Chemical ionization (CI) mass spectra of new macrolide antibiotics, mycinamicins are reported. Protonated molecules (MH+) are observed as base peaks in the CI mass spectra of all components. Fragmentations are mainly restricted to the glycosidic linkages and the resulting aglycone and sugar-derived ions appear regularly in their mass spectra. Moreover, characteristic fragment ions involving carbon-carbon bond fission are found in the CI mass spectra of the epoxyenone-containing components, mycinamicins I (1) and II (2). The mechanism for the formation of the ion species is also discussed.
The effect of oral administrations of nicotinohydroxamic acid (NHA) and neomycin (NM) in ammonia and urea metabolism were investigated in the rat. NHA and NM lowered the blood ammonia level, and NHA increased urinary excretion of urea and depressed urease activity in the stomach and the colon, while NM had a smaller effect on urinary excretion of urea but depressed urease activity in the colon.
A total of 71 cases with primary brain tumors (44 cases) and metastatic brain tumors (30 cases) were entered into our clinical studies with MCNU and radiation therapy or MCNU alone. With regard to tumor reduction on CT scan, the response rates obtained for MCNU and radiation were 21.7% for malignant gliomas and 50.0% for metastatic brain tumors. With regard to improvement of neurological signs, the response rates obtained for MCNU and radiation were 62.9% for malignant gliomas and 71.4% for metastatic brain tumors. The response rates for MCNU were 5.8% for malignant gliomas and 46.1% for metastatic brain tumors. In the improvement of performance status, the response rates for MCNU and radiation were 51.8% for malignant gliomas and 64.2% for metastatic brain tumors. The response rates for MCNU were 46.1% for malignant gliomas and 30.7% for metastatic brain tumors. A minimal degree of hematological toxicity occurred but this gradually disappeared. These results suggested that MCNU has relatively effective antitumor activity against metastatic brain tumors and an enhanced effect with radiation against malignant gliomas.

The value of glycosylated hemoglobin (HbA1) has been determined in diabetic patients with simple or proliferative retinopathy and in those without retinopathy. The mean value of HbA1 is significantly more highly elevated in patients with retinopathy than in those without retinopathy. Progressive augmentation of the value of HbA1 signifies a risk of deterioration of the retinopathy, whether it is a simple retinopathy or proliferative retinopathy.
Effect of training on basal metabolism in rats by means of long-term exercise loaded at a moderate intensity were studied. The Wistar-strain male rats were carefully bred at the room temperature of 23.0 +/- 1 degree C and humidity of 60%. The first physical training was carried out by motor driven treadmill for 8 weeks at a speed of 25 m/min for less than 15 min once daily and 6 times in a week after 4 weeks of birth. Continuously, the second training was carried out for 15 months at the same load of one time per week. Running ability and the recovery of glycogen in exhausted skeletal muscles on period of the first training, loaded from 4 weeks to 12 weeks after birth. There was no change on the basal metabolism between the trained and sedentary control rats. In general, the basal metabolism significantly fell by aging, for instance, 24 months rat's basal metabolism was 63% of 3-4 months rat's. The second training repressed a decline of running ability and rose the recovery of muscle glycogen in rats, though training could not be stopped lowering of basal metabolism in aged rats.
In order to clarify the conclusion that the change of basal metabolism affected by physical training, effect of endurance training for 8 weeks on basal metabolism of young adult rats were investigated. Results are as follows. Endurance training increased significantly running ability of rats, for instance the running time at a speed of 25 m/min on the control and training groups were 53.7 +/- 18.8 min, 232.8 +/- 32.8 min, respectively. The ratio of soleus's weight to the body weight in trained rats was high significantly (p less than 0.05). The glycogen contents of trained rats under the condition of feeding have higher than the control rats. Especially, glycogen contents of the soleus and red-gastrocnemius significant increased (p less than 0.05), and liver glycogen content under the same condition increased significantly (p less than 0.02). The oxygen consumption in trained rats increased significantly compared with control rats (p less than 0.03). The basal metabolism of trained rats showed 1.24-fold increase compared with those of control (p less than 0.02). Oxygen consumption of sliced ventricle in trained rats increased significantly (p less than 0.03), it's rate was 118% of control. However those of other tissues did not change significantly.
The amino acid sequence of the A3 subunit, an acidic subunit of the glycinin seed storage protein of the soybean (Glycine max (L.) Merrill), has been determined by sequence analysis of the peptides obtained by cyanogen bromide cleavage followed by enzymatic digestion and partial acid hydrolysis. The A3 subunit is composed of 410 amino acids (Mr approximately 46,000). It has been confirmed that there are conserved, repeated domains within the subunit. A comparison of the NH2-terminal sequence of the A4 subunit with the sequence of the A3 subunit reveals that a sequence highly homologous to the NH2-terminal sequence of the A4 subunit exists in internal regions of the A3 subunit. This suggests that the genes coding for the acidic subunits of glycinin are strongly related, and have the same evolutionary origin.
An isocratic high-performance liquid chromatographic method for the determination of tetracyclines is described using a mobile phase containing oxalic acid and C8- and C18-modified silica gel columns. For good separations of tetracyclines, oxalic acid concentrations of above 0.01 and 0.2 M respectively for parent tetracyclines (group I) and impurities in tetracycline (group II) are required. The optimum pH of the aqueous oxalic acids solution in the mobile phase is 2.0. The combinations of the C8-modified silica gel column with methanol-acetonitrile-0.01 M aqueous oxalic acid solution pH 2.0 (1:1.5:5) and the C18-modified silica gel column with methanol-acetonitrile-0.2 M aqueous oxalic acid solution pH 2.0(1:1:3.5) gave satisfactory results for groups I and II, respectively.
Lysophosphatidic acids having different fatty acyl moieties were directly analysed by gas chromatography/mass spectrometry after silylation. Trimethylsilyl derivatives of various LPAs escaped thermal degradation, and electron impact and chemical ionization of these derivatives yielded many ions which were useful for the structural determination. It was found by this new method that five species of lysophosphatidic acids were formed in rat plasma during incubation at 37 degrees C.
Lamola et al. (1974) reported that free erythrocyte protoporphyrin (FEP) is not present as free type protoporphyrin ( FPP ) but rather is chelated with zinc in lead poisoning and iron deficiency anemia. In our fluorometric study of FEP and zinc protoporphyrin ( ZnP ) in erythrocytes of lead-poisoned rabbits and lead workers, the co-existence of ZnP and FPP was observed in the severe stage of acute lead poisoning in rabbits. The ratio of ZnP to FEP in erythrocytes decreased with the progress of lead intoxication, and the amount of FPP was greater than that of ZnP in the severe stage of intoxication. This FPP accumulated in the erythrocytes was easily converted to ZnP by incubation of the hemolysate with zinc. On the other hand, increased protoporphyrin (PP) in bone marrow was present as FPP in lead intoxication. From these results, ZnP in peripheral erythrocytes is considered to be a secondary product, not a primary one. Therefore, the determination of total FEP ( FPP + ZnP ) by the acid solvent extraction method might be more reasonable than using ZnP -determination as the biological parameter of lead poisoning. We propose that erythrocyte porphyrin in lead intoxication should be expressed as FEP, not as ZnP , for the parameter. The present study also suggests that the form of erythrocyte FPP in erythropoietic protoporphyria (EPP) patients is a little different from that in lead-poisoning patients, because of its high chelation reactivity with zinc.
When plasma jet of Ar-arc plasma was blown into an aqueous solution containing organic compounds, oxidation reactions were induced in the solution. The plasma-induced reaction was a powerful oxidation which could convert a methyl to a carboxyl group and cleave a carbon-carbon bond without using any oxidizing reagent. This reaction could be regarded as a model for the solar plasma-induced reaction in the primitive hydrosphere.
Flames of flammable gases, when blown against a surface of an aqueous solution of organic compounds, were found to induce oxidation as well as other reactions in the solution. This reaction would be regarded as a new model for formation of bioorganic molecules in the primitive hydrosphere exposed to some radical-containing atmosphere.
We noted a marked improvement of lymphedema in a patient with recurrence of cancer 3 years after amputatio recti and with post-thrombotic syndrome of the leg, as the result of injection of a suspension of lymphocytes administrated as therapy against advanced cancer. Subsequently lymphocytes-injections were given 39 times to 7 patients with lymphedema of one limb, of various causes. A suspension of lymphocytes was injected repeatedly into the proximal artery of the affected limb. In one patient, lymphocytes were injected 4 times at intervals of 1 or 3 weeks. In most cases, this injection resulted in a marked reduction in the swelling of the affected limb and improvement was seen for several weeks or months. In all cases, there was a marked, continuous softening of the tissue.
