in vivo relevance of the in vitro observations was examined by determining the rates of eradication of endocardial L. plantarum by PNC versus PNC + GM. Mean vegetation L. plantarum titers were significantly lower (p less than 0.05) in PNC + GM treated rabbits versus both PNC-treated and control rabbits by 48 h of therapy. Also, PNC + GM more rapidly sterilized vegetations as compared to controls (p less than 0.025) and PNC-treated rabbits (p less than 0.05). Thus, in vivo antibiotic efficacy paralleled in vitro bactericidal studies, and may partially explain the relatively refractory nature of lactobacillary IE in humans treated with single beta-lactam antibiotics.

Esophageal echocardiographic left ventricular anterolateral wall motion in normal subjects and patients with coronary artery disease.

Esophageal echocardiography was developed for recording left ventricular anterolateral wall (LVAW) echocardiograms and was applied clinically to 14 normal subjects and 21 patients with coronary artery disease. LVAW echocardiograms were obtained satisfactorily in 11 of 14 normal subjects (75%) and 20 of 21 patients (95%) with coronary artery disease. LVAW echocardiograms were obtained by conventional anterior echocardiography in eight of 21 patients (38%) with coronary artery disease. In 11 normal subjects, mm/sec (mean 34.3 +/- 5.2 mm/sec); and diastolic wall thickness ranged from 9-12.5 mm (mean 11.2 +/- 0.7 mm). In 20 patients with coronary artery disease, LVAW motion obtained by esophageal echocardiography was classified into five groups according to the excursion, and the findings were in good agreement (80%) with those obtained by left ventriculography. Classification of LVAW motion by conventional echocardiography agreed with that of left ventriculography in only three of eight patients, although all eight patients had abnormal LVAW motion by the conventional method. In all patients except one, whose LVAW echocardiograms were obtained by conventional echocardiography, excursion was much less than that obtained by esophageal echocardiography. We conclude that the projection of an ultrasonic beam from the intraesophageal transducer is a better approach for accurate measurement of LVAW motion.

Regulation of vitamin B12 and bacteriochlorophyll biosynthesis in a facultative methylotroph, Protaminobacter ruber.

The regulation of vitamin B12 and bacteriochlorophyll formation was studied in a facultative methylotroph, Protaminobacter ruber, classified as a non-photosynthetic bacterium. Vitamin B12 was formed at an almost constant level under various cultivation conditions, while bacteriochlorophyll synthesis was drastically influenced by the growth conditions. Addition of B12 and hemin to the medium and the change of culture conditions from light to darkness in the early growth phases stimulated the pigment synthesis, but no pigment was formed under the continuous illumination and the addition of chloramphenicol inhibited the pigment formation. delta-ALA synthase and delta-ALA dehydratase activities were demonstrated in P. ruber. delta-ALA synthase formation was induced when cultures were transferred from light to dark conditions, which promoted the bacteriochlorophyll formation. Bacteriochlorophyll-protein complex had an absorption maximum at 870 nm and a very small peak at 800 nm.
