Transient retino-inferior collicular fibres in neonatal rats: their persistence after removal of one eye at birth.

Retinal projections to the inferior colliculus in the newborn albino rat were demonstrated by intraocular injection with horseradish peroxidase. During normal development, these retino-inferior collicular fibres disappeared by 26 days of age. However, after removal of one eye at birth, distinct retino-inferior collicular fibres arising from the remaining eye were preserved contralaterally in rats at 26 days of age.

Serum levels of macrophage colony-stimulating factor and aortic calcification in hemodialysis patients.

Hemodialysis (HD) patients have accelerated atherosclerosis. Recent reports have shown that aortosclerosis is more frequently observed in HD patients than in healthy subjects. Macrophage colony-stimulating factor (M-CSF) secreted by activated macrophages may be involved in the process of aortosclerosis in HD patients. To understand the mechanism behind the increased incidence of aortosclerosis in HD patients, we examined the relationships between serum M-CSF levels and aortic calcification index (ACI) estimated by CT scan. A significant increase in serum M-CSF concentrations was found in HD patients (3.8 +/- 0.2 ng/ml) as compared with controls (1.5 +/- 0.1 ng/ml). No significant differences were observed between chronic glomerulonephritis and diabetes mellitus groups of patients. We also found no significant differences between the groups using different membranes (triacetate 3.8 +/- 0.2 ng/ml vs. polysulfone 3.8 +/- 0.4 ng/ml). There was no correlation between serum M-CSF concentrations and clinical parameters such as age, duration of HD, blood pressure, serum concentrations of nitrogen, creatinine, cholesterol, triglyceride, LDL, Ca x P products, and intact parathyroid hormone. A positive correlation was observed between serum M-CSF levels and ACI in HD patients (r = 0.596, p < 0.01). These results suggest that M-CSF may be involved in the process of aortosclerosis in HD patients.

Amplification of DAT1 (human dopamine transporter gene) 3' variable region in the Japanese population.

Amplification of the human dopamine transporter gene (DAT1) was performed by the polymerase chain reaction, and polymorphism of the VNTR locus was analyzed in a population sample of 176 randomly selected Japanese individuals. Genomic DNAs were extracted from whole blood, and the 3' untranslated region of the human DAT1 gene was amplified. The amplification products were distinguished by agarose gel electrophoresis, and visualized directly by ethidium bromide staining. In this way, four alleles were visualized directly and designated A, B, C, and D in decreasing order according to size. The relative frequencies of DAT1 allels A through D were 0.0171, 0.0625, 0.9119, and 0.0085, respectively. Evidence of significant deviations from the Hardy-Weinberg equilibrium was observed from these Japanese population data. The DAT1 locus was estimated to have a heterozygosity index of 13% and polymorphic information content of 0.16.

Videolaryngoscopy. A comparison of fiberscopic and telescopic documentation.

Videolaryngoscopy is a most useful and effective method of evaluation and documentation of physiological and pathological conditions of the larynx. It is of great value for making accurate diagnosis and planning proper treatment. It permits instant and simultaneous voice and video recording and later analysis. Videolaryngoscopy can be accomplished with either a flexible fiberscope or a rigid right-angled telescope. Fiberscopic videolaryngoscopy is more useful for voice analysis of speech disorders and evaluation of such functions of the larynx as phonation, singing, and swallowing. Telescopic videolaryngoscopy is superior for critical evaluation of anatomical and pathological changes of the laryngeal structures as well as close-up examination of vocal cord function. While fiberscopic laryngoscopy is technically easy, fiberscopic video documentation is much more difficult than telescopic video documentation. Telescopic videolaryngoscopy provides clearer and sharper images of the larynx. Advantages and disadvantages of each method are discussed.

Linguistic models of F0 use, physiological models of F0 control, and the issue of "mean response time".

This paper evaluates "mean response time" (MRT), a method used in previous studies to relate physiological evidence (recordings of electromyographic activity in the cricothyroid and sternohyoid) to acoustic evidence (fundamental frequency). Rather than averaging over tokens before correlating these signals, we calculated the best response time (RT) for each token, and evaluated the pattern of variability across utterances. Furthermore, rather than correlating over whole utterances, we correlated electromyographic activity (EMG) to fundamental frequency (F0) only over intervals defined in terms of linguistically significant events in the F0 trace, identified using a linguistically motivated model of English intonation. Steep changes in the F0 tended to have better correlation coefficients than shallow ones, which we relate to the physiological model by noting the complex of components contributing to both signal types. Also, the distribution of lead times was easier to interpret when the two tones delimiting the analysis domain had some tight temporal relationship specified by the intonational phonology. Finally, lead times tended to vary as a function of what preceded the target rise or fall. In short, averaging over signals before analysis obscures patterns of variation in the data which may lead to new insights and to new directions for research.

Role of vertical larynx movement and cervical lordosis in F0 control.

The role of vertical larynx movement in vocal frequency (F0) change has attracted the attention of many researchers. Recently, Hirai, Honda, Fujimoto, and Shimada (1994) proposed a mechanism of F0 control by vertical larynx movement based on the measurement of magnetic resonance images (MRI). In F0 changes, the larynx moves vertically along the cervical spine, which displays anterior convexity (lordosis) at the level of the larynx. Therefore, the vertical larynx movement results in the rotation of the cricoid cartilage and vocal fold tension changes. The present study reexamines the above mechanism based on a qualitative analysis of midsagittal MRI data using three male subjects with evident cervical lordosis. Tracings of the jaw, hyoid bone, laryngeal cartilage, and cervical spine were compared in high and low F0 ranges. In the high F0 range, the hyoid bone moved horizontally while the larynx height remained relatively constant. In the low F0 range, the entire larynx moved vertically, and the cricoid cartilage rotated along the cervical lordosis. These results indicate that the vertical movement of the larynx comprises an effective F0 lowering mechanism, and suggest that the human morphologies of low larynx position and spinal curvature contribute to voluntary use of the vocal function.

Identification of odoriferous compounds from adults of a swallowtail butterfly, Papilio machaon (Lepidoptera: Papilionidae).

Adults, particularly males, of a papilionid butterfly, Papilio machaon hippocrates, emit a fairly strong scent perceivable by humans. We have identified a variety of volatile compounds (hydrocarbons, alcohols, aldehydes, ketones, esters, and so on) from the wings and bodies of both sexes of the butterfly. Male wings secreted n-dodecane, linalool and geranylacetone as major components together with small amounts of camphene, limonene, p-cymene, 2-phenylethanol, n-hexanal, n-decanal, isoamyl acetate, p-allylanisole, 2-pyrrolidone and other characteristic volatiles. The overall profile of volatile compounds detected from male body was quite different from that of the wings. Male body was devoid of camphene, 2-phenylethanol, n-hexanal but instead contained limonene, acetoin, a sesquiterpene hydrocarbon (C15H24, methyl n-octanoate, (E,E)-hepta-2,4-dienal, and another isomer of heptadienal as principal components, of which the last four compounds were specific to the body. All these substances seem to concurrently characterize the male odor. The chemical patterns of compounds found from female wings and body were essentially the same in quality as those of male wings and body, respectively, although their quantities in females were generally smaller than in males. Females, however, had a larger amount of acetamide than males. The chemical compositions of volatiles from the fore and hind wings of males were not greatly different from each other, and every component was considered to be present on all parts of the wings. This suggests that the scent-producing organs or scent-emitting pores are widely distributed on the whole wings. EAG responses of both sexes to 12 selected compounds identified from the butterfly were not strong at a dose of 1 microg, while both sexes showed relatively stronger responses to n-nonanal, methyl n-octanoate, D-limonene and linalool at a higher dose (10 microg). Although sexual difference in EAG response was not prominent, females appeared a little more sensitive, and n-nonanal and acetoin evoked significantly higher responses from females at 1 microg.

Use of frequency modulation combined with amplitude change to elicit frequency-specific whole-nerve response.

The averaged whole-nerve response to frequency modulation combined with amplitude change was recorded from the round window of the guinea pig. A downward shift of 20% in the frequency of a 500-Hz, 1-kHz or 2-kHz pure tone with an amplitude increase elicited the responses. The latency for each frequency was investigated at different intensities. The latency is greater for lower-frequency tones and this latency increase agrees well with the traveling wave delay for the intensity range used in this study. A downward shift with an amplitude decrease failed to yield the response. These findings suggest that the response to a downward shift of frequency with an amplitude increase results from new activation due to an apical extension of the envelope of the traveling wave and thus represents the activity in a restricted area of the basilar membrane. Thus, the frequency-modulated tone in downward direction with increased amplitude can be a frequency-specific stimulus.

Estrogen-producing ovarian mucinous cystadenomas in postmenopausal women.

Cells from benign and borderline malignant mucinous cystadenomas surgically resected from three postmenopausal women with elevated serum estradiol levels were found to have different abilities to synthesize estrogen from [14C]-cholesterol. Addition of gonadotropins to cultured cells with low levels of estrogen synthesis did not result in any significant increase in estrogen synthesis. Immunohistochemical studies of these mucinous cystadenomas showed that LDL receptors were distributed in the benign cystadenoma cells and in the stromal cells of borderline tumors, but not in the cystadenoma cells of borderline tumors. Sterol regulatory element binding protein was immunohistochemically shown to be distributed in the mucinous cystadenoma cells of borderline tumors, but not in the benign cystadenoma cells.

Diagnosis of metastasis of ovarian clear cell carcinoma to the peritoneum of the abdominal wall by positron emission tomography with (fluorine-18)-2-deoxyglucose.

A 43-year old woman, operated on for ovarian clear cell carcinoma (stage IIc) four years previously was found to have a small mass under the abdominal wall in the right lower quadrant of the abdomen. Neither diagnostic imaging (ultrasonography and MRI) nor tumor markers showed any evidence of recurrence, but positron emission tomography revealed a hot spot area, and it was diagnosed as recurrence of the ovarian carcinoma. The postoperative histopathological diagnosis was metastasis of ovarian carcinoma to the peritoneal wall.

Association between overexpression of cyclooxygenase-2 and suppression of apoptosis in advanced cancer of the uterine cervix after cyclic balloon-occluded arterial infusion.

Recent advances in cancer chemotherapy have drawn the attention of investigators to the usefulness of chemotherapy for cancer of the uterine cervix in general, and we ourselves have previously reported satisfactory therapeutic results of cisplatin-based cyclic balloon-occluded arterial infusion chemotherapy (BOAI), which enabled treatment by hysterectomy in patients with advanced cervical cancer. Nevertheless, there have been some patients in whom CDDP therapy by BOAI was ineffective, and hysterectomy was impracticable. In the present study we investigated the expression of cyclooxygenase-2 (COX-2) in these cases in an attempt to identify the cause of the lack of efficacy. The subjects of the study were 38 women with advanced cervical cancer classified as FIGO (International Federation of Gynecology and Obstetrics) stage III or higher who were admitted to the Department of Gynecology of Osaka City University Medical School Hospital between July 1993 and April 2000. All of these patients were treated by BOAI, and expression of COX-2, angiogenic factors, and cancer cell apoptosis, before and after BOAI were examined, and survival rates were compared. The 18 patients in whom BOAI was ineffective showed significantly higher expression of COX-2 after BOAI, and cancer cell apoptosis was inhibited. The 5-year survival rate of these patients was 16.8%, as opposed to 54.1% in the 20 patients in whom BOAI was effective. These results suggest that overexpression of COX-2 inhibits cancer cell apoptosis and adversely influences the prognosis.

Probucol decreases total body fat loss in VX2-carcinoma-induced cachectic rabbits.

We previously reported the continuous decrease of total body fat in VX2-carcinoma-bearing rabbits after tumor implantation, as well as changes in the serum lipid profile. Probucol, an antioxidant drug, has a cholesterol lowering effect against hyperlipidemic subjects. VX2-carcinoma-bearing rabbits fed with a diet containing 1% probucol did not show any difference in serum lipid compositions as compared with rabbits fed with a control diet. Similarly serum lipolytic activity showed no differences, whether probucol was administered or not, while the decrease in total body fat was significantly less when probucol was administered.

Central nervous system T-cell lymphoproliferative disorder in a patient with chronic active Epstein-Barr virus infection.

PURPOSE: Central nervous system (CNS)-T cell lymphoproliferative disorder (T-LPD) developing during the course of chronic active Epstein-Barr virus (CAEBV) infection is reported. PATIENTS AND METHODS: CAEBV was diagnosed in a 14-month-old boy with fever, cytopenia, hepatosplenomegaly, and abnormal high titers of anti-Epstein-Barr virus (EBV) antibodies. At 8 years of age, he had a splenectomy because of progressive disease. RESULTS: After 27 months of clinical remission, muscle weakness and paresthesia developed. Magnetic resonance imaging of his brain showed spotty T2 prolongation in left parietal, bilateral frontal, and temporal white matter with meningeal enhancement. Brain biopsy revealed the cerebral infiltration of CD3+, CD4+, CD8-, CD45RO+, CD56-, and EBV-encoded RNA 1+ cells. CONCLUSIONS: The CNS involvement of EBV-associated T-LPD is a rare but serious complication in CAEBV without known underlying immunodeficiency.

Effects of cyclic plasma perfusion on adipocyte apoptosis and lipolytic activity in VX2 carcinoma-bearing rabbits.

BACKGROUND: In an animal model we found that total body fat (TBF) decreased with progression of tumors, but that it rebounded after cyclic plasma perfusion (CPP). In this study, we investigated changes in adipocyte apoptosis and serum lipolytic activity (LA) after CPP. MATERIALS AND METHODS: TBF, LA, and adipocyte apoptotic index (AAI) in VX2 carcinoma-bearing rabbits were measured every ten days after implantation. CPP was performed every 5 days from post-implantation day (PID) 14. RESULTS: TBF decreased as the days passed and reached 43.2 +/- 10.0% on PID 50 in the non-CPP group, while it decreased 22.7 +/- 8.8% in the CPP group. LA on PID 50 was 17.8 +/- 14.0, 33.2 +/- 6.3 mumol glycerol/10(5) adipocytes in the non-CPP group, while LA did not increase in the CPP group. AAI was 28.8 +/- 16.4, 27.8 +/- 17.7, 10.3 +/- 9.9 and 0.4 +/- 1.1 on PID 10, 20, 30, and 40, respectively, in the non-CPP group. No change was noted in AAI after CPP. CONCLUSIONS: CPP removed a humoral factor that possesses TBF-decreasing LA, but it did not suppress adipocyte apoptosis.

Results of resection for hilar cholangiocarcinoma with analysis of prognostic factors.

BACKGROUND/AIMS: Resection for hilar cholangiocarcinoma remains a challenging procedure and recent published results continue to show that few patients are cured of their disease. The objective of this review was to determine the results of radical resection and to identify factors associated with long-term survival. METHODOLOGY: Retrospective review of resection for hilar cholangiocarcinoma in 29 consecutive patients with statistical analysis of prognostic factors, including p53 status. RESULTS: The mortality and morbidity rates were 6.9% and 34%, respectively. The overall 5-year survival was 20% with the median survival being 16 months. Univariate analysis identified the following factors associated with poor survival; involved lymph nodes, vascular invasion, advanced tumor stage, positive tumor margins, and p53 mutation. However, none of these factors was associated with poor survival by multivariate analyses. CONCLUSIONS: Radical resection for hilar cholangiocarcinoma can be performed with acceptable morbidity and mortality, but most patients succumb to their disease. p53 status may be a helpful adjunct to routine pathological staging.

Induction of estradiol synthesis by probucol in the adenocarcinoma cells of an ovarian clear cell tumor.

BACKGROUND: Clear cell tumor of Müllerian organs have characteristic intracellular structures with numerous small mitochondria, free ribosomes, lamellae of granular endoplasmic reticulum and intracytoplasmic glycogen granules, but synthesize no estrogens under ordinary conditions. MATERIALS AND METHODS: Cells from an ovarian clear cell tumor cultured with 10 micrograms/ml of the antioxidant probucol released more estradiol into the medium than cells cultured without probucol. Immunocytological analysis revealed the predominance of estradiol in the adenocarcinoma cells cultured with probucol. RESULTS: There was no difference in the distribution of low-density lipoprotein receptors in adenocarcinoma cells according to whether they had been cultured with or without probucol. Cytochemical analysis showed that hydroxylation of estrone was accelerated in the adenocarcinoma cells cultured with probucol as compared to the cells cultured without probucol. CONCLUSION: Probucol is thought to have an unknown function in the acceleration of hydroxylation of steroids in some types of cells with estrogen-producing potential.

Expression of skeletal muscle cells apoptosis regulatory proteins in plasma-perfused VX2 carcinoma-bearing rabbits.

BACKGROUND: In tumor-bearing animals we found that the skeletal muscle apoptosis might be involved in muscle wasting. In this study, we investigated changes in the skeletal muscle cell apoptosis regulatory proteins after cyclic plasma-perfusion (CPP). MATERIALS AND METHODS: We studied changes in body weight, lean body mass (LBM), apoptotic index (AI) and expression of Bax and Bcl-2 in skeletal muscle in VX2 carcinoma-bearing rabbits. RESULTS: 20 days after tumor implantation, LBM had decreased by 5.06+/-1.10%, while the AI had increased to 40.5+/-3.20%. By 40 days, LBM had decreased by 11.0+/-0.81% and the AI was only 0.93+/-0.96%. Bax expression was detected in proportion to the AI, but no Bcl-2 expression was detected in either the experimental or control groups. CPP improved LBM, but did not prevent Bax expression. CONCLUSION: Skeletal muscle cell apoptosis related to Bax was concluded to be the cause of muscle wasting in VX2 carcinoma-bearing rabbits. CPP appears to reduce muscle wasting and increase LBM, but it did not suppress Bax expression or skeletal muscle cell apoptosis.

Omental transplantation for chronic occlusive arterial diseases.

Problems of surgical indication and technique for the treatment of chronic occlusive arterial diseases remain unresolved, especially in the cases which show poor angiogram run-off. To establish the surgical technique of omental transplantation, anatomical findings of the omental vessels were studied. The anatomical findings of the omentum were classified into two types. Type I (single layer) and Type II (double layer), and subdivided into seven subtypes according to the number of omental arterial branches. Omental transplantation was performed in our clinic in 19 cases (20 limbs) and surgical results were satisfactory. The examination of microscopic findings of the implanted omentum and the ischemic muscle of the implanted limb by consecutive serial sections suggested that the vessels of the omentum might invade the ischemic muscular tissue and supply blood to the muscle. Omental transplantation should be considered one of the most useful surgical techniques in cases with poor angiogram run-off.
